Subcutaneous dirofilariosis in South-East Serbia--case report.

Human dirofilariosis is a relatively rare infection caused by filarial worms of the genus Dirofilaria. We herein report the first case of human subcutaneous dirofilariosis in the southeastern part of Serbia. A complete alive nematode was removed from a nodule in the periorbital region of a 50-year-old woman. The nematode was morphologically identified as a D. repens-like immature female. The diagnosis was confirmed with molecular methods. The patient was probably infected in the South-East Serbia as she had not travelled abroad, nor in other parts of Serbia such as Vojvodina, recently identifi

Human tapeworms in north Vietnam.

Sixty-five Taenia samples were collected from patients in a referral hospital in Hanoi, north Vietnam, for species identification by morphological and molecular techniques. PCR-RFLP of a mitochondrial 12S rDNA fragment, developed for this study, allowed direct differentiation between all Taenia spp., overcoming the disadvantages of classical morphological examination, which failed on disintegrated samples. Taenia saginata asiatica was the most common species (55.4%) followed by T. saginata (38.5%) and T. solium (6.2%). This report demonstrates the complexity of the epidemiology of Taenia spp.

Immunoprecipitation of in vitro translation products of Fasciola hepatica RNA.

Incubation of in vitro translation products of RNA isolated from Fasciola hepatica with immune rat serum and sera from infected sheep resulted in the immunoprecipitation of a number of polypeptides, most of which had similar electrophoretic mobilities. Although only one of these (a 30 kDa species) correlated precisely in mobility with that of biosynthetically labelled excretory-secretory (ES) antigens, this polypeptide (along with several others) was also immunoprecipitated by sheep sera raised against authentic ES antigens. This verified that the isolated RNA contained mRNA species encoding E

A single gene network accurately predicts phenotypic effects of gene perturbation in Caenorhabditis elegans.

The fundamental aim of genetics is to understand how an organism's phenotype is determined by its genotype, and implicit in this is predicting how changes in DNA sequence alter phenotypes. A single network covering all the genes of an organism might guide such predictions down to the level of individual cells and tissues. To validate this approach, we computationally generated a network covering most C. elegans genes and tested its predictive capacity. Connectivity within this network predicts essentiality, identifying this relationship as an evolutionarily conserved biological principle. Crit

Interleukin 12 and antigen independently induce substance P receptor expression in T cells in murine schistosomiasis mansoni.

Substance P (SP) regulates interferon-gamma (IFN-gamma) production through interaction with the SP receptor NK1 (SPr) on T cells at sites of inflammation. Using murine schistosomiasis, we evaluated whether SPr expression was subject to immunoregulation. Splenocytes from schistosome-infected mice cultured for < or =18 h did not express SPr, as determined by quantitative polymerase chain reaction assay. However, exposure to schistosome egg antigen (SEA) for < or =4 h induced strong receptor expression. Experiments using splenocytes fractionated with antibody-coupled, paramagnetic beads showed th

[Detection of Echinococcus granulosus and Echinococcus multilocularis in cyst samples using a novel single tube multiplex real-time polymerase chain reaction].

Cystic echinococcosis (CE) and alveolar echinococcosis (AE) caused by Echinococcus granulosus and Echinococcus multilocularis, respectively, are important helminthic diseases worldwide as well as in our country. Epidemiological studies conducted in Turkey showed that the prevalence of CE is 291-585/100.000. It has also been showed that the seroprevalence of AE is 3.5%. For the diagnosis of CE and AE, radiological (ultrasonography, computed tomography, magnetic resonance) and serological methods, in addition to clinical findings, are being used. The definitive diagnosis relies on pathological e

Issues of species identification within Paramphistomoidea Fischoeder, 1911 (Trematoda).

Cercariae of three Paramphistomoidea species were identified in the trematode fauna parasitizing the freshwater snails Gabbia fuchsiana from Vietnam and Helicorbis sujfunensis and Gyraulus centrifugops from the southern part of the Russian Far East. To elucidate their taxonomic status, the study included a morphological description accompanied by a molecular analysis using both nuclear and mitochondrial markers. An analysis of the evidence obtained showed that taxonomic identification of paramphistomes can only be possible on the basis of both morphological and molecular data, which are not al

DjCapon Affects the Neural Regeneration and Light Sensitivity via NO and Wnt5a Signaling in Planarian Dugesia japonica.

The carboxy-terminal postsynaptic density protein 95/discs large protein/zonula occludens 1 (PDZ) ligand of neuronal nitric oxide synthase (nNOS), known as CAPON, can interact with nNOS to influence neuropathological processes. However, the precise mechanism by which CAPON regulates neuronal regeneration remains poorly understood. In this study, we utilized the planarian Dugesia japonica, a model organism renowned for its remarkable regenerative abilities, to investigate this mechanism. First, we cloned the DjCapon gene and performed RNA interference (RNAi) to knock down DjCapon expression in

Mitochondrial genome, nuclear ribosomal RNA gene, and morphological-tegumental analysis of dermatitis-causing cercariae: Trichobilharzia sp. from Thailand.

Avian blood flukes of the genus Trichobilharzia, the largest subgroup of the Schistosomatidae family, are of medical and veterinary importance as causative agents of avian schistosomiasis and human cercarial dermatitis. Despite their significance, molecular data on Trichobilharzia species from Southeast Asia remain limited, and no mitochondrial genomes have been reported to date. This study presents the first complete mitochondrial genome and nuclear ribosomal RNA (rRNA) gene analysis of a Trichobilharzia sp. isolate (PYO) recovered from Radix (Lymnaea) rubiginosa in Phayao, Thailand, alongsid

Untangling the Derogenes varicus species complex in Scandinavian waters and the Arctic: description of Derogenes abba n. sp. (Trematoda, Derogenidae) from Hippoglossoides platessoides and new host records for D. varicus (Müller, 1784) sensu stricto.

