Ten-year cancer incidence in rescue/recovery workers and civilians exposed to the September 11, 2001 terrorist attacks on the World Trade Center.

BACKGROUND: Cancer incidence in exposed rescue/recovery workers (RRWs) and civilians (non-RRWs) was previously reported through 2008. METHODS: We studied occurrence of first primary cancer among World Trade Center Health Registry enrollees through 2011 using adjusted standardized incidence ratios (SIRs), and the WTC-exposure-cancer association, using Cox proportional hazards models. RESULTS: All-cancer SIR was 1.11 (95% confidence interval (CI) 1.03-1.20) in RRWs, and 1.08 (95% CI 1.02-1.15) in non-RRWs. Prostate cancer and skin melanoma were significantly elevated in both populations. Thyro

Effect of training on physiological, hematological, and chemical parameters in Belgian Malinois Dogs in Indonesia's tropical climate.

BACKGROUND: During exercise, dogs may experience heat stress and metabolic disorders, which can lead to physical health problems and impair their ability to work optimally. Furthermore, working at high ambient temperatures could result in blood chemistry parameter changes that differ from those at low ambient temperatures. It may even result in metabolic disorders in dogs. The biological responses of Belgian Malinois dogs to training in tropical areas with high temperatures and humidity, such as those found in Indonesia, have not been reported. AIM: This study aimed to investigate the biologi

Lumacaftor inhibits channel activity of rescued F508del cystic fibrosis transmembrane conductance regulator.

Lumacaftor, the corrector of Orkambi, enhances the processing of F508del cystic fibrosis transmembrane conductance regulator (CFTR), but its impact on the channel activity of rescued F508del CFTR (rF508del) is unclear. Using an electrode-based, real-time iodide efflux assay performed at room temperature, acute exposure to lumacaftor was shown to increase the processing of F508del CFTR without a proportional increase in channel activity in a CFBE41o-cell line stably expressing F508del CFTR (CFBE-DF). A similar effect was not observed on wild-type CFTR in a HEK293 cell line. At 37°C, rF508del ch

Retroperitoneal hand-assisted laparoscopic surgery for endoscopic adrenalectomy.

Although hand-assisted laparoscopic surgery (HALS) is very common in various laparoscopic procedures, it is rarely used for retroperitoneal endoscopic adrenalectomy because of the small working area. The authors evaluate HALS in endoscopic adrenalectomy with respect to its use as a rescue procedure in complicated cases. In their department, 47 patients underwent endoscopic adrenalectomies between 1998 and 2004. Mainly because of complicated anatomy, three primary aldosteronism cases were converted to retroperitoneal HALS. This involved making an additional 6 cm skin incision, into which the su

Canine tactical field care. Part two--Massive hemorrhage control and physiologic stabilization of the volume depleted, shock-affected, or heatstroke-affected canine.

Military and law enforcement agencies have seen a dramatic increase in the utilization of military working dogs (MWDs) and working canine officers, respectively both at home and in foreign deployments. Due to the fact that professional veterinary care is often distant from internal disaster or foreign deployment sites, the military medic, police tactical medic, or other first-response medical care providers may be charged with providing emergency or even basic, non-emergency veterinary care to working canines. The medical principles involved in treating canines are essentially the same as thos

Bone morphogenetic protein signaling pathway- Ethanol interactions disrupt palate formation independent of gata3.

Fetal Alcohol Spectrum Disorders (FASD) describes a wide array of neurological defects and craniofacial malformations, associated with ethanol teratogenicity. While there is growing evidence for a genetic component to FASD, little is known of the genes underlying these ethanol-induced defects. Along with timing and dosage, genetic predispositions may help explain the variability within FASD. From a screen for gene-ethanol interactions, we found that mutants for Bmp signaling components are ethanol-sensitive leading to defects in the zebrafish palate. Loss of Bmp signaling results in reductions

Ankle Sprains Risk Factors in a Sample of French Firefighters: A Preliminary Prospective Study.

CONTEXT: Firefighters participating in mandatory physical exercise sessions are exposed to a high risk of ankle sprain injury. Although both physiological and psychological risk factors have been identified, few prospective studies considered the complex interaction of these factors in firefighters. OBJECTIVE: To prospectively determine whether intrinsic physical risk factors and work-related environments predict ankle sprains occurring during on-duty physical exercise in firefighters during an 8-month follow-up period. DESIGN: Prospective. SETTING: Fire Department and Rescue Service. PART

Distinctive lipid signatures of bronchial epithelial cells associated with cystic fibrosis drugs, including Trikafta.

In recent years, a number of drugs have been approved for the treatment of cystic fibrosis (CF). Among them, newly released Trikafta, a combination of 3 drugs (VX-661/VX-445/VX-770), holds great promise to radically improve the quality of life for a large portion of patients with CF carrying 1 copy of F508del, the most frequent CF transmembrane conductance regulator (CFTR) mutation. Currently available disease-modifying CF drugs work by rescuing the function of the mutated CFTR anion channel. Recent research has shown that membrane lipids, and the cell lipidome in general, play a significant r

Ryanodine receptor antagonists adapt NPC1 proteostasis to ameliorate lipid storage in Niemann-Pick type C disease fibroblasts.

