Bacteria isolated from Aedes aegypti with potential vector control applications.

Highly anthropophilic and adapted to urban environments, Aedes aegypti mosquitoes are the main vectors of arboviruses that cause human diseases such as dengue, zika, and chikungunya fever, especially in countries with tropical and subtropical climates. Microorganisms with mosquitocidal and larvicidal activities have been suggested as environmentally safe alternatives to chemical or mechanical mosquito control methods. Here, we analyzed cultivable bacteria isolated from all stages of the mosquito life cycle for their larvicidal activity against Ae. aegypti. A total of 424 bacterial strains isol…

Process Evaluation of a Community-Based Microbial Larviciding Intervention for Malaria Control in Rural Tanzania.

Microbial larviciding can be an effective component of integrated vector management malaria control schemes, although it is not commonly implemented. Moreover, quality control and evaluation of intervention activities are essential to evaluate the potential of community-based larviciding interventions. We conducted a process evaluation of a larval source management intervention in rural Tanzania where local staff were employed to apply microbial larvicide to mosquito breeding habitats with the aim of long-term reductions in malaria transmission. We developed a logic model to guide the process

Wolbachia prevalence in Aedes aegypti across five comunas of Medellín, Colombia: Implications for post-release surveillance.

A promising sustainable strategy for reducing arbovirus transmission by the mosquito vector Aedes aegypti is the deployment of individuals transinfected with Wolbachia pipientis, a bacterium that inhibits replication of viruses such as dengue and is being used in population replacement programs aimed at blocking Aedes-borne virus transmission. Between 2017 and 2022, the World Mosquito Program released Ae. aegypti infected with the wMel Wolbachia strain throughout Medellín, Colombia. Long-term Wolbachia surveillance following population replacement requires approaches that balance accuracy and

Entomological Surveillance and Vector Competence: A Crucial Front in Dengue Prevention.

This chapter aims to bridge the gap between two fundamental topics in medical entomology-vector surveillance and vector competence-often seen as distinct areas of study. This perceived separation likely stems from the misconception that surveillance falls within the domain of field entomologists, while vector competence is primarily the focus of laboratory-based researchers. Here, we emphasize the necessity of integrating these two fields, highlighting the importance of monitoring the susceptibility of natural Aedes aegypti populations to dengue virus (DENV) and other arboviruses. By incorpora

Implementation fidelity of a community-based Aedes aegypti breeding site elimination program for dengue control in southeastern Mexico.

INTRODUCTION: Dengue is a globally significant arboviral disease in tropical regions such as southeastern Mexico. The "Healthy Environments and Communities" (E&CS) program, led by the Health Secretariat, promotes community practices to eliminate Aedes aegypti breeding sites. However, factors influencing its implementation and fidelity remain unclear. OBJECTIVE: To determine the factors affecting fidelity in implementing the E&CS program targeting breeding site elimination in Río Florido, Tapachula. METHODS: An observational mixed-methods study was conducted. The quantitative component evalua

Design and validation of a low-cost sugar-feeder for resource-poor insectaries.

BACKGROUND: The emergence of insecticide resistance in insects has led researchers to develop new control tools so that historic gains made in reducing disease transmission are not lost. Attractive targeted sugar baits (ATSBs) are a vector control tool being widely trialled to target insects that feed on plant sugars and blood. We designed a field-friendly, economical and more environmentally responsible sugar feeder for maintaining mosquito colonies and screening potential ATSB candidates. METHODS: We simultaneously tested, in both male and female Anopheles gambiae mosquitoes, the effect of

Potential interaction between clorsulon and ivermectin for malaria vector control.

Mass ivermectin (IVM) treatment of livestock (MITL) is under consideration as a malaria control tool as IVM-treated livestock are lethal to blood-feeding Anopheles mosquitoes. MITL is routinely used as a prophylaxis in livestock to reduce the burden and transmission of helminth infections. Recently, there has been a shift in the veterinary IVM market in Southeast Asia wherein nearly all standard IVM formulations are now co-formulated with clorsulon (CLO). CLO is used to treat the trematode liver fluke, Fasciola hepatica. Thus, the co-administration of IVM and CLO simultaneously targets multipl

Anti-CRISPR Anopheles mosquitoes inhibit gene drive spread under challenging behavioural conditions in large cages.

CRISPR-based gene drives have the potential to spread within populations and are considered as promising vector control tools. A doublesex-targeting gene drive was able to suppress laboratory Anopheles mosquito populations in small and large cages, and it is considered for field application. Challenges related to the field-use of gene drives and the evolving regulatory framework suggest that systems able to modulate or revert the action of gene drives, could be part of post-release risk-mitigation plans. In this study, we challenge an AcrIIA4-based anti-drive to inhibit gene drive spread in ag

Seasonal insights for integrative mosquito management from multi-year baseline entomological data on Aedes aegypti in Lee County, Florida.

