Leukaemia inhibitory factor mRNA is expressed in the brains of patients with subacute sclerosing panencephalitis.

We have examined the in situ transcription of leukaemia inhibitory factor (LIF) in brain tissue from 3 cases of subacute sclerosing panencephalitis (SSPE) and in 2 non-neurological control brains. This has been compared with expression of interleukin 2 (IL-2), interleukin 6 (IL-6) and tumour necrosis factor beta (TNF beta) in the same tissues. All of the cytokines in the study were expressed in cells in the inflammatory infiltrate as well as in glial cells. LIF mRNA was also found to be expressed in neurons, in foci where these cells were also virally infected. No hybridization was found with

An initial MRI picture of limbic encephalitis in subacute sclerosing panencephalitis.

BACKGROUND: Subacute sclerosing panencephalitis (SSPE) is a rare and severe long-term complication of measles. Hallmarks of this entity include progressive cognitive decline, myoclonia, a generalized periodic pattern on EEG and deep white matter abnormalities on MRI. However, imaging can be normal in early stages. AIM: We report herein the case of a previously healthy 13-years-old girl with an unusual radiological presentation. RESULTS: She presented with unilateral myoclonia, cognitive decline with memory impairment and a first brain MRI with swelling of both hippocampi mimicking limbic enc

Detection of interleukin 10 in cerebrospinal fluid of patients with subacute sclerosing panencephalitis.

Subacute sclerosing panencephalitis (SSPE) is a neurodegenerative disorder due to persistent measles virus infection, with high level of measles-specific antibodies in cerebrospinal fluid (CSF). To analyze whether such response arises from a TH2-biased response, the authors determined TH1 (interferon [IFN]-gamma) and TH2 (interleukin [IL]-4 and IL-10) cytokines in CSF, taken at diagnosis, of eight SSPE patients (median age, 57.5 month, range 42 to 76 months). All patients presented IL-10 (median 29.3 pg/ml, range 4.3 to 162 pg/ml), but not IL-4 (<10 pg/ml); only one case showed IFN-gamma (162

Antibodies to viral and non-viral antigens in subacute sclerosing panencephalitis and multiple sclerosis demonstrated by thin-layer polyacrylamide gel isoelectric focusing, antigen immunofixation and autoradiography.

Antibody activity in IgG zones separated by thin-layer polyacrylamide gel isoelectric focusing (PAG IEF) was determined in 3 patients with subacute sclerosing panencephalitis (SSPE), 4 patients with multiple sclerosis (MS) and 4 subjects with psychosomatic disorders, using antigen immunofixation and autoradiography. Viral (measles herpes simplex type 1, mumps) and non-viral (purified bovine myelin, bovine myelin basic protein, bovine oligodendrocytes, MS and normal human brain extract) were used as antigens. All oligoclonal and some of the polyclonal CSF IgG zones in the patients with SSPE con

[Changes in the fundus in subacute sclerosing panencephalitis. Apropos of 23 cases].

Subacute sclerosing panencephalitis (SSPE) is a fatal slow encephalitis. It is related to a chronic infection of the central nervous system by a defective measles virus. Among 41 patients with SSPE, 23 had ocular involvement. The ophthalmologic involvement preceded the onset of neurological signs in 4 cases, they were detected at first presentation in 9 cases, and they occurred in the course of the disease in 10 cases. The ocular involvement was bilateral in 19 cases. Various ophthalmologic features were reported: retinitis (16 eyes), macular pigment disturbances (4 eyes), optical neuritis (33

Regional Cerebral Blood Flow Patterns on ASL in Subacute Sclerosing Panencephalitis: Quantitative Analysis and Clinical Correlation.

BACKGROUND AND PURPOSE: Subacute sclerosing panencephalitis (SSPE) is a progressive, uniformly fatal neuroinflammatory disease caused by persistent measles virus encephalitis, characterised by chronic T-cell-mediated inflammation, progressive demyelination, and cortico-striatal-thalamic network dysfunction. Conventional MRI characterises established structural disease but does not capture early haemodynamic alterations. We aimed to characterise regional perfusion patterns using cerebellar-normalised arterial spin labeling (ASL) ratios, define qualitative perfusion phenotypes, and examine their

Pan-Immune Inflammation Value and Clinical Outcomes in Subacute Sclerosing Panencephalitis: A Retrospective Study.

Subacute sclerosing panencephalitis (SSPE) is a rare, progressive, and fatal neurological disorder caused by persistent measles virus infection. Reliable prognostic biomarkers remain limited. Systemic inflammation has been implicated in the pathogenesis of neuroinfectious diseases, and hematology-derived indices are increasingly recognized as accessible markers of inflammatory burden. This retrospective case-control study was conducted at İnönü University Faculty of Medicine, Malatya, Türkiye, between 2010 and 2025, including 40 pediatric patients with SSPE and 40 age- and sex-matched healthy

Synthesis of measles virus-specific IgM antibodies and IgM-class rheumatoid factor in relation to clinical onset of subacute sclerosing panencephalitis.

