Mg deficiency affects leaf Mg remobilization and the proteome in Brassica napus.

In order to cope with variable mineral nutrient availability, higher plants have developed numerous strategies including the remobilization of nutrients from source to sink tissues. However, such processes remain relatively unknown for magnesium (Mg), which is the third most important cation in plant tissues. Using Mg depletion of Brassica napus, we have demonstrated that Mg is remobilized from old leaves to young shoot tissues. Moreover, this study showed that Mg depletion induces modification of nutrient uptake, especially Zn and Mn. Finally, comparative proteomic analysis of old leaves (sou

Expression of Idh1R132H in the Murine Subventricular Zone Stem Cell Niche Recapitulates Features of Early Gliomagenesis.

Isocitrate dehydrogenase 1 mutations drive human gliomagenesis, probably through neomorphic enzyme activity that produces D-2-hydroxyglutarate. To model this disease, we conditionally expressed Idh1R132H in the subventricular zone (SVZ) of the adult mouse brain. The mice developed hydrocephalus and grossly dilated lateral ventricles, with accumulation of 2-hydroxyglutarate and reduced α-ketoglutarate. Stem and transit amplifying/progenitor cell populations were expanded, and proliferation increased. Cells expressing SVZ markers infiltrated surrounding brain regions. SVZ cells also gave rise to

The role of mutations in epigenetic regulators in myeloid malignancies.

Recent genomic studies have identified novel recurrent somatic mutations in patients with myeloid malignancies, including myeloproliferative neoplasms (MPNs), myelodysplastic syndrome (MDS) and acute myeloid leukaemia (AML). In some cases these mutations occur in genes with known roles in regulating chromatin and/or methylation states in haematopoietic progenitors, and in other cases genetic and functional studies have elucidated a role for specific mutations in altering epigenetic patterning in myeloid malignancies. In this Review we discuss recent genetic and functional data implicating muta

Ollier disease and Maffucci syndrome are caused by somatic mosaic mutations of IDH1 and IDH2.

Ollier disease and Maffucci syndrome are characterized by multiple central cartilaginous tumors that are accompanied by soft tissue hemangiomas in Maffucci syndrome. We show that in 37 of 40 individuals with these syndromes, at least one tumor has a mutation in isocitrate dehydrogenase 1 (IDH1) or in IDH2, 65% of which result in a R132C substitution in the protein. In 18 of 19 individuals with more than one tumor analyzed, all tumors from a given individual shared the same IDH1 mutation affecting Arg132. In 2 of 12 subjects, a low level of mutated DNA was identified in non-neoplastic tissue. T

Isolation and curing of the Klebsiella pneumoniae large indigenous plasmid using sodium dodecyl sulphate.

The ability of Klebsiella pneumoniae (NCTC, CL687/80) to produce and, in turn, excrete glutamate has been equated with the presence of a large indigenous plasmid with an apparent molecular mass in the region of 96 +/- 2 kbp (n = 6). Unlike mitomycin C, novobiocine and ethidium bromide (curing agents), the use of sodium dodecyl sulphate (SDS) proved very effective in curing the plasmid with a relatively high frequency (6.25 x 10(-4)). Furthermore, the absence of isocitrate dehydrogenase (ICDH) activity in the cured strain strongly suggests that the structural gene encoding ICDH in this organism

Dietary N-carbamylglutamate and l-arginine supplementation improves intestinal energy status in intrauterine-growth-retarded suckling lambs.

This study explores the roles of l-arginine (Arg) and N-carbamylglutamate (NCG) supplementation in the diet in intestine damage, energy state, as well as the associated protein kinase signaling pathways activated by AMP in intrauterine growth retarded (IUGR) suckling lambs. A total of 48 newborn Hu lambs with a normal birth weight (CON) and those with IUGR were randomly divided into four groups, CON, IUGR, IUGR + 1% Arg, and IUGR + 0.1% NCG, with 12 animals in each group. All animals were fed for 21 days, from day 7-28, following birth. Our results indicated that the IUGR suckling Hu lambs in

Metabolic zonation in teleost gastrointestinal tract. Effects of fasting and cortisol in tilapia.

Activities of several metabolic enzymes show distinct patterns of zonation along the intestinal tract of tilapia (Oreochromis niloticus), rainbow trout (Oncorhynchus mykiss) and copper rockfish (Sebastes caurinus). Zonation is species and enzyme specific, with different metabolic activities concentrated in specific areas, and few generalizations can be made. The rockfish show the smallest degree of zonation, with highest activities in the third quarter of the intestine, and shallow gradients to either side, and a general upswing in activity towards the distal end. In the trout, mitochondrial e

A Key Metabolic Protein in Active Mycobacterium tuberculosis: Insights into Carbon, Nitrogen, and Sulfur Metabolism.

Mycobacterium tuberculosis (Mtb) employs a highly adaptable network of metabolic pathways that are pivotal for its survival and pathogenesis within the host during both exponential growth and persistent infection phases. Central carbon metabolism in Mtb exhibits remarkable flexibility, enabling the bacterium to utilize diverse carbon sources efficiently. In the absence of glucose, Mtb preferentially metabolizes fatty acids as primary carbon substrates. This metabolic shift is supported by the glyoxylate shunt and methyl citrate cycle, which replenish tricarboxylic acid (TCA) cycle intermediate

Astrocytoma in Patients with HIV-Review of the Literature and Case Report.

