Ulcerative colitis (UC) involves a complex interplay of mucosal inflammation, intestinal barrier dysfunction, and gut dysbiosis, all exacerbated by elevated reactive oxygen species (ROS). Existing therapies often fail to concurrently address these interrelated pathologies. Here we present an orally administrable nanocage system that targets both host cells and gut microbiota and can simultaneously address immune dysregulation, epithelial damage, and microbial imbalances for UC treatment when loaded with antioxidant enzymes. This system features a poly(tertiary amine oxide) (PTAO)-based zwitter

Calcium peroxide (CaO2) is commonly used as a hydrogen peroxide (H2O2) donor to eliminate bacterial infections. However, the rapid dissociation of CaO2 and the explosive release of H2O2 have limited the development of CaO2 in the antibacterial field. Therefore, a series of silver nanoparticles (AgNPs) functionalized bacteria-triggered smart hydrogels (CSA-H) that integrate sustained release of nanoparticles and localized chemodynamic sterilization were constructed. The pH-responsive hydrogel formed through the Schiff base reaction enables the responsive release of CaO2 nanoparticles while simu

Design, synthesis, and biological evaluation of two series of O4'-benzyl-hispidol derivatives and the analogous corresponding O3'-benzyl derivatives aiming to develop selective monoamine oxidase-B inhibitors endowed with anti-neuroinflammatory activity is reported herein. The first O4'-benzyl-hispidol derivatives series afforded several more potentially active and MAO-B inhibitors than the O3'-benzyl derivatives series. The most potential compound 2e of O4'-benzyl derivatives elicited sub-micromolar MAO-B IC50 of 0.38 µM with a selectivity index >264 whereas most potential compound 3b of O3'-b

Snake venom serine proteases (SVSPs) are complex and multifunctional enzymes, acting primarily on hemostasis. In this work, we report the hitherto unknown inhibitory effect of a SVSP, named collinein-1, isolated from the venom of Crotalus durissus collilineatus, on a cancer-relevant voltage-gated potassium channel (hEAG1). Among 12 voltage-gated ion channels tested, collinein-1 selectively inhibited hEAG1 currents, with a mechanism independent of its enzymatic activity. Corroboratively, we demonstrated that collinein-1 reduced the viability of human breast cancer cell line MCF7 (high expressio…

Alpha-lipoic acid (ALA) is a disulphide compound with multifunctional antioxidant properties and is soluble in both water and lipid. Several recent studies evaluated efficacy of ALA in various diseases related to oxidative damage such as diabetes, multiple sclerosis and Alzheimer and concluded that ALA can reduce oxidative stress by quenching reactive oxygen and nitrogen species, restoring antioxidants such as glutathione, vitamins C and E, and/or improving activity of antioxidant enzymes. Varicocele, an enlargement of the veins in scrotum, is considered as the most common repairable cause of

Bee venom contains a variety of peptides and enzymes, including serine proteases. While the presence of serine proteases in bee venom has been demonstrated, the role of these proteins in bee venom has not been elucidated. Furthermore, there is currently no information available regarding the melanization response or the fibrin(ogen)olytic activity of bee venom serine protease, and the molecular mechanism of its action remains unknown. Here we show that bee venom serine protease (Bi-VSP) is a multifunctional enzyme. In insects, Bi-VSP acts as an arthropod prophenoloxidase (proPO)-activating fac

Differences in protein expression in midgut tissue of uninfected and Babesia bovis-infected southern cattle ticks, Rhipicephalus (Boophilus) microplus, were investigated in an effort to establish a proteome database containing proteins involved in successful pathogen transmission. The electrophoretic separation of midgut membrane proteins was greatly improved by using liquid-phase isoelectric focusing combined with one-dimensional or two-dimensional (2-D) gel electrophoresis. A selection of differentially expressed proteins were subjected to analysis by capillary-HPLC-electrospray tandem mass

Secretory ameloblasts have multiple functions including the synthesis and resorption of enamel matrix proteins and calcium transport during enamel formation. We have examined these functions by means of cytochemistry and immunocytochemistry. Enamel proteins, amelogenins and enamelins are localized in the biosynthetic pathways of ameloblasts and in the forming enamel. Sulfated glycoconjugates are present in secretory ameloblasts. The distal junctional complex of ameloblasts may act as a permeability barrier to enamel proteins, thereby confining the secreted proteins to the growing enamel front.

Inflammatory bowel disease (IBD) is a chronic gastrointestinal disorder characterized by oxidative stress, impaired antioxidant defense, and gut microbiota dysbiosis. Despite advances in treatment, current therapies remain limited by insufficient efficacy and systemic side effects. Here, we developed a multifunctional nanoplatform (BG/SOD@ZIF-zc) by encapsulating superoxide dismutase (SOD) within Cu-doped ZIF-zc and coating it with bacterial ghosts for targeted delivery. This system preserved enzymatic activity under gastrointestinal conditions, scavenged reactive oxygen species, and optimized

Toxoplasma bradyzoites reside in tissue cysts that undergo cycles of expansion, rupture, and release to foster chronic infection. The glycosylated cyst wall acts as a protective barrier, although the processes responsible for formation, remodeling, and turnover are not understood. Herein, we identify a noncanonical chitinase-like enzyme TgCLP1 that localizes to micronemes and is targeted to the cyst wall after secretion. Genetic deletion of TgCLP1 resulted in a thickened cyst wall that decreased cyst turnover, blocked the export of virulence effectors into host cells, and resulted in failure t

CAD encodes a multifunctional enzyme involved in de novo pyrimidine biosynthesis, and pyrimidine can be alternatively recycled from uridine. Trio whole-exome sequencing identified CAD compound heterozygous mutations in a new male patient with global developmental delay (DD), refractory epilepsy, and anemia with anisopoikilocytosis. We further reviewed all published cases with CAD deficiency. Five patients were collected from two publications, including three males and two females, and all presented with DD, drug-resistant epilepsy, and anemia with anisopoikilocytosis. Four out of six patients

Diabetes mellitus, especially Type 2 Diabetes (T2DM), appears to be a worldwide health challenge marked by progressing hyperglycemia and related complications. Postprandial Hyperglycemia (PPHG) is a critical therapeutic target due to its strong link with cardiovascular risks. Inhibition of carbohydrate-hydrolysing enzymes, α-glucosidase and α-amylase, is an established strategy to mitigate PPHG. Current Inhibitors, including acarbose, miglitol, and voglibose, are effective but limited by gastrointestinal side effects. Isatin (1H-indole-2,3-dione) is a privileged scaffold in medicinal chemistry

Biocatalytic conversion of CO-containing C1 gas to formate provides a route for C1 gas valorization, but purified carbon monoxide dehydrogenase (CODH)-formate dehydrogenase (FDH) enzyme cascades require enzyme purification and immobilization steps. As a practical alternative, this study developed an ethyl viologen (EV)-mediated intercellular whole-cell cascade by coupling CODH-expressing Escherichia coli with FDH-expressing Methylobacterium extorquens AM1. In this cascade, the two whole-cell biocatalysts performed the oxidative and reductive half-reactions, respectively, and were functionally
