Genetic analysis of plant disease resistance pathways.

Plant disease resistance (R) genes are introduced into high yielding crop varieties to improve resistance to agronomically important pathogens. The R gene-encoded proteins are recognitionally specific, interacting directly or indirectly with corresponding pathogen avirulence (avr) determinants, and are therefore under strong diversifying selection pressure to evolve new recognition capabilities. Genetic analyses in different plant species have also revealed more broadly recruited resistance signalling genes that provide further targets for manipulation in crop improvement strategies. Understan

Use of GFP to investigate expression of plant-derived vaccines.

Plants are low-cost bioreactors for the production of various biopharmaceuticals including oral vaccines. Plant-derived oral vaccines are potentially useful in combating viral infections involving mucosal immunity. Transgenic plants have been generated to successfully produce mucosal vaccines against cholera, hepatitis B, foot-and-mouth disease, and Norwalk virus. As a first step toward the generation of oral vaccines against the severe acute respiratory syndrome coronavirus (SARS-CoV), we have expressed a recombinant S1 protein of the SARS-CoV in transformed tobacco. Since plant transformatio

Rhizosphere triangular warfare: mechanistic insights for plant- beneficial microbe- pathogen interactions.

The rhizosphere is a living physical space that is constantly evolving and is characterized by a complex interplay between beneficial microbes, pathogens, and plants, mediated by molecular and ecological interactions. These three-way relationships constitute a rhizosphere "triangular warfare system," which controls plant health, nutrient uptake, disease resistance, and plant resilience. This review provides a mechanistic overview of the molecular, metabolic and ecological processes underlying these tripartite interactions. We discuss how plants integrate pattern-triggered immunity (PTI), effec

Leaf Developmental Stage Influences Disease Resistance in Tomato.

The processes of morphogenesis and differentiation are crucial for leaf development, with the duration of the morphogenetic window influencing final leaf shape. Leaves at different developmental stages exhibit distinct morphological and physiological characteristics that may influence their ability to resist pathogens, and disease resistance has been linked to developmental stage in many plant species. To understand how leaf development impacts disease resistance, we examined the immunity of leaves at distinct developmental stages, exploring the role of hormonal pathways and the impact of leaf

Histone variant H2A.Z is a conserved regulator of RBOH expression and ROS production during innate immunity in Arabidopsis thaliana and Solanum lycopersicum.

Histone variant H2A.Z, a conserved variant of core histone H2A, has been widely accept as a master regulator of plant gene transcription and plant-environment interactions including pathogen resistance. Yet the exact roles of H2A.Z in mediating stepwise plant immune such as ROS production are not known, which is a pivotal step in plant-pathogen interactions and is predominantly controlled by the RESPIRATORY BURST OXIDASE HOMOLOGUE (RBOH) family. In this research, we found that mutation of three and two H2A.Z genes in Arabidopsis and tomato both reduced the plant pathogen-associated molecular p

An atypical NLR protein modulates the NRC immune receptor network in Nicotiana benthamiana.

The NRC immune receptor network has evolved in asterid plants from a pair of linked genes into a genetically dispersed and phylogenetically structured network of sensor and helper NLR (nucleotide-binding domain and leucine-rich repeat-containing) proteins. In some species, such as the model plant Nicotiana benthamiana and other Solanaceae, the NRC (NLR-REQUIRED FOR CELL DEATH) network forms up to half of the NLRome, and NRCs are scattered throughout the genome in gene clusters of varying complexities. Here, we describe NRCX, an atypical member of the NRC family that lacks canonical features of

Pseudozyma aphidis Suppresses Microbe-Associated Molecular Pattern (MAMP)-Triggered Callose Deposition and Can Penetrate Leaf Tissue.

Beneficial microorganisms need to overcome the plant defense system to establish on or within plant tissues. Like pathogens, beneficial microbes can manipulate a plant's immunity pathways, first by suppressing and hiding to establish on the host and then by inducing resistance to protect the plant. In the current study, we demonstrated that although Pseudozyma aphidis can activate microbe-associated molecular pattern (MAMP)-associated genes, it does not activate MAMP-triggered callose deposition and can, moreover, suppress such deposition triggered by Flg22 or chitin. While MAMP-associated gen

Starvation as a weapon in fungal-plant warfare.

Fungal pathogens cause devastating diseases in staple crops and pose a tremendous threat to food security. Therefore, it is critical to unde … olase activity to provoke phosphate limitation in host plants. This novel effector strategy impairs plant immunity, thus favoring pathogen proliferation and disease. Intriguing new work also demonstrates that the phytohormones strigolactone and methyl jasmonate influence phosphate and carbon metabolism in fungi. As discussed in this review, these examples illust …

Oral delivery of human biopharmaceuticals, autoantigens and vaccine antigens bioencapsulated in plant cells.

Among 12billion injections administered annually, unsafe delivery leads to >20million infections and >100million reactions. In an emerging new concept, freeze-dried plant cells (lettuce) expressing vaccine antigens/biopharmaceuticals are protected in the stomach from acids/enzymes but are released to the immune or blood circulatory system when plant cell walls are digested by microbes that colonize the gut. Vaccine antigens bioencapsulated in plant cells upon oral delivery after priming, conferred both mucosal and systemic immunity and protection against bacterial, viral or protozoan pathogens

Using Clear Nail Polish to Make Arabidopsis Epidermal Impressions for Measuring the Change of Stomatal Aperture Size in Immune Response.

