Expanding the molecular understanding of Theileria infections in cattle and yaks from Gansu and Qinghai, China.

Piroplasmosis is a major tick-borne disease that causes significant economic losses in livestock production across various regions. The present study aimed to provide a comprehensive assessment of the prevalence of bovine piroplasmosis in Gansu and Qinghai provinces, China, offering crucial baseline data for the development of effective control and prevention strategies. A total of 736 bovine blood samples were collected from Tianshui, Pingliang, and Lanzhou in Gansu, and Xining and Haidong in Qinghai. These samples were analyzed using PCR with universal Piroplasma primers targeting the 18S rR

Comparative diagnostic performance of recombinant Tams1 protein based dot-ELISA in detecting tropical theileriosis in naturally infected cattle.

This study is the first to conduct a sero-surveillance of Bovine Tropical Theileriosis (BTT) caused by the protozoan parasite Theileria annulata (T. annulata) using a recombinant Tams1 protein-based dot-ELISA in cattle, and to compare its efficacy with plate-ELISA, single PCR, nested PCR, and blood microscopy. The goal was to identify the most effective method for the early and accurate detection of theileriosis, which significantly impacts livestock through reduced milk yield and increased mortality. A total of 101 field blood samples were examined using blood smear analysis, single PCR, nest

Linking tick and wildlife host distributions to map risk of tick-borne diseases.

BACKGROUND: Tick-borne pathogens threaten livestock worldwide, causing diseases such as anaplasmosis, babesiosis, heartwater and theileriosis in cattle. The epidemiology of each disease is complex, with multiple tick and/or host species interacting across variable environments, and disease risk has not been fully assessed. Here, we used a One Health approach that integrates ecological, livestock and wildlife factors to identify areas with potential risk of tick-borne pathogen circulation. METHODS: Using ecological data on tick and host distributions alongside environmental and anthropogenic v

Molecular identification, risk factor assessment, and phylogenetic analysis of tick-borne pathogens in symptomatic and asymptomatic cattle from South-Eastern Iran.

Tick-borne pathogens (TBPs) represent a substantial threat to cattle globally, exerting adverse impacts on production, health, and economic viability. This study delves into the prevalence and implications of TTBPs in cattle sourced from resource-limited smallholder livestock farms situated in southeastern Iran, proximate to Afghanistan and Pakistan. Blood and tick specimens were systematically collected from a cohort of 230 cattle, comprising 150 asymptomatic and 80 symptomatic individuals. Genomic DNA isolated from blood samples underwent rigorous examination for the presence of key TBPs, in

Molecular detection and characterization of tick-borne haemoparasites among cattle on Zanzibar Island, Tanzania.

Tick-borne diseases (TBDs) are serious constraints to livestock production in Tanzania and other tropical and subtropical countries and impact the livelihoods of resource-poor farming communities in the region. In Tanzania, detailed studies on tick-borne pathogens (TBPs) in cattle using sensitive molecular detection methods are scarce. The objective of this study was to investigate the occurrence and species composition of bovine TBPs in cattle kept in Zanzibar Island. A total of 236 blood samples were randomly collected in cattle population in June and July 2019. We used polymerase chain reac

Molecular assays reveal the presence of Theileria spp. and Babesia spp. in Asian water buffaloes (Bubalus bubalis, Linnaeus, 1758) in the Amazon region of Brazil.

Approximately 50% of buffalo herds in Brazil are located in Pará state in northern Brazil. There are several properties where cattle and buffalo live and graze together, and thus, buffalo pathogens may threaten the health of cattle and vice versa. Therefore, knowledge of infectious agents of buffalo is essential for maintaining healthy livestock. Clinical disease caused by Theileria and Babesia parasites in the Asian water buffalo is not common, although these animals may act as reservoir hosts, and the detection of these hemoparasites in buffaloes is as important as it is in cattle. Studies o

Seroprevalence of Babesia caballi and Theileria equi in five draught equine populated metropolises of Punjab, Pakistan.

Equine piroplasmosis (EP) caused by intraerythrocytic parasites (Theileria equi and Babesia caballi) is an emerging equine disease of world-wide distribution. In Pakistan, the prevalence and incidence of EP are unknown. In order to obtain the first insights into the prevalence of the disease, a total of 430 equids, including 33 mules, 65 horses and 332 donkeys, aging from ≤ 5 to ≥ 10 years of either sex, from five metropolises of Punjab, Pakistan, were serologically tested for the presence of antibodies directed against B. caballi and T. equi, using a competitive enzyme-linked immunosorbent as

Detection of Theileria equi in spleen and blood of asymptomatic piroplasm carrier horses.

This study aimed to determine whether asymptomatic horses naturally infected with Theileria equi retain infected erythrocytes in the spleen and whether the presence of the hemoparasite in this organ is associated with parasitemia. We collected samples from 25 adult horses without clinical signs of any disease. From each animal, we collected whole blood samples from the jugular vein and a splenic puncture blood sample. All samples were submited to blood cell counts and detection of Theileria or Babesia. DNA extraction and PCR were performed in all samples for identification of piroplasm infecti

Molecular detection and characterization of potentially new Babesia and Theileria species/variants in wild felids from Kenya.

