Differentiation of human leukemia cells and its usefulness for clinical diagnosis.

Various chemical inducers have effects on the induction of terminal differentiation of human myelogenous leukemia cell lines. We studied morphological and functional changes of human leukemia cells freshly obtained from patients using 12-0-tetradecanoyl phorbol-13-acetate (TPA), retinoic acid (RA) or dimethyl sulphoxide (DMSO). The myeloid leukemia cells cultured with TPA became adherent to plastic culture dishes, and then developed macrophage-like morphology with long filamentous pseudopods within 48 h incubation. They showed marked enhancement of the ability to phagocytose latex particles. B

Trisomy 13: a new recurring chromosome abnormality in acute leukemia.

A new recurring chromosome abnormality was identified in 8 of 621 consecutive successfully karyotyped adults with de novo acute leukemia. These eight patients had trisomy 13 as the sole cytogenetic abnormality. On central morphologic review, five cases were classified as subtypes of acute myeloid leukemia, one as acute mixed lymphoid and myeloid leukemia, one as acute lymphoid leukemia, and one as acute undifferentiated leukemia. Blasts of all eight cases expressed one or more myeloid differentiation antigens. Three also expressed T-lineage-associated antigens; however, none of these had rearr

Germline Predisposition to Pediatric Lymphoid Malignancies: Genetic Tumor Syndromes Identified in a Single-Center Study.

BACKGROUND: Germline predisposition (GP) is associated with a variety of hematolymphoid malignancies. While GP has been addressed mostly in myeloid malignancies, recent diagnostic systems have newly introduced GP in lymphoid malignancies; however, evidence and data are limited, particularly in pediatric patients. In this study, we investigated the frequency and characteristics of GP in pediatric lymphoid malignancies. METHODS: The study subjects were pediatric patients diagnosed with lymphoid malignancies by bone marrow (BM) study between April 2021 and August 2024 at Samsung Medical Center,

Lymphoid lesions of the oral cavity.

The lymphoid lesions of the oral cavity may be classified into three groups: 1. Benign lesions. 2. Malignant lesions. 3. Other lymphoid-like lesions. 1. Benign lymphoid lesions: In this group the following lesions must be differentiated: reactive lymph node hyperplasia, enlarged follicle of the lingual tonsil, lymphoepithelial cysts, lymphoepithelial lesions, lymphoid polyps, and cystadenolymphomas. The most frequent locations of benign lymphoid lesions are the lingual tonsil, buccal mucosa, tongue, and floor of mouth. 2. Malignant lesions: One subgroup includes the systemic lymphoid diseases,

Soft tissue aspiration cytopathology of malignant lymphoma and leukemia.

BACKGROUND: Malignant lymphoma (ML) and leukemia infrequently involve soft tissue and, to the authors' knowledge few reports exist regarding the role of fine-needle aspiration biopsy (FNAB) in their diagnosis. In the current study, the authors report their experience with FNAB in patients with soft tissue ML and leukemia. METHODS: All cases of ML, leukemia, or atypical lymphoid cells from soft tissue aspirates were reviewed. Masses from lymph node-rich sites, those adjacent to enlarged lymph nodes, or those associated with cutaneous lesions were excluded. RESULTS: Twenty-one patients (male:f

[Clinical course and prognosis in T-cell lymphoblastic tumors in children typed with Soviet-produced monoclonal antibodies].

Correlation analysis of the clinical course and prognosis was made in 56 children with T-cell lymphoblastic tumors (acute lymphoblastic leukemia and lymphosarcoma) typed with the use of monoclonal antibodies obtained in this country. The frequency of T-cell marker detection among all immunological subvariants in acute lymphoid leukemia and lymphosarcoma proved to be similar (36.6 and 37%, respectively). In acute lymphoid leukemia type T1 of tumor cells prevailed (58%), while in lymphosarcoma type T3 (63%). T2 and T3-cell lymphoblastic tumors were characterized by the most severe clinical cours

Acute interstitial nephritis in T-cell leukemia in a pediatric patient.

Acute lymphoid leukemia is the most frequently occurring malignancy in childhood, but acute tubulointerstitial nephritis with associated acute renal failure as the leading manifestation of leukemia is extremely rare. Only a few pediatric cases have been described in the literature. We present a surprising case in which physical examination and initial investigation were not typical for leukemia. Ultrasound showed only modest kidney enlargement while laboratory results indicated acute renal failure. Renal biopsy indicated tubulointerstitial nephritis, and subsequent steroid treatment led to sud

[New insights into acute lymphoblastic leukemia].

Acute lymphoblastic leukemia is a malignant disorder of lymphoid progenitor cells. Advances in our understanding of lymphoblastic leukemia have mainly come from new molecular technologies and genomics. This article describes recent advances in our understanding of maturation arrest of leukemic cells, initial and subsequent gene defects and rearrangements, the role of chemokines, and lymphoid cell homing. These advances point to new ways of targeting leukemic cells.

Lactate dehydrogenase isoenzymes in normal and malignant human lymphoid cells.