The spread of arboviruses like yellow fever, dengue, chikungunya, and Zika, transmitted by the invasive mosquito Aedes aegypti has led to the development of many strategies to suppress mosquito populations. Given the rapid development of resistance to common chemical larvicides and adulticides in some Ae. aegypti populations, as well as the ever-shrinking chemical options for mosquito control, there is a pressing need for new tools and deployment of those innovative tools as a component of integrative mosquito management programs. Prior to the adoption of any mosquito population intervention, …

Non-target effects of chemical malaria vector control on other biological and mechanical infectious disease vectors.

Public health insecticides play a crucial role in malaria control and elimination programmes. Many other arthropods, including mechanical and biological vectors of infectious diseases, have similar indoor feeding or resting behaviours, or both, as malaria mosquitoes, and could be exposed to the same insecticides. In this Personal View, we show that little is known about the insecticide susceptibility status and the extent of exposure to malaria interventions of other arthropod species. We highlight that there is an urgent need to better understand the selection pressure for insecticide resista

Evaluation of the effectiveness of the human-baited double net traps and BG traps compared with the human landing catches for collecting outdoor Aedes albopictus in China.

Dengue fever is one of the biggest threats to public health in China, causing huge disease burden and economic loss. Aedes-mosquito surveillance could be a cornerstone for predicting the risk of Aedes-borne diseases and evaluating the effect of vector management during diseases outbreaks. The human landing catch (HLC) method is regarded as the "gold standard" for catching Aedes mosquitoes, but it potentially exposes field professionals to vectors of known or unknown pathogens. Human-baited double net (HDN) was recommended to replace HLC for emergency monitoring in China when Aedes-borne diseas

RNAi-Mediated Screening of Selected Target Genes Against Culex quinquefasciatus (Diptera: Culicidae).

Culex quinquefasciatus, a member of the Culex pipiens complex, is widespread in Saudi Arabia and other parts of the world. It is a vector for lymphatic filariasis, Rift Valley fever, and West Nile virus. Studies have shown the deleterious effect of RNA interference (RNAi)-mediated knockdown of various lethal genes in model and agricultural pest insects. RNAi was proposed as a tool for mosquito control with a focus on Aedes aegypti and Anopheles gambiae. In this study, we examined the effect of RNAi of selected target genes on both larval mortality and adult emergence of Cx. quinquefasciatus th…

A potential global surveillance tool for effective, low-cost sampling of invasive Aedes mosquito eggs from tyres using adhesive tape.

BACKGROUND: The international movement of used tyres is a major factor responsible for global introductions of Aedes invasive mosquitoes (AIMs) (Diptera: Culicidae) that are major disease vectors (e.g. dengue, Zika, chikungunya and yellow fever). Surveillance methods are restricted by expense, availability and efficiency to detect all life stages. Currently, no tested method exists to screen imported used tyres for eggs in diapause, the life stage most at risk from accidental introduction. Here we test the efficiency of adhesive tape as an affordable and readily available material to screen ty

Evaluation of an ultraviolet LED trap for catching Anopheles and Culex mosquitoes in south-eastern Tanzania.

BACKGROUND: Improved surveillance techniques are required to accelerate efforts against major arthropod-borne diseases such as malaria, dengue, filariasis, Zika and yellow-fever. Light-emitting diodes (LEDs) are increasingly used in mosquito traps because they improve energy efficiency and battery longevity relative to incandescent bulbs. This study evaluated the efficacy of a new ultraviolet LED trap (Mosclean) against standard mosquito collection methods. METHODS: The study was conducted in controlled semi-field settings and in field conditions in rural south-eastern Tanzania. The Mosclean

Aedes albopictus and the world trade in used tires, 1988-1995: the shape of things to come?

In the decade since used tires were identified as the mode of introduction of Aedes albopictus to the United States, similar infestations have been reported from 10 other countries in the Americas and 2 in Europe. Millions of used tires are still being traded throughout the world and although a few governments have implemented inspection procedures to prevent further introductions, these are unlikely to be effective. Further introductions of mosquitoes of potential public health significance are inevitable.

A systematic review of entomological outcomes and sampling approaches used in the evaluation of cluster randomised controlled trials for malaria vector control products.

BACKGROUND: Entomological outcomes are critical for understanding the biological mechanisms and operational performance of malaria vector control interventions. However, the quality and consistency of how these outcomes are measured and reported in cluster randomised controlled trials (cRCTs) is unclear. We conducted a systematic review of design features, sampling methods, and metrics used for measuring entomological outcomes in malaria vector control cRCTs. Our aims were to assess the consistency and quality of entomological study designs and examine how key design features influence the pre
