Tenascin-XB plays a role in the infiltration of immune cells in tumor microenvironment.

We previously showed that tenascin-XB (TNXB) contributes to tumor suppressor function. The present study aimed to assess the tumor-suppressive mechanism of TNXB by focusing on immune cell infiltration into the tumor microenvironment (TME). We revealed that B16-OVA melanoma cells (MO5)-bearing TNXB-deficient (Tnxb-/-) mice exhibited significant tumor progression and a poor survival rate. Allogeneic mixed lymphocyte reaction showed reduced numbers and increased activation of both CD4+ and CD8+ T cells from Tnxb-/- spleens. Moreover, T cell activation assay further proved that CD4+ and CD8+ T cel

Gut virome plays an extended role with bacteriome in neurological health and disease.

The gut-brain axis (GBA) is a complex two-way communication system that links the gastrointestinal tract and the central nervous system (CNS) through neural, immune, hormonal, and microbial pathways. The microbiota-gut-brain axis (MGBA), a more specific concept, focuses on how gut microorganisms, including bacteria, viruses, and other microbes, modulate this communication and influence neurological health. This comprehensive review examines the intricate mechanisms through which gut microorganisms modulate neural function and contribute to neurological health and disease pathogenesis. The gut

StarD5 Plays a Critical Role in the Hepatocyte ER Stress Survival Response.

The unfolded protein response (UPR) is a highly orchestrated survival response initiated in cells under endoplasmic reticulum (ER) stress. Steroidogenic acute regulatory-related lipid transfer domain 5 (StarD5) is an ER stress-responsive, cholesterol-binding protein under the regulation of IRE1. Based upon in vitro findings, StarD5 delivers a protective response by translocating ER cholesterol to the plasma membrane (PM) and accompanying protective changes in PM fluidity. The study aimed to determine if StarD5's ability to provide in vitro hepatocyte protective responses is translatable to in

Monocytes Expressing IL-36G Play a Crucial Role in Atopic Dermatitis.

Atopic dermatitis (ad) is a chronic inflammatory skin disease, with recent studies indicating that immune cells, such as monocytes and inflammatory cytokines, play a crucial role. By retrieving datasets from public databases and analysing immune cell infiltration in lesional skin using CIBERSORT, we found that monocytes and M2 macrophages were significantly upregulated in atopic dermatitis. Differentially expressed gene (DEG) functional enrichment analysis revealed that cytokine-cytokine receptor interaction was the most significantly enriched pathway. Further analysis of cytokines and their r

Metabolic Activation of trans-Anethole Plays a Role in Hepatic Cytotoxicity in Mice.

trans-Anethole (1-methoxy-4-[1(E)-propenyl]benzene, tAT) is the main ingredient in the essential oil extracted from star anise fruits. The double bonds in the side chain of tAT are a type of alert structure that can be metabolized into epoxides possibly causing liver damage. This work investigated and identified the reactive metabolites of tAT that are chemically reactive to biothiols, such glutathione (GSH), N-acetyl-l-cysteine (NAC), and cysteine residues of proteins. Four GSH conjugates (M2-M5) and two NAC conjugates (M6 and M7), which are derived from epoxide metabolite of tAT, were found

Commensal bacteria play a fundamental role in maintaining gut immune homeostasis.

The intestinal microbiome, which is a key factor in the maintenance of host gut homeostasis, enhances intestinal mucosal barrier function and immune tolerance (Rooks and Garrett, 2016; Skelly et al., 2019). However, the specific immunomodulatory functions of microbiota-derived metabolites in mucosal inflammatory responses remain largely unknown. The effects of microbial metabolites may vary across different immune cell types and host homeostasis (Hu et al., 2023; Zhao et al., 2023). Hence, it is fundamental to understand how specific intestinal microbes and their metabolic small molecules caus

What role do miRNAs play in the injured heart of the infertile female?

Due to numerous overlapping risk factors between cardiovascular diseases (CVDs) and infertility, women of reproductive age with underlying CVD may increasingly seek reproductive assistance. miRNAs and exosomal miRNAs are well-known epigenetic modulators that regulate gene expression at the post-transcriptional level. A large body of evidence indicates the fundamental roles of various miRNAs in different aspects of both CVD and infertility. Dysregulation of miRNAs (including those carried by exosomes) could contribute to pathological alterations involved in CVD pathogenesis, potentially leading

Glutathione reductase plays a role in the metabolism of methylmercury degradation in Rhodotorula mucilaginosa.

