[Decline in semen quality from 1930 to 1991].

It has been suspected for years that semen quality has declined. However, the issue is still controversial since previous studies were criticized for methodological errors. We therefore attempted to systematically review the complete international literature on semen quality since 1930 with rigorous selection criteria and statistical analysis. Based on a literature search using Cumulated Index Medicus and Current List (1930-1965) and MEDLINE Silver Platter database (1966-august 1991), we identified 61 papers concerning sperm density in a total of 14,957 males without a history of infertility.

The Re-Addition of Seminal Plasma after Thawing Does Not Improve Buck Sperm Quality Parameters.

In order to achieve a higher post-thaw buck sperm quality, an approach in the thawing protocol of cryopreserved sperm doses under in vitro capacitation conditions mimicking the in vivo female environment was studied. Therefore, functional and kinetic characteristics of buck thawed sperm from males of different ages, the season of collection, and melatonin implanted males in the non-breeding season were assessed after 3 h of incubation in an in vitro fertilization (IVF) media with 20% of buck seminal plasma (SP). Previously, fresh ejaculates were collected via artificial vagina from eight males

Effect of various levels of dissolved oxygen on reactive oxygen species and cryocapacitation-like changes in bull sperm.

The present investigation was carried out to study the effect of various levels of dissolved oxygen (DO) on reactive oxygen species (ROS) and cryocapacitation-like changes in bull sperm. Egg yolk-Tris-glycerol (EYTG) extender was split into four subextenders; viz., Extender I (control; no flushing with liquid nitrogen (LN2 )), Extender II, Extender III and Extender IV were flushed with LN2 for 40, 16 and 8 min, respectively. The DO levels were standardized to 11.7, 2, 4 and 8 ppm, respectively, in control (Extender I), Extender II, Extender III and Extender IV. Ejaculates with mass motility of

Clinical assessment of the male fertility.

The evaluation of infertility in males consists of physical examination and semen analyses. Standardized semen analyses depend on the descriptive analysis of sperm motility, morphology, and concentration, with a threshold level that must be surpassed to be considered a fertile spermatozoon. Nonetheless, these conventional parameters are not satisfactory for clinicians since 25% of infertility cases worldwide remain unexplained. Therefore, newer tests methods have been established to investigate sperm physiology and functions by monitoring characteristics such as motility, capacitation, the acr

Sperm interaction in the hyperandrogenized female reproductive tract might contribute to female infertility: a hypothesis.

It is well known that hyperandrogenism (HA) in females is associated with oligo-ovulation or anovulation, leading to subfertility. The abnormality in ovulation is due to ovarian dysfunction. However, it remains unclear whether ovarian dysfunction is the sole cause of subfertility. The crosstalk between sperm and the hyperandrogenized female reproductive tract has yet to be explored in the context of subfertility or infertility. Therefore, in the present paper, we hypothesized that firm sperm binding to the hyperandrogenized female reproductive tract might be one of the reasons for subfertility

Improved approach for the cryopreservation of mouse sperm by combining monothioglycerol and l-glutamine.

The CryoPreservation Media (CPM) for mouse sperm using raffinose and skim milk have been improved by adding either monothioglycerol (MTG) or l-glutamine to reduce the oxidative damage during sperm freezing and thawing. The CARD-CPM utilizing l-glutamine, but not MTG, has been widely used to meet the rising demand for cryopreservation of genetically modified mice, as the CARD method also improved sperm capacitation and in vitro fertilization (IVF). However, the viability of sperm frozen in the CARD-CPM is highly variable, indicating a room for improvement. To develop a more dependable technique…

Changes in fertilization medium viscosity using hyaluronic acid impact bull sperm motility and acrosome status.

The female reproductive tract, in particular the composition of the uterine and oviduct fluids, is responsible, at least in part, for triggering sperm cell modifications, essential for the acquisition of fertilization ability. Hyaluronic acid (HA) is a glycosaminoglycan present in these fluids, and its role in the fertilization process and sperm functionality is still barely understood. This work was designed to (a) determine the rheological characteristics of the fertilization medium by the addition of HA and (b) determine the HA influence on sperm motility and functional status. To that end,

Detrimental effects of temephos on male fertility: An in vitro study on a mouse model.

The World Health Organization recommends temephos as a nonsystemic organophosphorus pesticide due to its low mammalian toxicity compared with other chemical compounds. Although several studies have reported that temephos may be toxic under certain conditions, little research effort has been made to evaluate its effects on mammalian fertility. Therefore, the present study was designed to evaluate the effect of temephos on sperm functions and male fertility. Initially, cauda epididymis from mouse spermatozoa was incubated with temephos (0, 0.1, 1, 10, and 100 μM). Then, sperm motility and motion

The functional integrity and fate of cryopreserved ram spermatozoa in the female tract.

Cryopreservation advances capacitation-like changes in ram spermatozoa. These changes are reflected in an increased fertilizing ability compared with fresh spermatozoa, followed by an accelerated decline in fertilizing ability after incubation in vitro or in vivo. Furthermore, frozen-thawed spermatozoa are released earlier than fresh spermatozoa after binding to oviduct cells in vitro, confirming their physiological readiness to participate in fertilization despite their short lifespan. After insemination large numbers of spermatozoa are lost from the female reproductive tract of the ewe via t

Presence, processing, and localization of mouse ADAM15 during sperm maturation and the role of its disintegrin domain during sperm-egg binding.

