Pigmentation phototype and prostate and breast cancer in a select Spanish population-A Mendelian randomization analysis in the MCC-Spain study.

INTRODUCTION: Phototype has been associated with an increased risk of prostate cancer, and it is yet unknown if it is related to other hormone-dependent cancers, such as breast cancer or whether this association could be considered causal. METHODS: We examined the association between the phototype and breast and prostate cancers using a Mendelian randomization analysis. We studied 1,738 incident cases of breast cancer and another 817 cases of prostate cancer. To perform a Mendelian randomization analysis on the phototype-cancer relationship, a genetic pigmentation score was required that met

X-linked alpha-thalassemia/mental retardation (ATR-X) syndrome: localization to Xq12-q21.31 by X inactivation and linkage analysis.

We have examined seven pedigrees that include individuals with a recently described X-linked form of severe mental retardation associated with alpha-thalassemia (ATR-X syndrome). Using hematologic and molecular approaches, we have shown that intellectually normal female carriers of this syndrome may be identified by the presence of rare cells containing HbH inclusions in their peripheral blood and by an extremely skewed pattern of X inactivation seen in cells from a variety of tissues. Linkage analysis has localized the ATR-X locus to an interval of approximately 11 cM between the loci DXS106

Genetic Risk, Healthy Lifestyle, and Risk Stratification for Diverticulitis.

BACKGROUND: A polygenic risk score approximates an individual's genetic susceptibility to disease, whereas the Life's Essential 8 (LE8) score captures a healthy lifestyle. This study applied the LE8 score and a polygenic risk score to severe diverticulitis. Multiple variations in lifestyle scores were compared. We hypothesized that genetics would complement lifestyle information in risk stratification. STUDY DESIGN: A polygenic risk score for severe diverticulitis (operative intervention or at least 2 inpatient admissions) was optimized in BioVU, followed by application to an independent pros

Integrating amyloid imaging and genetics for early risk stratification of Alzheimer's disease.

INTRODUCTION: Alzheimer's disease (AD) initiates years prior to symptoms, underscoring the importance of early detection. While amyloid accumulation starts early, individuals with substantial amyloid burden may remain cognitively normal, implying that amyloid alone is not sufficient for early risk assessment. METHODS: Given the genetic susceptibility of AD, a multi-factorial pseudotime approach was proposed to integrate amyloid imaging and genotype data for estimating a risk score. Validation involved association with cognitive decline and survival analysis across risk-stratified groups, focu

Novel Potential Risk Loci for Migraine in the Portuguese Population.

Common forms of migraine are complex disorders characterized by significant clinical diversity. Their genetic basis has been extensively studied but remains unclear. This study represents the first pilot genome-wide association study (GWAS) integrating a polygenic risk score (PRS) in the Portuguese population, designed to identify migraine susceptibility loci through a case-control study and unravel population-specific variants. Genotyping data was acquired with Applied Biosystems Axiom™ PMDA array, producing 12,035,248 single-nucleotide polymorphisms (SNPs) post-imputation, providing a compre

GWAS for systemic sclerosis identifies six novel susceptibility loci including one in the Fcγ receptor region.

Here we report the largest Asian genome-wide association study (GWAS) for systemic sclerosis performed to date, based on data from Japanese subjects and comprising of 1428 cases and 112,599 controls. The lead SNP is in the FCGR/FCRL region, which shows a penetrating association in the Asian population, while a complete linkage disequilibrium SNP, rs10917688, is found in a cis-regulatory element for IRF8. IRF8 is also a significant locus in European GWAS for systemic sclerosis, but rs10917688 only shows an association in the presence of the risk allele of IRF8 in the Japanese population. Furthe

Genome-wide polygenic risk scores for hypertensive disease during pregnancy can also predict the risk for long-term cardiovascular disease.

BACKGROUND: Hypertensive disorders during pregnancy are associated with the risk of long-term cardiovascular disease after pregnancy, but it has not yet been determined whether genetic predisposition for hypertensive disorders during pregnancy can predict the risk for long-term cardiovascular disease. OBJECTIVE: This study aimed to evaluate the risk for long-term atherosclerotic cardiovascular disease according to polygenic risk scores for hypertensive disorders during pregnancy. STUDY DESIGN: Among UK Biobank participants, we included European-descent women (n=164,575) with at least 1 live

Identifying potential causal effects of age at menarche: a Mendelian randomization phenome-wide association study.

BACKGROUND: Age at menarche has been associated with various health outcomes. We aimed to identify potential causal effects of age at menarche on health-related traits in a hypothesis-free manner. METHODS: We conducted a Mendelian randomization phenome-wide association study (MR-pheWAS) of age at menarche with 17,893 health-related traits in UK Biobank (n = 181,318) using PHESANT. The exposure of interest was the genetic risk score for age at menarche. We conducted a second MR-pheWAS after excluding SNPs associated with BMI from the genetic risk score, to examine whether results might be due …

"It's Not Deterministic and It Will Never Be Deterministic": A Qualitative Study on Stakeholder Perspectives of Polygenic Risk Score Testing for Post-Traumatic Stress Disorder.

Post-traumatic stress disorder (PTSD) causes significant mental and physical distress, yet only a small subset of individuals exposed to trauma develop the disorder. Scientists and clinicians are still unable to predict who will get the disorder or how it will manifest. Genetic factors contribute to PTSD risk, but integrating genetic findings into clinical practice has been challenging. Military veterans are especially impacted by PTSD and have different treatment outcomes when compared to civilians. Thus, genetic findings related to PTSD may be particularly valuable to this population. This q

Global multi-ancestry genome-wide analyses identify genes and biological pathways associated with thyroid cancer and benign thyroid diseases.