Plant stomata are an essential route for bacterial pathogens to entry inside host tissue and cause diseases. As an important defense mechanism, plant stomata can actively restrict bacterial invasion by dynamically regulating the opening, closing, and reopening of stomatal guard cells. Therefore, accurately measuring the stomatal aperture size during the bacterial pathogenesis is an important approach to study the stomata related immunity. Several methods have been developed for stomatal aperture measurement. Here, we described a detailed protocol of using clear nail polish to make Arabidopsis

Plant-derived immuno-adjuvants in vaccines formulation: a promising avenue for improving vaccines efficacy against SARS-CoV-2 virus.

The SARS-CoV-2 outbreak has posed a plethora of problems for the global healthcare system and socioeconomic burden. Despite valiant efforts to contain the COVID-19 outbreak, the situation has deteriorated to the point that there are no viable preventive therapies to treat this disease. The case count has skyrocketed globally due to the newly evolved variants. Despite vaccination drives, the re-occurrence of recent pandemic waves has reinforced the importance of innovation/utilization of immune-booster to achieve appropriate long-term vaccine protection. Plant-derived immuno-adjuvants, which ha

A wheat lncRNA-encoded peptide promotes antiviral resistance by targeting viral NIb for autophagic degradation.

Long noncoding RNAs (lncRNAs) are increasingly recognized as sources of functional peptides that regulate plant development and immunity, yet little is known about whether lncRNA-encoded peptides participate in antiviral defense. Through transcriptomic analysis combined with peptide-coding potential prediction, we identified wheat lncRNA19864, which is highly expressed in wheat yellow mosaic virus (WYMV)-resistant cultivars and encodes a 39-amino-acid peptide, TaLEP1. Genetic manipulation of TaLEP1 expression revealed that TaLEP1 acts as a positive regulator of wheat resistance to WYMV. Mechan

NAC transcription factors transcriptionally fine-tune signal homeostasis in plant systemic acquired resistance.

Small signalling molecules, such as salicylic acid (SA) and N-hydroxypipecolic acid (NHP), act synergistically to activate systemic acquired … 1, that negatively regulates NHP and SA biosynthesis. NAC90 and NAC36 act as negative regulators of plant immunity by repressing transcription of ALD1, FMO1, and ICS1, the major NHP and SA biosynthetic genes, via direct binding to their promoters. These TFs, along with another NAC TF, NAC61, form heterodimers, further enhancing their repressive effects on NHP an …

Membrane remodelling mediates lipopeptide-induced immunity in Arabidopsis.

Bacteria-derived lipopeptides are immunogenic triggers of host defences in metazoans and plants. Root-associated rhizobacteria produce cyclic lipopeptides that activate induced systemic resistance against microbial infection in various plant species. Whether and how these molecules are perceived at the plant cell surface remains elusive. Here we reveal that immune activation in Arabidopsis thaliana by the lipopeptide elicitor surfactin is mediated via a specific interaction with membrane sphingolipids. It relies on host membrane remodelling and subsequent activation of mechanosensitive ion cha

Influence of source and quantity of protein on the development of immunity and resistance to African trypanosomiasis.

Although it is well documented that severe protein deprivation inhibits the development of the immune response and exacerbates certain infections, little has been done to study the effects of native diets on endemic diseases or immunity. Therefore, protein-restricted diets were formulated for mice to mimic the sources and amounts measured in human diets of the Batouri region of Cameroon, endemic for African trypanosomiasis. Weanling C57BL/6 female mice were fed a diet that contained 73% of the recommended daily allowance (RDA) of protein. The sources of protein were all plant (cornmeal), all a

MeMLO12 Impairs Cassava Resistance to Xanthomonas axonopodis pv. manihotis by Repressing Reactive Oxygen Species, Callose Deposition, and Defense Hormone Pathways.

Cassava (Manihot esculenta Crantz) is a vital food and energy crop in tropical regions, yet its yield is significantly threatened by cassava bacterial blight, caused by Xanthomonas axonopodis pv. manihotis (Xpm). Mildew resistance locus O (MLO) genes are known negative regulators of plant immunity, but their roles in cassava disease susceptibility remain unclear. In this study, we identified the MeMLO12 gene from cassava and investigated its function in response to Xpm infection. Subcellular localization revealed that MeMLO12 is specifically localized in the nucleus. Functional analysis via tr…

An apoplastic peptide activates salicylic acid signalling in maize.

Localized control of cell death is crucial for the resistance of plants to pathogens. Papain-like cysteine proteases (PLCPs) regulate plant defence to drive cell death and protection against biotrophic pathogens. In maize (Zea mays), PLCPs are crucial in the orchestration of salicylic acid (SA)-dependent defence signalling. Despite this central role in immunity, it remains unknown how PLCPs are activated, and which downstream signals they induce to trigger plant immunity. Here, we discover an immune signalling peptide, Z. mays immune signalling peptide 1 (Zip1), which is produced after salicyl…

Bacterial Leaf Infiltration Assay for Fine Characterization of Plant Defense Responses using the Arabidopsis thaliana-Pseudomonas syringae Pathosystem.

In the absence of specialized mobile immune cells, plants utilize their localized programmed cell death and Systemic Acquired Resistance to defend themselves against pathogen attack. The contribution of a specific Arabidopsis gene to the overall plant immune response can be specifically and quantitatively assessed by assaying the pathogen growth within the infected tissue. For over three decades, the hemibiotrophic bacterium Pseudomonas syringae pv. maculicola ES4326 (Psm ES4326) has been widely applied as the model pathogen to investigate the molecular mechanisms underlying the Arabidopsis im