Piroplasms frequently infect domestic and wild carnivores. At present, there is limited information on the occurrence and molecular identity of these tick-borne parasites in wild felids in Kenya. In 2009, a pair of captive lions (Panthare leo) was diagnosed with suspected babesiosis and mineral deficiency at an animal orphanage on the outskirts of Nairobi, Kenya. Blood smears indicated presences of haemoparasites in the erythrocytes, however, no further investigations were conducted to identify the infecting agent. The animals recovered completely following diet supplementation and treatment w

Occurrence of Theileria parva infection in cattle on a farm in the Ladysmith district, KwaZulu-Natal, South Africa.

Theileria parva causes widespread morbidity and mortality in cattle in endemic regions. An outbreak of theileriosis occurred on a farm near Ladysmith in KwaZulu-Natal, South Africa, which is not a declared Corridor disease-infected area. A survey of Red Brangus cattle from all age groups and areas of the farm was performed. Transmission of the parasite from infected animals on the farm to susceptible animals by tick transmission and tick-stabilate injection, was attempted. The survey indicated high numbers of animals with antibody titres to T. parva but only 6 infected animals, based on real-t

Piroplasmids of livestock in Tunisia.

Several species of piroplasms of livestock are present in Tunisia; some of them are of high veterinary importance. This paper reviews the species already reported in Tunisia on the basis of clinical observations, parasitological routine diagnostic and serological surveys, as well as those considered as potentially present according to epidemiological argumentations. The genus Theileria includes four species reported in Tunisia: T. annulata, T. buffeli, T. ovis, and T. equi. The ovine malignant theileriosis agent, T. lestoquardi, appears to be absent in Tunisia. Five species belonging to the ge

Use of an in vitro infectivity assay in comparison with histological techniques in the study of Theileria parva sporozoite maturation.

Adult male and female Rhipicephalus appendiculatus ticks infected with Theileria parva (Muguga 3087) were fed on rabbits and the development of infection was monitored daily using light microscopy and an in vitro titration technique able to quantify the infectivity of sporozoite suspensions. The salivary glands stained with methyl green pyronine showed presence of infection in some unfed ticks. The intensity of staining was shown to increase with the number of days the ticks had fed. The in vitro technique, on the other hand, could detect infection only in ticks which had fed for 3-5 days. Fee

Vaccines against hemoparasitic diseases in Israel with special reference to quality assurance.

Four vaccines against hemoparasitic diseases (anaplasmosis, babesiosis and theileriosis) and a vaccine against besnoitiosis are currently used in Israel. These vaccines contain live attenuated parasites derived from cell culture (Theileria annulata and Besnoitia besnoiti) or from blood of infected, splenectomized calves (Babesia bigemina, B. bovis and Anaplasma centrale). Cryopreserved master seed is used to initiate production of the vaccines. Quality control performed during the preproduction period is particularly important with blood-derived vaccines. Post production quality control compri

Elimination of Theileria buffeli infections from cattle by concurrent treatment with primaquine phosphate and halofuginone lactate.

Fifty splenectomised calves naturally infected with Theileria buffeli were treated with primaquine phosphate (ICI, UK) and halofuginone lactate (Hoechst, Australia) either separately or in combination. Infections in treated calves were monitored for up to 26 weeks by examining Giemsa stained peripheral blood films for piroplasms and by an immunofluorescent antibody test. When used alone neither of the drugs eliminated infection. The most successful results were obtained when two treatments of halofuginone lactate, at a rate of 1 mg kg-1 body weight and six treatments of primaquine phosphate, a

Salivary gland of the tick vector (R. appendiculatus) of East Coast fever. I. Ultrastructure of the type III acinus.

The brown ear tick Rhipicephalus appendiculatus is the vector for East Coast fever, a disease that seriously limits livestock production in East Africa. The sporozoites of the infectious agent Theileria parva develop in the tick salivary gland. This paper describes the organization of the type III acinus of the gland and establishes unambiguous ultrastructural criteria for identification of the three secretory cell types: the d-cell, e-cell and f-cell. These observations are basic to exploration of possible cell-type specificity of the invading theileria and other aspects of host-parasite rela

Theileria equi sensu lato genetic diversity and phylogeny: new insights from mitochondrion, apicoplast and nucleus multilocus gene analysis.

Equine piroplasmosis is a significant tick-borne disease with a global distribution. Among the two causative agents, Theileria equi has undergone numerous taxonomic reclassifications over time, reflecting advances in our understanding of its biology, molecular characteristics, and phylogenetic relationships. Formerly named Babesia equi, now classified in the Equus group apart from Babesia and Theileria species, its genus name will again certainly change. The genetic diversity also within T. equi lead to the renaming of one lineage as Theileria haneyi. Theileria equi should better now be consid
