Particle deposition in the human lung: Health implications of particulate matter from different sources.

Although ambient particulate matter or particles have been found to be associated with morbidity and mortality all over the world, specific health effects of particles from different sources need further elucidation. The objective of this work is to predict the deposition of particles from different sources in the human lung. The whole lung, consisting of 24 generations of branches from trachea to alveoli, was approximated using a one-dimensional lumped "trumpet" model with a variable cross-sectional area. The aerosol dynamics equation was numerically solved using a finite difference method to…

Short-Term Elevation of Fine Particulate Matter Air Pollution and Acute Lower Respiratory Infection.

RATIONALE: Nearly 60% of U.S. children live in counties with particulate matter less than or equal to 2.5 μm in aerodynamic diameter (PM2.5) concentrations above air quality standards. Understanding the relationship between ambient air pollution exposure and health outcomes informs actions to reduce exposure and disease risk. OBJECTIVES: To evaluate the association between ambient PM2.5 levels and healthcare encounters for acute lower respiratory infection (ALRI). METHODS: Using an observational case-crossover design, subjects (n = 146,397) were studied if they had an ALRI diagnosis and resi…

Exposure to Concentrated Ambient Fine Particulate Matter Induces Vascular Endothelial Dysfunction via miR-21.

Vascular endothelial permeability transition does not cause significant lesions, but enhanced permeability may contribute to the development of vascular and other diseases, including atherosclerosis, hypertension, heart failure and cancer. Therefore, elucidating the effect of Particulate Matter 2.5 (PM2.5) on vascular endothelial permeability could help prevent disease that might be caused by PM2.5. Our previous study and the present one revealed that PM2.5 significantly increased the permeability of vascular endothelial cells and disrupted the barrier function of the vascular endothelium in S…

Regulation of particulate matter-induced mucin secretion by transient receptor potential vanilloid 1 receptors.

Exposure to airborne particulate matter (PM) is a worldwide health problem. Previous studies have reported that PMs induced depolarizing currents and increased intracellular Ca(2+) in human bronchial epithelial cells. Ca(2+) plays important role in the regulation of mucus exocytosis, and mucin hypersecretion is a key pathological feature of inflammatory respiratory diseases. To explore more mechanisms underlying PM toxicity, we measured PM-induced mucin secretion in human bronchial epithelial (16HBE) cells. MUC5AC secretion and cyclic adenosine monophosphate (cAMP) level were detected by ELISA…

The potential for delivery of particulate matter through positive airway pressure devices (CPAP/BPAP).

BACKGROUND: Airborne particulate matter may induce health risk with inhalation. Special concerns exist for deployed military personnel with inhaled particulate matter in desert environments. Continuous positive airway pressure (CPAP) used in obstructive sleep apnea may facilitate inhalation of particulate matter. We evaluated the ability of commercial CPAP filter systems to eliminate inhalation of particulate matter. METHODS: An ultrasonic medical nebulizer (DeVilbliss Ultraneb, DeVilbliss, Somerset, PA) atomized liquid producing "respirable" aerosol. Technetium-99m diethylene triamine pentaa…

Gender differences in fetal growth of newborns exposed prenatally to airborne fine particulate matter.

Our primary purpose was to assess sex-specific fetal growth reduction in newborns exposed prenatally to fine particulate matter. Only women 18-35 years of age, who claimed to be non-smokers, with singleton pregnancies, without illicit drug use and HIV infection, free from chronic diseases were eligible for the study. A total of 481 enrolled pregnant women who gave birth between 37 and 43 weeks of gestation were included in the study. Prenatal personal exposure to fine particles over 48 h during the second trimester was measured using personal monitors. To evaluate the relationship between the …

Midpolarity and nonpolar wood smoke particulate matter fractions deplete glutathione in RAW 264.7 macrophages.

Wood smoke particulate matter (PM) is a complex mixture of components falling in a spectrum of highly polar to nonpolar species. Wood smoke PM is a likely factor in pulmonary disease and induces oxidative damage. Most toxicity studies focus upon nonpolar species such as polycyclic aromatic hydrocarbons (PAHs). However, the role of more polar PM constituents as toxicants is not clear. In this work, we evaluated the ability of multiple fractions of varying polarity to deplete glutathione (GSH) in RAW 264.7 macrophages and BEAS-2B bronchial epithelial cells. We utilized hot pressurized (subcritic…

Chemical analysis of World Trade Center fine particulate matter for use in toxicologic assessment.

The catastrophic destruction of the World Trade Center (WTC) on 11 September 2001 caused the release of high levels of airborne pollutants into the local environment. To assess the toxicity of fine particulate matter [particulate matter with a mass median aerodynamic diameter < 2.5 microm (PM2.5)], which may adversely affect the health of workers and residents in the area, we collected fallen dust samples on 12 and 13 September 2001 from sites within a half-mile of Ground Zero. Samples of WTC dust were sieved, aerosolized, and size-separated, and the PM2.5 fraction was isolated on filters. Her…

Particulate matter, sulfur dioxide, and pulmonary function in never-smoking adults in Chongqing, China.

Chongquing is one of the most polluted cities in China. To study the respiratory health effects of air pollution for this city, the authors monitored the ambient levels of particulate matter (PM2.5) and sulfur dioxide (SO2) in urban and suburban areas and performed pulmonary function testing on 1,075 adults 35 to 60 years of age who had never smoked and did not use coal stoves for cooking or heating. The mean concentration of SO2 in the urban areas (213 micrograms/m3) was twice as high as that in suburban areas (103 micrograms/m3). Mean PM2.5 levels were high in both urban (143 micrograms/m3) …

Fine Particulate Matter (PM2.5), the Exposome, and Cumulative Health Risk: A Life-course Perspective.