Successful fertilization requires gametes to complete several stages, beginning with maturation and transport along the male and female reproductive tracts and ending with the interaction between the sperm and the egg. This last step involves sperm-egg adhesion and membrane fusion. ADAMs (disintegrin and metalloprotease domain proteins) are a family of membrane-anchored glycoproteins that are thought to play diverse roles in cell-cell adhesion through their interaction with integrins. This study analyzes the presence, location, processing, and possible role of ADAM15 in mouse sperm. The presen

Localization and significance of molecular chaperones, heat shock protein 1, and tumor rejection antigen gp96 in the male reproductive tract and during capacitation and acrosome reaction.

Although the molecular basis of sperm-oocyte interaction is unclear, recent studies have implicated two chaperone proteins, heat shock protein 1 (HSPD1; previously known as heat shock protein 60) and tumor rejection antigen gp96 (TRA1; previously known as endoplasmin), in the formation of a functional zona-receptor complex on the surface of mammalian spermatozoa. The current study was undertaken to investigate the expression of these chaperones during the ontogeny of male germ cells through spermatogenesis, epididymal sperm maturation, capacitation, and acrosomal exocytosis. Signaling mechanisms in mammalian sperm motility.

The goal of sperm is to fertilize the oocyte. To achieve that purpose, it must acquire motility in the epididymis and hyperactivated motility in the female reproductive tract. Motility is only achieved when the sperm presents a fully functional flagellum, is capable of producing energy to fuel the movement, and suffers epididymal maturation and capacitation. Since sperm is a transcriptionally silent cell, motility depends on the activation and/or inhibitions of key signaling pathways. This review describes and discusses the main signaling pathways involved in primary and hyperactivated motilit

Spink13, an epididymis-specific gene of the Kazal-type serine protease inhibitor (SPINK) family, is essential for the acrosomal integrity and male fertility.

Sperm maturation involves numerous surface modifications by a variety of secreted proteins from epididymal epithelia. The sperm surface architecture depends on correct localization of its components and highlights the importance of the sequence of the proteolytic processing of the sperm surface in the epididymal duct. The presence of several protease inhibitors from different families is consistent with the hypothesis that correctly timed epididymal protein processing is essential for proper sperm maturation. Here we show that the rat (Rattus norvegicus) epididymis-specific gene Spink13, an an

Proteomic identification of sperm from mice exposed to sodium fluoride.

Fluoride is a widespread environmental pollutant which can induce low sperm quality and fertilizing ability. However, effect of fluoride on proteomic changes of sperm is unknown. In this study, two-dimensional electrophoresis (2DE) and mass spectrometry (MS) were used to investigate the differently expressed proteins of sperm from mice exposed to fluoride. 180 male mice were randomly divided into three groups, and were administrated with the distilled water containing 0, 25, and 100 mg L-1 NaF, respectively. After 45, 90 and 180 day's exposure, mice were sacrificed and sperm from the cauda epi

Hyperactivation and Tyrosine Phosphorylation of Cryopreserved Stallion Spermatozoa in Relation to Conventional In Vitro Fertilization Blastocyst Production.

BACKGROUND: Challenges in establishing clinical conventional in vitro fertilization (IVF) in horses include variation in sperm and oocyte quality, which can be affected by mare age. Extended preincubation of stallion spermatozoa has resulted in successful IVF. However, there is limited research on the impact of preincubation on sperm characteristics and IVF success. OBJECTIVES: Responses of frozen-thawed stallion spermatozoa over time under IVF preincubation conditions were evaluated, and samples based on specific responses were tested for their ability to fertilize oocytes, including those f

Seminal plasma proteins as modulators of the sperm function and their application in sperm biotechnologies.

Seminal plasma (SP) is known to play an important role in mammalian fertilization. However, the variability found in its composition among species, males and even fractions of the same ejaculate has made difficult to completely understand its effect in sperm function. Proteins are one of the major SP components that modulate sperm functionality. During the last years, intensive work has been performed to characterize the role of these proteins. They have been found to influence sperm capacitation, formation of the oviductal sperm reservoir and sperm-oocyte interaction. Sperm biotechnologies, s…

Involvement of cholesterol, calcium and progesterone in the induction of capacitation and acrosome reaction of mammalian spermatozoa.

The aim of this paper was to review the effects of some important substances involved in the induction of sperm plasma membrane changes referred to as acrosome reaction, namely cholesterol (C), calcium (Ca(2+)) and progesterone (P(4)). For this purpose, mechanisms of capacitation and acrosome reaction (AR) as well as the processes, C, Ca(2+) and P(4) are involved in, are described. Subsequently, to get a better insight into possible beneficial and detrimental effects on sperm function, the occurrence of these molecules in semen extenders is discussed.

WD repeat-containing protein 41 (WDR41) is involved in fertilization and interacts with IZUMO1 and JUNO.

During mammalian fertilization, sperm-oocyte recognition, binding, and subsequent plasma membrane fusion are facilitated by specific protein-protein interactions. Among these interactions, the IZUMO1-JUNO receptor pair is considered important for the adhesion and fusion of sperm and oocyte. However, the IZUMO1-JUNO interaction alone is insufficient to mediate membrane fusion, suggesting that the coordinated interactions of other proteins are required. In this study, we aimed to identify novel molecular players that interact with IZUMO1 and JUNO and investigate their functional roles in fertili

[Regulation of sperm hyperactivation by transporters involved in Na+ homeostasis].

Mammalian sperm have to undergo several physiological and biochemical changes to be fertilization-competent. These changes are collectively called "capacitation". Capacitated sperm show frantic whiplash-like flagellar movement called "hyperactivation". Recently, it has been reported that treatments of sperm to enhance hyperactivation improve in vitro fertilization (IVF) and development of the fertilized embryos. This fact indicates that hyperactivation is an attractive target to improve assisted reproductive technologies. However, the detailed mechanisms which regulate hyperactivation have not
