Stem Cells: Innovations, Applications, and Future Directions.

Stem cells offer great promise in dentistry by regenerating critical oral tissues. Derived from sources such as dental pulp, periodontal ligament, and dental follicle, these cells can be pluripotent or multipotent and are capable of differentiating into various cell types, including odontoblasts and osteoblasts. This regenerative potential could transform treatments for dental conditions like periodontal disease and pulpitis. Advancing this field requires understanding stem cell differentiation and exploring tissue engineering and biomaterial scaffolds. Despite the potential, evolving legal an…

Healing of pulp exposures: an ultrastructural study.

The fine structure of tissue changes during the first 14 days following pulp exposure and capping with a hard setting Ca(OH)2 cement has been studied. The early changes included hemorrhage and moderate inflammation which were largely resolved during the first week. During the second week differentiation of cells occurred at the wound surface. These cells had the characteristic features of odontoblasts and formed a predentin-like collagen matrix. The capping material was closely adapted to cellular structures at the wound surface or to the predentin-like matrix at all observation periods. Denti…

Effect of solvent pretreatments on trans-dentinal cytotoxicity of bioactive materials.

This study evaluated the effect of solvent-based dentin pretreatments containing dimethyl sulfoxide (DMSO) and/or ethanol on trans-dentinal cell viability of bioactive materials in simulated deep clinical cavities. Dentin discs (300 μm) were prepared, balanced among groups according to dentin permeability and autoclaved. Three-dimensional cultures of odontoblast-like cells were transferred to the pulpal aspect of dentin discs inside perfusion split-chambers designed for trans-dentinal cytotoxicity test following ISO 7405 standards. Dentin pretreatments were composed of ethanol (EtOH), DMSO, th

Dental Wear and Molar Pulp Volume Reduction in Macaca fascicularis.

OBJECTIVES: Odontoblasts lining the pulp cavity deposit dentine throughout life, meaning the volume of the pulp cavity decreases with age. Primates with more abrasive diets have relatively higher molar pulp volume in their unworn molars than those with less abrasive diets. We propose that species with more abrasive diets deposit additional dentine across their lifespans to help resist wear and extend the effective lifespan of their molars. Whereas both age and wear affect dentine deposition, it is unknown which of these two variables has the greater impact. MATERIALS AND METHODS: We measured …

Long-Term Fluoride Release from Dental Resins Affects STRO-1+ Cell Behavior.

Fluoride-releasing restorative dental materials can be beneficial to remineralize dentin and help prevent secondary caries. However, the effects of fluoride release from dental materials on the activity of dental pulp stem cells are not known. Here we investigate whether different fluoride release kinetics from dental resins supplemented with modified hydrotalcite (RK-F10) or fluoride-glass filler (RK-FG10) could influence the behavior of a human dental pulp stem cell subpopulation (STRO-1(+) cells) known for its ability to differentiate toward an odontoblast-like phenotype. The 2 resins, char

Intratubular crystal deposition and remineralization of carious dentin.

An analysis of accumulated experimental results, the mechanism of intratubular crystal deposition in the transparent layer and remineralization of the matrix of carious dentin has led to a more rounded concept and understanding of dentin caries relevant to clinical practice. The transparent layer of carious dentin is not sclerotic but is part of the demineralized softened dentin. The crystal precipitation in the tubules is an intermediate product in a slow demineralization process. Physiological remineralization occurs in matrix of the inner carious dentin which has crossbanded collagen fibers

Single-cell sequencing systematically analyzed the mechanism of Emdogain on the restoration of delayed replantation periodontal membrane.

The repair of the periodontal membrane is essential for the successful management of periodontal disease and dental trauma. Emdogain® (EMD) is widely used in periodontal therapy due to its ability to promote repair. Despite substantial research, the cellular and molecular mechanisms underlying EMD's effects, particularly at the single-cell resolution, remain incompletely understood. This study established a delayed tooth replantation model in rats to investigate these aspects. Tooth loss rate and degree of loosening were evaluated at 4 and 8 weeks. Micro-CT, HE staining, TRAP staining, and imm

MicroRNA-155 expression is associated with pulpitis progression by targeting SHIP1.

BACKGROUND: Pulpitis is a commonly seen oral inflammation condition in clinical practice, it can cause much pain for the patient and may ind … verse the effects. Bioinformatics analysis demonstrated that SHIP1 is a direct target of miR-155 in odontoblasts, this result was further verified at both mRNA and protein level. Inhibition of miR-155 resulted in the downregulation of inflammation factors, while co-transfection of si-SHIP1 and miR-155 inhibitor promoted the inflammatory responses. Treatment wi …

Pulpal response to a fluoride-releasing all-in-one resin bonding system.

Pulp tissue reactions to a fluoride-releasing all-in-one resin bonding system (Reactmer Bond and Reactmer Paste) in non-exposed monkey teeth were histopathologically evaluated at three, 30, and 90 days after restoration. No serious inflammatory reactions of the pulp, such as necrosis or abscess formation, were observed. At 90 days in the Reactmer group, odontoblastic change and inflammatory cell infiltration were not observed, and slight irritation dentin formation was formed. The pulpal response of the Reactmer group was minimally different from that of the control group. Consequently, the Re

The Reparative Function of MMP13 in Tertiary Reactionary Dentinogenesis after Tooth Injury.

MMP13 gene expression increases up to 2000-fold in mineralizing dental pulp cells (DPCs), with research previously demonstrating that global MMP13 deletion resulted in critical alterations in the dentine phenotype, affecting dentine-tubule regularity, the odontoblast palisade, and significantly reducing the dentine volume. Global MMP13-KO and wild-type mice of a range of ages had their molar teeth injured to stimulate reactionary tertiary dentinogenesis. The response was measured qualitatively and quantitatively using histology, immunohistochemistry, micro-CT, and qRT-PCR in order to assess ch

Extracellular processing of bone and dentin proteins in matrix mineralization.

There are two steps in the process of matrix-mediated bone and dentin mineralization. First, as in other soft tissues, osteoblasts/odontoblasts synthesize collagenous matrices and second, mineral deposits in these matrices at a location distant from the cells that synthesized the matrices. We suggest a sequence of events that lead the matrix to mineralization: the phosphoproteins of bone and dentin are posttranslationally processed by limited proteolysis, then they are extracellularly processed into a more phosphorylated species that, we believe, facilitates mineralization. Our in situ phospho…

Effects of Porphyromonas gingivalis lipopolysaccharide on the behavior of human dental pulp stem cells in vitro and their inflammation-related transcriptomics profile.

OBJECTIVE: The inflammatory microenvironment in pulpitis and periapical periodontitis critically impairs dental pulp stem cells (DPSCs) functionality, yet the underlying molecular mechanisms remain poorly defined. This study aimed to investigate the effects of Porphyromonas gingivalis lipopolysaccharide (P. gingivalis LPS) on human DPSCs behavior and elucidate the transcriptomic changes underlying LPS-induced inflammation. DESIGN: Human DPSCs were exposed to 1 µg/mL LPS for 24 hours to establish an inflammatory model, with subsequent evaluation of proliferation (CCK-8), migration (Transwell),