Thyroid diseases are common and highly heritable. We performed a meta-analysis of genome-wide association studies from 19 biobanks for five thyroid diseases: thyroid cancer (ThC), benign nodular goiter, Graves' disease, lymphocytic thyroiditis and primary hypothyroidism. We analyzed genetic association data from ~2.9 million genomes and identified 313 known and 570 new independent loci linked to thyroid diseases. We discovered genetic correlations between ThC, benign nodular goiter and autoimmune thyroid diseases (rg = 0.16-0.97). Telomere maintenance genes contributed to benign and malignant

Sleep patterns, genetic susceptibility, and risk of new-onset gout: The UK Biobank prospective cohort study.

OBJECTIVE: The association between sleep behaviors and gout risk remains uncertain. We aimed to evaluate the relationship of sleep patterns based on a combination of five major sleep behaviors with the risk of new-onset gout, and to explore whether genetic risks of gout may modify this association in the general population. METHODS: 403,630 participants without gout at baseline in UK Biobank were included. A healthy sleep score was created by combining five major sleep behaviors, including chronotype, sleep duration, insomnia, snoring, and daytime sleepiness. Genetic risk score for gout was c…

A large-scale genome-wide association meta-analysis for nevus count provides direct insights into the genetics of melanoma.

A greater understanding of the biology of nevi will provide insights into the etiology of melanoma. Our large-scale meta-analysis of 14 nevus genome-wide association studies (GWAS) includes 85,965 individuals of European ancestry. We identify 29 nevus-associated loci (p < 5 × 10-8), of which 24 have not been previously reported in a GWAS conducted for nevus count alone. We further identify 255 candidate genes for nevus loci, including SIKE1 which is involved in immune response regulation. This is of interest because immune response regulation influences the formation of nevi and melanoma susce

The predictive value of polygenic risk scores for depression in gene-environment interaction studies: a systematic review.

BACKGROUND: According to the diathesis-stress model, genetic liability and environmental exposures interact in the pathogenesis of depression. Polygenic risk scores for depression (PRSD) based on large-scale genome-wide association studies have opened new avenues for investigating gene-environment interaction (GxE) beyond candidate gene studies. To the best of our knowledge, this is the first systematic review of studies that have taken a polygenic score approach to study GxE interaction effects on depression phenotypes. METHODS: Based on a preregistered, systematic literature search accordin

Polygenic scores for executive functioning as predictors of performance improvements after repeated testing in major psychiatric disorders.

Executive Functions (EF) control goal-oriented behaviors and are disrupted in patients with psychiatric disorders (PPD). Repeated EF testing in clinical practice and research aims to evaluate disease progression and treatment efficacy. Performance improvements (or “practice effects”) are expected, while deficiencies had been proposed as markers of conversion to dementia. Given the high conversion risk in PPD, understanding the determinants of practice effects is of high relevance. However, the role of genetic predisposition remains unknown. We aim to test the influence of polygenic scores on p

Polygenic and spatial insights into the genetic uniqueness of essential tremor using common variants.

INTRO: The heterogeneity of Essential Tremor (ET) complicates how it is diagnosed and studied. ET is often misdiagnosed as other neurological disorders and vice versa. We aimed to better understand ET by comparing brain maps informed by ET common variants to those derived from related phenotypes. A further goal was to enhance the diagnostic precision of ET by accounting for shared genetic signals between ET and genetically overlapping phenotypes. METHODS: Phenotype variant association mapping to the brain was done for ET, Parkinson's disease (PD), dystonia, and cognition across adult mouse wh

Assessing measurement bias in substance withdrawal symptoms attributable to childhood adversity and genetic liability.

BACKGROUND: Substance withdrawal results from the abrupt cessation or reduction of substance use in physically dependent individuals. Diagnosis of withdrawal requires endorsing multiple substance-specific symptoms, implicitly assuming that all symptoms are equally indicative of underlying withdrawal severity. However, some criteria may be more informative than others, and their endorsement may be impacted by genetic and environmental factors. METHODS: We examined differential item functioning (DIF) of tobacco, alcohol, and opioid withdrawal symptoms as a function of substance use disorder (SU

A partitioned polygenic risk score reveals distinct contributions to psoriasis clinical phenotypes across a multi-ethnic cohort.

Psoriasis is a chronic, immune-mediated inflammatory skin disease associated with a polygenic mode of inheritance. There are few studies that explore the association of a psoriasis Polygenic Risk Score (PRS) with patient clinical characteristics, and to our knowledge there are no studies examining psoriasis PRS associations across different ethnicities. In this study, we used a multi-racial psoriasis cohort to investigate PRS associations with clinical phenotypes including age of onset, psoriatic arthritis, other comorbidities, psoriasis body location, psoriasis subtype, environmental triggers

Sarcopenic obesity is part of obesity paradox in dementia development: evidence from a population-based cohort study.

BACKGROUND: Sarcopenic obesity, a clinical and functional condition characterized by the coexistence of obesity and sarcopenia, has not been investigated in relation to dementia risk and its onset. METHODS: We included 208,867 participants from UK biobank, who aged 60 to 69 years at baseline. Dementia diagnoses were identified using hospital records and death register data. Hazard ratios (HRs) and 95% confidence intervals (CIs) were estimated using Cox proportional hazards models to evaluate the associations of obesity, sarcopenia, and sarcopenic obesity with dementia risk, stratified by sex.
