Shear injuries of the brain.

A blow to the head will impart rotational velocity to the brain and, depending on its magnitude, will produce effects ranging from concussio … vessel injury in all; this was localized to the basal and midsagittal areas of the diencephalon and mesencephalon, particularly in those less severely injured. Such changes represent the basic pathology of all head injury. Data from this study suggest that concussion depends upon varying degrees of damage to the axon as well as the neuron. The current definitio …

Convulsive-Like Movements in Posterior Circulation Large Vessel Occlusion: Associations With Midbrain Injury and Poor Outcome.

BACKGROUND AND PURPOSE: Convulsive-like movements are an uncommon and frequently misinterpreted manifestation of posterior circulation stroke. Although they are often regarded as non-epileptic brainstem motor phenomena, their neuroanatomical substrates and prognostic implications in posterior circulation large vessel occlusion (LVO) remain poorly characterized. This study aimed to define the clinical and imaging correlates of convulsive-like movements and to evaluate their association with functional outcomes. METHODS: We performed a retrospective analysis of prospectively collected data from

Disruption of lysosomal proteolysis in astrocytes facilitates midbrain organoid proteostasis failure in an early-onset Parkinson's disease model.

Accumulation of advanced glycation end products (AGEs) on biopolymers accompanies cellular aging and drives poorly understood disease processes. Here, we studied how AGEs contribute to development of early onset Parkinson's Disease (PD) caused by loss-of-function of DJ1, a protein deglycase. In induced pluripotent stem cell (iPSC)-derived midbrain organoid models deficient for DJ1 activity, we find that lysosomal proteolysis is impaired, causing AGEs to accumulate, α-synuclein (α-syn) phosphorylation to increase, and proteins to aggregate. We demonstrated these processes are at least partly dr

Delayed levodopa-responsive parkinsonism following acute midbrain injury.

Acute midbrain injury may cause both hyperkinetic movement disorders and parkinsonism. The temporal interval between the insult and the emergence of hyperkinetic disorders can last years. A delayed appearance of parkinsonism, on the other hand, was rarely described. We present three cases of male patients (50-, 58- and 28-year-old) who developed levodopa-responsive parkinsonism 20, 8 and two years, respectively, after acute brain insult involving the midbrain. Insults included subcortical intracerebral hemorrhage dissecting into the midbrain, embolic basilar occlusion and trauma. A fluorodopa

Brain MRI in Progressive Supranuclear Palsy with Richardson's Syndrome and Variant Phenotypes.

BACKGROUND: Brain magnetic resonance imaging (MRI) is used to support the diagnosis of progressive supranuclear palsy (PSP). However, the value of visual descriptive, manual planimetric, automatic volumetric MRI markers and fully automatic categorization is unclear, particularly regarding PSP predominance types other than Richardson's syndrome (RS). OBJECTIVES: To compare different visual reading strategies and automatic classification of T1-weighted MRI for detection of PSP in a typical clinical cohort including PSP-RS and (non-RS) variant PSP (vPSP) patients. METHODS: Forty-one patients (2

Stimulus-independent and stimulus-dependent neural networks underpin placebo analgesia responsiveness in humans.

The neural circuits that regulate placebo analgesia responsivity are unknown, although engagement of brainstem pain modulatory regions is likely critical. Here we show in 47 participants that differences are present in neural circuit connectivity's in placebo responders versus non-responders. We distinguish stimulus-independent and stimulus-dependent neural networks that display altered connections between the hypothalamus, anterior cingulate cortex and midbrain periaqueductal gray matter. This dual regulatory system underpins an individual's ability to mount placebo analgesia.

Nothnagel Syndrome.

The internist Hermann Nothnagel (1841-1905) took a special interest in the cerebellum. In an early experimental study on rabbits conducted in 1876, he demonstrated the involvement of the vermis in the pathophysiology of motor ataxia. Between 1879 and 1889, he reported four cases of tectal tumors that clinically manifested with bilateral ophthalmoplegia and unilateral gait ataxia, culminating in the Cerebellar Classic highlighted here. Nothnagel attributed this clinical syndrome to lesions of the colliculi ("quadrigeminal bodies") and compression of the nuclei of the third cranial nerves, but a

In vitro modeling of the human dopaminergic system using spatially arranged ventral midbrain-striatum-cortex assembloids.

