Opposite effects of tor1 and tor2 on nitrogen starvation responses in fission yeast.

The TOR protein kinases exhibit a conserved role in regulating cellular growth and proliferation. In the fission yeast two TOR homologs are present. tor1(+) is required for starvation and stress responses, while tor2(+) is essential. We report here that Tor2 depleted cells show a phenotype very similar to that of wild-type cells starved for nitrogen, including arrest at the G(1) phase of the cell cycle, induction of nitrogen-starvation-specific genes, and entrance into the sexual development pathway. The phenotype of tor2 mutants is in a striking contrast to the failure of tor1 mutants to init

N, S-doped graphene derived from graphene oxide and thiourea-formaldehyde resin for high stability lithium-sulfur batteries.

Although lithium-sulfur (Li-S) batteries with a high theoretical energy density and low cost have attracted extensive research attention, their commercialization is still unsuccessful due to the poor cycle life caused by the dissolution of polysulfides. It is the key challenge to overcome polysulfide shuttling for achieving long-term cycling stability in Li-S batteries. Here we report a novel strategy for the synthesis of N, S-doped graphene with high nitrogen and sulfur contents via in situ self-assembly of graphene oxide and thiourea-formaldehyde resin and calcination. The N, S-doped graphen

Air pollution and fecundability: Results from a Danish preconception cohort study.

BACKGROUND: Animal and epidemiologic studies indicate that air pollution may adversely affect fertility. Epidemiologic studies have been restricted largely to couples undergoing fertility treatment or have retrospectively ascertained time-to-pregnancy among pregnant women. OBJECTIVES: We examined the association between residential ambient air pollution and fecundability, the per-cycle probability of conception, in a large preconception cohort of Danish pregnancy planners. METHODS: During 2007-2018, we used the Internet to recruit and follow women who were trying to conceive without the use

Proteomic Analysis of Paracoccidioides brasiliensis During Infection of Alveolar Macrophages Primed or Not by Interferon-Gamma.

Although members of the Paracoccidioides complex are not obligate intracellular pathogens, they present the ability to survive and multiply inside epithelial cells and phagocytes of mammals, which may favor the spread of the fungus in host tissues. Macrophages resident in the lung are the first line of defense against paracoccidioidomycosis (PCM), presenting mechanisms to control the pathogen dissemination through the granuloma formation or eliminating the fungus through phagocytosis. Phagocytosis triggers an oxidative burst, in which there is an increase in the production of toxic elements, d

In-situ algal control by two-stage nanobubble technology in Taihu Lake: Efficacy and ecological impact.

Hydrodynamic cavitation and ozone nanobubble-coupled hydrodynamic cavitation have demonstrated effective algae control in laboratories, but their in-situ potential, especially impact on nutrient salt degradation and microbial communities remain unclear. This study applied two-stage nanobubble technology, combining hydrodynamic cavitation and ozone nanobubbles, in a 3300 m2 semi-enclosed area of Taihu Lake to address these gaps. Results show that the technology efficiently controls algae, reduces odors, improves anaerobic conditions, and lowers ammonia nitrogen. Over 20 days, chlorophyll-a conc

Genetic variation in the urea cycle: a model resource for investigating key candidate genes for common diseases.

The urea cycle is the primary means of nitrogen metabolism in humans and other ureotelic organisms. There are five key enzymes in the urea cycle: carbamoyl-phosphate synthetase 1 (CPS1), ornithine transcarbamylase (OTC), argininosuccinate synthetase (ASS1), argininosuccinate lyase (ASL), and arginase 1 (ARG1). Additionally, a sixth enzyme, N-acetylglutamate synthase (NAGS), is critical for urea cycle function, providing CPS1 with its necessary cofactor. Deficiencies in any of these enzymes result in elevated blood ammonia concentrations, which can have detrimental effects, including central ne

The influence of combined carbon and nitrogen application on the growth and metabolism of Ilex rotunda.

