Ancistrocladinium A Induces Apoptosis in Proteasome Inhibitor-Resistant Multiple Myeloma Cells: A Promising Therapeutic Agent Candidate.

The N,C-coupled naphthylisoquinoline alkaloid ancistrocladinium A belongs to a novel class of natural products with potent antiprotozoal activity. Its effects on tumor cells, however, have not yet been explored. We demonstrate the antitumor activity of ancistrocladinium A in multiple myeloma (MM), a yet incurable blood cancer that represents a model disease for adaptation to proteotoxic stress. Viability assays showed a potent apoptosis-inducing effect of ancistrocladinium A in MM cell lines, including those with proteasome inhibitor (PI) resistance, and in primary MM cells, but not in non-mal

The Endoplasmic Reticulum Unfolded Protein Response in Neurodegenerative Disorders and Its Potential Therapeutic Significance.

In eukaryotic cells, the endoplasmic reticulum (ER) is the cell compartment involved in secretory protein translocation and quality control of secretory protein folding. Different conditions can alter ER function, resulting in the accumulation of unfolded or misfolded proteins within the ER lumen. Such a condition, known as ER stress, elicits an integrated adaptive response known as the unfolded protein response (UPR) that aims to restore proteostasis within the secretory pathway. Conversely, in prolonged cell stress or insufficient adaptive response, UPR signaling causes cell death. ER dysfun

Response mechanisms of Polygonatum kingianum to temperature stress: implications for cultivation practices.

Polygonatum kingianum, a medicinally significant herb indigenous to Yunnan, China, exhibits pronounced sensitivity to temperature affecting its bioactive compound accumulation and growth. However, the molecular mechanisms underlying its response to temperature stress remain poorly characterized. To elucidate these mechanisms, we integrated physiological assessments with RNA-seq analysis of P. kingianum under controlled temperatures (10°C, 25°C, 30°C, and 35°C). The results indicate that P. kingianum exhibits optimal root growth at 10-25°C but shows poor adaptation to high temperature. Temperat

Integrative RNA-Seq Analysis Reveals Stress Type-Dependent lncRNA-Centered Co-Expression Networks Across Human Cellular Stress Contexts.

Long non-coding RNAs (lncRNAs) are emerging as important regulators of cellular adaptation to environmental and molecular stress, but the extent to which their responses remain reproducible and stress-type-dependent across human stress conditions remains unclear. Here, we performed an integrative transcriptomic meta-analysis of human stress-response datasets from ASTRA and GEO, focusing on normal, non-cancerous, wild-type human cell lines exposed to oxidative stress (H2O2), hypoxia, heat stress, or UV-induced DNA damage. Gene Ontology (GO) enrichment analysis of differentially expressed (DE) p

Bronopol and benzisothiazolinone exhibit synergistic toxicity with enhanced cell stress responses exceeding the impacts of individual compounds.

Bronopol and benzisothiazolinone (BIT) constitute antimicrobial-preservative co-formulations in consumer products, creating mixture exposure scenarios. Current risk assessments evaluate individual compounds, potentially underestimating mixture toxicity. This study investigated individual and combined toxicological effects using stress granule formation as an early biomarker. A549 lung epithelial cells were exposed to Bronopol, BIT, or combinations, assessed through real-time stress granule monitoring, viability assays, oxidative stress measurements, and integrated stress response pathway analy…

Cardiac-brain integration and stress: a mechanistic role for the heartbeat-evoked potential?

The heartbeat-evoked potential (HEP) is an electrophysiological marker of cortical heartbeat processing that offers an informative index of neural interoception (i.e. the central sensing and integration of internal bodily signals). Stress is known to influence both peripheral and central physiology, and the HEP provides a metric of heart-brain integration with explanatory power beyond that of either the cardiac response or the brain alone. Emerging evidence suggests that the HEP can provide informative insight into individual differences across a range of clinical and emotional measures, with

Global Adam17 Deficiency Preserves Renal Function and Modulates Integrated Pathogenic Responses in Experimental Diabetic Kidney Disease.