Niemann-Pick type C disease is a lysosomal storage disorder most often caused by loss-of-function mutations in the NPC1 gene. The encoded multipass transmembrane protein is required for cholesterol efflux from late endosomes and lysosomes. Numerous missense mutations in the NPC1 gene cause disease, including the prevalent I1061T mutation that leads to protein misfolding and degradation. Here, we sought to modulate the cellular proteostasis machinery to achieve functional recovery in primary patient fibroblasts. We demonstrate that targeting endoplasmic reticulum (ER) calcium levels using ryano

Bridging the Evidence Gap: A Call to Action for Anticoagulation Research in Pediatric Burned Patients on Extracorporeal Membrane Oxygenation.

Although early extracorporeal membrane oxygenation (ECMO) improves respiratory complications in unburned populations, it remains a rarely utilized rescue therapy in pediatric burn care, averaging just 3 to 6 cases annually worldwide despite a promising 53-62% survival rate. This underutilization stems from a profound therapeutic paradox: the high bleeding risks and need for coagulation inherent to burn wounds with necessary surgical debridement directly conflict with the systemic anticoagulation required to prevent ECMO circuit thrombosis. Importantly, emerging evidence demonstrates that syste

An Insectified Caco-2 Cell-Based Assay to Monitor Pesticide Transport Processes and Pharmacokinetics.

In vitro intestinal models such as Caco-2 cells are integral to pharmacokinetic research. However, insect intestinal cells have proven less tractable for pharmacokinetic studies limiting our knowledge of pesticide pharmacokinetics in these species. To overcome this, we have developed a Caco-2 cell line that has been genetically modified to express insect P-glycoprotein (Pgp), providing a robust 'insectified' screening platform to monitor xenobiotic transport. This platform consists of wild-type (Pgpwild type) cells, a Pgp knockout (PgpKO) line which eliminates endogenous background interferenc

Ribosomal frameshifting selectively modulates the assembly, function, and pharmacological rescue of a misfolded CFTR variant.

The cotranslational misfolding of the cystic fibrosis transmembrane conductance regulator chloride channel (CFTR) plays a central role in the molecular basis of CF. The misfolding of the most common CF variant (ΔF508) remodels both the translational regulation and quality control of CFTR. Nevertheless, it is unclear how the misassembly of the nascent polypeptide may directly influence the activity of the translation machinery. In this work, we identify a structural motif within the CFTR transcript that stimulates efficient -1 ribosomal frameshifting and triggers the premature termination of tr

mGlu5 positive allosteric modulation normalizes synaptic plasticity defects and motor phenotypes in a mouse model of Rett syndrome.

Rett syndrome (RS) is a neurodevelopmental disorder that shares many symptomatic and pathological commonalities with idiopathic autism. Alterations in protein synthesis-dependent synaptic plasticity (PSDSP) are a hallmark of a number of syndromic forms of autism; in the present work, we explore the consequences of disruption and rescue of PSDSP in a mouse model of RS. We report that expression of a key regulator of synaptic protein synthesis, the metabotropic glutamate receptor 5 (mGlu5) protein, is significantly reduced in both the brains of RS model mice and in the motor cortex of human RS a

PPARGC1A/PGC-1α, TFEB and enhanced proteostasis in Huntington disease: defining regulatory linkages between energy production and protein-organelle quality control.

Huntington disease (HD) results from CAG repeats that encode expanded polyglutamine tracts in the HTT/huntingtin protein. HD belongs to a large category of inherited and sporadic neurodegenerative disorders in which production of a misfolded protein initiates the pathogenic cascade. Previous studies have shown that misfolded proteins become resistant to cellular protein turnover pathways by eluding and disabling the ubiquitin-proteasome system (UPS) and autophagy-lysosome pathway. Based upon earlier work implicating impaired PPARGC1A function in HD, we derived inducible PPARGC1A mice and cross

Early anti-C5 therapy in NMOSD: rationale and clinical evidence.

Complement-mediated astrocyte injury is central to aquaporin-4 immunoglobulin G-positive neuromyelitis optica spectrum disorder (AQP4-IgG+ NMOSD). Although C5 inhibitors are approved for relapse prevention, their role as rescue therapy during severe acute attacks remains unclear. We describe a 39-year-old woman with AQP4-IgG+ NMOSD presenting with longitudinally extensive myelitis, optic neuropathy, and a basal ganglia lesion. Despite two courses of intravenous methylprednisolone, five plasma exchanges, and intravenous immunoglobulin, disability progressed to an Expanded Disability Status Scale

Rescuing cognitive deficits in alcohol-dependent mice by targeting physiological adaptations in prefrontal cortex K V 7 channels.

Chronic alcohol misuse causes cognitive deficits and functional adaptations in the prefrontal cortex (PFC) that facilitate excessive drinking and increase relapse probability during prolonged withdrawal. Positive K V 7 channel modulation reduces alcohol intake in high-drinking rodents, but the impact of alcohol dependence on cortical K V 7 physiology and its role in cognitive impairments are understudied. Here, we demonstrate spatial working memory deficits and physiological adaptations in intrinsic excitability and K V 7 channel function in intratelencephalic PFC projection neurons in alcohol
