Causal effects of circulating cathepsins on cholecystitis, cholelithiasis, primary biliary cholangitis, primary sclerosing cholangitis, acute pancreatitis, and chronic pancreatitis: A Mendelian randomization study.

Gallbladder, biliary, and pancreatic disorders are nonneoplastic diseases marked by inflammation, epithelial injury, and fibrosis. Cathepsins are lysosomal proteases involved in inflammatory activation, immune regulation, extracellular matrix turnover, and pancreatic enzyme activation. Observational links between dysregulated cathepsins and hepatopancreatobiliary pathology could reflect confounding or reverse causation. This study used Mendelian randomization (MR) to assess whether circulating cathepsins causally influence gallbladder, biliary, and pancreatic diseases. We conducted a 2-sample

Shared genetic architecture of gastroesophageal reflux disease and age related phenotypes.

Increasing age is a risk factor of gastroesophageal reflux disease. This study aims to uncover the shared genetic architecture of gastroesophageal reflux disease (GERD) and age-related phenotypes. Based on publicly available GWAS statistics, this genome-wide pleiotropic association research was performed with multiple genetic approaches sequentially to explore the pleiotropic associations from single-nucleotide polymorphism (SNP) and gene levels, to reveal the underlying shared genetic etiology between GERD and age-related phenotypes. This study featured shared genetic mechanisms between GERD

Genetic analysis of comorbidities between osteoarthritis, sarcopenia, and osteoporosis.

BACKGROUND: Osteoarthritis (OA), sarcopenia (SCP), and osteoporosis (OP) pose a substantial global morbidity and mortality burden, and previous studies have observed potential associations among them. This study aims to comprehensively characterize the common genetic structure, biological basis, and underlying causal relationship among OA, SCP, and OP. METHODS: We used pooled statistics from the largest European genome-wide association study to investigate the genetic overlap and underlying causal relationships among OA, SCP, and OP. LD Score Regression (LDSC) was first used for estimating gl

Dissecting the genetic overlap between severe mental disorders and markers of cellular aging: Identification of pleiotropic genes and druggable targets.

Patients with severe mental disorders such as bipolar disorder (BD), schizophrenia (SCZ) and major depressive disorder (MDD) show a substantial reduction in life expectancy, increased incidence of comorbid medical conditions commonly observed with advanced age and alterations of aging hallmarks. While severe mental disorders are heritable, the extent to which genetic predisposition might contribute to accelerated cellular aging is not known. We used bivariate causal mixture models to quantify the trait-specific and shared architecture of mental disorders and 2 aging hallmarks (leukocyte telome

Pleiotropic and Adverse Effects of Statins-Do Epigenetics Play a Role?

Statins are widely used to prevent major cardiovascular events by lowering serum cholesterol. There is evidence that statins have pleiotropic effects-that is, cholesterol-independent effects-that may also confer protection from cardiovascular disease and potentially numerous other pathologies, including cancer. Statins also have a number of well described adverse effects, including myopathy, rhabdomyolysis, liver damage, and type 2 diabetes. This paper examines the evidence of epigenetic modifications as a contributory factor to the pleiotropic and adverse effects of statins. In vitro and anim

No Genetic Causal Relationship between COVID-19 and Herpes Zoster: A Bidirectional Mendelian Randomization Analysis.

PURPOSE: Increased incidences of herpes zoster (HZ) have been reported among COVID-19 patients, but the underlying causal mechanisms remain unclear. Inspired by an atypical case of HZ in a COVID-19 patient, we conducted a bidirectional Mendelian randomization (MR) analysis to investigate potential causal relationships. PATIENTS AND METHODS: The genetic statistics were extracted from the COVID19-hg GWAS meta-analyses and the IEU GWAS database. MR analyses were performed using the inverse-variance weighted (IVW) method as the primary approach, with MR-Egger, weighted median, simple mode, and we

Childhood asthma and type 1 diabetes mellitus: A meta-analysis and bidirectional Mendelian randomization study.

BACKGROUND: Worldwide incidence and prevalence of both asthma and type 1 diabetes mellitus (T1DM) in children have been increasing in past decades. Association between the two diseases has been found in some but not in other studies. OBJECTIVE: We conducted a meta-analysis to verify such an association, and bidirectional Mendelian randomization analysis to examine the potential cause-effect relationships. METHODS: Three databases (PubMed, Embase, and Web of Science) were searched from their inception to February 1, 2021. Pooled hazard ratios (HR) or odds ratios (OR), and 95% confidence inter

MR-PheWAS: exploring the causal effect of SUA level on multiple disease outcomes by using genetic instruments in UK Biobank.

OBJECTIVES: We aimed to investigate the role of serum uric acid (SUA) level in a broad spectrum of disease outcomes using data for 120 091 individuals from UK Biobank. METHODS: We performed a phenome-wide association study (PheWAS) to identify disease outcomes associated with SUA genetic risk loci. We then implemented conventional Mendelianrandomisation (MR) analysis to investigate the causal relevance between SUA level and disease outcomes identified from PheWAS. We next applied MR Egger analysis to detect and account for potential pleiotropy, which conventional MR analysis might mistake for

Non-Alcoholic Fatty Liver Disease and Vitamin D in the UK Biobank: A Two-Sample Bidirectional Mendelian Randomisation Study.

