A novel class of Saccharomyces cerevisiae mutants specifically UV-sensitive to "petite" induction.

A mutant of Saccharomyces cerevisiae has been isolated which, though exhibiting a normal response to nuclear genetic damage by ultraviolet light (UV), is more sensitive than its wild type specifically in the production of the cytoplasmic (rho-) mutation by this agent. Some of the features of this mutation which has been designated uvsrho 5 are: i) The mutation is recessive, it exhibits a Mendelian, and hence presumably nuclear, pattern of segregation, but manifests its effects specifically and pleiotropically on mitochondrial functions. ii) Mutant cells resemble their wild type parents in a) g…

Biocontrol and bioprotection potential of selected non-Saccharomyces yeasts in grape and wine production.

In recent years, a key objective of the wine industry is the reduction of fungicide application in vineyard and sulphur dioxide (SO₂) during wine production, thereby encouraging alternative preservation methods. To address this objective, four non-Saccharomyces yeast species were evaluated in two independent experiments for their potential use as biocontrol and bioprotective agents. First, the selected yeasts were sprayed on Vitis vinifera L. cv. Nebbiolo grapes and evaluated as potential biocontrol agents against post-harvest grey mould caused by Botrytis cinerea. In parallel, the same strain…

An integrated cell and medium engineering approach for production of a nanobody fusion in Saccharomyces cerevisiae.

Saccharomyces cerevisiae is an established production host for therapeutic proteins; many of those are small proteins such as insulin or glucagon-like peptide-1 (GLP-1) analogs. Contrastingly, proteins of higher molecular weight, foremost antibodies, did not reach the market due, among other factors, to limiting productivity. Here we addressed the loss of product to protein degradation through a combination of genetic engineering of the host and medium optimization. We screened target genes that either directly or indirectly can lead to proteolytic degradation. We identified four deletions tha…

Rtt107 cooperates with Rad55 to limit specific types of genome instability in Saccharomyces cerevisiae.

A range of genome maintenance factors respond to endogenous and exogenous DNA damage to prevent mutations and cell death. The scaffold protein, Rtt107, is important for the growth of cells exposed to DNA-damaging agents in the budding yeast Saccharomyces cerevisiae. Rtt107 binds to a diverse array of partner proteins and responds to DNA damage by localizing to phosphorylated histone H2A. Rad55-Rad57, a heterodimer involved in DNA repair, also binds to Rtt107, but the function of the Rtt107-Rad55-Rad57 complex remains unclear. In addition to their sensitivity to DNA-damaging agents, rtt107∆ mut…

Sti1 participates in the dynamics of protein aggregation triggered by glucose signaling in Saccharomyces cerevisiae.

Environmental changes put stress on living organisms. We find that nutrient starvation induces dynamic protein aggregations in yeast cells, and many chaperones are involved in this process. Among them, Sti1/HOP, the co-chaperone of Hsp70 and Hsp90, plays roles in the formation of protein quality control (PQC) compartments and protein stasis (or proteostasis) maintenance, and it co-localizes to insoluble protein deposits (IPOD) by liquid-liquid phase separation (LLPS). Notably, the subcellular localization and cytoplasmic aggregation of Sti1 are rigorously regulated by the PQC machinery, includ

The effect of spontaneous mutations on CUP1 copy number and copper tolerance in Saccharomyces cerevisiae.

Genomic regions containing tandem duplications may be subject to particularly high rates of copy number mutations due to recombinational repair of DNA damage. Consequently, changes in copy number may be among the most accessible beneficial mutations during evolution in a new environment. In the budding yeast Saccharomyces cerevisiae, the gene CUP1 occurs in multiple tandem copies and encodes a metallothionein relevant for copper homeostasis. We examined CUP1 copy number in 220 mutation accumulation lines and their ancestors to quantify spontaneous genetic change at this locus. We also measured…

Moderate expression and activity of flocculins underlie the characteristic flocculation phenotype of Saccharomyces pastorianus.

Flocculation is a key technological trait in lager brewing, governing fermentation performance, yeast recovery, and beer quality. In the allo-aneuploid hybrid yeast Saccharomyces pastorianus, the genetic basis of flocculation remains poorly resolved due to its complex dual sub-genome architecture. Here, we systematically re-annotated and functionally characterized the complete FLO gene repertoire of the Group II strain CBS 1483. Thirteen FLO genes were identified, including allelic variants and a previously uncharacterized adhesin, Flo12, containing a Hyphal_reg_CWP domain instead of the canon…

Specific impact of nitrogen supplementation on the metabolism of Saccharomyces and hybrids during wine fermentation.

Nitrogen is essential for yeast metabolism and is often added to avoid slow or stuck fermentations in the wine industry. This study aimed to investigate the impact of different nitrogen supplementation strategies on the metabolism of various Saccharomyces yeast strains, with a particular focus on their fermentative performance, primary metabolite production, and aromatic profiles, including polyfunctional mercaptans (PFMs) varietal aromas. Two nitrogen supplements, organic (amino acids) and inorganic (NH4Cl), were evaluated under conditions and addition limits representative of the oenological…

Optogenetic Control the Activity of Pyruvate Decarboxylase in Saccharomyces cerevisiae for Tunable Ethanol Production.

Saccharomyces cerevisiae is a widely used chassis in metabolic engineering. Due to the Crabtree effect, it preferentially produces ethanol under high-glucose conditions, limiting the synthesis of other valuable metabolites. Conventional metabolic engineering approaches typically rely on irreversible genetic modifications, making it insufficient for dynamic metabolic control. In contrast, optogenetics offers a reversible and tunable method for regulating cellular metabolism with high temporal precision. In this study, we engineered the pyruvate decarboxylase isozyme 1 (Pdc1) by inserting the ph…

Research progress on the fermentation characterization of non-Saccharomyces yeasts in fruit wines: a review.

