Biomimetic Anisotropic Polyvinyl Alcohol/Sodium Alginate/MXene Aerogel for Efficient Uremic Toxin Adsorption in Hemoperfusion.

The accumulation of uremic toxins, which ultimately leads to renal failure, results in systemic complications and necessitates efficient and biocompatible removal strategies. This paper presents the preparation of a biomimetic anisotropic aerogel composed of polyvinyl alcohol (PVA), sodium alginate (SA), and MXenes (Ti3C2TX) via directional freezing. Inspired by plant vascular bundles, the resulting vertically aligned porous structure enhances the mechanical stability of and mass transfer in the material. The aerogel with its high MXene loading and high surface utilization offers remarkable ad

Enhancing therapeutic effects alginate microencapsulation of thyme and calendula oils using ionic gelation for controlled drug delivery.

This study focuses on encapsulating and characterizing essential oils such as thyme and calendula oils, which are known for their therapeutic properties but are limited in pharmaceutical formulations due to their low water solubility and instability, with alginate microspheres. Alginate presents an excellent option for microencapsulation due to its biocompatibility and biological degradability. The ionic gelation (IG) technique, based on the ionic binding between alginate and divalent cations, allows the formation of hydrogel materials with high water content, mechanical strength, and biocompa

Probing insulin bioactivity in oral nanoparticles produced by ultrasonication-assisted emulsification/internal gelation.

Alginate-dextran sulfate-based particles obtained by emulsification/internal gelation technology can be considered suitable carriers for oral insulin delivery. A rational study focused on the emulsification and particle recovery steps was developed in order to reduce particles to the nanosize range while keeping insulin bioactivity. There was a decrease in size when ultrasonication was used during emulsification, which was more pronounced when a cosurfactant was added. Ultrasonication add-on after particle recovery decreased aggregation and led to a narrower nanoscale particle-size distributio

Studies of craniofacial development in rotating bioreactors.

Several studies in our laboratory assessed the effect of 3-D culture in various rotating bioreactors on craniofacial development. Initially, mouse first branchial arches were cultured. Molar and incisor development occurred in both upper and lower jaws, but maxilla development was deficient because no brain was present. In a second study using excised whole heads, the oral epithelia fused and teeth did not develop. External structure of the face was obliterated, although internally, eye development was excellent. To preserve both internal spaces and external face structure, subsequent experime

Unusual donor site reactions to calcium alginate dressings.

Calcium alginate dressings have been used as the standard dressing for split skin donor sites at the Women's and Children's Hospital burn unit for the last 12 yr. This method has proven satisfactory with good haemostasis, reliable healing, low infection rate and is well accepted by the children and nursing staff. We have recently had 5 cases with the unusual and unexplained phenomenon of dermal calcification in the donor site following the use of two new varieties of calcium alginate dressing.

Human iPSC-derived platelets generated in alginate scaffolds exhibit functional haemostasis in vitro and in vivo.

The manufacture of platelets from induced pluripotent stem cells (iPSCs) in a cost-effective, controlled and reproducible manner is a critical challenge for transfusion and regenerative medicine applications. Here we employ a high throughput, combinatorial screening method (CombiCult®) to discover new efficient protocols for generation of mature megakaryocytes (MKs) and platelets from human iPSC (hiPSC) lines. The CombiCult® platform was adapted to encapsulate hiPSCs cells in alginate by electro-spraying and cell differentiation was initiated in alginate beads, circumventing the need for strom

Efficient removal of methylene blue by sodium alginate microspheres immobilizing Aspergillus niger and Fe3O4/biochar.

The extensive discharge of synthetic dyes from textile and pharmaceutical industries has created an urgent demand for eco-friendly and cost-effective purification technologies. In this study, a magnetic bio-based hybrid microgel adsorbent (SAFBCA) was fabricated by incorporating Fe3O4/biochar (FBC) and Aspergillus niger (A. niger) into a sodium alginate (SA) hydrogel matrix. Physicochemical characterization confirmed the formation of porous microspheres with a moderate specific surface area (8.22 m2 g⁻1) and sufficient magnetic responsiveness (9.31 emu g⁻1) for facile recovery. The adsorbent a

In vitro space of Disse model for exploration of drug induced hepatotoxicity.

The limited predictive power of animal models remains a major bottleneck in drug development, particularly in assessing drug-induced liver injury (DILI). To address this, we developed a novel in vitro liver-on-a-chip platform focused on modelling the space of Disse (sD) - a key microenvironment, yet insufficiently studied, which mediates cross-talk between endothelial and hepatic systems. The system integrates a biocompatible sodium alginate hydrogel whose mechanical properties were optimised to mimic physiological liver stiffness, enabling molecular cross-talk between human liver sinusoidal e

Biomimetic recombinant humanized type III collagen peptide-RGD loaded alginate hydrogels enhance proliferation of mesenchymal-lineage cells via Integrin α5β1 activation.