Several studies have shown that the euryxenic trematode Derogenes varicus (Müller, 1784) represents a species complex. Four lineages have been designated (DV1-4) with the DV1 clade corresponding to D. varicus sensu stricto. Herein, we investigate newly collected specimens of D. varicus sensu lato from Scandinavian and Arctic waters using integrative taxonomy. The trematodes were collected from Melanogrammus aeglefinus, Eutrigla gurnardus, Trachinus draco, and Merluccius merluccius off the Atlantic coast of Sweden and from Hippoglossoides platessoides from Arctic Svalbard. 28S sequences of dero

Morphological and molecular identification of helminths of the greater bulldog bat Noctilio leporinus (Quiroptera: Noctilionidae) from Campeche, Mexico.

Surveys on parasites of bats from the Americas have been conducted, but information on helminths is still scarce, especially in the Neotropical region. In Mexico, there are species of bats that lack of a record for helminth species, such as members of the family Noctilionidae. The present study describes for the first time the helminths of Noctilio leporinus in Campeche, Mexico. In 2017, six specimens of N. leporinus were studied for helminths. The species identification of helminths was based on morphological studies and molecular analysis of fragments of the 28S rDNA. All bat specimens were

Ascarids exposed: a method for in vitro drug exposure and gene expression analysis of anthelmintic naïve Parascaris spp.

Ascarid parasites infect a variety of hosts and regular anthelmintic treatment is recommended for all species. Parascaris spp. is the only ascarid species with widespread anthelmintic resistance, which allows for the study of resistance mechanisms. The purpose of this study was to establish an in vitro drug exposure protocol for adult anthelmintic-naïve Parascaris spp. and report a preliminary transcriptomic analysis in response to drug exposure. Live worms were harvested from foal necropsies and maintained in RPMI-1640 at 37 °C. Serial dilutions of oxibendazole (OBZ) and ivermectin (IVM) were

Potential of Nucleic Acid Receptor Ligands to Improve Vaccination Efficacy against the Filarial Nematode Litomosoides sigmodontis.

More than two-hundred-million people are infected with filariae worldwide. However, there is no vaccine available that confers long-lasting protection against filarial infections. Previous studies indicated that vaccination with irradiated infective L3 larvae reduces the worm load. This present study investigated whether the additional activation of cytosolic nucleic acid receptors as an adjuvant improves the efficacy of vaccination with irradiated L3 larvae of the rodent filaria Litomosoides sigmodontis with the aim of identifying novel vaccination strategies for filarial infections. Subcutan

Primary Infection by E. multilocularis Induces Distinct Patterns of Cross Talk between Hepatic Natural Killer T Cells and Regulatory T Cells in Mice.

The larval stage of the helminthic cestode Echinococcus multilocularis can inflict tumor-like hepatic lesions that cause the parasitic disease alveolar echinococcosis in humans, with high mortality in untreated patients. Opportunistic properties of the disease have been established based on the increased incidence in immunocompromised patients and mouse models, indicating that an appropriate adaptive immune response is required for the control of the disease. However, cellular interactions and the kinetics of the local hepatic immune responses during the different stages of infection with E. m

Genomics and proteomics of the liver fluke Opisthorchis felineus.

The causative agent of opisthorchiasis, the liver fluke Opisthorchis felineus (Rivolta, 1884) is one of the helminths of humans and animals in Russia. Together with closely related species of trematodes O. viverrini (Poirier, 1886) and Clonorchis sinensis (Loos, 1907), O. felineus is a part of a triad of epidemiologically important trematodes in the family Opisthorchiidae. Adult O. felineus worms infest the hepatobiliary system of warm-blooded animals and might provoke the development of severe pathologies, including malignancy of bile duct epithelium. The high medical importance of O. felineu

Fimbriarioides intermedia and F. lintoni (Cestoda: Hymenolepididae): molecular phylogeny, species status, and transmission in Arctic coastal ecosystems.

Cestodes of the genus Fimbriarioides (Hymenolepididae, Fimbriariinae) parasitise waterfowl and shorebirds in the Holarctic. Using molecular and morphological methods, we studied two Fimbriarioides spp. from sea ducks of the European North of Russia: the type species F. intermedia from eiders Somateria mollissima and S. spectabilis 1758 and a closely related species F. lintoni from a previously unrecognised host, Velvet Scoter Melanitta fusca 1758. Our analysis also involved metacestodes Fimbriarioides sp. from barnacles Semibalanus balanoides of the Barents Sea and the Sea of Okhotsk. Metacest

Development and field evaluation of a multiplex qPCR assay for environmental DNA detection of Schistosoma mekongi and its intermediate snail host Neotricula aperta in the Mekong River Basin.

BACKGROUND: Schistosoma mekongi remains an important cause of intestinal schistosomiasis in the lower Mekong River Basin. Sensitive environmental surveillance tools are needed to support transmission monitoring, particularly where conventional snail surveys are seasonally constrained. This study developed and evaluated a multiplex quantitative PCR (qPCR) assay for simultaneous detection of S. mekongi and its intermediate snail host, Neotricula aperta, in aquatic environmental DNA (eDNA) samples. METHODS: Singleplex and multiplex qPCR assays targeting S. mekongi and N. aperta were compared to