Intracellular lactate dehydrogenase (LD) isoenzyme patterns were studied in the malignant cells of patients with a variety of lymphoid malignancies. These were compared with intracellular LD isoenzyme patterns of normal lymphoid cells and were also correlated with immunologic cell surface marker characteristics. Results showed that, in general, the malignant B cells of Burkitt's lymphoma and the lymphoblasts of T-cell acute lymphoblastic leukemia had isoenzyme patterns similar to those of normal B and T cells, respectively. The isoenzyme patterns of malignant lymphoid cells from patients with

Causal association between endocrine diseases and lymphoid malignancies explored through two-sample Mendelian randomization analysis.

Endocrine diseases are suspected contributors to lymphoid malignancies, but their precise association is unclear. This study aimed to investigate the causal relationship between various endocrine diseases-specifically type 2 diabetes, obesity, diabetic hypoglycemia, hyperlipidemia, and hyperthyroidism-and lymphoid malignancies, including lymphocytic leukemia and non-Hodgkin's lymphoma with subtypes like diffuse large B-cell lymphoma and follicular lymphoma. Utilizing data from genome-wide association studies, a two-sample Mendelian randomization analysis was performed. The primary approach inv

[Patterns in the lesions of different sections of the hemopoietic system in leukemia].

Morphological examinations of the hemopoietic system in 43 patients died of different forms of leukemias revealed irregular involvement into the leukemic processes of its different parts. Differences in the spread of leukemic lesions in the spleen, lymph nodes, tonsils and other lymphoid formations in lympho- and myeloleukemias were demonstrated. They are discussed from the point of view of the concept on metastatic involvement of different parts of the hemopoietic system in leukemias and on the importance in the development of these lesions of the cellular microenvironment. CD7+, CD4-, CD8- acute leukemia: a syndrome of malignant pluripotent lymphohematopoietic cells.

Following our initial observation of in vivo conversion of CD7+, CD4-, CD8- acute lymphoblastic leukemia (ALL) cells from lymphoid to myeloid lineages (Proc Natl Acad Sci (USA) 81:253, 1984) we have studied eight additional cases of ALL with this leukemic cell phenotype. The CD7+, CD4-, CD8- phenotype was associated with a distinct clinical entity with those affected predominantly male (either less than 35 years or greater than 65 years of age), with frequent mediastinal and/or thymic masses, skin and CNS disease, high peripheral WBC counts, and bone marrow blasts that were morphologically L1

Abnormalities of the retinoblastoma gene in the pathogenesis of acute leukemia.

The retinoblastoma-susceptibility (Rb) gene is an antioncogene that is frequently altered in retinoblastomas, sarcomas, and some epithelial tumors. We examined the structure of the Rb gene by Southern blotting in 215 cases of leukemias and lymphomas of diverse phenotype and in 15 leukemic cell lines. In selected cases Rb protein expression was examined with specific monoclonal antibodies. Structural abnormalities of the Rb gene with absent protein expression were frequent in all types of human acute leukemia, but were particularly common (27% incidence) in M4 and M5 myeloid leukemia with monoc

In Situ Leukemia as Manifestation of Relapse After Allogeneic Transplantation of Bone Marrow of ALL B: A Case Report.

Acute lymphoid leukemia (ALL) is a malignant proliferation of abnormal lymphoid precursors in the bone marrow. It is the most common acute leukemia in childhood, accounting for only 20% of acute leukemia in adults. The clinical presentation is heterogeneous including pallor, fatigue and weakness. Diagnosis is performed through myelogram, bone marrow biopsy and immunophenotyping. Chemotherapy and, in some cases, bone marrow transplantation (BMT) are used to treat the disease. However, the efficacy of treatment in adult patients is still below expected and therapeutic innovations have not increa

Geographical correlation between ambient UVB level and mortality risk of leukemia in Japan.

As a preliminary epidemiological study, we evaluated the geographical correlation between estimated ambient ultraviolet B (UVB) levels and the mortality risk of leukemia in Japan. Ambient UVB levels were estimated from meteorological data for several successive periods. The standardized mortality ratio (SMR) was calculated for the 11 regions or 38 large cities and different times for nine types of leukemia [the International Classification of Diseases, ninth revision (ICD-9), ICD 200-208]. The ecological relationship was assessed by calculating Spearman's correlation coefficient. Among the nin

Clinical characteristics of hybrid leukemia: report of five cases.

We studied clinical and biological features of five cases of hybrid leukemia. Three of the five patients were classified as biphenotypic leukemia because of the coexpression of myeloid/B lymphoid markers in patients 1 (FAB M2) and 2 (FAB CMMoL) and myeloid/T lymphoid markers in patient 3 (FAB M4). Patient 4 was identified as bilineal-biphenotypic leukemia because acute myelogenous leukemia (AML) (FAB M4) and acute lymphoblastic leukemia (ALL) (FAB L1) coexisted and each population coexpressed myeloid and T lymphoid markers. Patient 5 was identified as bilineal leukemia due to the conversion fr
