Middle aged turn point in parameters of oxidative stress and glucose catabolism in mouse cerebellum during lifespan: minor effects of every-other-day fasting.

The cerebellum is considered to develop aging markers more slowly than other parts of the brain. Intensification of free radical processes and compromised bioenergetics, critical hallmarks of normal brain aging, may be slowed down by caloric restriction. This study aimed to evaluate the intensity of oxidative stress and the enzymatic potential to utilize glucose via glycolysis or the pentose phosphate pathway (PPP) in the cerebellum of mice under ad libitum versus every-other-day fasting (EODF) feeding regimens. Levels of lipid peroxides, activities of antioxidant and key glycolytic and PPP en

Regulation of autophagy, glucose uptake, and glycolysis under dengue virus infection.

We previously reported that dengue virus (DENV)-induced autophagy plays a promoting role in viral replication and pathogenesis both in vitro and in vivo. Although it is known that DENV infection increases glycolysis, which promotes viral replication, the role of glucose metabolism together with autophagic activity in DENV replication remains unclear. In this study, we reveal that DENV2 infection increased autophagic activity, glucose uptake, protein levels of glucose transporter-1 (GLUT1), and glycolysis rate-limiting enzyme hexokinase-2 (HK2) in cells. Furthermore, the protein levels of LC3-I

Metabolic engineering of Corynebacterium glutamicum for hyperproduction of polymer-grade L- and D-lactic acid.

Strain development is critical for microbial production of bio-based chemicals. The stereo-complex form of polylactic acid, a complex of poly-L- and poly-D-lactic acid, is a promising polymer candidate due to its high thermotolerance. Here, we developed Corynebacterium glutamicum strains producing high amounts of L- and D-lactic acid through intensive metabolic engineering. Chromosomal overexpression of genes encoding the glycolytic enzymes, glucokinase, glyceraldehyde-3-phosphate dehydrogenase, phosphofructokinase, triosephosphate isomerase, and enolase, increased L- and D-lactic acid concent

Blockade of glycolysis-dependent contraction by oroxylin a via inhibition of lactate dehydrogenase-a in hepatic stellate cells.

BACKGROUND: Contraction of hepatic stellate cells (HSCs) plays an important role in the pathogenesis of liver fibrosis by regulating sinusoidal blood flow and extracellular matrix remodeling. Here, we investigated how HSC contraction was affected by the natural compound oroxylin A, and elucidated the underlying mechanism. METHODS: Cell contraction and glycolysis were examined in cultured human HSCs and mouse liver fibrosis model upon oroxylin A intervention using diversified cellular and molecular assays, as well as genetic approaches. RESULTS: Oroxylin A limited HSC contraction associated w

Copy number variation as a genetic basis for heterotaxy and heterotaxy-spectrum congenital heart defects.

Genomic disorders and rare copy number abnormalities are identified in 15-25% of patients with syndromic conditions, but their prevalence in individuals with isolated birth defects is less clear. A spectrum of congenital heart defects (CHDs) is seen in heterotaxy, a highly heritable and genetically heterogeneous multiple congenital anomaly syndrome resulting from failure to properly establish left-right (L-R) organ asymmetry during early embryonic development. To identify novel genetic causes of heterotaxy, we analysed copy number variants (CNVs) in 225 patients with heterotaxy and heterotaxy-

T-cell metabolism in autoimmune disease.

Cancer cells have long been known to fuel their pathogenic growth habits by sustaining a high glycolytic flux, first described almost 90 years ago as the so-called Warburg effect. Immune cells utilize a similar strategy to generate the energy carriers and metabolic intermediates they need to produce biomass and inflammatory mediators. Resting lymphocytes generate energy through oxidative phosphorylation and breakdown of fatty acids, and upon activation rapidly switch to aerobic glycolysis and low tricarboxylic acid flux. T cells in patients with rheumatoid arthritis (RA) and systemic lupus ery

Pathogenesis, laboratory diagnosis, and clinical implications of erythrocyte enzyme deficiencies in dogs, cats, and horses.

Deficiencies of enzymes involved in erythrocyte metabolism can have significant effects on erythrocyte function and survival. Animals with pyruvate kinase (PK) or phosphofructokinase (PFK) deficiencies have shortened erythrocyte life spans and regenerative anemia. PK-deficient dogs (but not PK-deficient cats) develop progressive myelofibrosis and osteosclerosis of bone marrow and hemochromatosis and cirrhosis of the liver. PFK-deficient dogs have sporadic episodes of hyperventilation-induced intravascular hemolysis and hemoglobinuria. Cytochrome b5 reductase (Cb5R) deficiency in dogs and cats

Metabolic effects and cellular volume responses induced by noradrenaline in nucleated erythrocytes.

The mechanism of adrenergic swelling, cAMP accumulation and associated Na+ and K+ concentration changes was investigated in amphibian Rana ridibunda erythrocytes. The addition of noradrenaline to an isotonic suspension of red cells of frog Rana ridibunda in the presence of the phosphodiesterase inhibitor isobutyryl-methylxanthine (IBMX), induced a significant increase of the cell volume. Forskolin treatment showed analogous results. The removal of the Na+ from the incubation medium, inhibited the volume changes caused by either noradrenaline or forskolin. Under the same conditions a more than

Cyclosporin A antagonizes phenylephrine, oxytocin and angiotensin effects on glucose metabolism in rat thymus lymphocytes.

