Healing fats of the skin: the structural and immunologic roles of the omega-6 and omega-3 fatty acids.

Linoleic acid (18:2omega6) and alpha-linolenic acid (18:3omega3) represent the parent fats of the two main classes of polyunsaturated fatty acids: the omega-6 (n-6) and the omega-3 (n-3) fatty acids, respectively. Linoleic acid and alpha-linolenic acid both give rise to other long-chain fatty acid derivatives, including gamma-linolenic acid and arachidonic acid (omega-6 fatty acids) and docosahexaenoic acid and eicosapentaenoic acid (omega-3 fatty acids). These fatty acids are showing promise as safe adjunctive treatments for many skin disorders, including atopic dermatitis, psoriasis, acne vu

Oil production by Mortierella isabellina from whey treated with lactase.

Whey, a by-product of cheese manufacturing is rich in nutrients such as lactose, proteins, and mineral salts. The fungus Mortierella isabellina was used for production of oil containing γ-linoleic acid (GLA) during fermentation on deproteinized whey permeate (DP-WP) with and without lactase addition. The maximum oil concentration was 3.65 g/L in DP-whey (16.0% lactose) without enzyme treatment. Treatment of DP-WP with lactase resulted in an increase in oil content to 17.13 g/L. Palmitic (22.50-25.80%) and oleic acids (37.60-48.56%) were the major fatty acids along with GLA (2.18-5.48%), linole

Biochemical changes accompanying brefeldin A formation in Curvularia lunata.

Extracellular brefeldin A was detected in 4% glucose--peptone--mineral salts cultures of Curvularia lunata at the start of the exponential growth phase. Some fluctuations in brefeldin A levels occurred during the exponential growth phase followed by a significant reduction in level at the stationary growth phase. Broth glucose levels decreased according to a sigmoid relationship with time whereas broth pH remained fairly constant during the exponential growth phase followed by a gradual increase into the stationary growth phase. Mycelial brefeldin A levels were low throughout the various growt

Radical scavenger can scavenge lipid allyl radicals complexed with lipoxygenase at lower oxygen content.

Lipoxygenases have been proposed to be a possible factor that is responsible for the pathology of certain diseases, including ischaemic injury. In the peroxidation process of linoleic acid by lipoxygenase, the E,Z-linoleate allyl radical-lipoxygenase complex seems to be generated as an intermediate. In the present study, we evaluated whether E,Z-linoleate allyl radicals on the enzyme are scavenged by radical scavengers. Linoleic acid, the content of which was greater than the dissolved oxygen content, was treated with soya bean lipoxygenase-1 (ferric form) in the presence of radical scavenger,

Serum fatty acid concentrations in patients on postoperative parenteral nutrition with and without fat.

To examine changes in fatty acid composition of serum lipids, sixteen patients with gastric cancer were maintained on total parenteral nutrition (TPN) or intravenous feeding immediately after total gastrectomy. Subjects receiving no fat showed decreases in linoleic acid, linolenic acid and arachidonic acid and increases in palmitoleic acid and oleic acid, whereas those receiving fat emulsion showed no detectable changes. Decrease in the linoleic acid content was greater in subjects on a higher carbohydrate intake, but less in those on a higher fat intake. Multiple regression analysis of the re

Nontargeted metabolomics of Korean and Chinese Platycodon grandiflorum using GC-TOF-MS: geographical-origin discrimination and metabolic pathway analysis via chemometrics.

Platycodon grandiflorum is an economically significant medicinal and edible plant in East Asia, whose chemical composition varies with cultivation location. In this study, a nontargeted metabolomics approach based on gas chromatography-time-of-flight mass spectrometry was combined with multivariate chemometrics to differentiate P. grandiflorum roots from Korea and China. Following sample derivatization, 53 primary metabolites, including amino acids, organic acids, sugars, and fatty acids, were putatively annotated. Principal component analysis showed origin-related clustering, and orthogonal p

Copper deficiency-induced changes in the fatty acid composition of mitochondrial and microsomal membranes of rat liver.

The effects of copper deficiency on the fatty acid composition of mitochondrial and microsomal phospholipids in rat liver were studied. Copper deficiency was induced by a milk powder diet. To evaluate the effect of the milk diet on the fatty acid pattern of mitochondrial and microsomal phospholipids, one group of rats was fed Cusupplemented powdered milk. A decrease in the relative proportion of linoleic acid and an increase in the level of oleic and docosahexaenoic acids in membrane phospholipids were found in this group. However, no changes in the fatty acid pattern characteristic of essenti

Unbalanced long-chain fatty acid beta-oxidation in newborns with cystic fibrosis and congenital hypothyroidism.

