Cyst fluid from human autosomal dominant polycystic kidneys promotes cyst formation and expansion by renal epithelial cells in vitro.

Autosomal dominant polycystic kidney disease (ADPKD) is characterized by progressive renal enlargement, culminating in renal insufficiency in over one half of affected individuals. The highly variable onset and clinical course of ADPKD may be due to factors extrinsic to the genetically defined renal cysts. In this study, cyst fluid samples from 12 nonazotemic and 18 azotemic ADPKD subjects were examined for in vitro biologic activity that promotes cellular proliferation and the secretion of fluid by renal epithelial monolayers, two pathogenetic mechanisms that have critical roles in the format

Exogenous Eugenol Alleviates Salt Stress in Tobacco Seedlings by Regulating the Antioxidant System and Hormone Signaling.

Salt stress seriously affects crop growth, leading to a decline in crop quality and yield. Application of exogenous substances to improve the salt tolerance of crops and promote their growth under salt stress has become a widespread and effective means. Eugenol is a small molecule of plant origin with medicinal properties such as antibacterial, antiviral, and antioxidant properties. In this study, tobacco seedlings were placed in Hoagland's solution containing NaCl in the presence or absence of eugenol, and physiological indices related to stress tolerance were measured along with transcriptom

Plant growth regulation by seed coating with films of alginate and auxin-intercalated layered double hydroxides.

Auxins are a class of organic substances known as plant-growth regulators, which act on plant physiology, promoting its full development. However, due to the great instability of these substances among the diversity of crops and cultivation environments, it is necessary to seek more efficient modes of application, which lead to a homogeneous distribution and promote a sustained release according to the plants demand. Seed coating, using films containing a biodegradable polymer and auxins intercalated into layered compounds, emerges as a very promising approach to a new form of growth regulator

Toxicity of trichloroethylene (TCE) on some algae and cyanobacteria.

The toxicity of trichloroethylene was tested in both glass enclosures and in polystyrene immunological plates, and resulted in insignificant correlation of EC₅₀ (evaluated as OD 750 nm). In enclosures, EC₅₀ evaluated as O₂ evolution and as pH increment was highly significant. The optimal time for the assay of TCE in enclosures was 48-72 h; and in plates ca 200 h. After a longer time (e.g. 300 h) TCE diffused from the plates and growth was revived. TCE in low concentrations (ca. 0.1-0.2 g/L) stimulated the growth of some tested organisms, both in plates and in enclosures. Toxicity occurred from

[The peritoneal environment: physiopathogenesis of endometriosis].

In this report an hypothetical model of the pathophysiology of endometriosis is reviewed based on recent literature, focusing a variety of factors within the specific environment confined by peritoneum, whose alteration has repercussion among endometriotic and immune response cells relationships. At this point vasoactive substances, cytokines (interleukines and growth factors), and menstrual cycle hormones may act as soluble mediators that are able to induce several effects over cellular proliferation, growth and differentiation; and expression of new antigenic epitopes and cell adhesion molec

Cripto-1: an oncofetal gene with many faces.

Human Cripto-1 (CR-1), a member of the epidermal growth factor (EGF)-CFC family, has been implicated in embryogenesis and in carcinogenesis. During early vertebrate development, CR-1 functions as a co-receptor for Nodal, a transforming growth factor beta (TGFbeta) family member and is essential for mesoderm and endoderm formation and anterior-posterior and left-right axis establishment. In adult tissues, CR-1 is expressed at a low level in all stages of mammary gland development and expression increases during pregnancy and lactation. Overexpression of CR-1 in mouse mammary epithelial cells le

Profiles of cytokine expression in radicular cyst-lining epithelium examined by RT-PCR.

Levels of messenger RNA (mRNA) extracted from five samples of radicular cyst-lining epithelium were analyzed for cytokines, growth factors and epithelial cell growth-related receptors by RT-PCR. All five samples expressed IL-1alpha, -1beta, IL-6, IL-8, IL-11, TGF-beta1, PDGF-A and aFGF, and receptors for EGF (c-erbB), KGF, HGF (c-met) and IL-6. Some of the specimens expressed MIP-1alpha, RANTES, GM-CSF, M-CSF, TNF-alpha, PDGF-B and bFGF, but no expression of IL-2, IL-4, IFN-gamma, IGF-I, EGF and KGF was detected. These results indicate that radicular cyst-lining epithelium, which is considered

Decreased mitogenic and osteogenic responsiveness of calvarial osteoblasts isolated from aged rats to basic fibroblast growth factor.

Osteoporosis seen in aged individuals is represented by the reduced bone mass most likely resulting from decreased bone formation by osteoblasts. To examine whether aging causes a decrease in osteoblast activity, calvarial osteoblasts were isolated from aged rats (AOB) and studied for the capacity of the cells to form mineralized bone-like nodules in comparison with that of fetal calvarial osteoblasts (FOB). There were no significant differences in basal mineralized bone-like nodule formation determined by quantifying the size of the nodules which were formed in the cultures of AOB and FOB. Ho

Oncogenes and erythroid differentiation.

Committed erythroid progenitors are the cellular targets of oncogene-carrying avian and murine retroviruses that induce acute erythroleukemia. Normally, these cells undergo a fixed program of 5-10 cell divisions while terminally differentiating into erythrocytes. The sustained self-renewal observed in the context of their retrovirus-mediated leukemic transformation has been viewed as an abnormal, oncogene-induced effect. Recently, however, it was found that certain combinations of growth/differentiation hormones (e.g. TGF-alpha and estradiol) can induce normal avian erythroid progenitors to un

Transforming growth factor-beta promotes deposition of collagen VII in a modified organotypic skin model.

