Effect of melatonin on cerebral edema in rats.

OBJECTIVE: Melatonin (5-methoxy-N-acetyltrypamine), a chemical naturally produced in the pineal gland, has been suggested to be a free radical scavenger and an antioxidant. In the present study, the effect of melatonin on cold-induced brain edema was evaluated by determination of cerebral water content, blood-brain barrier permeability, and areas of infarct; the effects were also studied histopathologically. METHODS: Brain edema was produced in rats by creating a lesion via cortical freezing. Animals were separated into four groups: sham-operated (craniectomy only); control (cold injury); sha…

Melatonin for preventing and treating jet lag.

BACKGROUND: : Jet-lag commonly affects air travellers who cross several time zones. It results from the body's internal rhythms being out of step with the day-night cycle at the destination. Melatonin is a pineal hormone that plays a central part in regulating bodily rhythms and has been used as a drug to re-align them with the outside world. OBJECTIVES: : To assess the effectiveness of oral melatonin taken in different dosage regimens for alleviating jet-lag after air travel across several time zones. SEARCH STRATEGY: : We searched the Cochrane Controlled Trials Register, MEDLINE, EMBASE, P…

Melatonin and its physiological and therapeutic properties.

Melatonin is a hormone produced mainly by the pineal gland in most vertebrate species, including humans. Recent metabolic, receptor and functional studies created a picture of the melatoninergic system(s) in living organisms, its organization, physiology and a role in some pathologic conditions. The melatonin-generating system is characterized by three basic features: (1) photosensitivity, (2) diurnal (or circadian) rhythmicity (with highest levels of melatonin production occurring at night in darkness), and (3) age-related decline in its activity. Cyclic nocturnal increases of melatonin level…

The role of melatonin in the human fetus (review).

Melatonin, an indole amine, primarily derived from the pineal gland is secreted during the hours of darkness. Melatonin acts as a hormonal transduction of photoperiod influencing the timing of seasonal and daily (circadian) physiological rhythms. Maternal melatonin crosses the placenta and enters the fetal circulation providing photoperiodic information to the fetus influencing the subsequent circadian and seasonal rhythms of the offspring. The function of melatonin in humans is more obscure. However, melatonin has attained prominence as a treatment for disturbed circadian rhythms and sleep pa…

[Melatonin and reproduction in domestic farm animals].

Melatonin, synthetized by the pineal gland, is the chemical messenger which allows seasonally reproductive animals to perceive day length changes. In the ewe, the neural message, transformed into a hormonal one, triggers pulsatile activity of the LHRH neurons. About 40 days are necessary for melatonin to centrally stimulate the pulsatile LHRH activity. Its sites and mode of action are not yet known completely, but a precise hypothalamic zone has been defined in which radioactive melatonin binds specifically and where cold melatonin delivered locally stimulates LHRH activity. In the veterinary …

[Melatonin--clinical perspectives in prevention and therapy].

Melatonin is a hormone produced by the pineal gland mainly during the night. Its primary function is to coordinate biological rhythms. It may be used for therapy of chronobiological disorders, such as occur in jet lag, shift work, blind persons or in specific forms of insomnia. Due to its anti-gonadotropic effect, melatonin in high doses has been used as a contraceptive. In-vitro and animal studies have demonstrated cancer inhibition, immun-stimulating and rejuvenating effects, but none of these claims have been confirmed in clinical studies. Melatonin used over a short period is not associate…

New actions of melatonin and their relevance to biometeorology.

Melatonin is not only produced by the pineal gland, retina and parietal but also by various other tissues and cells from vertebrates, invertebrates, fungi, plants, multicellular algae and by unicells. In plants, many invertebrates and unicells, its concentration often exceeds that found in vertebrate blood by several orders of magnitude. The action of melatonin is highly pleiotropic. It involves firstly, direct effects, via specific binding sites in various peripheral tissues and cells of vertebrates, including immunomodulation; secondly, systemic influences on the cytoskeleton and nitric oxid…

[Melatonin and ubiquinone as endogenous antioxidant factors].

We studied the effect of restraint stress upon the degree of lipidic peroxidation in the fronto-temporal cerebral cortex, diencephalon, thymus and gastric mucosa. We determined the malon-dialdehyde (MDA) by spectrocolorimetry, the percentage of animals with stress gastric ulcer and the surface of gastric ulcerations. The results demonstrated an important increase of the MDA correlated with frequency and intensity of the gastric ulcer. In the second stage of the experiment, we studied the influence of the ubiquinone and melatonin on the same parameters studied in the first part. In every struct…

Melatonin in relation to physiology in adult humans.

