Tuberculosis enhanced transmission due to interterritorial mobility and prolonged diagnostic delay: a call for an integrated genomic analysis.

OBJECTIVES: Genomic epidemiology has revolutionized how we can delineate tuberculosis (TB) transmission. However, TB surveillance is restricted to the analysis of geographically limited populations, which impairs the identification of cross-regional transmission. This study aimed to integrate genomic data from independent populations, targeted strain sequencing, and refined epidemiological research to enhance the analysis of tuberculosis transmission. DESIGN: Integration of sequencing data from Madrid and Almería, evolutionary analysis of the single-nucleotide polymorphisms (SNPs) identified

Converging infectious disease threats in the post-COVID era: surveillance fragility, pandemic risk, and global preparedness.

The COVID-19 pandemic exposed profound weaknesses in global infectious disease surveillance yet, in its aftermath, pandemic-prone threats have intensified rather than receded. This narrative review synthesizes contemporary evidence on three converging drivers of pandemic risk zoonotic emergence, antimicrobial resistance, and climate-sensitive disease expansion and examines how structural failures in surveillance and global health governance amplify their impact. Drawing on recent epidemiological analyses, policy reports, and One Health evaluations, the review maps post-COVID surges in zoonotic

Uncovering the genomic landscape of Mycobacterium bovis in Wales.

Bovine tuberculosis (bTB), caused by the bacterium Mycobacterium bovis, is one of the most pressing animal health issues in Wales today. It negatively impacts cattle health, affects profitability and trade, and can decimate years of genetic improvement towards desirable production traits. It also imposes substantial financial, social, and psychological burdens on farming communities. Eradication of bTB requires an understanding of local transmission pathways to target effective disease-control interventions. In this study, we characterised the genomic diversity of M. bovis across Wales by anal

A Systematic Investigation of the Common Genetic Architecture of Substance Use Traits and the Relationship with Mental Health.

INTRODUCTION: Understanding shared and unique genetic vulnerability for substance use traits will give insight into the etiology of their comorbidity with psychiatric and other health outcomes. METHODS: We applied genomic structural equation modeling to summary statistics from genome-wide association studies (GWASs) to map the genetic architecture of fourteen substance use traits across five substances (tobacco, alcohol, cannabis, cocaine, and opioids) and three stages (initiation, frequency, and dependence). We systematically compared data-driven and theory-based models and conducted sensiti

Family-based genome-wide association analysis of novelty seeking in a Korean schizophrenic population: A pilot study.

Schizophrenia (SPR) is the most devastating mental illness that causes severe deterioration in social and occupational functioning, but, the etiology remains unknown. The objective of this study is to explore the genetic underpinnings of novelty seeking behavior in schizophrenic family within the Korean population. By conducting a family-based genome-wide association study, we aim to identify potential genetic markers and variations associated with novelty seeking traits in the context of SPR. We have recruited 27 probands (with SPR) with their parents and siblings whenever possible. DNA was e

Genetic diversity of the Chagas vector Triatoma dimidiata s.l. (Hemiptera: Reduviidae) across geographic scales in a top-priority area for control.

Population genetic structure of arthropod disease vectors provides important information on vector movement and climate or other environmental variables that influence their distribution. This information is critical for data-driven vector control. In the first comprehensive study of the genetic structure of T. dimidiata s.l. (Latreille, 1811) we focus on an area of active transmission designated as a top priority for control. We examined a high number of specimens across a broad geographic area along the border of Guatemala and El Salvador including multiple spatial scales using a high number

Replication of previous autism-GWAS hits suggests the association between NAA1, SORCS3, and GSDME and autism in the Han Chinese population.

BACKGROUND: Autism is a severe neurodevelopmental disorder characterized by social interaction deficits, impairments in communication, and restricted and repetitive stereotyped behavior and activities. Family and twin studies suggested an essential role of genetic factors in the etiology of autism spectrum disorder (ASD). Also, other studies found SORCS3 and GSDME (DFNA5) might be involved in brain development and susceptible to ASD. METHODS: In this study, 17 genome-wide significant SNPs reported in previous ASD genome-wide association studies (GWAS) and 7 SNPs in strong linkage disequilibri

Dictyostelium discoideum: An Alternative Nonanimal Model for Developmental Toxicity Testing.