Alginate hydrogels (HG) are widely utilized in tissue engineering due to their biocompatibility and tunable physical properties, however, their bio-inert nature limits direct cellular interactions. Moreover, the receptor-level interactions of recombinant humanized type-III collagen-peptide (rCP) and RGD with mesenchymal-lineage cells such as mesenchymal stem cells (MSCs), human osteoblast-like cells (hFOB), and fibroblasts (FB) remain unexplored. To address these limitations, alginate hydrogels were incorporated with RGD (R-HG), rCP (rCP-HG), and both peptides (CPR-HG), and systematically eval

Dual-responsive chitosan/alginate magnetic nanoparticles for sequential chemo-photothermal therapy of glioblastoma.

Glioblastoma (GBM) is among the most aggressive malignancies, with limited treatment success due to its invasiveness and resistance to conventional therapies. To address these challenges, we developed a dual-functional nanoplatform composed of curcumin diethyl disuccinate (CDD)-loaded chitosan/alginate-coated magnetic nanoparticles (CS/ALG-MNPs) for sequential chemo-photothermal therapy. This system integrates near-infrared (NIR)-responsive photothermal therapy (PTT) with pH-triggered drug release, using superparamagnetic iron oxide nanoparticles as the core. The nanoparticles, averaging 260 n

Quality-by-Design Optimization of Mucoadhesive Trimethyl Chitosan-Coated Alginate/Dextran Sulfate Nanoparticles for Oral Insulin Delivery.

Trimethyl chitosan (TMC)-coated alginate/dextran sulfate (ADS) nanoparticles were developed as mucoadhesive nanocarriers for oral insulin delivery using a Quality-by-Design strategy. In a first screening step, a two-level factorial design was applied to evaluate the influence of ADS concentration, TMC concentration, insulin concentration, and poloxamer® concentration on particle size and encapsulation efficiency. The screening design identified the ADS-TMC pair as the main formulation parameter for particle size, while TMC and poloxamer® were the most influential factors for encapsulation effi

Exploring the anticancer potential of epigallocatechin gallate-loaded sodium alginate nanoparticles: impact of size variation on head and neck cancer cells.

INTRODUCTION: Head and neck cancer is a common malignancy, and its current treatments face major challenges, including recurrence, drug resistance, side effects, and high costs. Epigallocatechin gallate (EGCG) from green tea has anticancer properties, but its low stability and bioavailability limit its clinical use. These limitations may be addressed by nanocarrier-based delivery systems. METHODS: Five different sizes of sodium alginate (SA) nanoparticles (NPs) were synthesized, and EGCG was loaded into the selected particles. Characterizations of SA NPs with and without EGCG were conducted u

Multifunctional hydrogel based on oxidized sodium alginate and carboxymethyl chitosan delivers minoxidil loaded liposomes for the treatment of androgenetic alopecia.

Minoxidil (MXD) is a widely used drug to treat androgenetic alopecia (AGA), the most prevalent hair loss disorder in humans. However, the therapeutic effect of common methods of administration, such as tinctures or liniments, is limited. This study aims to develop an antibacterial, anti-inflammatory, injectable, nanofiber-reinforced hydrogel platform based on oxidized sodium alginate (OSA) and carboxymethyl chitosan (CMC). This will enable the precise, point-specific, long-term, continuous delivery of minoxidil-loaded liposomes (MXD@Lip), enhance the transdermal rate and drug utilization of MX

Tissue engineering of articular cartilage using nasal chondrocytes on alginate-chitosan-based composite scaffolds.

Articular cartilage defects present a significant clinical challenge due to the tissue's limited self-repair capacity. Tissue-engineered scaffolds that support cell survival, phenotype maintenance, and extracellular matrix (ECM) formation offer a promising strategy for restoring hyaline cartilage. In this study, we synthesized a composite scaffold composed of naturally derived biomaterials alginate and chitosan (AC), and a variant supplemented with type II collagen (CAC), and evaluated their pro-chondrogenic potential. Rabbit nasal septal chondrocytes (NSCs) were selected as the cellular compo

Rapidly self-crosslinking sodium alginate hydrogel for infected wounds.

The risks associated with wound infections are significant, making a snug-fitting hydrogel dressing an optimal choice for wound management. For it, we employed the self-cross-linking method of oxidized sodium alginate (SCSA), incorporating clarithromycin (Cla) and basic fibroblast growth factor (bFGF) to formulate a rapidly forming, bacteriostatic, and wound-healing hydrogel (SCSA@C/b). Bacteriostatic and cytocompatibility assays demonstrated that SCSA@C/b exhibits exceptional antibacterial activity alongside strong biocompatibility. A fractional infected wound model showed that SCSA@C/b accel
