Pre- and post-stroke memantine administration attenuates neurological deficits, infarct size, and astrogliosis, with a concomitant increase in MAP2 protein in a rat stroke model.

Over the past 30 years, the global incidence of stroke and related disability-adjusted life years has risen significantly, underscoring the urgent need for effective therapeutic strategies. Memantine (Mem), a non-competitive antagonist of N-methyl-d-aspartate (NMDA) glutamate receptors, has been investigated as a potential treatment in stroke models. By blocking NMDA receptor activity, Mem can prevent excessive calcium influx and excitotoxic neuronal death, thereby promoting neuroprotection with good tolerability. Because early intervention is crucial, we evaluated a sequential memantine regim…

Modulation of Lingo-1 and NMDA receptor expression by memantine and vitamin D3 co-therapy attenuates motor and non-motor symptoms in essential tremor.

Essential tremor (ET) is a common movement disorder, characterized by bilateral postural or kinetic tremor and associated non-motor symptoms including anxiety and cognitive impairment. Current treatments offer limited efficacy and significant side effects, highlighting the need for novel therapeutic approaches. This study investigated the therapeutic potential of memantine and vitamin D3 (vitD3) combination therapy in a harmaline-induced mouse model of essential tremor. Adult male Swiss mice were divided into eight groups (n = 8/group): control, sham, harmaline-induced ET (10 mg/kg, i.p. on da…

The anti-craving compound acamprosate acts as a weak NMDA-receptor antagonist, but modulates NMDA-receptor subunit expression similar to memantine and MK-801.

NMDA-receptor-mediated mechanisms may be crucial in addictive states, e.g. alcoholism, and provide a target for the novel anti-craving compound acamprosate. Here, the pharmacological effects of acamprosate on NMDA-receptors were studied using electrophysiological techniques in different cell lines in vitro. Additionally, a possible modulation of brain NMDA-receptor subunit expression was examined in vivo in rats, and compared to two effective non-competitive NMDA-receptor antagonists, memantine and MK-801. Electrophysiology in cultured hippocampal neurons (IC(50) approx. 5.5mM) and Xenopus ooc…

Memantine Use in Prolonged Cognitive Sequalae in Pediatric Anti-N-methyl-D-aspartate Receptor Encephalitis: A Three-year Follow-up Case Report and Brief Review.

Anti-N-methyl-D-aspartate receptor encephalitis (anti-NMDAR encephalitis) is an autoimmune encephalopathy increasingly recognized in the pediatric population. Although conventional immunotherapy-focused treatments have significantly improved outcomes, a substantial proportion of patients experience enduring neuropsychiatric and cognitive sequelae. Here, we present a three-year longitudinal follow-up report of a female case with confirmed anti-NMDAR encephalitis, focusing on the neuropsychiatric trajectory and cognitive sequelae management with memantine. A previously healthy 10-year-old girl p…

Antenatal blockade of N-methyl-D-aspartate receptors by Memantine reduces the susceptibility to diabetes induced by a high-fat diet in rats with intrauterine growth restriction.

Intrauterine growth retardation (IUGR) is closely related to the later development of type 2 diabetes in adulthood. Excessive activation of N-methly-D-aspartate receptors (NMDARs) causes excitatory neurotoxicity, resulting in neuronal injury or death. Inhibition of NMDARs enhances the glucose-stimulated insulin secretion and survival of islet cells in type 2 diabetic mouse and human islets. Here, we examined whether antenatal blockade of NMDARs by Memantine could decrease the risk of diabetes induced by a high-fat (HF) diet at adulthood in IUGR rats. Pregnant SD rats were assigned to four grou…