A critical aspect of toxicity evaluation is developmental and reproductive toxicity (DART) testing. Traditionally, DART testing has been conducted in vivo in mammalian model systems. New legislation aimed at reducing animal use and the prohibitive costs associated with DART testing, together with a need to understand the genetic pathways underlying developmental toxicity means there is a growing demand for alternative model systems for toxicity evaluation. Here we explore the potential of the eukaryotic social amoeba Dictyostelium discoideum, which is already widely used as a simple model syst

Imbalanced social-communicative and restricted repetitive behavior subtypes of autism spectrum disorder exhibit different neural circuitry.

Social-communication (SC) and restricted repetitive behaviors (RRB) are autism diagnostic symptom domains. SC and RRB severity can markedly differ within and between individuals and may be underpinned by different neural circuitry and genetic mechanisms. Modeling SC-RRB balance could help identify how neural circuitry and genetic mechanisms map onto such phenotypic heterogeneity. Here, we developed a phenotypic stratification model that makes highly accurate (97-99%) out-of-sample SC = RRB, SC > RRB, and RRB > SC subtype predictions. Applying this model to resting state fMRI data from the EU-A

MSX ₁ gene variant and non-syndromic clefting: association or rejection?

CONTEXT: Non-syndromic cleft lip/palate (NSCL/P) is a congenital anomaly with significant medical, psychological and social ramifications. There is sufficient evidence to hypothesize that locus for this condition can be identified by candidate genes. AIMS: The aim of this study is to amplify the chosen region (799 G >T) of MSX 1 gene, investigate the degree of association and perform a mutation research from Raichur cleft lip and palate patient sample. SETTINGS AND DESIGN: Case history and clinical examination of the patient were recorded to rule. Written consent was obtained from patients a

An atypical deletion of the Williams-Beuren syndrome interval implicates genes associated with defective visuospatial processing and autism.

BACKGROUND: During a genetic study of autism, a female child who met diagnostic criteria for autism spectrum disorder, but also exhibited the cognitive-behavioural profile (CBP) associated with Williams-Beuren syndrome (WBS) was examined. The WBS CBP includes impaired visuospatial ability, an overly friendly personality, excessive non-social anxiety and language delay. METHODS: Using array-based comparative genomic hybridisation (aCGH), a deletion corresponding to BAC RP11-89A20 in the distal end of the WBS deletion interval was detected. Hemizygosity was confirmed using fluorescence in situ

Brain structures mediate the causal effects of socioeconomic status on mental disorders: A multi-stage Mendelian randomization study.

BACKGROUND: The associations between socioeconomic status (SES), brain structural alterations, and mental disorders have been well established, but the causal nature and underlying mechanisms of such associations are elusive. METHODS: By performing multi-stage Mendelian randomization analyses of multi-source summary data from genome-wide association studies, we explored the causal associations of 3 SES indicators with 12 mental disorders and 13 types of brain structures, pursued by characterization of the mediating roles of brain structures in accounting for the causal effects of SES on menta

Exploring socio-economic, biochemical, and genetic factors influencing thyroid status in Indian school-going adolescents.

Thyroid hormones are central to regulating metabolism, growth, and development, yet their complex interactions with socioeconomic, metabolic, and genetic factors remain understudied in diverse populations. We compared thyroid profiles - free triiodothyronine (FT3), free thyroxine (FT4), and thyroid-stimulating hormone (TSH) in Indian adolescents with anthropometric traits, metabolic markers, and socioeconomic status (SES). We observed that adolescents from higher SES backgrounds exhibited greater metabolic dysregulation, altered thyroid profiles, and abnormalities in lipid and adipokine levels

Multiancestry Genome-Wide Association Study of Early Childhood Caries.