The role exerted by melatonin in human physiology has not been completely ascertained. Melatonin levels have been measured in different physiopathological conditions, but the effects induced by melatonin administration or withdrawal have been tested only recently. Some effects have been clearly documented. Melatonin has hypothermic properties, and its nocturnal secretion generates about 40% of the amplitude of the circadian body temperature rhythm. Melatonin has sleep inducing properties, and exerts important activities in the regulation of circadian rhythms. Melatonin is capable of phase shif…

Duodenum is not a consistent source of melatonin in rats.

Markedly increased melatonin levels in plasma have been observed in response to tryptophan administration. This post-tryptophan melatonin increase has been attributed to the duodenum. Because extra-pineal sources of melatonin may be important in interpreting the meaning of altered melatonin production observed in patient populations, this work was undertaken to confirm whether melatonin is produced in the duodenum and to know whether the duodenum need be considered when investigating the circadian control of melatonin production. We measured melatonin in rat duodenum by HPLC both under basal c…

The role of melatonin and serotonin in aging: update.

It has been proposed that aging occurs because of a failure of the pineal gland to produce melatonin from serotonin each day beginning at sunset and throughout the night. This lack leads to a nighttime deficiency of melatonin both absolutely and also relatively to serotonin. As melatonin has wide-spread integrative and regenerative effects, its lack may lead to disturbances normally associated with aging. The present paper reviews the pertinent literature which appeared since our first publication, but earlier articles are also included. Evidence is presented for a role of melatonin and seroto…

Melatonin levels are decreased in rheumatoid arthritis.

The hormone, melatonin, is a product of the pineal gland. This methoxy-indole, also known as N-acetyl-5-methoxy-tryptamine, is structurally related to indomethacin, a derivative of methylated indole. The fact that indomethacin has been used successfully in patients suffering from certain chronic inflammatory conditions (such as rheumatoid- and osteoarthritis) gives rise to the question as to whether melatonin also possesses anti-inflammatory virtues. This hypothesis has been tested by determining melatonin concentration levels by means of a radioimmunoassay in patients with rheumatoid arthriti…

Melatonin and petit-mal epilepsy: an hypothesis.

Intraventricular administration of the opioid peptide leucine-enkephalin has been reported to induce petit-mal-like seizures in rats. These seizures have been found to be an age-dependent phenomenon. In rats, the full manifestation of these seizures develops after 4 weeks of age during which time ethosuximide was effective in aborting these seizures, while phenytoin and phenobarbital were ineffective. The period associated with the development of enkephalin-induced seizures in rats coincides with an important milestone in pineal chronobiology. In rats, melatonin plasma levels peak at 3 weeks o…

What are the implications of melatonin in the peritoneal fluid?

The Authors evidence the presence of melatonin (MT) in the peritoneal fluid (PF). They researched possible correlations between MT and other protein and steroid hormones in the PF and in the plasma. The only significant correlation found was a "negative" one between plasma MT and plasma E2. The Authors exclude an ovarian source for peritoneal fluid MT. They believe that MT may have a role in menstrual cycle regulation, directly modulating ovarian steroid production. To determine if MT has indeed such a role, they feel it important to study the possible presence of ovarian MT receptors.

Melatonin biosynthesis in the mammalian pineal gland.

Rhythmic production of melatonin by the mammalian pineal occurs in response to noradrenergic stimulation which produces a cascade of biochemical events within the pinealocyte. In the rat, massive changes in NAT activity result from an increase in intracellular c-AMP levels produced by a synergistic interaction whereby an alpha 1 activation amplifies beta-adrenergic stimulation. The intracellular events mediating this effect are described. A major aspect of the temporal control of melatonin production is the programmed down-regulation of responses to noradrenergic stimulation once the initial s…

Alterations of melatonin secretion in atopic eczema.

The circadian rhythm of melatonin secretion of the pineal gland can be disturbed in a variety of clinical conditions and diseases including some psychic disorders. To study the rhythmic behaviour of melatonin secretion in atopic eczema (AE), melatonin serum levels were measured every 2 h, starting at 8 a.m. in 18 patients suffering from severe AE. In 6 patients exhibiting low serum levels of melatonin, the circadian melatonin rhythm was found to be abolished. In 8 patients a diminished nocturnal melatonin increase was observed compared with the controls (n = 40). Only 4 patients showed a norma…