Astrocytomas are among the most common primary tumors of the central nervous system, arising from astrocytic glial cells and encompassing a wide spectrum of histopathological grades and clinical behaviors. Human immunodeficiency virus (HIV) infection is characterized by chronic immune dysregulation, neuroinflammation, and increased susceptibility to both opportunistic infections and malignancies. The management of astrocytomas in patients living with HIV presents unique clinical challenges but should, whenever feasible, follow standard neuro-oncological principles. We report the case of a 34-y

Potential Prognostic Markers for Glioblastoma Associated with the Glioma Immune Microenvironment.

Glioblastoma (GBM) is a highly aggressive and lethal brain tumor with very poor prognosis despite recent progress in multimodal treatments. Within glioma tissue, various niche cells such as macrophages and neutrophils form a unique glioma immune microenvironment (GIME) by interacting with heterogenous cancer cells, and this has been implicated in disease progression and responsiveness to immunomodulatory therapies. This study explores novel potential prognostic markers associated with the GIME using integrated bioinformatics analyses, including single-cell RNA-sequencing (scRNA-seq), and spati

IDH mutations may not preclude distant, trans-tentorial spread in gliomas: a case report and review of the literature.

BACKGROUND: IDH mutations have been demonstrated to confer prolonged survival in patients suffering from gliomas, but the mechanisms underlying the improved prognosis are unclear. While some studies have attributed these observations to an enhanced sensitivity to genotoxic therapies, others have postulated that IDH-mutated gliomas exhibit less aggressive intrinsic biological behavior, including the propensity to invade distant sites. Although most gliomas recur local to the site of initial presentation, some tumors demonstrate distant recurrence, the vast majority of which involve the contrala

Maffucci syndrome and neoplasms: a case report and review of the literature.

BACKGROUND: Maffucci syndrome is characterized by the sporadic occurrence of multiple enchondromas together with multiple hemangiomas. Patients with Maffucci syndrome are at increased risk of developing different kinds of malignant tumors. CASE PRESENTATION: We report on a 39-year-old woman who was diagnosed with Maffucci syndrome together with intrahepatic cholangiocarcinoma (IHCC). Heterozygous somatic mutations in the isocitrate dehydrogenase 1 and 2 (IDH1/IDH2) genes are associated with a number of different tumor types (e.g. IHCC) and also with Maffucci syndrome. For IHCC, mutations in I

An unresectable cholangiolocarcinoma diagnosed by integrating biopsy-based histopathology and genomic findings with clinical and imaging data.

Cholangiolocarcinoma is a rare malignant liver tumor typically diagnosed on pathological evaluation of surgically resected specimens. Establishing a definitive diagnosis in unresectable cases is therefore challenging. A case of a large unresectable liver tumor, extending across the hepatic hilum into both lobes, is presented. Despite the advanced stage, the patient's serum total bilirubin and carbohydrate antigen 19-9 levels remained within normal ranges. On dynamic computed tomography, characteristic imaging features of CLC, including intrahepatic vessel penetration, were seen. Histopathologi

METTL3 regulates α-KG-dependent mitophagy and apoptosis in nucleus pulposus cells through the MALAT1/miR-23c/IDH1 axis.

Alpha-ketoglutarate (α-KG), a key intermediate in the tricarboxylic acid (TCA) cycle, was found to be significantly decreased in nucleus pulposus (NP) tissues of patients with intervertebral disc degeneration (IVDD). Supplementation with α-KG restored nucleus pulposus cell (NPC) proliferation, reduced apoptosis, and reestablished extracellular matrix (ECM) metabolic homeostasis. Mechanistically, α-KG enhanced mitophagy and suppressed reactive oxygen species (ROS) accumulation, effects that were abolished by the mitophagy inhibitor Mdivi-1. Further investigation identified isocitrate dehydrogen

In vitro and in silico analysis reveals antifungal activity and potential targets of curcumin on Paracoccidioides spp.

The search for new compounds with activity against Paracoccidioides, etiologic agents of Paracoccidioidomycosis (PCM), is extremely necessary due to the current scenario of the available therapeutic arsenal. Treatment is restricted to three classes of antifungals with side effects. Curcumin is a polyphenol with antifungal effects that is extracted from Curcuma longa. The present work aimed to evaluate the activity of curcumin in different species of Paracoccidioides and to evaluate the potential molecular targets of curcumin using computational strategies. In addition, interactions with classi

[Glioblastoma Mimicking Meningioma Recurrence: A Case Report].

Meningioma and glioma represent two common primary intracranial tumors. However, the coexistence of these two lesions in the same patient at the same location is rare. Here, we present a case of a fibroblastic meningioma with a secondary glioblastoma occurring at the same location. A 67-year-old woman underwent surgery for a left frontal parasagittal meningioma, and the tumor was subtotally removed. Two years and 11 months after the surgery, the patient had a tumor at the same location with invasion into the adjacent brain, suggesting recurrent meningioma with malignant transformation. The res

Brain Tumors of Glial Origin.

Gliomas are a heterogeneous group of tumors with evolving classification based on genotype. Isocitrate dehydrogenase (IDH) mutation is an early event in the formation of some diffuse gliomas, and is the best understood mechanism of their epigenetic dysregulation. Glioblastoma may evolve from lower-grade lesions with IDH mutations, or arise independently from copy number changes in platelet-derived growth factor receptor alpha (PDGFRA) and phosphatase and tensin homolog (PTEN). Several molecular subtypes of glioblastoma arise from a common proneural precursor with a tendency toward transition t