Thirty-seven serum and 37 cerebrospinal fluid (CSF) specimens from 27 patients with subacute sclerosing panencephalitis (SSPE) were tested for measles virus (MV) IgM antibodies and IgM-class rheumatoid factor (RF) to determine if a temporal relationship exists between SSPE onset and these immunoglobulins. Sensitive solid-phase radioimmunoassays were used for immunoglobulin detection. Five of six MV IgM-positive CSF, both MV IgM-positive sera and six out of eight sera with elevated RF levels were collected within 1 yr of SSPE onset. Also collected during this time were six other sera having hig

Structural defect linked to nonrandom mutations in the matrix gene of biken strain subacute sclerosing panencephalitis virus defined by cDNA cloning and expression of chimeric genes.

Biken strain, a nonproductive measles viruslike agent isolated from a subacute sclerosing panencephalitis (SSPE) patient, contains a posttra … ding and noncoding regions of the Biken M gene were identical to those in an independently isolated SSPE virus strain with a similar defect. These results offer insights into the basis of the defect in Biken strain and pose intriguing questions about the evolutionary origins of SSPE viruses in general.

Serological identification of viral antigens after electrophoretic transfer.

The method described permits detection of specific antibodies to antigenic material, the protein components of which can first be separated by electrophoresis and then transferred to a supporting medium where their reactivity with antibody may be demonstrated using peroxidase-labelled anti-Ig. Results are described for measles virus and measles-like virus isolated from subacute sclerosing panencephalitis (SSPE) brain. Dilutions, incubation time and conditions, detection with peroxidase-labelled immunoglobulins and selection of substrate are described and discussed.

Comparison of lytic and persistent measles virus matrix proteins by competition radioimmunoassay.

Measles virus matrix (M) proteins were compared by competitive monoclonal antibody-binding studies. Three strains of measles and of subacute sclerosing panencephalitis viruses were found to be identical in this way. The matrix protein formed by Edmonston strain virus during a persistent infection could be distinguished from that made in the lytic virus infection. It is concluded that structural alterations in the M peptide can occur during persistence.

Mutant fusion proteins with enhanced fusion activity promote measles virus spread in human neuronal cells and brains of suckling hamsters.

Subacute sclerosing panencephalitis (SSPE) is a fatal degenerative disease caused by persistent measles virus (MV) infection in the central nervous system (CNS). From the genetic study of MV isolates obtained from SSPE patients, it is thought that defects of the matrix (M) protein play a crucial role in MV pathogenicity in the CNS. In this study, we report several notable mutations in the extracellular domain of the MV fusion (F) protein, including those found in multiple SSPE strains. The F proteins with these mutations induced syncytium formation in cells lacking SLAM and nectin 4 (receptors

The role of the nucleus in the replication of measles virus.

The replication of measles and SSPE viruses in enucleate BSC-1 cells was measured by plaque assay, and the synthesis of virus-specific antigen was measured by indirect immunofluorescence. Titres of infectious virus produced in enucleate cells at 24 hours post inoculation (p.i.) were consistently 100-fold less, and the number of enucleate cells containing virus antigen at 30 hours p.i. was 10-fold less, than in nucleate control cultures. Enucleate cells at this time had 30% of the protein synthetic capacity of control nucleate cells, and supported the growth of Semliki Forest virus to titres eq…

Pathogenic aspects of measles virus infections.

Measles virus (MV) infections normally cause an acute self limiting disease which is resumed by a virus-specific immune response and leads to the establishment of a lifelong immunity. Complications associated with acute measles can, on rare occasions, involve the central nervous system (CNS). These are postinfectious measles encephalitis which develops soon after infection, and, months to years after the acute disease, measles inclusion body encephalitis (MIBE) and subacute sclerosing panencephalitis (SSPE) which are based on a persistent MV infection of brain cells. Before the advent of HIV,

Subacute sclerosing panencephalitis (SSPE): a report of 16 cases.

Sixteen cases of subacute sclerosing panencephalitis (SSPE) were diagnosed at the Royal Children's and Mater Misericordiae Children's Hospitals in Brisbane from September 1974 to June 1983. Although the incidence of the disease with regard to age at onset, previous exposure to measles virus, and longevity was similar to that reported in the United States and Europe, the frequency of the illness was substantially higher in the present study. The high frequency of the disease in Queensland appears to reflect a relatively low level of immunity to measles virus in the population at risk.

Subacute measles encephalitis with AIDS diagnosed by fine needle aspiration biopsy. A case report.

BACKGROUND: Subacute measles encephalitis (SME), or measles inclusion body encephalitis (MIBE), is a rare central nervous system (CNS) complication following infection by the measles (rubeola) virus. It usually occurs in young patients with defective cellular immunity from either congenital or acquired causes. In the United States, with the resurgence of measles infection and the human immunodeficiency virus (HIV) pandemic, subacute measles encephalitis should be added to the differential diagnosis in acquired immune deficiency syndrome (AIDS) patients presenting with unexplained CNS manifesta

Subacute sclerosing panencephalitis (SSPE) agent in hamsters.

A hamster-adapted SSPE agent was shown to cause a productive infection in weanling hamster brain, which changed to a cell-associated or defective infection coincident with the appearance of measles antibodies in serum. Antibodies to measles hemagglutinin, hemolysin and neucleocapsid antigens developed in serum, which also contained neutralizing activity for regular measles virus. The agent recovered from the brains prior to the appearance of serum antibodies was infectious in cell-free media, capable of rapidly destroying Vero-cell cultures and able to progressively destroy primary hamster bra
