Structural optimization based on 4,5-dihydropyrazolo[1,5-a]quinazoline scaffold for improved insecticidal activities.

The long-term and irrational application of insecticides has increased the rate of development of pest resistance and caused numerous environmental issues. To address these problems, our previous work reported that 4,5-dihydropyrazolo[1,5-a]quinazoline (DPQ) is a class of gelled heterocyclic compounds that act on insect γ-aminobutyric acid receptors (GABAR). DPQ scaffold has no cross-resistance to existing insecticides, so the development of this scaffold is an interesting task for integrated pest management. In the present study, a novel series of 4,5-dihydropyrazolo[1,5-a]quinazolines (DPQs)

Bacterial cellulase from the intestinal tract of the sugarcane borer.

Cellulolytic enzymes have wide use in several industrial segments (e.g. biofuels, pulp and paper, food, and cosmetics). However, one of the challenges is their large-scale production with high specific activity to eliminate the dependence of the purchase of enzymatic cocktails produced by commercial parties. The aims of this study were (1) isolation, selection, and partial characterization of bacterial cellulases present in the intestinal tract of the sugarcane borer and (2) to identify cellulase-producing bacteria by analyzing the 16S rDNA gene. Cellulase production and purification assays re

Exposure route mediates toxicological effects of sulphur and fluxapyroxad fungicides in a non-target butterfly.

Agricultural intensification and the associated increase in pesticide use have raised concerns about impacts on non-target insects. However, … icant differences among fungicides. These findings demonstrate that fungicide effects on non-target Lepidoptera are route-dependent, resulting in sublethal impacts via ingestion and lethal effects via contact exposure. Our results thus emphasize the need to consider (1) the dual risk posed by fungicides through both lethal and sublethal effects, (2) exposure …

Ecoinformatics Can Infer Causal Effects of Crop Variety on Insect Attack by Capitalizing on 'Pseudoexperiments' Created When Different Crop Varieties Are Interspersed: A Case Study in Almonds.

Capturing the complementary strengths of observational and experimental research methods usually requires the researcher to gather separate … effect of different commercial varieties of almond, Prunus dulcis (Mill.) on the impact of two key lepidopteran pests, the navel orangeworm Amyelois transitella (Walker) (Lepidoptera: Pyralidae), and the peach twig borer Anarsia lineatella Zeller (Lepidoptera: Gelechiidae). Almonds are universally planted as polycultures of different varieties to obtain effi …

HarmGR13 mediates myo-inositol taste perception in Helicoverpa armigera larvae.

Myo-inositol, a sugar alcohol produced by most plants, serves as a nutrient and feeding stimulant for many phytophagous insects. Inositol-sensitive taste sensilla have been characterized in many Lepidoptera larvae, but their molecular bases remain unclear. In this study, we determined the gustatory receptors (GRs) for myo-inositol in larva of Helicoverpa armigera, a worldwide crop pest. First, electrophysiological analyses revealed that medial sensilla styloconica strongly responded to myo-inositol and ribose, with weaker responses to xylose, and one GRN inside sensillum may mediate the respon…

Attraction of Dibrachys cavus (Hymenoptera: Pteromalidae) to its host frass volatiles.

The European grapevine moth, Lobesia botrana (Lepidoptera: Tortricidae), is a polyphagous insect able to develop on grapes and wild plants. We tested the hypothesis that the parasitoid Dibrachys cavus (Hymenoptera: Pteromalidae) uses the larval frass in its host search. A two-armed olfactometer was used to measure the attractiveness of L. botrana larvae, their silk, or their frass after larvae were fed on different host plants. Frass of three Lepidoptera (L. botrana, Eupoecillia ambiguella, Sphinx ligustri) and one Orthoptera (Chorthippus brunneus) was assayed, but only L. botrana was used to …

The Diversity of Bacterial Symbionts in Major Agricultural Lepidopteran Pests in Australia.

Lepidopteran pests are among the most destructive insects in agriculture, causing major yield losses across a wide range of crops worldwide. With growing emphasis on reducing chemical pesticide use, there is increasing interest in understanding their associated microbiomes and endosymbiotic bacteria. These microbial associates can influence host physiology, fitness, and pesticide/thermal resistance traits, and may provide novel opportunities for developing sustainable pest management strategies. Here, we characterized the diversity and distribution of various endosymbionts and other microbiota…

Constraints on chromosome evolution revealed by the 229 chromosome pairs of the Atlas blue butterfly.

Chromosomal arrangements are important for processes including genetic recombination, adaptation, and speciation. Related taxa often possess similar numbers of chromosomes, but some groups show remarkable variation in chromosome numbers. Most Lepidoptera, the butterflies and moths, have 31 or 32 chromosomes, but some species deviate from this norm. We present a chromosome-level genome assembly of a heterogametic female Atlas blue butterfly (Polyommatus atlantica; Lycaenidae) and find that it has 227 pairs of autosomes and four sex chromosomes, the highest recorded chromosome number in non-poly…

Sphingidae moths as a new host record for Ophryocystis aff. elektroscirrha (Apicomplexa: Neogregarinida) in Brazil.

Ophryocystis elektroscirrha is a parasite primarily described in Danaini butterflies. We investigated the occurrence of Ophryocystis-like cysts in lepidopterans from collections and field samplings in Mato Grosso, Brazil. We report the first record of Ophryocystis aff. elektroscirrha in Lepidoptera from Brazil and the first worldwide record in the Sphingidae (Eumorpha analis and Agrius cingulata). No infections were detected in Danaus species. Our findings suggest that the known host range of Ophryocystis is underestimated. We highlight the importance of examining non-traditional host groups a…

Engineered expression of the invertebrate-specific scorpion toxin AaHIT reduces adult longevity and female fecundity in the diamondback moth Plutella xylostella.

BACKGROUND: Previous genetic pest management (GPM) systems in diamondback moth (DBM) have relied on expressing lethal proteins ('effectors') … d the feasibility of using non-cell-autonomous effectors within a GPM context for the first time in Lepidoptera, one of the most economically damaging orders of insects. These findings provide a framework for extending this system to other pest Lepidoptera and to other secreted effectors. © 2021 The Authors. Pest Management Science published by John Wiley & S …

Monitoring and Behavioral Control of Lepidopteran Pests in Soybean: Evaluation of Food-Based Attractants, Trap Performance, and Selectivity on Non-Target Organisms.

Chemically mediated behavioral strategies, including semiochemicals and food-based attractants, have gained prominence in Integrated Pest Management (IPM) programs because of their potential for selective and environmentally friendly pest control. The objective of this study is to evaluate the use of food-based attractants and two trap models (delta-ajar and funnel-ball) for monitoring adult lepidopteran pests in soybean fields. Additionally, the field performance and selectivity of an attract-and-kill (attractant + insecticide) strategy were assessed. Field trials were conducted in commercial…

Caterpillar dermatitis.

Lepidopterism is the term used to describe the aggregate adverse medical effects resulting from contact with adult or larval forms of butterflies and moths. Histamine, histamine-releasing substances, kinin activators, and other as yet undefined proteins are responsible for cutaneous, cardiovascular, neurologic, and constitutional signs and symptoms of lepidopterism. Insect venom is likely injected into the skin through specialized caterpillar hairs when contact occurs with the insect (or vegetation laden with insect debris). Mild cases of lepidopterism will resolve spontaneously; systemic cort