BACKGROUND: Immediately after birth, adaptation to the extrauterine environment includes an upregulation of fatty acid catabolism. Cystic fibrosis and untreated hypothyroidism exert a life-long impact on fatty acid metabolism, but their influence during this transitional period is unknown. Children and adults with cystic fibrosis exhibit unbalanced fatty acid composition, most prominently a relative deficit of linoleic acid. Lipid catabolism is downregulated in hypothyroidism. METHODS: We analyzed acylcarnitine data in newborn screening blood spot samples from infants with cystic fibrosis, wi

Changes in plasma and erythrocyte omega-6 and omega-3 fatty acids in response to intravenous supply of omega-3 fatty acids in patients with hepatic colorectal metastases.

BACKGROUND: Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) are functionally the most important omega-3 polyunsaturated fatty acids (PUFAs). Oral supply of these fatty acids increases their levels in plasma and cell membranes, often at the expense of the omega-6 PUFAs arachidonic acid (ARA) and linoleic acid. This results in an altered pattern of lipid mediator production to one which is less pro-inflammatory. We investigated whether short term intravenous supply of omega-3 PUFAs could change the levels of EPA, DHA, ARA and linoleic acid in plasma and erythrocytes in patients with h

Fatty acid changes in rat brain ethanolamine phosphoglycerides during and following chronic exposure to trichloroethylene.

Rats were exposed by continuous inhalation to a moderate level of trichloroethylene (1720 mg/m3). The fatty acid pattern of brain ethanolamine phosphoglycerides was examined during exposure and after an additional exposure-free period. Alterations in the fatty acid pattern were noted after 30 days of exposure. An increased ratio of linoleic acid-derived (n-6) to linolenic acid-derived (n-3) fatty acids was observed in the cerebral cortex, the hippocampus, and the brain stem. Of the major fatty acids, arachidonic acid (20:4(n-6)) was increased in the cerebral cortex and the brain stem, while do

Comparison of the Thermal Behavior and Chemical Composition of Milk Powders of Animal and Plant Origin.

The present study aims to perform a comparative analysis of the chemical composition and thermal behavior of two distinct milk types, namely animal and plant-based. The thermal analysis revealed the presence of the following classes of compounds: hydrocarbons, heterocycles, aldehydes, ketones, amines and alcohols. All types of milk contain saturated fatty acids (SFAs), monounsaturated fatty acids (MUFAs) and polyunsaturated fatty acids (PUFAs), though the relative proportions of these vary depending on the specific milk type. Animal milk powders contain SFAs, including palmitic, stearic, and m

Essential fatty acid deficiency in premature infants.

To better characterize essential fatty acid (EFA) deficiency in neonates, we assessed 63 premature infants by serial determinations of plasma fatty acids for the level of linoleic acid, the presence of an abnormal trienoic acid (5,8,11-eicosatrienoic acid [20:3 omega 9]), and the ratio of this compound to arachidonic acid, ie, the triene-tetraene ratio. The data indicated that at age 7 d, 67% of these infants had low plasma linoleic acid levels, 62% showed readily detectable 20:3 omega 9, and 44% had a high triene-tetraene ratio. Infants fed by age 2 d had a normal mean linoleate level at 7 d

The effects of diet and zinc treatment on the fatty acid composition of serum lipids and adipose tissue and on serum lipoproteins in two adolescent patients with acrodermatitis enteropathica.

In two adolescent patients with acrodermatitis enteropathica fatty acid spectra in serum lipids and adipose tissue and serum lipoprotein concentrations were followed for about 7 yr. One patient was treated by diet and iv infusions of high amounts of linoleic acid and later by different doses of zinc. The other boy was given only varying doses of zinc. Extra supply of linoleic acid raised its concentrations in serum triglycerides, cholesterol esters, phospholipids, and adipose tissue lipids from low to normal or high levels. In both patients linoleic acid in serum lipids was sensible to the dos

Localization of arachidonate 12-lipoxygenase in canine brain tissues.

The cytosol fraction from a thoroughly irrigated canine cerebrum was subjected to immunoaffinity chromatography using a monoclonal antibody against porcine leukocyte 12-lipoxygenase. Arachidonate 12-lipoxygenase eluted from the column with some retardation. The enzyme, with a specific activity of 9 nmol/min/mg of protein, converted arachidonic acid to 12(S)-hydroperoxy-5,8,10,14-eicosatetraenoic acid. The enzyme was active not only with arachidonic acid, but also with linoleic and alpha-linolenic acids. In contrast, 12-lipoxygenase of canine platelets was almost inactive with linoleic and alph

Plasma fatty acid changes and increased lipid peroxidation in patients with adult respiratory distress syndrome.

OBJECTIVE: There is a strong evidence that adult patients with acute respiratory distress syndrome (ARDS) are under severe oxidative stress, which leads to molecular damage. Using gas chromatography-mass spectrometry, our objective was to sequentially monitor changes, in intensive care unit (ICU) patients, characteristic of the oxidative loss of plasma unsaturated fatty acids and formation of the highly specific oxidation product of linoleic acid, 4-hydroxy-2-nonenal. DESIGN: Prospective, nonintervention, descriptive study. Limited statistics were applied to facilitate interpretation of the d
