Kinetics of glutamate and gamma-aminobutyric acid in cerebrospinal fluid in a canine model of complex partial status epilepticus induced by kainic acid.

An imbalance of excitatory and inhibitory transmitters in the brain has been suggested to cause epileptic seizures. In this study, we investigated the kinetics of glutamate (GLU) and gamma-aminobutyric acid (GABA) in cerebrospinal fluid (CSF-GLU and CSF-GABA, respectively) using a high performance liquid chromatography (HPLC) in a canine model of complex partial status epilepticus (CPSE) induced by the microinjection of kainic acid (KA) into the unilateral amygdala. During the acute phase (3, 6, 12 and 48 hr after the onset of CPSE), CSF-GLU was significantly increased, while CSF-GABA was decr

Accuracy of glutamic acid decarboxylase antibodies for the identification of adult-onset type 1 diabetes mellitus: a systematic review and meta-analysis.

INTRODUCTION: The glutamic acid decarboxylase autoantibody (GADA) test is a widely used marker to differentiate type 1 diabetes and type 2 diabetes in adults. We conducted a systematic review to estimate the sensitivity and specificity of the presence of GADA for T1D in adults with newly diagnosed diabetes. METHODS: We conducted a PubMed and Embase search for studies that report both the GADA result and type 1 versus type 2 diabetes in adults diagnosed with diabetes. We calculated the sensitivity and specificity for each study. RESULTS: We identified 19 studies involving 11,760 patients from…

Leucine and glutamic acid attenuate the dextran sulfate sodium-induced colitis in mice by modulating the inflammatory response and energy metabolism processes.

Ulcerative colitis, a multifactorial chronic condition of the gastrointestinal tract, has seen increasing interest in dietary intervention as part of its treatment strategy. In this study, we utilized a Dextran Sodium Sulfate (DSS)-induced acute colitis mouse model to assess the impact of leucine and glutamic acid on mucosal ulceration, inflammatory cell infiltration, and proteomic alterations in colon tissue. The results indicated that both glutamic acid and leucine alleviate the symptoms of colitis by reducing the histological scores of mice colon, promoting the expression of anti-inflammato…

Graft failure after allogeneic islet transplantation in a patient with type 1 diabetes and a high anti-glutamic acid decarboxylase antibody titer.

Pancreatic islet transplantation is a β-cell replacement therapy for people with insulin-deficient diabetes who have difficulty in glycemic … detected. Autoimmunity relapse was also not observed. However, the patient had a high titer of anti-glutamic acid decarboxylase antibody from before the islet transplantation, and autoimmunity might thus have affected the β-cells in the transplanted islet. The evidence is still scarce to reach conclusions, and further data accumulation is required to enable pro …

Exercise training modulates glutamic acid decarboxylase-65/67 expression through TrkB signaling to ameliorate neuropathic pain in rats with spinal cord injury.

Neuropathic pain is one of the most frequently stated complications after spinal cord injury. In post-spinal cord injury, the decrease of gamma aminobutyric acid synthesis within the distal spinal cord is one of the main causes of neuropathic pain. The predominant research question of this study was whether exercise training may promote the expression of glutamic acid decarboxylase-65 and glutamic acid decarboxylase-67, which are key enzymes of gamma aminobutyric acid synthesis, within the distal spinal cord through tropomyosin-related kinase B signaling, as its synthesis assists to relieve ne…

Near-Infrared Activatable Phthalocyanine-Poly-L-Glutamic Acid Conjugate: Enhanced in Vivo Safety and Antitumor Efficacy toward an Effective Photodynamic Cancer Therapy.

We previously developed a new zinc(II) phthalocyanine (ZnPc) derivative (Pc 1) conjugated to poly-L-glutamic acid (PGA) (1-PG) to address the limitations of ZnPc as part of an antitumor photodynamic therapy approach, which include hydrophobicity, phototoxicity, and nonselectivity in biodistribution and tumor targeting. During this study, we discovered that 1-PG possessed high near-infrared (NIR) light absorptivity (λmax = 675 nm), good singlet oxygen generation efficiency in an aqueous environment, and enhanced photocytotoxic efficacy and cancer cell uptake in vitro. In the current study, we d…

Construction of a chitosan/γ-poly(glutamic acid) composite hydrogel crosslinked by ZIF-8 for enhanced α-glucosidase immobilization and inhibitor screening.

Herein, a composite hydrogel based on two natural macromolecules-chitosan (CS) and γ-poly(glutamic acid) (γ-PGA)-was developed, crosslinked by zeolitic imidazolate framework-8 (ZIF-8) nanoparticles, which serve as reactive crosslinking hubs to covalently connect the biopolymer chains for efficient immobilization of α-glucosidase (α-Glu). A carboxyl-terminated poly(ethylene glycol)-poly(glutamic acid) copolymer was first synthesized and its activated carboxyl groups underwent amidation with amino groups of CS to rapidly form a three-dimensional PC/ZIF-8 hydrogel. The effects of ZIF-8 content an…