Effects of phenylephrine, oxytocin and angiotensin on fructose 2,6-bisphosphate (Fru 2,6-P2) content and glycolytic parameters were studied in incubated thymus lymphocytes. These hormones modified Fru 2,6-P2 content dependent upon the energetic status of the cells. In non-preincubated thymus lymphocytes (with relatively high levels of glycogen and ATP), phenylephrine, oxytocin and angiotensin depressed Fru 2,6-P2 content in a dose-dependent manner. The opposite was found when the cells were preincubated for 2 h without substrates (low levels of ATP and glycogen). Changes in lactate release wer

Effects of dehydration on organ metabolism in the frog Pseudacris crucifer: hyperglycemic responses to dehydration mimic freezing-induced cryoprotectant production.

The metabolic effects of evaporative water loss at 5 degrees C were assessed for both fall- and spring-collected spring peepers Pseudacris crucifer. Frogs readily endured the loss of 50% of total body water. During dehydration organ water content was defined with no change in water content in skeletal muscle, gut, and kidney of 50% dehydrated frogs and reduced water content in liver, brain and heart. Dehydration stimulated a rapid and massive increase in liver glucose production. In fall-collected frogs liver glucose rose by 120-fold to 2690 +/- 400 nmol.mg protein-1 or 220 mumol.g ww-1 in 50%

Carbohydrate utilization by tilapia (Oreochromis niloticus x O. aureus) as influenced by different chromium sources.

An experiment was conducted to study the influence of three different forms of chromium (CrCl3.6H2O, Na2CrO4.4H2O and Cr2O3) on the utilization of two carbohydrates (glucose and cornstarch) by juvenile hybrid tilapia (Oreochromis niloticus x O. aureus). Average initial body weight of the fish was 1.13 +/- 0.02 g. Fish were fed 5% body wt/d. Significantly (P < 0.05) greater body weight gain, food intake, protein retention, energy retention and body lipid concentration were observed in fish fed the starch diet than in those fed the glucose diet. Fish fed the glucose diet supplemented with any ty

Naftidrofuryl oxalate improves impaired brain glucose metabolism after microsphere-induced cerebral embolism in rats.

The present study was designed to elucidate possible therapeutic effects of naftidrofuryl on the brain glucose metabolism after cerebral ischemia. Cerebral ischemia was induced by injecting 680 microspheres with a diameter of 48 microns into the right internal carotid artery of the rat. After ensuring the onset of symptoms of stroke on the first day after the operation, the rats were treated with intraperitoneal injections of 15 mg/kg naftidrofuryl oxalate twice a day. The behavioral and metabolic changes of operated rats were monitored up to the 5th day after surgery. The symptoms gradually f

[Exercise-induced muscular weakness, myalgia and contractures. I. A clinical review].

In the differential diagnosis of intermittent claudication some rare myopathies have to be considered. The most frequent is phosphorylase deficiency (McArdle's disease). Exercise-induced muscular pain, weakness, contractures and occasionally myoglobinuria are the most prominent clinical signs. Serum creatine phosphokinase, aldolase and lactic dehydrogenase may be elevated after exertion. In the ischemic forearm test there is no rise of serum lactic acid. The enzyme deficiency can be demonstrated by histochemical and biochemical examination of a muscle specimen. Further, but more infrequent, en

PFKFB3-involved glycolytic metabolism supports sepsis-induced neutrophil extracellular trap formation.

Uncontrolled formation of neutrophil extracellular traps (NETs) is one of the main mechanisms leading to death in septic patients. PFKFB3, a notable enzyme of glycolysis metabolism, is involved in the inflammatory activation of granulocytes. Our previous studies demonstrated that PFKFB3-involved glycolytic metabolism supports CXCR4hi neutrophil inflammatory activation, which is a key mechanism for the inflammatory injury of the lungs induced by sepsis. However, the specific mechanism by which PFKFB3 supports NET formation in sepsis remains unclear. Here, we found that high expression of neutro

Loss of eIF3m aggravates myocardial ischemia/reperfusion injury via enhancing nitrative damage and inhibiting glycolytic reprogramming.

The reprogramming of myocardial energy metabolism is critical for cardiac repair after ischemia/reperfusion (I/R) injury, yet the upstream translational control mechanisms remain elusive. Here, through multi-omics analysis of human failing hearts, we identify the mRNA regulator eIF3m as an essential factor for maintaining cardiac glycolytic flux post-I/R. We show that eIF3m, independently of the canonical eIF3 complex, stabilizes Mt2 and related stress-response mRNAs, ultimately protecting the key glycolytic activator Pfkfb3. Loss of eIF3m impairs the heart's adaptive shift toward utilizing gl

Quercetin alleviates acute pancreatitis by modulating glycolysis and mitochondrial function via PFKFB3 inhibition.

OBJECTIVE: Acute pancreatitis (AP) is a severe inflammatory disease associated with dysregulated glycolysis and mitochondrial dysfunction. This study investigates the therapeutic potential of quercetin, a novel PFKFB3 inhibitor, in modulating glycolysis and mitochondrial function to alleviate AP. METHODS: We conducted homology analysis of the PFKFB3 protein and identified quercetin as a potential inhibitor through molecular docking. In vitro experiments using a cerulein-induced inflammatory pancreatic cell model assessed the effects of quercetin on PFKFB3 expression, glycolysis, and mitochond

PFKFB3 Mediated Glycolytic Reprogramming Drives Vascular Endothelial Injury Under Chronic Intermittent Hypoxia.

Metabolic-inflammatory crosstalk is a hallmark of cardiovascular pathogenesis. Obstructive sleep apnea (OSA), characterized by chronic intermittent hypoxia (CIH), is an independent risk factor for cardiovascular diseases. While endothelial inflammation driven by CIH is pivotal in disease progression, the underlying metabolic mechanisms remain poorly defined. Our research shows that phosphofructo-2-kinase/fructose-2,6-biphosphatase 3 (PFKFB3), a key glycolytic activator, is markedly upregulated in endothelial cells (ECs) exposed to CIH, correlating with enhanced glycolysis, suppressed mitochond
