Bradykinin B2 receptors mediate contraction in the normal and inflamed human gallbladder in vitro.

BACKGROUND & AIMS: The components of the kinin system, including kinongens, kininogenases, and B(2) and B(1) receptors, are expressed and activated during inflammation. Here, we investigated the expression of the kinin B(2) receptor messenger RNA, kininogen and kallikrein immunoreactivity, and the ability of kinins to contract control and inflamed gallbladders in vitro. METHODS: Human gallbladders, obtained from patients undergoing cholecystectomy either for acute cholecystitis secondary to gallstone disease or during elective gastro-entero-pancreatic surgery (controls), were processed for re…

Protective actions of human tissue kallikrein gene in transgenic rat hearts.

In this study, we used an experimental model of cardiac hypertrophy to explore the role of the kallikrein-kinin system (KKS) in cardiac protection in transgenic rats harboring the human tissue kallikrein gene, TGR(hKLK1). Tissue kallikrein cleaves low-molecular-weight kininogen to produce kinin peptides, which bind to kinin receptors and trigger a wide spectrum of biological effects. The transgene, under the control of the zinc-inducible metallothionein promoter, was expressed in most tissues including the heart, kidney, lung and brain. These animals were subjected to treatment with thyroid ho

[The use of the kallikrein-kininogen system indices in the prognosis of the development of hemorrhagic erysipelas].

A number of the kallikrein-kinin system parameters (kallikrein, prekallikrein, total arginine esterase activity, alpha 1 protease inhibitor, and alpha 2 macroglobulin) were measured in 59 patients with erythematous erysipelas and in 51 ones with hemorrhagic erysipelas over the course of the disease. Marked activation of the blood kallikrein-kinin system was seen in all the patients during the initial period of the disease, manifesting by elevated levels of kallikrein, total arginine esterase activity, alpha 1 protease inhibitor, alpha 2 macroglobulin, and a lowered prekallikrein concentration.

Rat uterine contraction by kallikrein and its dependence on uterine kininogen.

Smooth muscle responses to kallikrein (EC 3.4.21.8) are generally considered to result from kinin formation. In the present study, this premise was reexamined with respect to the isolated rat uterus. Rat submandibular gland kallikrein produced contractions of the rat uterus but the contractions disappeared after successive additions of the same dose of the enzyme to the preparation. Kallikrein-induced rat uterine contractions as well as bradykinin-induced contractions were enhanced by rat submandibular gland bradykinin potentiating factor. The incubation of kallikrein with rat uterine extract

Mechanism of antihypertensive effect of atenolol in patients with borderline hypertension during short-term treatment. A comprehensive study.

Short-term treatment with 1-(p-carbamoyl-methylphenoxy)-3-isopropylamino)-2-propanol (atenolol, Tenormin) (100 mg/d for 5 days) was conducted in 12 patients with labile essential hypertension. Before and at the end of the treatment, cardiohemodynamics, renal hemodynamic and excretory function, and renal pressor (renin-angiotensin-aldosterone) and depressor (renal kallikrein-kinin and prostaglandin) systems were examined. Atenolol decreased cardiac output (CO) without affecting total peripheral resistance. Atenolol also decreased plasma renin activity, plasma aldosterone concentration, urinary

Bradykinin levels in inflammatory exudates.

An enzyme immunoassay was used to measure the levels of bradykinin (BK)-like immunoreactivity in inflammatory exudates provoked by calcium p … ssolved in saline, were also negative. The present results indicate that an increased production of kinins is detectable only in exudates induced by CaPP and not in those provoked by carrageenin. Our data also suggest that the kinins are rapidly metabolized, both in vitro (in the exudate sample) and in vivo (in the rat pleural cavity).

Plasma kallikrein and diabetic macular edema.