When cultivating Ilex rotunda Thunb., the blind application of nitrogen fertilizer often resulted in poor growth and negatively impacted yield and quality. To explore the balancing effect of carbon fertilizer on excessive nitrogen fertilizer and improve the yield and quality of I. rotunda, two-year-old cuttings were sprayed with different concentrations of urea and α-ketoglutarate (α-KG) in 12 treatment groups. After 12 weeks of treatment, the growth, photosynthetic indexes, main physiological indexes and metabolome of I. rotunda were determined and analyzed. The results showed that applying a

Hyperammonemic encephalopathy due to essential amino acid hyperalimentation.

Hyperammonemic encephalopathy has occasionally been reported in uremic patients receiving hyperalimentation with essential amino acid (EAA) as a source of nitrogen as one of the remaining treatment options when the enteric routes were prohibited. We encountered this complication in a patient with normal renal function. A rat animal model was designed to elucidate the mechanism of hyperammonemia resulting from hyperalimentation with EAA as a source of nitrogen. Sixty-four male Long-Evan rats were divided into eight groups receiving feeds ad libitum or different formula of hyperalimentation. Hyp

Genome-resolved metagenomics reveals depth-related patterns of microbial community structure and functions in a highly stratified, AMD overlaying mine tailings.

Acid mine drainage (AMD) is a worldwide environmental problem, yet bioremediation is hampered by a limited knowledge of the reductive microbial processes in the AMD ecosystem. Here, we generate extensive metagenome and geochemical datasets to investigate how microbial populations and metabolic capacities driving major element cycles are structured in a highly stratified, AMD overlaying tailings environment. The results demonstrated an explicit depth-dependent differentiation of microbial community composition and function profiles between the surface and deeper tailings layers, paralleling the

Nannochloris sp. Microalgae Strain for Treatment of Dairy Wastewaters.

This paper focuses on a process for dairy wastewater treatment by mixotrophic cultivation of microalgae Nannochloris sp., using cheese whey obtained as a side flow from cheese production as an organic carbon source. The microalgae samples were prepared by adding to the standard growth medium increasing amounts of cheese whey, calculated to ensure a lactose concentration between 0 and 10 g/L. The samples were incubated at a constant temperature of 28 °C and 175 rpm stirring speed for a total time of seven days. Two LED (Light Emitting Diode) illumination schemes were applied in order to assess

Observations of nitrogen and phosphorus biogeochemistry in a surface flow constructed wetland.

Free surface water constructed wetlands (CWs) provide a buffer between domestic wastewater treatment plants and natural waterways. Understanding the biogeochemical processes in CWs is crucial to improve their performance. In this study we measured a range of water and sediment parameters, and biogeochemical processes, in an effort to describe the processing of nutrients within two wetland cells in series. As a whole the studied CW effectively absorbed both nitrogen (N) and phosphorus (P) emanating from the waste treatment plant. However the two individual cells showed marked differences relate

Nitrate accumulation and transformation processes within a karst conduit system: Insights from hydrochemistry, stable isotopes, metagenomics, and tracer tests.

Nitrate (NO3-) pollution in karst groundwater threatens drinking water security. However, limited subsurface accessibility makes tracing its migration and transformation within karst conduits challenging. This study integrated hydrochemistry, multiple isotopes, functional genes, and tracer tests to elucidate NO3- sources and transformation in the Lanfeng watershed, a representative karst conduit system in southwestern China. Nitrification dominated the watershed nitrogen cycle, with soil organic nitrogen (SON) as the primary NO3- source, contributing 69.4% and 74.0% to surface and groundwater,…

Phase shifts in the stoichiometry of rifamycin B fermentation and correlation with the trends in the parameters measured online.

Antibiotic fermentation processes are raw material cost intensive and the profitability is greatly dependent on the product yield per unit substrate consumed. In order to reduce costs, industrial processes use organic nitrogen substrates (ONS) such as corn steep liquor and yeast extract. Thus, although the stoichiometric analysis is the first logical step in process development, it is often difficult to achieve due to the ill-defined nature of the medium. Here, we present a black-box stoichiometric model for rifamycin B production via Amycolatopsis mediterranei S699 fermentation in complex mul

Regulation of ammonium loss under contrasting upwelling conditions: sensitivity of Feammox to environmental drivers.

