Several highly conserved genes play a role in anterior neural plate patterning of vertebrates and in head and brain patterning of insects. However, head involution in Drosophila has impeded a systematic identification of genes required for insect head formation. Therefore, we use the red flour beetle Tribolium castaneum in order to comprehensively test the function of orthologs of vertebrate neural plate patterning genes for a function in insect head development. RNAi analysis reveals that most of these genes are indeed required for insect head capsule patterning, and we also identified severa

The strategic use of refuge areas is a well-known method for delaying the development of pest resistance to Bacillus thuringiensis (Bt) crop. A lesser-known method to control against resistance development is sterile insect releases. In this article, an agent-based simulation model is used to test the effectiveness of combining the use of Bt sugarcane, refuge areas, and sterile insect releases as an integrated strategy against Eldana saccharina Walker (Lepidoptera: Pyralidae) infestation and resistance development. Individual insects are modeled with their own genetic traits on a simulated sug

In this paper, in-line phase-contrast synchrotron microtomography was used to study the morphology of adult Aedes aegypti. These specimens are vectors of several arboviruses, causing dengue, chikungunya, Zika and yellow fever. The morphological details of this insect species are still incomplete and insufficient. To address this gap, this study examined whole specimens of Aedes aegypti in the adult phase at high resolution. For this, the adult samples were scanned in the microtomography beamline (SYRMEP) at the Italian Synchrotron Light Laboratory (ELETTRA). The phase-contrast technique allowe

Chemical pesticides are still commonly used in Thailand for control of agricultural pests and disease vectors. Organophosphates, carbamates and synthetic pyrethroids are commonly used for agricultural purposes, whereas synthetic pyrethroids have become more popular and predominate for public health use. The genetic selection of insecticide resistance (whether physiological, biochemical or behavioral) in pests and disease vectors has been extensively reported worldwide (Brown and Pal, 1971). The long-term intensive use of chemical pesticides to control insect pests and disease vectors is often

Lack of tools for detailed, real-time observation of mosquito behavior with high spatio-temporal resolution limits progress towards improved malaria vector control. We deployed a high-resolution entomological lidar to monitor a half-kilometer static transect positioned over rice fields outside a Tanzanian village. A quarter of a million in situ insect observations were classified, and several insect taxa were identified based on their modulation signatures. We observed distinct range distributions of male and female mosquitoes in relation to the village periphery, and spatio-temporal behaviora

Control of malaria uses a variety of tools including drugs, vaccines, and insecticidal strategies to reduce parasite transmission by mosquito vectors. A new tool that eliminates parasites in mosquitoes by delivering antimalarials to the insects rather than infected people is emerging as a simple, relatively inexpensive malaria control strategy. In this Chapter, we review antimalarials that show efficacy in mosquitoes, effective methods to deliver compounds into the insects, and strategies to optimize this new tool as a useful component of the arsenal of strategies to combat malaria.

The sterile insect technique (SIT) is an effective, environmentally friendly method for insect control whose success depends on the sexual performance and survival of sterile males. These two parameters are influenced by environmental conditions of target areas, and releasing insects with a higher tolerance to stressful environments can improve SIT efficiency. Directional selection can be used to produce insect strains with higher tolerance to extreme environmental conditions, such as low humidity, for extended periods. We evaluated, under field cage conditions, the sexual competitiveness, sex…

Organic and inorganic forms of mercury are highly neurotoxic environmental contaminants. The exact mechanisms involved in mercury neurotoxicity are still unclear. Oxidative stress appears to play central role in this process. In this study, we aimed to validate an insect-based model for the investigation of oxidative stress during mercury poisoning of lobster cockroach Nauphoeta cinerea. The advantages of using insects in basic toxicological studies include the easier handling, rapid proliferation/growing and absence of ethical issues, comparing to rodent-based models. Insects received solutio

The entomopathogenic nematode Heterorhabditis bacteriophora (Nematoda: Rhabditidae) is used in biological insect control. Their dauer juveniles (DJs) are free-living and developmentally arrested, invading host insects. They carry cells of their bacterial symbiont Photorhabdus spp. in the intestine. Once inside the insect´s hemolymph the DJs perceive a food signal, triggering them to exit the DJ stage and regurgitate the Photorhabdus cells into the insect's haemocoel, which kill the host and later provide essential nutrients for nematode reproduction. The exit from the DJ stage is called "recov…

