The isolation of Central European encephalitis (tick-borne encephalitis) virus from Ixodes ricinus (L.) ticks in southern-Germany.

During the years 1972--1976 a total of 14.692 ticks of the species Ixodes ricinus (L.), (2.738 adults and 11.954 nymphs) was collected. They were taken from five areas where human Central European encephalitis virus (CEE) infections had occurred. By means of, intracerebral (IC) and subcutaneous (SC) isolation experiments with suckling mice 19 agents, exhibiting the pathogenic characteristics of CEE virus, were isolated from these ticks. In neutralization tests with suckling mice these isolates could be identified as CEE virus strains. From the 14.692 ticks at least 1.29% were virus carriers as…

Ixodes ricinus infesting snakes: Insights on a new tick-host association in a Borrelia burgdorferi sensu lato endemic area.

The castor bean tick Ixodes ricinus is one of the most abundant tick species in Europe, being able to parasitize a wide number of vertebrate hosts, including mammals, birds, and reptiles. This tick species has an important role as vector of zoonotic pathogens, including the causative agents of Lyme borreliosis (i.e. Borrelia burgdorferi sensu lato). Here, we provide insights on a new tick-host association (i.e. I. ricinus infesting snakes) in an area recently recognized as endemic for reptile-associated zoonotic species of Borrelia burgdorferi s.l.

Two ways of experimental infection of Ixodes ricinus ticks (Acari: Ixodidae) with spirochetes of Borrelia burgdorferi sensu lato complex.

A previously reported procedure for the introduction of Borrelia spirochetes into tick larvae by immersion in a suspension of spirochetes was tested on Ixodes ricinus (L.) ticks and three of the most medically important European Borrelia genomic species, B. burgdorferi sensu stricto, B. garinii and B. afzelii. The procedure was compared with "classical" infection of nymphs by feeding on infected mice. Both methods yielded comparable results (infection rate 44-65%) with the exception of B. afzelii, which produced better results using the immersion method (44%) compared with feeding on infected …

Rickettsia monacensis sp. nov., a spotted fever group Rickettsia, from ticks (Ixodes ricinus) collected in a European city park.

We describe the isolation and characterization of Rickettsia monacensis sp. nov. (type strain, IrR/Munich(T)) from an Ixodes ricinus tick collected in a city park, the English Garden in Munich, Germany. Rickettsiae were propagated in vitro with Ixodes scapularis cell line ISE6. BLAST analysis of the 16S rRNA, the citrate synthase, and the partial 190-kDa rickettsial outer membrane protein A (rOmpA) gene sequences demonstrated that the isolate was a spotted fever group (SFG) rickettsia closely related to several yet-to-be-cultivated rickettsiae associated with I. ricinus. Phylogenetic analysis …

Ticks on migrating birds in southwestern Poland: occurrence of Ixodes ricinus and the first Polish record of Haemaphysalis concinna on birds.

During ornithological research conducted at the Milicz Fishponds Nature Reserve (Barycz Valley Landscape Park, southwestern Poland) from May to July 2024, a total of 245 birds were captured. Four ticks were collected: two Ixodes ricinus larvae and two Haemaphysalis concinna nymphs. We present the first confirmed record of H. concinna parasitising a great reed warbler (Acrocephalus arundinaceus) in Poland. Identification of H. concinna was based on both morphological identification keys and molecular analysis of the COI gene. This finding expands current knowledge on the biodiversity of ticks p…

Differential Impact of Simultaneous or Sequential Coinfections With Borrelia afzelii and Tick-Borne Encephalitis Virus on the Ixodes ricinus Microbiota.

Ticks, particularly Ixodes ricinus, are significant vectors of pathogens such as Borrelia spp. and tick-borne encephalitis virus (TBEV), which cause Lyme borreliosis (LB) and tick-borne encephalitis (TBE), respectively. Understanding how these pathogens interact within the tick microbiome is essential for developing vector control strategies. This study investigates the impact of Borrelia afzelii and TBEV, as well as their coinfection, on the microbiota composition and structure of I. ricinus nymphs. Using a network-based approach, we analyzed the microbial communities of ticks exposed to infe…

Effects of Live and Peptide-Based Antimicrobiota Vaccines on Ixodes ricinus Fitness, Microbiota, and Acquisition of Tick-Borne Pathogens.

This study explored the effects of antimicrobiota vaccines on the acquisition of Borrelia and Rickettsia, and on the microbiota composition of Ixodes ricinus ticks. Using a murine model, we investigated the immunological responses to live Staphylococcus epidermidis and multi-antigenic peptide (MAP) vaccines. Immunized mice were infected with either Borrelia afzelii or Rickettsia helvetica, and subsequently infested with pathogen-free I. ricinus nymphs. We monitored the tick feeding behavior, survival rates, and infection levels. Additionally, we employed comprehensive microbiota analyses, incl…

Prevalence of Babesia canis DNA in Ixodes ricinus ticks collected in forest and urban ecosystems in west-central Poland.

Babesia canis, a widely distributed European tick-borne protozoan haemoparasite, causes canine babesiosis, the most important tick-borne disease afflicting dogs worldwide. The meadow tick, Dermacentor reticulatus, is considered to be the primary vector of this parasite in central Europe. Females of the more broadly distributed and medically important castor bean tick, Ixodes ricinus, also commonly feed upon dogs, but their role in the enzootic transmission cycle of B. canis is unclear. Here, we screened 1,598 host-seeking I. ricinus ticks collected from two different ecosystems, forest stands …

Infection with Borrelia afzelii and manipulation of the egg surface microbiota have no effect on the fitness of immature Ixodes ricinus ticks.

Arthropod vectors carry vector-borne pathogens that cause infectious disease in vertebrate hosts, and arthropod-associated microbiota, which consists of non-pathogenic microorganisms. Vector-borne pathogens and the microbiota can both influence the fitness of their arthropod vectors, and hence the epidemiology of vector-borne diseases. The bacterium Borrelia afzelii, which causes Lyme borreliosis in Europe, is transmitted among vertebrate reservoir hosts by Ixodes ricinus ticks, which also harbour a diverse microbiota of non-pathogenic bacteria. The purpose of this controlled study was to test…

Clinical/serological outcome in humans bitten by Babesia species positive Ixodes ricinus ticks in Sweden and on the Åland Islands.

The risk of contracting babesiosis after a tick bite in Sweden and on the Åland Islands, Finland, is unknown. We investigated clinical and serological outcomes in people bitten by Ixodes ricinus ticks positive for Babesia species. Ticks, blood and questionnaires were obtained from study participants in Sweden and on the Åland Islands. Sixty-five of 2098 (3.1 %) ticks were positive by real-time PCR. Three Babesia species were detected, Babesia microti (n = 33), B. venatorum (n = 27) and B. capreoli (n = 5), the latter species not known to cause human infection. Half (46 %) of the Babesia PCR-po…

Seasonal cycles of the TBE and Lyme borreliosis vector Ixodes ricinus modelled with time-lagged and interval-averaged predictors.

Ticks of the species Ixodes ricinus (L.) are the major vectors for tick-borne diseases in Europe. The aim of this study was to quantify the influence of environmental variables on the seasonal cycle of questing I. ricinus. Therefore, an 8-year time series of nymphal I. ricinus flagged at monthly intervals in Haselmühl (Germany) was compiled. For the first time, cross correlation maps were applied to identify optimal associations between observed nymphal I. ricinus densities and time-lagged as well as temporal averaged explanatory variables. To prove the explanatory power of these associations,…