Fine particulate matter (PM2.5; aerodynamic diameter ≤2.5 μm) is a leading contributor to the global burden of disease and is associated with substantial morbidity and mortality. However, its health effects are often evaluated within single-exposure frameworks, limiting causal inference and reducing the translational relevance of evidence for policy development. In this study, we reconceptualise PM2.5 within the exposome framework by integrating external exposures, internal biological responses, and structural determinants of health across the life course. PM2.5 comprises a complex and dynamic…

Ursodeoxycholic acid ameliorates particulate matter-induced cutaneous inflammation via the AhR/ERK signaling pathway.

Particulate matter (PM) and its constituent polycyclic aromatic hydrocarbons (PAHs) are major environmental hazards that induce skin inflammation by activating the aryl hydrocarbon receptor (AhR) and its downstream mitogen-activated protein kinase (MAPK) signaling pathways. Ursodeoxycholic acid (UDCA) is a hydrophilic bile acid, originally identified in bear bile, and has been used for the treatment of cholesterol gallstones and primary biliary cholangitis. Although recent pharmacological studies have highlighted its roles in metabolic and autoimmune diseases, its specific effect on the PM-ind…

Multilevel toxicological impacts of industrial settleable particulate matter in a freshwater crustacean, Macrobrachium rosenbergii.

As integral components of aquatic food webs, crustaceans serve as pivotal conduits for energy transfer and sensitive bioindicators of environmental quality. This study characterizes the toxicological consequences of settleable atmospheric particulate matter (SePM)-a byproduct of industrial iron-ore processing-utilizing the freshwater shrimp Macrobrachium rosenbergii as a model organism. Experimental exposure demonstrated that SePM significantly enhances the aqueous solubility of both conventional (Fe, Al, Zn, Cu, Mn) and emergent (Cr, Ni, Sr, Sn, La, Ce) metals, establishing it as a potent vec…

Particulate Matter Exposure and Diabetic Kidney Dysfunction: Insights from Integrated Transcriptomic and Bioinformatics Analyses.

Exposure to ambient particulate matter (PM) has been linked to renal dysfunction, particularly in diabetic populations, but the underlying mechanisms remain unclear. We performed bidirectional Mendelian randomization to assess causal relationships between PM exposure and estimated glomerular filtration rate (eGFR), integrated transcriptomic datasets to identify PM-related genes overlapping with diabetic kidney disease (DKD) differentially expressed genes, and applied machine learning approaches to select key feature genes and construct diagnostic models. Single-cell and spatial transcriptomic …

Seasonal particulate matter exposure is associated with upper respiratory microbiota restructuring in dairy heifers.

BACKGROUND: Particulate matter (PM), particularly PM2.5 and PM10, is a major environmental health concern linked to respiratory diseases in humans and animals. Northern Thailand, especially Chiang Mai, experiences recurrent seasonal air pollution from biomass burning, exposing outdoor-housed livestock to elevated ambient PM levels. The bovine upper respiratory tract (URT) harbors both commensal and opportunistic microorganisms, and disruption of this microbiota may influence susceptibility to bovine respiratory disease (BRD). However, the impact of natural PM exposure on the bovine URT microbi…

Impact of particulate matter 2.5 on placental ultrastructure including mitochondrial damage through oxidative stress.

INTRODUCTION: Particulate matter 2.5 (PM2.5) refers to fine particles with a diameter of less than 2.5μm, associated with adverse pregnancy outcomes. The study aims to determine whether elevated prenatal PM2.5 levels were associated with alterations in the ultrastructure of the placenta and mitochondria, potentially linked to oxidative stress. METHODS: The placental samples were collected from the Air Pollution on Pregnancy Outcome (APPO) cohort and were classified into two groups based on PM2.5 exposure levels: the High group (> 15 μg/m³; n = 9) and the Low group (≤ 15 μg/m³; n = 8). Transmi…

Fine particulate matter promoted vascular calcification through the miR-27b-5p/HES1 signaling pathway.

Fine particulate matter (PM2.5) remains to be the leading cause of global disease burden and is linked to arterial calcification in humans. This study revealed that PM2.5 facilitated the phenotype switching of mouse aortic smooth muscle (MOVAS) cells and promoted in vitro calcification in the present of phosphate calcification medium. Mechanistic investigations revealed that PM2.5 may inhibit the transcription of miR-27b-5p in an XBP1 dependent manner, which subsequently promoted the interaction of HES1 and Runx2, eventually leading to the osteogenic transformation of MOVAS cells. More importa…

Combined exposure of sleep deprivation and environmental particulate matter drives aging in multiple systems.

Sleep disturbance accelerates aging, with accompanying exposure to air pollution. However, most studies ignore the combined exposure. This study aimed to investigate the combined effects of sleep deprivation and PM2.5 exposure on multi-system aging and to explore the damage mechanisms. The sleep deprivation instrument and the Shanghai Meteorological and Environmental Animal Exposure System (Shanghai-METAS) were used to construct a combined exposure model for one month. Our study used multiple behavioral, imaging, and molecular biological examinations to describe the aging characteristics in th

One-Point Calibration of Low-Cost Sensors for Particulate Air Matter (PM) Concentration Measurement.

The use of low-cost sensors has dramatically increased in recent years in all engineering sectors. In the buildings and automotive field, low-cost sensors open very interesting perspectives, because they allow one to monitor temperature and humidity distributions together with air quality in a widespread and punctual way and allow for the control of all energy parameters. The main issue remains the validation of the measurements. In this work, we propose an innovative approach to verify the measurements given by some low-cost systems built ad hoc for automotive applications. Two independent lo
