Sporotrichosis is usually a localized, lymphocutaneous disease, but its disseminated type was rarely reported. The main objective of this study was to identify specific DNA sequence variation and virulence of a strain of Sporothrix schenckii isolated from the lesion of disseminated cutaneous sporotrichosis. We confirmed this strain to be S. schenckii by(®) tubulin and chitin synthase gene sequence analysis in addition to the routine mycological and partial ITS and NTS sequencing. We found a 10-bp deletion in the ribosomal NTS region of this strain, in reference to the sequence of control strai…

BACKGROUND: In order to study the genotype variation of Sporothrix schenckii, we previously analyzed the genomic sequences of the DNA topoisomerase II genes of this fungus and S. schenckii-related species, such as S. schenckii var. luriei and Ceratocystis stenoceras.
OBJECTIVE: To develop a PCR-based identification system that can distinguish S. schenckii from its related species for clinical diagnosis of sporotrichosis.
METHODS: Based on the nucleotide sequences of the DNA topoisomerase II genes of S. schenckii, S. schenckii var. luriei and C. stenoceras, three gene-specific primers (SSHF31…

A murine model was used to evaluate the virulence of Sporothrix schenckii conidia cultured for 4, 7, 10 or 12 days in Sabouraud's dextrose broth (SDB). A correlation was observed between length of culture and virulence. Mice infected intravenously with S. schenckii conidia cultured for 4 or 7 days showed 40-100% cumulative mortality. In contrast, mice infected with conidia from cultures grown for 10 or 12 days in SDB showed no mortality (100% survival). A much greater accumulation of fungal colony forming units (cfu) was observed in the lungs, livers and spleens of mice inoculated with conidia…

Infection with Sporothrix schenckii causes a localized lymphocutaneous disease in the immunocompetent host, while it frequently results in disseminated disease in the immunocompromised patient. There are a growing number of reports of S. schenckii infection in the human immunodeficiency virus (HIV)-infected population, where the disease usually starts as a localized cutaneous lesion and subsequently disseminates. The optimal treatment of systemic sporotrichosis in HIV-positive patients is as yet unknown. This article presents a case report of disseminated sporotrichosis in an HIV-infected pati…

Isolates of Sporothrix schenckii were examined for their infectivity in BALB/c mice. The mice were injected with yeast forms of S. schenckii isolates differing in clinical source (human cutaneous lesions and pulmonary lesions), and fungal growth was determined at intervals in the footpad and visceral organs. After subcutaneous injection of approximately 10 colony forming units (cfu) of S. schenckii into the footpad, locally restricted fungal infection developed gradually. At the peak of the infection (3-4 weeks post-inoculation), viable fungal counts reached 102-106 cfu/footpad. Dissemination …

Sporothrix schenckii is the etiologic agent of sporotrichosis, a mycosis of world-wide distribution more commonly occurring in tropical regions. The immunological mechanisms involved in the prevention and control of sporotrichosis are not fully understood but apparently include both the humoral and cellular responses. In the present investigation, cellular immunity was evaluated by in vivo and in vitro tests in mice infected with yeast-like forms of S. schenckii. The disease developed systemically and cellular immunity was evaluated for a period of 10 weeks. The soluble antigen utilized in the…

Sporothrix cyanescens has been recovered from blood and a finger lesion at several medical centers in the United States. The morphology and physiology of these and three additional isolates were studied. S. cyanescens was distinguished from S. schenckii and S. fungorum by white to lavender colonial pigmentation and from S. schenckii also by the formation of secondary conidia. All isolates of S. cyanescens grew well at 37 degrees C, were cycloheximide susceptible, strongly urease positive, and benomyl resistant, failed to hydrolyze starch, and were inhibited by sodium chloride in vitro at a con…

In 1979 Kwon Chung described two varieties of Sporothrix schenckii based on the thermotolerance of isolates from fixed cutaneous (35 degrees C) and that of disseminated cutaneous forms (37 degrees C) of sporotrichosis. Since we had not noted such a difference previously in a study of 100 cases of this disease (55% localized and 45% disseminated) wherein all the isolates grew at 37 degrees C, we decided to repeat this work. Our results differ from those reported by Kwon Chung, since the isolates of both the fixed and disseminated forms of sporotrichosis grew at 37 and 38 degrees C, even when we…

The rapid expansion of zoonotic sporotrichosis in South America necessitates innovative surveillance strategies to identify natural ecological niches. Roadkill provides a unique, underutilized opportunity to monitor Sporothrix circulation within human-impacted landscapes. We conducted a molecular survey via a triplex probe quantitative real-time PCR (qPCR) assay targeting pathogenic Sporothrix species in 81 roadkilled vertebrates (mammals, birds, and reptiles) collected along highways BR-376 and PR-445 traversing the Atlantic Forest in Paraná, Brazil (2017-2023). Genomic DNA from visceral orga…

Hyperendemic zoonotic sporotrichosis, attributed to Sporothrix brasiliensis, presents a significant public health challenge in Brazil. Cats exhibit severe symptoms and high fungal loads, though their susceptibility is unclear. Sporothrix schenckii can also cause feline disease, primarily seen in Asia. This study is the first to report an in vitro model for examining cat immune cell responses to S. brasiliensis or S. schenckii. We investigated the phagocytic activity of blood cells (FMdP) from healthy domestic cats, challenged with yeast cells of S. brasiliensis and S. schenckii. The survival o…

Sporotrichosis is a subcutaneous infection caused by fungi from the genus Sporothrix, among which Sporothrix brasiliensis displays high virulence and transmissibility. So far, classical antifungal agents have been efficient against S. brasiliensis, but here we describe the first case of therapeutic failure and a high minimum inhibitory concentration (MIC) in relation to itraconazole.