Many animals use their tactile sense for active exploration and tactually guided behaviors such as near-range orientation. In insects, tactile sensing is often intimately linked to locomotion, resulting in the orchestration of several concurrent active movements, including turning of the entire body, rotation of the head, and searching or sampling movements of the antennae. The present study aims at linking the sequence of tactile contact events to associated changes of all three kinds of these active movements (body, head and antennae). To do so, we chose the Indian stick insect Carausius mor

Agricultural intensification, driven by increasing food demand, is one of the main drivers of global biodiversity loss. Pesticides, including fungicides, may increase crop yield but pose substantial threats to the environment. However, our current knowledge on the effects of fungicides on biodiversity suffers from less research interest compared to other pesticides and a neglect of insect herbivores. We here investigate the effects of a commercially available, ready-to-use difenoconazole fungicide on a non-target insect, the butterfly Pieris napi, under controlled laboratory conditions. When b

The World Health Organization recently declared a global initiative to control arboviral diseases. These are mainly caused by pathogenic flaviviruses (such as dengue, yellow fever and Zika viruses) and alphaviruses (such as chikungunya and Venezuelan equine encephalitis viruses). Vaccines represent key interventions for these viruses, with licensed human and/or veterinary vaccines being available for several members of both genera. However, a hurdle for the licensing of new vaccines is the epidemic nature of many arboviruses, which presents logistical challenges for phase III efficacy trials.

Susceptibility to insecticides is one of the limiting factors preventing wider adoption of natural enemies to control insect pest populations. Identification and selective breeding of insecticide tolerant strains of commercially used biological control agents (BCAs) is one of the approaches to overcome this constraint. Although a number of beneficial insects have been selected for increased tolerance to insecticides the molecular mechanisms underpinning these shifts in tolerance are not well characterised. Here we investigated the molecular mechanisms of enhanced tolerance of a lab selected st

Tools that could be used to subvert the insect diapause response offer potential for insect pest management as well as for the experimental manipulation of insects and the facilitation of mass rearing procedures. In some cases, it is desirable to break diapause on demand and in other cases, it may be attractive to exploit diapause for long-term storage of biocontrol agents or valuable experimental lines. This review highlights some of the diapause disruptors reported in the literature, as well as chemical and physical manipulations that can be used to extend diapause or even induce diapause in

In landscape ecology, the waterscape refers to permanent or temporary, running or stagnant surface waters within a terrestrial area. Across ecosystem boundaries, aquatic organisms and nutrients can reach terrestrial ecosystems, as formalised by the meta-ecosystem theory. Recent studies on aquatic insects emerging from temperate streams suggest that the extent of their biomass and fluxes across agricultural landscapes may have been neglected until now. Following a conceptual and empirical approach, we presently discuss how the temporal dynamics of floods coupled with the emergence and aerial fl

Baculoviruses provide alternatives to chemicals for controlling insect pests and can be applied by spraying. Baculoviruses have a limited host range, but work relatively slowly. They are dissolved in the midgut of insect larvae to release infectious virions which enter gut epithelial cells and begin to replicate. Replication in other organs causes extensive tissue damage and eventually death. This process can take 4-5 days, but in the field may last for more than a week, allowing the larvae to feed for longer and thereby damaging the host plant. Baculovirus expression vectors expressing foreig

BACKGROUND: Onchocerciasis, or river blindness, has historically been an important cause of blindness, skin disease and economic disruption in Africa and the Americas. It is caused by the filarial parasite Onchocerca volvulus, which is transmitted by black flies in the genus Simulium. Over the past decade, several international programs have been formed to control, or more recently eliminate onchocerciasis, using mass drug administration (MDA) of ivermectin. However, in many areas of Africa (particularly those which are endemic for the eyeworm, Loa loa, or where vector densities are very high)

Visceral leishmaniasis (VL) is a deadly vector-borne disease that causes an estimated 500 000 new cases a year. In India, Nepal and Bangladesh, VL is caused by Leishmania donovani, which is transmitted from man to man by the sandfly Phlebotomus argentipes. In 2005, these three countries signed a memorandum of understanding to eliminate VL from the region. Integrated vector management is one of the pillars of this elimination strategy, alongside early case detection and treatment. We reviewed the evidence of effectiveness of different vector control methods, to examine the potential role of ins

The aerial larvicidng operation of the Onchocerciasis Control Programme of the World Health Organization which began in February 1975 resulted in a sharp reduction in Simulium damnosum numbers. However, at the onset of the rainy season the fly population increased in certain areas. Detailed surveys both on the ground and by helicopter did not reveal any significant failures of treatments that could account for the fly densities observed, and it was concluded that the flies must originate from sources outside the controlled zone. This reinvasion proved to be an annual occurrence which has been