Ventral midbrain dopaminergic neurons project to the striatum as well as the cortex and are involved in movement control and reward-related cognition. In Parkinson's disease, nigrostriatal midbrain dopaminergic neurons degenerate and cause typical Parkinson's disease motor-related impairments, while the dysfunction of mesocorticolimbic midbrain dopaminergic neurons is implicated in addiction and neuropsychiatric disorders. Study of the development and selective neurodegeneration of the human dopaminergic system, however, has been limited due to the lack of an appropriate model and access to hu

A D2 to D1 shift in dopaminergic inputs to midbrain 5-HT neurons causes anorexia in mice.

Midbrain dopamine (DA) and serotonin (5-HT) neurons regulate motivated behaviors, including feeding, but less is known about how these circuits may interact. In this study, we found that DA neurons in the mouse ventral tegmental area bidirectionally regulate the activity of 5-HT neurons in the dorsal raphe nucleus (DRN), with weaker stimulation causing DRD2-dependent inhibition and overeating, while stronger stimulation causing DRD1-dependent activation and anorexia. Furthermore, in the activity-based anorexia (ABA) paradigm, which is a mouse model mimicking some clinical features of human ano

Organization of the galaninergic neuronal system in the brain of the gecko Hemidactylus frenatus.

Galanin (GAL) is a 29 amino acid peptide present in the central nervous system (CNS) as well as peripheral tissues in vertebrates. However, the brain distribution pattern of GAL is understudied in reptiles. The aim of this study was to determine the organization of galaninergic neuronal system in the brain of the gecko Hemidactylus frenatus, a tropical and sub-tropical lizard, using rabbit anti-galanin antibody. In the telencephalon, GAL-ir perikarya and fibres were found in the lateral septal nucleus, but only GAL-ir fibres were observed in the striatum, nucleus accumbens, anterior commissure

Cytoarchitectonic study of the brain of a perciform species, the sea bass (Dicentrarchus labrax). II. The diencephalon.

The cytoarchitecture of nuclei in the preoptic area, ventral thalamus, dorsal thalamus, epithalamus, hypothalamus, posterior tuberculum, synencephalon, and pretectum and the accessory optic nuclei was analyzed in a perciform teleost, the sea bass Dicentrarchus labrax, by using serial sections stained with cresyl-violet. In general, the cytoarchitecture of the preoptic area, ventral and dorsal thalamus, epithalamus, and synencephalon resembles the histological pattern of other teleosts. However, the parvocellular preoptic nucleus of sea bass has been subdivided into parvocellular and anterovent

Comparative development of the mammalian isocortex and the reptilian dorsal ventricular ridge. Evolutionary considerations.

There has been a long debate about a possible homology between parts of the dorsal ventricular ridge (DVR) of reptiles and birds, and parts of the mammalian isocortex. Correspondence between these structures was originally proposed on the basis of connectional similarities between the DVR of birds and the mammalian auditory and extrastriate visual isocortical areas. Furthermore, the proposal of homology includes the possible embryological similarity of cells that give rise to the DVR and cells that give rise to the isocortex. Against this concept it has been claimed that the DVR and the isocor

A case of Tourette's syndrome with midbrain involvement.

Several hypotheses have attempted to incriminate a particular anatomical region responsible for the complex symptomatology of Tourette's syndrome (TS). Recently, Devinsky speculated that damage to the peri-aqueductal gray matter (PAG) and midbrain tegmentum may represent the major anatomical site of dysfunction in TS. A patient with TS in whom radiological evidence indicated midbrain involvement may support Devinsky's hypothesis. A 7-year-old boy exhibited motor and vocal tics since the age of four. These were associated with hyperactive behavior, stereotype body movements, abnormal sexual beh

Central projections of the mesencephalic nucleus of the fifth nerve: an autoradiographic study.

Projections of cells of the mesencephalic nucleus of the fifth nerve (Mes V) to brainstem structures in the cat were studied by labelling Mes V cells with tritiated leucine. Before the leucine injections were made, however, large kainic acid lesions were produced in the vicinity of Mes V cells because these neurons are resistant to being killed or injured by this neurotoxin. Thus Mes V cells were selectively labelled by the leucine even though they are scattered among many other neurons. Leucine injections near Mes V cells located in the mesencephalon, which are primarily the somata of jaw mus…