This review highlights the pivotal role of non-Saccharomyces yeasts in enhancing fruit wine quality, addressing limitations of traditional Saccharomyces cerevisiae monocultures. It systematically elaborates their mechanisms in improving color stability (via anthocyanin derivatives and enzymatic actions), enriching aroma (through ethanol, esters, higher alcohol, terpenoids, and volatile acid regulation), and modulating mouthfeel (via glycerol, polysaccharides, and organic acid degradation). The findings underscore strain-specific metabolic traits and mixed fermentation dynamics, providing criti…

Engineering of Saccharomyces cerevisiae as a platform strain for microbial production of sphingosine-1-phosphate.

BACKGROUND: Sphingosine-1-phosphate (S1P) is a multifunctional sphingolipid that has been implicated in regulating cellular activities in mammalian cells. Due to its therapeutic potential, there is a growing interest in developing efficient methods for S1P production. To date, the production of S1P has been achieved through chemical synthesis or blood extraction, but these processes have limitations such as complexity and cost. In this study, we generated an S1P-producing Saccharomyces cerevisiae strain by using metabolic engineering and introducing a heterologous sphingolipid biosynthetic pat…

Metabolic profiles and fingerprints for the investigation of the influence of nitisinone on the metabolism of the yeast Saccharomyces cerevisiae.

Nitisinone (2-(2-nitro-4-trifluoromethylbenzoyl)-1,3-cyclohexanedione, NTBC) is considered a potentially effective drug for the treatment of various metabolic diseases associated with disorders of L-tyrosine metabolism however, side-effects impede its widespread use. This work aimed to broaden the knowledge of the influence of NTBC and its metabolites 2-amino-4-(trifluoromethyl)benzoic acid (ATFA), 2-nitro-4-(trifluoromethyl)benzoic acid (NTFA), and cyclohexane-1,3-dione (CHD) on the catabolism of L-tyrosine and other endogenous compounds in Saccharomyces cerevisiae. Methyl and Ethylmercury elicit oxidative stress and unbalance the antioxidant system in Saccharomyces cerevisiae.

Methylmercury (MeHg) and Ethylmercury (EtHg) are toxic to the central nervous system. Human exposure to MeHg and EtHg results mainly from the consumption of contaminated fish and thimerosal-containing vaccines, respectively. The mechanisms underlying the toxicity of MeHg and EtHg are still elusive. Here, we compared the toxic effects of MeHg and EtHg in Saccharomyces cerevisiae (S. cerevisiae) emphasizing the involvement of oxidative stress and the identification of molecular targets from antioxidant pathways. Wild type and mutant strains with deleted genes for antioxidant defenses, namely: γ-…

Genomewide screening for genes involved in biofilm formation and miconazole susceptibility in Saccharomyces cerevisiae.

Infections related to fungal biofilms are difficult to treat due to the reduced susceptibility of sessile cells to most antifungal agents. Previous research has shown that 1-10% of sessile Candida cells survive treatment with high doses of miconazole (a fungicidal imidazole). The aim of this study was to identify genes involved in fungal biofilm formation and to unravel the mechanisms of resistance of these biofilms to miconazole. To this end, a screening of a Saccharomyces cerevisiae deletion mutant bank was carried out. Our results revealed that genes involved in peroxisomal transport and th…

Maf1, repressor of tRNA transcription, is involved in the control of gluconeogenetic genes in Saccharomyces cerevisiae.

Maf1 is a negative regulator of RNA polymerase III (Pol III) in yeast. Maf1-depleted cells manifest elevated tRNA transcription and inability to grow on non-fermentable carbon source, such as glycerol. Using genomic microarray approach, we examined the effect of Maf1 deletion on expression of Pol II-transcribed genes in yeast grown in medium containing glycerol. We found that transcription of FBP1 and PCK1, two major genes controlling gluconeogenesis, was decreased in maf1Δ cells. FBP1 is located on chromosome XII in close proximity to a tRNA-Lys gene. Accordingly we hypothesized that decrease

Gpd1 and Gpd2 fine-tuning for sustainable reduction of glycerol formation in Saccharomyces cerevisiae.

Gpd1 and Gpd2 are the two isoforms of glycerol 3-phosphate dehydrogenase (GPDH), which is the rate-controlling enzyme of glycerol formation in Saccharomyces cerevisiae. The two isoenzymes play crucial roles in osmoregulation and redox balancing. Past approaches to increase ethanol yield at the cost of reduced glycerol yield have most often been based on deletion of either one or two isogenes (GPD1 and GPD2). While single deletions of GPD1 or GPD2 reduced glycerol formation only slightly, the gpd1Δ gpd2Δ double deletion strain produced zero glycerol but showed an osmosensitive phenotype and abo…

The YEATS domain of Taf14 in Saccharomyces cerevisiae has a negative impact on cell growth.

The role of a highly conserved YEATS protein motif is explored in the context of the Taf14 protein of Saccharomyces cerevisiae. In S. cerevisiae, Taf14 is a protein physically associated with many critical multisubunit complexes including the general transcription factors TFIID and TFIIF, the chromatin remodeling complexes SWI/SNF, Ino80 and RSC, Mediator and the histone modification enzyme NuA3. Taf14 is a member of the YEATS superfamily, conserved from bacteria to eukaryotes and thought to have a transcription stimulatory activity. However, besides its ubiquitous presence and its links with …