UNLABELLED: Mercury pollution is a kind of heavy metal pollution with great harm and strong toxicity which exists worldwide. Some microorganisms can convert highly toxic methylmercury into inorganic mercury compounds with significantly reduced toxicity. This is an effective means of methylmercury pollution remediation. As a safe microorganism with great potential in the remediation of heavy metal pollution, Rhodotorula mucilaginosa has not been studied in the remediation of methylmercury. Here, a R. mucilaginosa strain Rm4 with high methylmercury resistance was obtained by wild-strain screenin

Mitochondrial Dysfunction Plays a Relevant Role in Heart Toxicity Caused by MeHg.

The effects of methylmercury (MeHg) on exposed populations are a public health problem. In contrast to widely studied neurological damage, few cardiovascular changes have been described. Our group evaluated the cardiotoxicity of a cumulative dose of 70 mg.kg-1 fractioned over a 14-day exposure period in mice (MeHg70 group). The effects of MeHg on proteins relevant to cardiac mitochondrial function were also investigated. The results obtained showed a reduction in oxygen consumption in the two settings. In cardiac tissue samples in oxygraphy studies, this reduction was related to a lower effici

Does the kynurenine pathway play a pathogenic role in autism spectrum disorder?

Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by deficits in communication, sociability, and repetitive/stereotyped behavior. The etiology of autism is diverse, with genetic susceptibility playing an important role alongside environmental insults and conditions. Human and preclinical studies have shown that ASD is commonly accompanied by inflammation, and inhibition of the inflammatory response can ameliorate, or prevent the phenotype in preclinical studies. The kynurenine pathway, responsible for tryptophan metabolism, is upregulated by inflammation. Hence, thi

Mesenchymal stem cell transplantation plays a role in relieving cancer pain.

Tumors can invade, compress, and damage nerves, leading to persistent pain and seriously affecting the quality of life of patients. However, their treatment is challenging. Sensitization of peripheral receptors, abnormal activity of primary sensory neurons, activation of glial cells, enhanced inflammatory responses, and sensory information transmission contribute towards cancer pain. Therefore, considerable attention has been paid to exploring prospective methods to inhibit the occurrence of these factors and relieve cancer pain. Studies on different types of pains have revealed that the trans

Angiogenesis-associated pathways play critical roles in neonatal sepsis outcomes.

Neonatal sepsis is a major cause of childhood mortality. Limited diagnostic tools and mechanistic insights have hampered our abilities to develop prophylactic or therapeutic interventions. Biomarkers in human neonatal sepsis have been repeatedly identified as associated with dysregulation of angiopoietin signaling and altered arachidonic acid metabolism. We here provide the mechanistic evidence in support of the relevance for these observations. Angiopoetin-1 (Ang-1), which promotes vascular integrity, was decreased in blood plasma of human and murine septic newborns. In preclinical models, ad

Vision plays a calibrating role in discriminating threat-related vocal emotions.

The ability to reliably discriminate vocal expressions of emotion is crucial to engage in successful social interactions. This process is arguably more crucial for blind individuals, since they cannot extract social information from faces and bodies, and therefore chiefly rely on voices to infer the emotional state of their interlocutors. Blind have demonstrated superior abilities in several aspects of auditory perception, but research on their ability to discriminate vocal features is still scarce and has provided unclear results. Here, we used a gating psychophysical paradigm to test whether

Could emulated trials play a key role in cardiogenic shock trials?

Temporary mechanical circulatory support (tMCS) using extracorporeal life support (ECLS), has been widely implemented in patients with cardiogenic shock (CS), although evidence regarding its efficacy and safety remains unclear. This lack of clarity has recently raised concerns about the role of tMCS in CS management. Conducting randomized controlled trials (RCTs) in the context of CS poses significant challenges due to ethical considerations and logistical complexities. In response to these challenges, emulated trials (ETs) are emerging as a promising alternative. By incorporating design featu

Gut microbiota in neurological diseases: Melatonin plays an important regulatory role.

Melatonin is a highly conserved molecule produced in the human pineal gland as a hormone. It is known for its essential biological effects, such as antioxidant activity, circadian rhythm regulator, and immunomodulatory effects. The gut is one of the primary known sources of melatonin. The gut microbiota helps produce melatonin from tryptophan, and melatonin has been shown to have a beneficial effect on gut barrier function and microbial population. Dysbiosis of the intestinal microbiota is associated with bacterial imbalance and decreased beneficial microbial metabolites, including melatonin.

TB and HIV induced immunosenescence: where do vaccines play a role?

This paper tackles the complex interplay between Human Immunodeficiency virus (HIV-1) and Mycobacterium tuberculosis (M. tuberculosis) infections, particularly their contribution to immunosenescence, the age-related decline in immune function. Using the current literature, we discuss the immunological mechanisms behind TB and HIV-induced immunosenescence and critically evaluate the BCG (Bacillus Calmette-Guérin) vaccine's role. Both HIV-1 and M. tuberculosis demonstrably accelerate immunosenescence: M. tuberculosis through DNA modification and heightened inflammation, and HIV-1 through chronic