Early childhood caries (ECC) is the most common noncommunicable childhood disease-an important health problem with known environmental and social/behavioral influences lacking consensus genetic risk loci. To address this knowledge gap, we conducted a genome-wide association study of ECC in a multiancestry population of U.S. preschool-age children (N = 6,103) ages 3 to 5 y participating in a community-based epidemiologic study of early childhood oral health. Calibrated examiners used International Caries Detection and Assessment System criteria to measure ECC; the primary trait was the number o

Identification of SF-1 and FOXL2 and Their Effect on Activating P450 Aromatase Transcription via Specific Binding to the Promoter Motifs in Sex Reversing Cheilinus undulatus.

The giant wrasse Cheilinus undulatus is a protogynous socially hermaphroditic fish. However, the physiological basis of its sex reversal remains largely unknown. cyp19 is a key gender-related gene encoding P450 aromatase, which converts androgens to estrogens. cyp19 transcription regulation is currently unknown in socially sexually reversible fish. We identified NR5A1 by encoding SF-1, and FOXL2 from giant wrasse cDNA and cyp19a1a and cyp19a1b promoter regions were cloned from genomic DNA to determine the function of both genes in cyp19a1 regulation. Structural analysis showed that SF-1 contai

Generation and analysis of novel Reln-deleted mouse model corresponding to exonic Reln deletion in schizophrenia.

AIM: A Japanese individual with schizophrenia harboring a novel exonic deletion in RELN was recently identified by genome-wide copy-number variation analysis. Thus, the present study aimed to generate and analyze a model mouse to clarify whether Reln deficiency is associated with the pathogenesis of schizophrenia. METHODS: A mouse line with a novel RELN exonic deletion (Reln-del) was established using the CRISPR/Cas9 method to elucidate the underlying molecular mechanism. Subsequently, general behavioral tests and histopathological examinations of the model mice were conducted and phenotypic

Sociological variety and the transmission efficiency of Mycobacterium tuberculosis: a secondary analysis of qualitative and quantitative data from 15 communities in Zambia.

OBJECTIVES: Selected Zambian communities formed part of a cluster randomised trial: the Zambia and South Africa TB and AIDS Reduction study (ZAMSTAR). There was wide variability in the prevalence of Mycobacterium tuberculosis infection and tuberculosis (TB) disease across these communities. We sought to clarify whether specific communities could have been more/less vulnerable to M. tuberculosis transmission as a result of sociological variety relevant to transmission efficiency. DESIGN: We conducted a mixed methods secondary analysis using existing data sets. First, we analysed qualitative da

Multi-trait GWAS identifies pleiotropic loci shared between early pregnancy bleeding and psychiatric traits.

INTRODUCTION: Early pregnancy bleeding is a common pregnancy complication, yet its genetic basis and potential links with psychiatric traits remain poorly understood. This study aimed to characterize the shared genetic architecture between early pregnancy bleeding and reproductive, psychiatric, and cardiometabolic traits. METHODS: We integrated linkage disequilibrium score regression (LDSC), local genetic correlation analysis (LAVA), and multi-trait genome-wide association analysis (MTAG). LDSC was used to estimate genome-wide genetic correlations, including sex-stratified analyses. LAVA was

Chromatin Accessibility Profiling of the Canine Brain Reveals Regulatory Programs Associated With Dog-Wolf Divergence and Human Psychiatric Risk.

The domestic dog has emerged as a valuable model for investigating the biology of social behavior and neuropsychiatric disease, yet a comprehensive map of gene regulatory elements across the canine brain is still lacking. Here, we generated an ATAC-seq atlas spanning seven major brain regions and the pituitary gland from four adult beagle dogs. We identified 251,492 putative cis-regulatory elements (CREs) and found that chromatin accessibility profiles resolve anatomically distinct regions into discrete regulatory modules, including striatal, cortical-associated, hypothalamic/habenular, and pi

The life history of recessive deleterious alleles as seen through the eyes of a honey bee (Apis mellifera).

Deleterious mutations arise continually in natural populations, yet their full life histories-from origin to elimination-are rarely observed directly. Our understanding of mutational fate in the wild comes from indirect inference rather than from tracking a single mutation through time and space. Haplodiploid systems provide a rare opportunity to observe negative selection in action: because males are haploid, recessive alleles are immediately exposed to selection rather than masked by heterozygosity. Haplodiploidy has long been hypothesized to accelerate the purging of deleterious variation.