Recent proteomic studies have identified components of the kallikrein kinin system, including plasma kallikrein, factor XII, and kininogen, in vitreous obtained from individuals with advanced diabetic retinopathy. In rodent models, activation of plasma kallikrein in vitreous increases retinal vascular permeability; whereas inhibition of the kallikrein kinin system reduces retinal leakage induced by diabetes and hypertension. These findings suggest that intraocular activation of the plasma kallikrein pathway may contribute to excessive retinal vascular permeability that can lead to diabetic mac

Kininase I drives angiotensin II-induced hypertension through activation of the bradykinin B1 receptor.

BACKGROUND AND PURPOSE: Kininase I, comprising carboxypeptidase N (CPN) and carboxypeptidase M, regulates kinin activity and is up-regulated during injury, inflammation and immune activation. Under these conditions, kininase I converts bradykinin and kallidin into des-Arg kinins, primary endogenous agonists of the bradykinin B1 receptor (R). B1R signalling promotes vasoconstriction and inflammatory responses in the pathogenesis of hypertension. The roles of kininase I in regulating kinin-dependent signalling and inter-system interactions that contribute to blood pressure control and hypertensi…

Neurocysticercosis and pharmacoresistant epilepsy: possible role of calcified lesions in epileptogenesis.

Neurocysticercosis is a neglected and usually poverty-related disease of high public importance. The mechanisms by which the calcified lesions cause epilepsy are not known, but have been attributed to residual perilesional gliosis or an inflammatory process. This case shows that an inflammatory response to a calcified granuloma may be associated with the development of epilepsy. The increase in glutamate and kinin B1 (pro-epileptogenic) receptors added by reduced expression of kinin B2 (anti-epileptogenic) receptors may explain the chronic epileptogenesis associated with the lesion, corroborat

[Vasoactive polypeptides in rosacea].

Plasma kininogenesis components were studied in 110 patients with rosacea and in 3 groups of subjects at risk of developing this condition because of chronic gastrointestinal diseases, nervous system stress, or endocrine dysfunctions, conducive to rosacea. Changes in kininogenesis parameters similar to those detected in all rosacea patients were detected in all the 3 risk groups. Vasoactive polypeptide hyperproduction is a common component of rosacea pathogenesis. Vasoactive trend of the kinin action in kinin hyperproduction in rosacea induces vascular changes in the face and general vascular

Cutting edge: bradykinin induces IL-12 production by dendritic cells: a danger signal that drives Th1 polarization.

Dendritic cells play a major role in the induction of both innate and acquired immune responses against pathogenic invaders. These cells are also able to sense endogenous activation signals liberated by injured tissues even in the absence of infection. In the present work, we demonstrate that kinins mobilize dendritic cells to produce IL-12 through activation of the B(2) bradykinin receptor subtype and that bradykinin-induced IL-12 responses are tightly regulated both by angiotensin-converting enzyme, a kinin-degrading peptidase, and by endogenous IL-10. Using a mouse model of allergic inflamm…

Peptidylarginine Deiminase of Porphyromonas gingivalis Modulates the Interactions between Candida albicans Biofilm and Human Plasminogen and High-Molecular-Mass Kininogen.

Microorganisms that create mixed-species biofilms in the human oral cavity include, among others, the opportunistic fungus Candida albicans … ninogen to C. albicans cells. Competitive radioreceptor assays tested the affinity of citrullinated kinins to their specific cellular receptors. The citrullination of surface-exposed fungal proteins reduced the level of unmodified plasminogen binding but did not affect unmodified kininogen binding. However, the modification of human proteins did not disr …

Effects of antihistamines on inflammatory mediators.

The nasal antigen challenge model has proved useful in assessing the roles of inflammatory mediators in the clinical allergic response. Studies using this model have revealed that the acute allergic response is associated with increased concentrations of histamine, prostaglandin D2 (PGD2), leukotrienes, tryptase, and kinins, and with increased TAME-esterase activity. The effects of antihistamines on the clinical response and inhibition of mediator release have also been examined with this model. Premedication with terfenadine caused a marked reduction in sneezing as well as decreased histamine…

Lipopolysaccharide injection into the cerebral ventricle evokes kininogen induction in the rat brain.

