A workflow to reduce red blood cell autofluorescence for imaging IL-4/IL-4R interactions in the inflamed lung.

MOTIVATION: Autofluorescence is a common challenge in fluorescence microscopy of inflamed tissue, particularly where red blood cell (RBC) autofluorescence can mimic true fluorescence signals. This complicates image interpretation, reduces counting accuracy, and undermines reproducibility. Reliable methods are needed to distinguish true signals from autofluorescence, while preserving tissue architecture. In this study, we address this challenge by developing an optimized workflow that integrates antigen retrieval, selective fluorophore choice, and spectral separation. SUMMARY: Autofluorescence

Composite tissue transplantation in rats: fusion of donor muscle to the recipient site.

UNLABELLED: Little information currently exists on the repair of muscular tissue at the site of an amputation stump. This study examined the healing process of muscular tissue following composite limb transplantation using transgenic rat models. METHODS: Green fluorescent protein (GFP) transgenic rats were used to study the process of connection of donor muscle with the recipient. A DsRed2/GFP double-reporter transgenic rat and an NCre transgenic rat were used to study cell fusion. These rats have the unique characteristic of changing red fluorescence to green fluorescence by Cre/LoxP recombi

Mechanism of polarization of Listeria monocytogenes surface protein ActA.

The polar distribution of the ActA protein on the surface of the Gram-positive intracellular bacterial pathogen, Listeria monocytogenes, is required for bacterial actin-based motility and successful infection. ActA spans both the bacterial membrane and the peptidoglycan cell wall. We have directly examined the de novo ActA polarization process in vitro by using an ActA-RFP (red fluorescent protein) fusion. After induction of expression, ActA initially appeared at distinct sites along the sides of bacteria and was then redistributed over the entire cylindrical cell body through helical cell wal…

Human flexor tendon tissue engineering: in vivo effects of stem cell reseeding.

BACKGROUND: Tissue-engineered human flexor tendons may be an option to aid in reconstruction of complex upper extremity injuries with significant tendon loss. The authors hypothesize that human adipose-derived stem cells remain viable following reseeding on human tendon scaffolds in vivo and aid in graft integration. METHODS: Decellularized human flexor tendons harvested from fresh-frozen cadavers and reseeded with green fluorescent protein-labeled pooled human adipose-derived stem cells were examined with bioluminescent imaging and immunohistochemistry. Reseeded repaired tendons were compare

Recombinant Cell-Permeable Puromycin N-Acetyltransferase Confers Puromycin Resistance by Direct Protein Transduction.

Puromycin N-acetyltransferase (PAC) is an enzyme that catalyzes the acetylation of puromycin, an inhibitor of protein synthesis. The PAC gen … r delivery of TAT-PAC was confirmed using confocal microscopy and flow cytometry, employing an RFP (red fluorescent protein)-tagged TAT-PAC fusion protein. Treatment of HEK293 and SY5Y cells with TAT-PAC resulted in increased cell viability in the presence of puromycin, demonstrating its functionality as a selection marker. This study suggests the potential application o …

Improved method to track and precisely count Schwann cells post-transplantation in a peripheral nerve injury model.

BACKGROUND: To optimize survival evaluation of Schwann cells (SCs) in vivo, we tested fluorescent labeling of the nucleus as an improved method of tracking and counting the transplanted SCs at sciatic nerve injury sites in rodents. We also investigated if co-administering cells with the glial growth factor Neuregulin-1 β (NRG1β) improves in vivo survival. NEW METHOD: We transduced SCs using a Lentiviral vector with a nuclear localization signal (NLS) fused with mCherry and transplanted them in the sciatic nerve of rat post-crush injury (bilateral) either in the presence or absence of NRG1β in

Organization of multisynaptic inputs to the dorsal and ventral dentate gyrus: retrograde trans-synaptic tracing with rabies virus vector in the rat.

Behavioral, anatomical, and gene expression studies have shown functional dissociations between the dorsal and ventral hippocampus with regard to their involvement in spatial cognition, emotion, and stress. In this study we examined the difference of the multisynaptic inputs to the dorsal and ventral dentate gyrus (DG) in the rat by using retrograde trans-synaptic tracing of recombinant rabies virus vectors. Three days after the vectors were injected into the dorsal or ventral DG, monosynaptic neuronal labeling was present in the entorhinal cortex, medial septum, diagonal band, and supramammil

Bottom-Up Creation of Virus-like Particles via Post-Insertion of Protein-Lipid Conjugates.

Virus-like particles (VLPs) are widely used as noninfectious platforms for vaccines, drug delivery, and synthetic biology. Here we report a bottom-up approach to generate liposomal VLPs via post-insertion of protein-lipid conjugates into preformed liposomes. Proteins were covalently coupled to a benzylguanine (BG) lipid via the bioorthogonal and irreversible SNAP-tag/BG reaction, and the resulting protein-lipid conjugate enabled detergent-free insertion into liposomal lipid bilayers. Using a SNAP-tagged GFP conjugate, we first confirmed membrane association by liposome cosedimentation and dete

Far-Red Turn-On Fluorogenic Probe for Versatile Detection of Inorganic Polyphosphates.

Inorganic polyphosphate (polyP) is a ubiquitous phosphate biopolymer involved in diverse cellular processes. Despite its significance, selective detection of polyP remains challenging because of its simple and highly charged structure. Here, we report a far-red fluorogenic turn-on chemosensor for selective polyP detection and imaging, SiX-DPA-Zn. It integrates a silicon-xanthene fluorophore with a Zn(II)-coordinated 2,2'-dipicolylamine recognition unit. Replacing the bridging oxygen in the xanthene scaffold with silicon shifts fluorescence into the far-red region and confers up to 100-fold sel

Sensory innervation of the anal canal and anorectal line in Hirschsprung's disease: histological evidence from mouse models.

AIM: We used non-Hirschsprung's disease (HD) Sox10-Venus Transgenic mice (non-HDSV-mice), an endothelin receptor-B knockout mouse model of HD (HD-mice), and C57B6C3 wild controls (C-mice) to identify the correlation between the anorectal line (ARL) and successful transanal pull-through (TAPT). METHODS: In non-HDSV-mice, intestinal neural crest-derived cells can be visualized with Venus,-a green fluorescent protein-without histochemical staining. We exposed the anal canal in each non-HDSV-mouse and marked the ARL directly with red ink. Specimens of anus and rectum from HD- and C-mice were immu

Inhibitory effect of red ginseng acidic polysaccharide from Korean red ginseng on phagocytic activity and intracellular replication of Brucella abortus in RAW 264.7 cells.

Korean red ginseng (KRG) has long been used in traditional Korean and Oriental medicine. However, the anti-bacterial mechanism and therapeutic efficiency of KGR for intracellular Brucella infection are still unclear. In this study, the bactericidal activity of Korean red ginseng acidic polysaccharide (RGAP) on Brucella (B.) abortus and its cytotoxic effects on RAW 264.7 cells were evaluated. In addition, B. abortus internalization and intracellular replication in macrophages were investigated after RGAP treatment. RGAP-incubated cells displayed a marked reduction in the adherence, internalizat