BACKGROUND: Anchoring fibrils, which consist mainly of collagen VII, form a three-dimensional network below stratified squamous epithelia thus attaching the epithelial basement membrane to the underlying connective tissue. In skin, collagen VII is mainly produced by keratinocytes, and in vitro findings of keratinocyte-fibroblast cocultures suggest that mesenchymal-epithelial signaling plays a major role in the regulation of its expression. The matrix cytokine transforming growth factor-beta (TGF-beta) is a candidate mediator of such interactions, and exogenous TGF-beta has been shown to stimul

Calcineurin B1 Deficiency Reduces Proliferation, Increases Apoptosis, and Alters Secretion in Enteric Glial Cells of Mouse Small Intestine in Culture.

To investigate the roles of calcineurin (CN) in glial cells, we previously generated conditional knockout (CKO) mice lacking CNB1 in glial cells. Because these CKO mice showed dysfunction and inflammation of the small intestine in addition to growth impairment and postweaning death, we have focused on enteric glial cells (EGCs) in the small intestine. In this study, we examined the effects of CNB1 deficiency on the proliferation and survival of EGCs and the expression and secretion of EGC-derived substances in culture to reveal the mechanisms of how CNB1 deficiency leads to dysfunction and inf

Biotransformation of sulfamethoxazole (SMX) by aerobic granular sludge: Removal performance, degradation mechanism and microbial response.

Aerobic granular sludge (AGS) is a promising biotechnology for the treatment of antibiotic-rich wastewater. However, little is known about the antibiotics degradation mechanism and microbial response in a sulfamethoxazole (SMX)-loaded AGS system. Herein, the results of a continuous 240 days test suggested that 0.5-5 mg/L of SMX could be thoroughly removed by AGS via adsorption and degradation. The degradation pathway of SMX involved the hydrolysis of the sulfonamide bond and cleavage of NS or CS bonds, subsequently leading to the production of small molecular substances (e.g. benzene and 5-met

Comparison of Different Intraarticular Injection Substances Followed by Temporomandibular Joint Arthroscopy.

Nowadays, temporomandibular joint (TMJ) arthroscopies are often finished with intraarticular injections. There are a couple of different substances that can be injected into the articular cavity following arthroscopy; however, it is still unclear which one can help to reach the best clinical results. The aim of this systematic review was to compare different substances that can be injected intraarticularly following TMJ arthroscopy to enhance clinical success.A review protocol was registered in PROSPERO (Registration number: CRD42018114833). An electronic search was performed using PubMed, Coch

Epidemic of gynecomastia among haitian refugees: exposure to an environmental antiandrogen.

OBJECTIVE: To investigate an observed epidemic of gynecomastia among Haitian refugees in US detention centers in 1981 and 1982. METHODS: All identifiable environmental exposures were investigated for estrogenic and antiandrogenic activity. RESULTS: A high incidence of gynecomastia was observed among Haitian refugees in five detention centers in the United States. Of 284 men screened, 20 (from 18 to 53 years old) demonstrated new-onset gynecomastia (Tanner stages 2 to 5) in June 1982. The mean onset of gynecomastia was 130 +/- 12 days after arrival in the United States. Other symptoms include

Molecular neuro-oncology and development of targeted therapeutic strategies for brain tumors. Part 1: Growth factor and Ras signaling pathways.

Brain tumors are a diverse group of malignancies that remain refractory to conventional treatment approaches, including radiotherapy and cytotoxic chemotherapy. Molecular neuro-oncology has now begun to clarify the transformed phenotype of brain tumors and identify oncogenic pathways that may be amenable to targeted therapy. Growth factor signaling pathways are often upregulated in brain tumors and may contribute to oncogenesis through autocrine and paracrine mechanisms. Excessive growth factor receptor stimulation can also lead to overactivity of the Ras signaling pathway, which is frequently

Sprouting of non-sympathetic myelinated and unmyelinated fibres in response to chronic sympathetic denervation in the pineal gland of the Chinese hamster, Cricetulus griseus.

We have examined the effects of chronic sympathetic denervation on non-sympathetic myelinated and unmyelinated fibres in the superficial pineal gland of the Chinese hamster (Cricetulus griseus), using LM, EM and immunohistochemistry. The results suggest that non-sympathetic, myelinated and unmyelinated fibres enter the superficial pineal gland at its distal portion by way of the nervi conarii, and that these fibres are immunoreactive for calcitonin gene-related peptide or substance P. Non-sympathetic, myelinated and unmyelinated fibres in the superficial pineal gland increased in number follow

Mucinous (colloid) adenocarcinoma.

We describe a case of primary mucinous (colloid) adenocarcinoma of the lung. The patient was a 61-year-old male with a peripheral mass of the left lung. Retrospectively, the mass, which showed slow growth later, could be seen in a chest X-ray 3 years before surgery. The tumor was filled with a gelatinous substance resembling strawberry jelly, macroscopically. Microscopically, it was mucin with a small amount of malignant cells (adenocarcinoma). Diagnosis was mucinous (colloid) adenocarcinoma, which is a very rare intrapulmonary neoplasm. Mucinous (colloid) adenocarcinoma is suspected when a ge

Genetic alterations and growth factors in the pathogenesis of von Recklinghausen neurofibromatosis.

This paper is a summary review of recent developments with regard to the pathogenesis of von Recklinghausen neurofibromatosis, or NF-1. In the section on molecular biology of NF-1, the focus is on genetic linkage, with a note about prenatal diagnosis, tumor genetics and animal models. In the section on growth factors and receptors, the first element deals with neurofibromatosis as a model of cellular interaction, and particularly the issues of dysplasia versus neoplasia, cell-cell interactions and 'vicious cycle' positive feedback mechanisms; additional elements focus on nerve growth factor-be