Diabetic kidney disease (DKD) progression results from complex interactions between metabolic stress, inflammatory activation, maladaptive intracellular signalling, and fibrotic remodelling. While previous studies demonstrated renoprotective effects of cell-specific Adam17 deletion, the impact of global Adam17 deficiency on the integrated renal response to diabetes remains incompletely understood. Here, we investigated the effects of tamoxifen-induced global Adam17 deletion in a streptozotocin-induced murine model of type 1 diabetes. Renal function, structural injury, inflammatory responses, s

The Ionic and Metabolic Response Mechanisms of Kochia scoparia in Response to Saline-Alkaline Stress.

Saline-alkaline stress is a critical environmental issue that limits plant growth and crop production. With the expansion of salinized land, investigating the response mechanisms of plants to salt-alkali stress is crucial. Integrated ionomic and metabolomic analyses were employed to investigate the response mechanisms of Kochia scoparia in our studies. Compared with the halophyte Suaeda salsa, K. scoparia exhibits distinct ionic and metabolic strategies for coping with saline-alkaline stress. Ca, Mg, and B were significantly accumulated in K. scoparia to alleviate ion toxicity and oxidative da

Perception of response governance and collaborative attitude of hospital infection outbreak and their association with perceived stress among healthcare workers: a cross-sectional study in North-Eastern China.

OBJECTIVES: Healthcare-acquired infection outbreaks impose significant psychological stress on healthcare workers, yet limited research has examined stress perception and protective factors during institutional-level outbreak scenarios. Grounded in the Job Demands-Resources model, this study examines the relationships among healthcare workers' perceptions of response governance, collaborative attitudes and perceived stress, and tests the moderating role of collaborative attitudes. DESIGN: A cross-sectional study was conducted using an online questionnaire from May to October 2025. SETTING: H

Disentangling non-specific and condition-specific transcriptomic responses: A framework from heat stress in yeast.

A central challenge in biology is to distinguish transcriptional responses that recur across heterogeneous stress conditions from those that are specific to particular experimental contexts. Here, using Saccharomyces cerevisiae as a model system, we present an integrative transcriptomic framework to address this issue from bulk population-level datasets. We combined 13 carefully curated public transcriptomes with two in-house datasets generated under controlled heat ramp and heat shock conditions. By integrating co-expression network analysis, functional enrichment, and promoter motif discover

Evaluation of anti-leukemic activity and underlying mechanisms of the novel GSPT1 degrader AB138 in acute myeloid leukemia.

Acute myeloid leukemia (AML) is a relapsing and drug-resistant hematologic malignancy. We report AB138, a novel molecular glue degrader that recruits G1-to-S phase transition protein 1 (GSPT1) to cereblon (CRBN). In AML cell lines, AB138 induces rapid, sustained GSPT1 degradation. qPCR and immunoblotting revealed activation of the integrated stress response, as evidenced by eIF2α phosphorylation and the upregulation of ATF3 and CHOP. The subsequent depletion of the oncoproteins MCL1 and c-Myc coincides with the accumulation of cleaved caspase-3 and cleaved PARP and marked apoptosis. Flow cytom…

Molecular evolution of GPCRs: CRH/CRH receptors.

Corticotrophin-releasing hormone (CRH) is the pivotal neuroendocrine peptide hormone associated with the regulation of the stress response in vertebrates. However, CRH-like peptides are also found in a number of invertebrate species. The origin of this peptide can be traced to a common ancestor of lineages leading to chordates and to arthropods, postulated to occur some 500 million years ago. Evidence indicates the presence of a single CRH-like receptor and a soluble binding protein system that acted to transduce and regulate the actions of the early CRH peptide. In vertebrates, genome duplica

Redox control at the ER-mitochondria interface in kidney transplantation: MAM-centered stress signaling and translational organoid platforms.

Kidney transplantation is inevitably accompanied by ischemia-reperfusion injury in which oxidative stress and endoplasmic reticulum (ER) stress act as tightly interconnected drivers of mitochondrial dysfunction, inflammation, and long-term graft failure. Excessive reactive oxygen species disrupt mitochondrial homeostasis, while unresolved ER stress activates maladaptive unfolded protein response signaling, together shaping tubular cell fate. Although these processes have been extensively studied, their spatial and functional integration remains incompletely understood. Growing evidence indicat
