Intranasal administration of in silico designed Rivastigmine mucoadhesive nanoparticles ameliorates scopolamine-induced Alzheimer's symptoms in mice: Pharmacokinetic and pharmacodynamic evidences.

The low oral bioavailability (36 %) of rivastigmine tartrate (RT), and the inconspicuous pharmacological target site has necessitated the need to explore alternative routes of delivery for effective management of Alzheimer's disease (AD). The current study aimed to develop in silico designed polymeric nanoparticles (NPs) for the intranasal delivery of RT. Eudragit RL 100 (EDRL) was selected as mucoadhesive polymer because of its optimum binding affinity with RT and nasal mucin. These NPs were prepared using spray drying method by utilizing QbD approach. Yield, particle size, zeta potential, en

Effects of a Natural Polyherbal Extract on Alleviating Scopolamine-Induced Memory Deficits in C57BL/6 Mice via Enhancing Cholinergic Function.

Alzheimer's disease (AD) is a progressive neurological condition with limited effective pharmaceutical treatments, often accompanied by side effects. This has increased interest in plant-based alternatives. This study examined the cognitive effects of a Natural Polyherbal Extract (NPX) on scopolamine-induced memory deficits in mice. Male C57BL/6 mice (10 weeks old, n = 36) were divided into four groups: control (saline), scopolamine (1 mg/kg, i.p.), tacrine (10 mg/kg, oral), and NPX (1000 mg/kg, oral). NPX and tacrine were administered daily by oral gavage for two weeks. Cognitive function was

Kinetic and analytical characterization of a new tropinone oxidase enzyme and its application to the simultaneous determination of the tropane alkaloids atropine and scopolamine.

A spectrophotometric enzymatic method for the determination of atropine (Atp) and scopolamine (Scp), two tropane alkaloids (TAs), has been developed. The method is based on a previous basic hydrolysis to tropine (Trp) and scopine (Sci) respectively, and a subsequent enzymatic oxidation catalyzed by a tropinone reductase 1 (TRase) using NAD as oxidant; the absorbance of NADH (340 nm) is monitored during the reaction. First, the enzyme kinetics of both substrates are studied; it is concluded that both TAs follow a compulsory order ternary complex mechanism and the Michalis-Menten constant is cal

Coadministration of scopolamine and mGlu2 NAM VU6001966 as a novel antidepressant approach: Lowering effective dose and reducing cognitive side effects.

RATIONALE: Scopolamine, whose rapid and sustained antidepressant effects were first described nearly two decades ago, is associated with significant adverse effects that limit its clinical use. A safer alternative may involve coadministration of scopolamine at lower doses with other compounds that exhibit antidepressant properties. Growing evidence highlights the potential of group II metabotropic glutamate (mGlu) receptors as targets for depression treatment. OBJECTIVES: This study aimed to evaluate whether the mGlu2 receptor NAM, VU6001966, can enhance the antidepressant-like effects of a s

The potential antidepressant action and adverse effects profile of scopolamine co-administered with the mGlu7 receptor allosteric agonist AMN082 in mice.

Scopolamine, a muscarinic cholinergic receptor antagonist, exerts fast and prolonged antidepressant effects in the clinic. In contrast, the current treatments for major depressive disorder (MDD) require long-term drug administration. On the other hand, the sole use of scopolamine might be related to the high risk of adverse effects. Therefore, it may be preferable to reduce its therapeutic dose. A new approach might include the co-administration of low-dose scopolamine with selected ligands of metabotropic glutamate (mGlu) receptors, which are known to possess antidepressant-like activity in s

Selective 5-HT1A antagonists WAY 100635 and NAD-299 attenuate the impairment of passive avoidance caused by scopolamine in the rat.

Systemic administration of the muscarinic-receptor antagonists atropine and scopolamine produces cognitive deficits in humans, nonhuman primates and rodents. In humans, these deficits resemble symptoms of dementia seen in Alzheimer's disease. The passive avoidance (PA) task has been one of the most frequently used animal models for studying cholinergic mechanisms in learning and memory. The present study examined the ability of two selective 5-HT(1A) receptor antagonists WAY 100635 and NAD-299 (robalzotan) and two acetylcholinesterase (AChE) inhibitors tacrine and donepezil to attenuate the im

Effects of scopolamine on extracellular acetylcholine and choline levels and on spontaneous motor activity in freely moving rats measured by brain dialysis.

The present study demonstrates the feasibility of measuring acetylcholine (ACh) and choline in perfusate samples collected by in vivo brain dialysis in the frontal cortex and hippocampus of freely moving rats in which spontaneous motor activity could be measured simultaneously. Systemically administered scopolamine increased the output of ACh about 10-fold and 20-fold in the frontal cortex and hippocampus, respectively. By contrast, scopolamine decreased the choline level in the extracellular fluid about 2-fold in both brain regions, possibly owing to enhanced choline uptake into the presynapt

Joint cholinergic-serotonergic control of neocortical and hippocampal electrical activity in relation to behavior: effects of scopolamine, ditran, trifluoperazine and amphetamine.

Previous research has indicated that low voltage fast activity (LVFA) in the neocortex and rhythmical slow activity (RSA) in the hippocampus can result from activity in either (or both) the cholinergic corticipetal projections from the basal forebrain and the serotonergic corticipetal projections from the brainstem raphe. These inputs appear to give rise, respectively, to atropine-sensitive LVFA and RSA and atropine-resistant LVFA and RSA. The atropine-sensitive and atropine-resistant waveforms have been shown to have distinctive behavioral correlates. The present experiments extend these find

A reduction of vestibulo-visual integration during transdermally administered scopolamine and dimenhydrinate. A presentation of gain control theory in motion sickness.

The effect of transdermally administered scopolamine (TTS scopolamine) (release rate 5 micrograms/h) and dimenhydrinate (100 mg) was examined on optovestibular nystagmus in 16 volunteers in a randomized double-blind trial. A statistically significant decrease in the optokinetic part of nystagmus was observed during all treatments. Most profound reduction was found during treatment with two TTS scopolamine. The vestibular part was reduced by treatment with two TTS scopolamine, only. The results indicate that scopolamine and dimenhydrinate exert their effect in motion sickness by reducing the ve
