Injectable reactive biocomposites for bone healing in critical-size rabbit calvarial defects.

Craniofacial injuries can result from trauma, tumor ablation, or infection and may require multiple surgical revisions. To address the challenges associated with treating craniofacial bone defects, an ideal material should have the ability to fit complex defects (i.e. be conformable), provide temporary protection to the brain until the bone heals, and enhance tissue regeneration with the delivery of biologics. In this study, we evaluated the ability of injectable lysine-derived polyurethane (PUR)/allograft biocomposites to promote bone healing in critical-size rabbit calvarial defects. The bio

Emerging roles of platelet concentrates and platelet-derived extracellular vesicles in regenerative periodontology and implant dentistry.

Platelet concentrates (PCs) are easily obtained from autogenous whole blood after centrifugation and have evolved through three generations of development to include platelet-rich plasma, platelet-rich fibrin, and concentrated growth factor. Currently, PCs are widely used for sinus floor elevation, alveolar ridge preservation, periodontal bone defects, guided bone regeneration, and treatment of gingival recession. More recently, PCs have been leveraged for tissue regeneration to promote oral soft and hard tissue regeneration in implant dentistry and regenerative periodontology. PCs are ideal f

Advanced concepts in implant dentistry: creating the "aesthetic site foundation".

To obtain optimal and predictable aesthetics, deficiencies caused by soft and particularly hard tissue loss can be managed by various methods, such as orthodontic tooth eruption, socket preservation, and guided bone regeneration. However, in complex cases, these methods are often insufficient. Here, the authors introduce advanced concepts in aesthetic implant dentistry, such as "Aesthetic Site Foundation", "Aesthetic Guided Bone Regeneration" and "Implant Rectangle" that will guide the clinician in the quest to optimal aesthetic outcomes.

Shear Flow-Assembled Janus Membrane with Bifunctional Osteogenic and Antibacterial Effects for Guided Bone Regeneration.

Guided bone regeneration (GBR) membranes that reside at the interface between the bone and soft tissues for bone repair attract increasing attention, but currently developed GBR membranes suffer from relatively poor osteogenic and antibacterial effects as well as limited mechanical property and biodegradability. We present here the design and fabrication of a bifunctional Janus GBR membrane based on a shear flow-driven layer by a layer self-assembly approach. The Janus GBR membrane comprises a calcium phosphate-collagen/polyethylene glycol (CaP@COL/PEG) layer and a chitosan/poly(acrylic acid)

In vivo effectiveness of hybrid membranes with osteogenic growth peptide for bone regeneration.

Guided bone regeneration (GBR) technique helps to restore bone tissue through cellular selectivity principle. Currently no osteoinductive membrane exists on the market. Osteogenic growth peptide (OGP) acts as a hematopoietic stimulator. This association could improve the quality of bone formation, benefiting more than 2.2 million patients annually. The objective of this work was to develop membranes from ureasil-polyether materials containing OGP. The membranes were characterized by differential scanning calorimetry (DSC) and small angle X-ray scattering (SAXS). OGP was synthesized by the soli

Guided bone regeneration using a flexible hydroxyapatite patch.

Guided bone regeneration (GBR) is a new method of promoting new bone formation by blocking the proliferation of regenerated connective tissue or providing additional interventions such as direct drug delivery and mechanical support. This in vivo study of bone regeneration in radius compound fractures in rabbits was conducted using a highly flexible scaffold of nanoscale hydroxyapatite (nHAp)/chitosan, termed a "bone patch". A solidification-assisted compression (SAC) method was utilized to fabricate the bone patch, and its in vivo cytotoxicity, bio-absorption, and bone regeneration capacity we

Engineered Nanofiber-Based Nerve Guidance Conduit Facilitates the Restoration of Peripheral Nerve Injury through Enhanced Vascularization.

