Feeding after fourth ventricular administration of neuropeptide Y receptor agonists in rats.

Neuropeptide Y (NPY) and peptide YY (PYY) stimulate food intake after injection into the fourth cerebral ventricle, suggesting that NPY receptors in the hindbrain are targets for the stimulatory effect of these peptides on food intake. However, the NPY/PYY receptor subtype mediating the feeding response in the hindbrain is not known. To approach to this question we compared dose-effect of several NPY receptor agonists to stimulate food intake in freely-feeding rats 60- and 120-min after injection into the fourth cerebral ventricle. At the 120-min time point, PYY was 2- to 10-times as potent as

Alterations in cranial morphogenesis in the Lp mutant mouse.

The effects of exencephaly on cranial morphogenesis were studied at 10 to 12 days of gestation in the loop-tail (Lp) mutant mouse in which the hindbrain and spinal cord fail to close. At the level of the hindbrain, the otocysts became displaced ventrally in abnormal (Lp/Lp) embryos, and the everted neuroepithelium showed a diminished "luminal" reaction to the lectins WGA and Con A, as compared with normal embryos. Also, occasional clusters of rounded cells that resembled presumptive neural crest cells and that labeled with WGA, Con A, and anti-N-CAM were observed at the everted tips of the ope

Caudal hindbrain participation in the suppression of feeding by central and peripheral bombesin.

We have previously demonstrated that bombesin (BBS) infused into the fourth cerebral ventricle in rats suppressed feeding at doses 10-1,000 times less than those required after lateral ventricular administration. These results suggest that centrally administered BBS acts in the caudal hindbrain to suppress food intake. To investigate the participation of caudal hindbrain structures in fourth-ventricular BBS-induced suppression of feeding, we made lesions that destroyed the following: 1) area postrema (AP) and 2) and medial nucleus of the solitary tract (NST). Additionally, we examined the role

Segmental patterns of neuronal development in the chick hindbrain.

Identification of specific neuronal populations and their projections in the developing hindbrain reveals a segmental organization in which pairs of metameric epithelial units cooperate to generate the repeating sequence of cranial branchiomotor nerves. Neurogenesis also follows a two-segment repeat, suggesting parallels with insect pattern formation.

Contact Lens-Based Management of Neurotrophic Keratopathy in Rhombencephalosynapsis Without Alopecia (a Gómez-López-Hernández Syndrome Variant).

A child with rhombencephalosynapsis, part of the Gómez-López-Hernández spectrum, developed bilateral corneal hypoesthesia and neurotrophic keratopathy. Recurrent epithelial defects and progressive corneal haze occurred despite conservative therapy with lubrication, serum drops, and topical treatment. A persistent epithelial defect healed rapidly after application of a bandage contact lens. This case highlights the importance of early ophthalmologic surveillance and corneal sensitivity assessment in children with rhombencephalosynapsis. Bandage contact lenses are a safe and effective treatment

Kinematic Analysis of the Hindbrain via Dynamic Neck Motion in Adult Patients with Chiari Malformation Type I: A Radiological Study with Clinical Implications.

BACKGROUND: The influence of cervical motion on the hindbrain in patients with Chiari malformation type I (CMI) remains under investigation. In this study, the kinematic effects of cervical extension and flexion on cerebellar tonsillar herniation were evaluated. METHODS: The clinical data of 27 adult CMI patients were retrospectively studied, and sex- and age-matched patients with cervical spondylopathy were included as controls. Several linear, angular variables related to the hindbrain and mobility of the neck were assessed on midsagittal views of neutral and dynamic cervical magnetic reson

FGF19 in the Hindbrain Lowers Blood Glucose and Alters Excitability of Vagal Motor Neurons in Hyperglycemic Mice.

Fibroblast growth factor 19 (FGF19) is a protein hormone that produces antidiabetic effects when administered intracerebroventricularly in the forebrain. However, no studies have examined how FGF19 affects hindbrain neurons that participate directly in autonomic control of systemic glucose regulation. Within the dorsal hindbrain, parasympathetic motor neurons of the dorsal motor nucleus of the vagus (DMV) express fibroblast growth factor receptors and their activity regulates visceral homeostatic processes, including energy balance. This study tested the hypothesis that FGF19 acts in the hindb

Spatial structure and anti-fatigue of polysaccharide from Inonotus obliquus.

The aim of this study was to evaluate the spatial structure and potential antifatigue activity of polysaccharide fractions which was extracted from Inonotus obliquus. The first polysaccharide fractions of Inonotus obliquus (PIO-1) were obtained after hot-water extraction and purification by DEAE cellulose-52 chromatography. Results of the forced swimming test showed that the doses (50 mg/kg) of PIO-1 could increase the climbing duration and swimming time as well as reduced the immobility time in the PIO treated mice. The fatigue related metabolic parameters showed that PIO-1 decreased the leve

Cerebrospinal Fluid Flowmetry in Pediatric Patients with Chiari Malformation I with Surgical Implications.