Evidence for a role for vitamin D in non-alcoholic fatty liver disease (NAFLD) pathogenesis is conflicting. As Mendelian randomisation (MR) avoids many limitations of conventional observational studies, this two-sample bidirectional MR analysis was conducted to determine the following: (i) whether genetically predicted 25-hydroxyvitamin D [25(OH)D] levels are a risk factor for NAFLD, and (ii) whether genetic risk for NAFLD influences 25(OH)D levels. Single-nucleotide polymorphisms (SNPs) associated with serum 25(OH)D levels were obtained from the European ancestry-derived SUNLIGHT consortium.

Association Between Serum Galectin-3 and Parkinson's Disease: A Two-Sample Mendelian Randomization Study.

BACKGROUND: Parkinson's disease (PD) is a prevalent neurodegenerative disorder with poor prognosis. Observational studies have demonstrated a significant correlation between serum galectin-3 and PD, suggesting a potential role of galectin-3 as a biomarker for PD. However, it is still unclear whether galectin-3 contributes to the risk of the disease. METHODS: A two-sample Mendelian randomization (MR) approach was used in this study. Genetic instruments for serum galectin-3 level were selected from a genome-wide association study (GWAS), including 30,931 European individuals. Summary-level stat

Nutrient-sensitive, antagonistically pleiotropic genes and their contribution to malignant behavior.

Recent evidence from microarray data argues that much of the malignant potential of neoplastic cells is a result of either somatic initiating mutations or constitutional polymorphisms present in normal tissue. Consequently, we reasoned that the initiating genetic lesions seen in some of the Phakomatoses, disorders associated with predominantly benign neoplasms, may somehow fundamentally limit malignant behavior (invasion and metastasis). The function of these genes reveals a consistent pattern of antagonistic pleiotropy; they inhibit proliferation but are critical for motility. We argue that i

Movement disorders: a brief overview from an evolutionary perspective.

Evolutionary approach tries to explain several aspects of neurological diseases under the perspective of evolution. Evolutive bottle-neck may explain some aspects of neurodegenerative diseases. Additionally, a number of genetic alleles may reflect an evolutionary trade-off between benefits in early life and late-life disadvantages. Antagonistic pleiotropy theory suggests that some adaptive evolutionary changes increase individual fitness early in life, but can exacerbate detrimental aging-related processes We discuss three diseases with movement disorders from an evolutionary perspective: Park

Multi-trait analysis of pigmentation traits in relation to skin and ocular cancers: Relevance for evolutionary medicine.

BACKGROUND AND OBJECTIVES: Pigmentary traits (skin, hair, eye colour, tanning response) are epidemiologically linked to skin and ocular cancers, but the extent and nature of shared genetic architecture remain incompletely defined. In this study we explored to what extent pigmentation- and cancer-associated variants exhibit pleiotropy. METHODOLOGY: We analyzed genome-wide association studies summary statistics from European populations for skin colour; hair colour (blonde, light brown, red, dark brown, black); eye colour; tanning response; and four cancers: cutaneous malignant melanoma (CMM),

A two-sample Mendelian Randomization study on the causal relationship between obesity and breast cancer.

BACKGROUND: The relationship between obesity and breast cancer risk remains complex and inconsistently observed in epidemiological studies due to potential confounding and reverse causation. Comprehensive Mendelian randomization analyses of multiple anthropometric traits, particularly central adiposity measures, and their stratified effects by estrogen receptor (ER) subtype are still lacking. METHODS: We conducted a two-sample MR study to assess the causal relationship between nine obesity-related traits (e.g., Body Mass Index (BMI), Waist circumference (WC), basal metabolic rate) and breast

Causal Relationship Between Sjögren's Syndrome and Atherosclerosis: A Bidirectional, Two-Sample, Mendelian Randomization Study.

BACKGROUND: There is growing evidence that Sjögren's syndrome (SS) and atherosclerosis (AS) might share underlying immunological and inflammatory processes. Observational data have pointed toward a potential association between SS and a heightened likelihood of developing AS, though the causal direction and specific dynamics of this relationship have not been clearly verified. This Mendelian randomization (MR) investigation opts to investigate potential bidirectional causality between SS and three types of AS: coronary, cerebral, and peripheral. METHODS: Genetic variants significantly linked

No genetic causal association between periodontitis and ankylosing spondylitis: a bidirectional two-sample mendelian randomization analysis.

BACKGROUND: Observational studies that reveal an association between periodontitis (PD) and ankylosing spondylitis (AS) exist. However, observational research is prone to reverse causality and confounding factors, which make it challenging to infer cause-and-effect relationships. We conducted a two-sample Mendelian randomization (MR) study to examine the causal relationship between the genetic prediction of PD and AS. METHODS: In our study, single-nucleotide polymorphisms (SNPs) were defined as instrumental variables (IVs). The genetic association with PD came from the Gene-Lifestyle Interact