Peripheral nerve injuries (PNI) can disrupt the innervation of target organs, leading to diverse dysfunctions. While autologous nerve grafts are often regarded as the gold standard for treatment, their use is limited by donor nerve availability and potential complications at the injury site. Therefore, nerve guidance conduits (NGCs) constructed from biomaterials are favoured as an alternative for nerve repair. Angiogenesis provides the necessary prerequisites and microenvironmental support for neural regeneration. In this study, the extracted endothelial cell-derived matrix (ECd) was modified

Schwann cell strip for peripheral nerve repair.

Many strategies have been investigated to provide an ideal substitute to treat a nerve gap injury. Initially, silicone conduits were used and more recently conduits fabricated from natural materials such as poly-3-hydroxybutyrate (PHB) showed good results but still have their limitations. Surgically, a new concept optimising harvested autologous nerve graft has been introduced as the single fascicle method. It has been shown that a single fascicle repair of nerve grafting is successful. We investigated a new approach using a PHB strip seeded with Schwann cells to mimic a small nerve fascicle.

Melt electrowriting of hydrophilic/hydrophobic multiblock copolymers for bone tissue regeneration.

Bone-healing complications can occur due to large bone defects or an insufficient bone regeneration capacity. Melt electrowriting (MEW) is a potential candidate for manufacturing synthetic scaffolds that may resolve bone-healing complications. MEW can exploit various biocompatible polymers with a wide range of tissue engineering applications. Poly (ethylene oxide terephthalate)-poly(butylene terephthalate) (PEOT/PBT), a multiblock copolymer family, has emerged as a promising biomaterial to guide cell behavior, particularly in promoting bone differentiation. The polymer is known for its tunabil

Leveraging Oriented Lateral Walls of Nerve Guidance Conduit with Core-Shell MWCNTs Fibers for Peripheral Nerve Regeneration.

Peripheral nerve regeneration and functional recovery rely on the chemical, physical, and structural properties of nerve guidance conduits (NGCs). However, the limited support for long-distance nerve regeneration and axonal guidance has hindered the widespread use of NGCs. This study introduces a novel nerve guidance conduit with oriented lateral walls, incorporating multi-walled carbon nanotubes (MWCNTs) within core-shell fibers prepared in a single step using a modified electrohydrodynamic (EHD) printing technique to promote peripheral nerve regeneration. The structured conduit demonstrated

Multi-dimensional bioinspired tactics using an engineered mussel protein glue-based nanofiber conduit for accelerated functional nerve regeneration.

Limited regenerative capacity of the nervous system makes treating traumatic nerve injuries with conventional polymer-based nerve grafting a challenging task. Consequently, utilizing natural polymers and biomimetic topologies became obvious strategies for nerve conduit designs. As a bioinspired natural polymer from a marine organism, mussel adhesive proteins (MAPs) fused with biofunctional peptides from extracellular matrix (ECM) were engineered for accelerated nerve regeneration by enhancing cell adhesion, proliferation, neural differentiation, and neurite formation. To physically promote con

Tubulation repair mitigates misdirection of regenerating motor axons across a sciatic nerve gap in rats.

The repair of peripheral nerve laceration injury to obtain optimal function recovery remains a big challenge in the clinic. Misdirection of regenerating axons to inappropriate target, as a result of forced mismatch of endoneurial sheaths in the case of end-to-end nerve anastomosis or nerve autografting, represents one major drawback that limits nerve function recovery. Here we tested whether tubulation repair of a nerve defect could be beneficial in terms of nerve regeneration accuracy and nerve function. We employed sequential retrograde neuronal tracing to assess the accuracy of motor axon r

An Experimental Study to Bridge a Nerve Gap with a Decellularized Allogeneic Nerve.

BACKGROUND: The authors evaluated the efficacy of decellularized nerve as a scaffold for nerve regeneration. METHODS: Sciatic nerves harvested from Sprague-Dawley rats were decellularized in combination with sodium dodecyl sulfate and Triton X-100, and examined with scanning electron microscopy and immunofluorescence staining. A graft into the sciatic nerve in Wistar rats was performed with the decellularized Sprague-Dawley rat sciatic nerves [allograft: 10 mm long (n = 3) for short term and 15 mm long (n = 5) for long term]. As a control, a portion of sciatic nerve of Wistar rats was cut, re