BACKGROUND: Chiari malformations include a large spectrum of anomalies of hindbrain formation that appear at different stages of development of the central nervous system. Chiari described these malformations as congenital anomalies of the hindbrain characterized by downward elongation of the brain stem and cerebellum into the cervical portion of spinal cord. Much research regarding Chiari malformation I (CMI) has focused on the cerebrospinal fluid (CSF) dynamics rather than the anatomic relationships of the tonsils and brainstem. The aim of the present study was to measure the peak CSF veloci

Unsolved recognizable patterns of human malformation: Challenges and opportunities.

Due to the efforts of the clinical and scientific communities and boosted by recent advances in genetic technologies, we now understand the molecular mechanisms underlying most of the frequent and recognizable human malformation syndromes. However, some well-established human malformation syndromes remain without a molecular diagnosis despite intensive investigation. This issue of Seminars mines the phenotypic entries in OMIM and estimates that of the documented 2,034 unsolved entries likely to represent a rare genetic disease, only 160 are well-established and possibly amenable to investigati

Closed-loop neural stimulation for pentylenetetrazole-induced seizures in zebrafish.

Neural stimulation can reduce the frequency of seizures in persons with epilepsy, but rates of seizure-free outcome are low. Vagus nerve stimulation prevents seizures by continuously activating noradrenergic projections from the brainstem to the cortex. Cortical norepinephrine then increases GABAergic transmission and increases seizure threshold. Another approach, responsive nervous stimulation, prevents seizures by reactively shocking the seizure onset zone in precise synchrony with seizure onset. The electrical shocks abort seizures before they can spread and manifest clinically. The goal of

Rhombencephalosynapsis: association with single umbilical artery.

A 6-year-old girl who presented with developmental delay and non-progressive ataxia is described. MRI of brain showed agenesis of cerebellar vermis with fusion of cerebellar hemispheres and dentate nuclei. MRI findings were characteristic of rhombencephalosynapsis. Partial agenesis of corpus callosum and absent septum pellucidum were also seen. The child had also been noted to have a single umbilical artery at birth: a hitherto undescribed association.

Aetiology of herniation of the hindbrain in craniosynostosis. An investigation incorporating intracranial pressure monitoring and magnetic resonance imaging.

The occurrence and extent of herniation of the hindbrain has been evaluated in a population of children with craniosynostosis by means of magnetic resonance imaging of the craniocervical junction. The role of intracranial pressure (ICP), posterior fossa size and hydrocephalus in the development of this deformity has also been assessed. Magnetic resonance imaging (Siemens Magnetom 1.5T) was reviewed in 27 cases of craniosynostosis in whom there had been no previous cranial vault surgery. The position of the cerebellar tonsils in relation to the plane of the foramen magnum was measured and an in

Development of midbrain and anterior hindbrain ocular motoneurons in normal and Wnt-1 knockout mice.

The effect of homozygotic Wnt-1-/- mutations on the development of ocular motoneurons was examined with the lipophilic dye DiI and compared to control and phenotypic wild-type mouse embryos. A piece of DiI-soaked filter paper was inserted into the orbit, the midbrain, or rhombomere 5 of the hindbrain in six paraformaldehyde-fixed litters (10.5, 12.5, and 14.5 days postcoitum) containing Wnt-1, Wnt+/-, and Wnt-1+/+ individuals and three control litters. We labeled all ocular motoneurons retrogradely and all relevant nerves anterogradely in all control and phenotypic wild-type animals. In all ph

Hyperglycemia delays rostral initiation sites during neural tube closure.

Neural tube defects contribute greatly to perinatal loss, physical handicap, mental retardation and other developmental defects, yet the mechanisms through which they occur are poorly understood. One hindrance to the study of these defects at the cellular and molecular levels is the low frequency with which they arise in susceptible animals. The present study utilizes a culture system for the study of rodent exencephaly, an animal model of human anencephaly, in which a high frequency of affected animals are obtained by culture in hyperglycemic rat serum. Rat embryos were dissected at day 9.5 f

Gomez-López-Hernández syndrome: A case report with clinical and molecular evaluation and literature review.

Gomez-López-Hernández syndrome (GLHS) is characterized by rhombencephalosynapsis (RES), alopecia, trigeminal anesthesia and a distinctive phenotype, including brachyturricephaly. It has been suggested that GLHS should be considered as part of the spectrum of RES-associated conditions that include alopecia, trigeminal anesthesia, and craniofacial anomalies, rather than a distinct entity. To the best of our knowledge, 57 patients with GLHS have been described. Despite its first description in 1979, the etiology of this syndrome remains unknown. Here, we describe, to our knowledge, the first case