BACKGROUND: Sporotrichosis is a neglected tropical disease and the most common subcutaneous mycosis, mainly caused by Sporothrix species, particularly S. brasiliensis, S. schenckii and S. globosa, which exhibit varying biological behaviours and virulence. The epidemic of sporotrichosis in Brazil, initiated in Rio de Janeiro in the late 1990s, rapidly spread to other states, including Amazonas in 2021. This study aimed to identify the specific Sporothrix species responsible for the human sporotrichosis outbreak in the Brazilian Amazon.
METHODS: A cross-sectional study was conducted by enrollin…

Sporotrichosis is a subcutaneous mycosis resulting from the traumatic implantation of pathogenic Sporothrix species. In Brazil, zoonotic transmission plays an important role in the epidemiology of the disease, involving especially cats. The objective of this study was to isolate Sporothrix spp. from cats with signs of sporotrichosis, determining the causative species, clinical and epidemiological aspects, and the in vitro susceptibility profile of the isolates against antifungal drugs. From September 2017 to February 2019, 245 samples of lesions were collected from symptomatic cats in São José…

Introduction. Sporotrichosis, caused by species of the Sporothrix schenckii complex, is the most prevalent subcutaneous mycosis in many areas of Latin America. Statins are a class of drugs widely used for lowering high sterol levels through their action on 3-hydroxy-3-methylglutaryl-CoA reductase, a key enzyme in the synthesis of sterol.Aim. In this study, the antifungal activity of statins (simvastatin, atorvastatin, pravastatin) against planktonic cells and biofilms of S. schenckii complex species was evaluated, as well as the interaction of pravastatin with classical antifungals (amphoteric…

The metropolitan region of Rio de Janeiro is hyperendemic for cat-associated sporotrichosis. This study aimed to assess the virulence of serial Sporothrix isolates from a 61-year-old male patient with chronic, destructive disseminated sporotrichosis. Five Sporothrix isolates were cultured from skin exudates and bone samples over a 5-year period, and all were molecularly identified as Sporothrix brasiliensis. The final isolate was significantly more virulent in Galleria mellonella larvae compared to earlier isolates. We conclude that S. brasiliensis has the capacity to increase in virulence in …

Sporotrichosis and cutaneous leishmaniasis are skin infections with similar clinical manifestations but different treatment methods. The present study aimed to evaluate qPCR and conventional PCR for differential detection of the etiological agents of both infections in multiplex format. Assays were designed using two sets of reported primers: SS1/SS2, designed on the 18S ribosomal RNA gene from Sporothrix spp., and JW11/JW12, designed on the kinetoplast DNA (kDNA) minicircles of Leishmania spp. qPCR detected 200 fg of DNA per reaction for both Sporothrix and Leishmania. Melting curve analysis …

Sporothrix (Sp.) schenckii is a pathogenic fungus that infects humans and animals, and is responsible for the disease named sporotrichosis. The cell wall of this fungus has glycoproteins with a high content of mannose and rhamnose units, which are synthesized by endoplasmic reticulum- and Golgi-localized glycosyltransferases. Little is known about the enzymic machinery involved in the synthesis of these oligosaccharides in Sp. schenckii, or the genes encoding these activities. This is in part because of the lack of an available genome sequence for this organism. Using a partial genomic DNA lib…

Sporothrix schenckii isolates of fixed and lymphocutaneous clinical forms from Mexico (MX), Guatemala (GT), and Colombia (CO) as well as environmental isolates from MX were studied by analyzing their phenotypic characteristics (conidial length, thermotolerance by percent growth inhibition [GI] at 35 and 37 degrees C, median lethal dose [LD(50)]) and genotypic characteristics (by random amplified polymorphic DNA [RAPD] analysis-PCR). A significant difference (P < 0.01) in the mean conidial length of S. schenckii clinical isolates from CO ( = 4.03 +/- 1.04 microm) compared with those of clinical…

A 58-year-old, alcoholic, diabetic man presented with multiple, ulcerated skin lesions and polymicrobial septicemia. Sporothrix schenckii was recovered from blood cultures and was resistant to amphotericin B by in vitro testing. Amphotericin B therapy failed, but the patient responded dramatically to itraconazole therapy, only to relapse 3 months after therapy was stopped. Reinstitution of itraconazole therapy has produced another dramatic response. This report is noteworthy for three reasons. First, to our knowledge, it represents only the second reported instance of fungemia with S schenckii…