Understanding the microbial processes involved in ammonium loss in highly productive marine systems is crucial for reconstructing the global nitrogen cycle. This study is the first to examine the anaerobic ammonium oxidation coupled with dissimilatory Fe(III) reduction (Feammox) and the abundance of iron-reducing bacteria (IRB) in a seagrass-dominated coastal lagoon exposed to two contrasting upwelling conditions. Potential Feammox rates varied from 6.0 to 39.2 mg N per m2 per day and were positively correlated with the abundance of IRB (Acidomicrobiaceae A6 spp. and Geobacteraceae spp.), sugg…

Ocean warming interacts with elevated nitrogen to modulate carbon storage-excretion balance in Gracilariopsis lemaneiformis.

Macroalgae farming represents a proposed approach for carbon dioxide removal (CDR), as macroalgae exhibit distinct advantages over other blue carbon taxa in cultivation scalability and cost effectiveness. Yet how future warming and coastal nitrogen enrichment will jointly affect macroalgal carbon fixation and sequestration remains poorly understood. Here, we investigated physiological and biochemical responses (growth, photosynthesis, pigment content, and carbon accumulation and excretion) of the commercially important seaweed Gracilariopsis lemaneiformis under a cross-factorial design of two

Fungal traits that drive ecosystem dynamics on land.

Fungi contribute extensively to a wide range of ecosystem processes, including decomposition of organic carbon, deposition of recalcitrant carbon, and transformations of nitrogen and phosphorus. In this review, we discuss the current knowledge about physiological and morphological traits of fungi that directly influence these processes, and we describe the functional genes that encode these traits. In addition, we synthesize information from 157 whole fungal genomes in order to determine relationships among selected functional genes within fungal taxa. Ecosystem-related traits varied most at r

Hyperammonemia Encephalopathy due to Urea Cycle Disorder Precipitated by Gastrointestinal Bleed in the Setting of Prior Bariatric Surgery.

The urea cycle is a metabolic pathway that excretes nitrogenous waste products from the body. Urea cycle disorders (UCDs) result from enzymatic deficiencies within this pathway, which can lead to life-threatening hyperammonemia. Gastric bypass-related hyperammonemia in patients who have undergone Roux-en-Y gastric bypass surgery has been previously reported. UCDs have been implicated as a cause of gastric bypass-related hyperammonemia. In this report, we present the case of a patient with a history of bariatric surgery who experienced severe hyperammonemia encephalopathy triggered by a gastroi

Revealing Mechanisms of Lithium-Mediated Nitrogen Reduction Reaction from First-Principles Simulations.

Recently, lithium-mediated nitrogen reduction reaction (Li-NRR) in nonaqueous electrolytes has proven to be an environmentally friendly and feasible route for ammonia electrosynthesis, revealing tremendous economic and social advantages over the industrial Haber-Bosch process which consumes enormous fossil fuels and generates massive carbon dioxide emissions, and direct electrocatalytic nitrogen reduction reaction (NRR) which suffers from sluggish kinetics and poor faradaic efficiencies. However, reaction mechanisms of Li-NRR and the role of solid electrolyte interface (SEI) layer in activatin

Marine antifouling biocide 4,5-dichloro-2-n-octyl-4-isothiazolin-3-one disrupts sediment microbiome structure and function: insights from absolute quantification and enzyme activity dynamics.

The organic booster biocide DCOIT (4,5-dichloro-2-n-octyl-4-isothiazolin-3-one) is widely used in marine antifouling systems, yet its ecological impacts on sediment microbiomes remain poorly understood. Here, we integrated absolute quantitative 16S rRNA gene sequencing, metagenomics, and enzyme activity assays to examine microbial responses to DCOIT exposure (0-50 μg/g sediment) over 30 days. DCOIT induced oxidative stress and bioenergetic impairment, accompanied by reduced microbial activity and inhibition of key enzyme-mediated processes involved in organic matter turnover and nitrogen trans