Kinins, such as bradykinin and Lys-bradykinin, are important mediators in peripheral inflammation. Although the existence of the components necessary for generating kinins has been demonstrated in the brain, a functional role of the kinin-generating system in cerebral inflammation remains to be defined. The aim of the present study was to elucidate whether inflammatory stimuli alter the mRNA levels of components for the kallikrein-kinin system, including kallikreins, kininogens and bradykinin type 2 (B(2)-) receptor in rat brain using the reverse transcription polymerase chain reaction. The in…

Human kallikrein gene delivery attenuates hypertension, cardiac hypertrophy, and renal injury in Dahl salt-sensitive rats.

The tissue kallikrein-kinin system has been documented to be involved in the pathogenesis of hypertension and renal diseases. To investigate the protective effects of kallikrein gene delivery on salt-induced renal damage, cardiac dysfunction, and hypertension, adenovirus harboring the human tissue kallikrein gene under the control of the cytomegalovirus promoter Ad.CMV-cHK was delivered into Dahl salt-sensitive (Dahl-SS) rats fed to a high-salt (4% NaCl) diet. A single intravenous injection of Ad.CMV-cHK resulted in a significant reduction of blood pressure beginning 2 days post injection and

Mode of action of a staphylococcal anti-inflammatory factor.

A mucopeptide protein residue, DOCR, is an impedin, isolated from virulent strains of Staphylococcus aureus, which has been shown previously to potentiate staphylococcal infection by inhibiting the early phase of the inflammatory response. Comparison of the effects of DOCR with those of various pharmacological inhibitors on inflammation in vivo and on the guinea-pig ileum in vitro suggests that DOCR prevents the release of kinins by acting somewhere in the Hageman factor-kinin pathway. DOCR did not prevent the more massive fluid exudate which occurred in previously infected animals. It had no …

Effects of aldosterone and deoxycorticosterone on the urniary excretion of kallikrein and of prostaglandin E-like substance in the rat.

Deoxycorticosterone (5 mg) and aldosterone (0.25 mg), given to rats for 14 days, increased the urinary excretion of kallikrein and of prostaglandin E-like substance and produced polyuria, but affected neither sodium excretion nor blood pressure. These results suggest that interactions of mineralocorticoid hormones, kinins and of prostaglandins may be important in the maintenance of salt-water homeostasis.

Effects of captopril and bradykinin on chorda tympani-induced salivation in cat.

The effects of captopril and bradykinin on chorda tympani-induced salivation were studied in the submandibular gland of anesthetized cat. Captopril and bradykinin, but not des-Arg9-bradykinin, increased salivation evoked by electrical stimulation of chorda tympani. These potentiated effects of captopril and bradykinin were suppressed by Hoe-140, a kinin B2 receptor antagonist, or by concomitant addition of N(G)-nitro arginine methyl ester (L-NAME), a nitric oxide synthase inhibitor, and aspirin, a cyclooxygenase inhibitor. It appears that captopril potentiates chorda tympani-induced salivation

Kallikrein gene delivery attenuates myocardial infarction and apoptosis after myocardial ischemia and reperfusion.

The tissue kallikrein-kinin system is present in the heart, and kinin has been shown to have cardioprotective effects. In this study, we investigated the potential role of tissue kallikrein in myocardial ischemia/reperfusion injury through adenovirus-mediated human kallikrein gene delivery. One week after gene delivery, the rats were subjected to a 30-minute coronary occlusion followed by a 2-hour reperfusion. Kallikrein gene delivery caused significant decreases in the ratio of infarct size to ischemic area at risk (from 69.6% to 44.5%, n=10 and 8, P<0.01) and in the incidence of ventricular
