The ablation cycle drives glacier microbiome dynamics and downstream dissemination risk of the resistome.

Glacial ecosystems on the Tibetan Plateau undergo pronounced hydrological shifts across the glacial ablation cycle, driven by the onset and retreat of the Indian summer monsoon. To elucidate how transitions between four distinct hydrological ablation stages (pre-ablation, early ablation, late ablation, and frozen) shape microbial community structures and antibiotic resistance gene (ARG) profiles, we analyzed 112 samples collected across four stages from multiple glacier catchments on the southeastern Tibetan Plateau using metagenomic sequencing. Our results indicated that warmer stages favored…

Mineral and Microbial Properties Drive the Formation of Mineral-Associated Organic Matter and Its Response to Increased Temperature.

A comprehensive understanding of the formation of mineral-associated organic matter (MAOM) is a prerequisite for the sustainable management of soil carbon (C) and the development of effective long-term strategies for C sequestration in soils. Nevertheless, the precise manner by which microbial and mineral properties drive MAOM formation efficiency and its subsequent response to elevated temperature at the regional scale remains unclear. Here, we employed isotopically labelled laboratory incubations (at 15°C and 25°C) with soil samples from a ~3000 km transect across the Tibetan Plateau to eluc…

Consistent differences in brain structure and functional connectivity in high-altitude native Tibetans and immigrants.

It has been well-established that high-altitude (HA) environments affect the human brain; however, the differences in brain structural and functional networks between HA natives and acclimatized immigrants have not been well clarified. In this study, native HA Tibetans were recruited for comparison with Han immigrants (average of 2.3 ± 0.3 years at HA), with lowland residents recruited as controls. Cortical gray matter volume, thickness, and functional connectivity were investigated using magnetic resonance imaging data. In addition, reaction time and correct score in the visual movement task,…

Impaired Neurological Activity in the Mental Rotation Ability of Tibetan Indigenous Residents After Chronic Exposure to High Altitude.

Mental rotation is a core indicator of spatial ability, and a threshold for cognitive impairment may exist at approximately 4,000 m above sea level, but the specific thresholds for the severity of hypoxia in Tibetan indigenous populations in mental rotation ability remain largely unknown. To determine whether a threshold for mental rotation impairment exists in indigenous residents, we related a mental rotation task to inter-individual differences in a range of behavioral performance and neuropsychological characteristics across 51 indigenous Tibetan highlanders and 34 matched controls at thre…

Transcriptomic Study of Spermatogenesis in the Testis of Hu Sheep and Tibetan Sheep.

Numerous genes involved in male reproduction regulate testis development and spermatogenesis. In this study, the testis tissue transcriptome was used to identify candidate genes and key pathways associated with fecundity in sheep. Histological analysis of testis tissue using hematoxylin-eosin (HE) routine staining was performed for two sheep breeds. Overall, 466 differentially expressed genes (DEGs) were identified between Hu sheep (HS) and Tibetan sheep (TS) through RNA sequencing technology (RNA-Seq), including 226 upregulated and 240 downregulated genes. Functional analysis showed that seve…

How hypoxia slows fetal growth: insights from high altitude.

A continuous oxygen supply is vital for fueling fetal growth, yet we know surprisingly little as to the precise mechanisms by which hypoxia … qually affected. Dolma et al., have added valuable information now showing that that Ladakhis, like Tibetans and Andeans, have a lower-than-expected frequency of small-for-gestational age (SGA) infants. Consistent with previous reports, these authors also found that uterine artery diameters at mid pregnancy (week 26–28) were larger in women giving birth …

Assessment of a 10-year dog deworming programme on the transmission of Echinococcus multilocularis in Tibetan communities in Sichuan Province, China.

Human alveolar echinococcosis (AE) is considered a neglected zoonotic disease by the World Health Organization (WHO). The causative pathogen, Echinococcus multilocularis, lives as an adult tapeworm in the intestinal tract of canines. AE was identified as an emerging public health issue in Tibetan communities of Shiqu County 20 years ago. On St. Lawrence Island, Alaska (USA), in the 1980s peri-domestic transmission of E. multilocularis was controlled by regular deworming of owned dogs over a 10-year period. In Tibetan communities, on the Tibetan Plateau, control of E. multilocularis transmissio…

Microbial mercury methylation profile in terminus of a high-elevation glacier on the northern boundary of the Tibetan Plateau.

The Qinghai-Tibet Plateau glaciers are an important carrier of mercury (Hg). With global warming, Hg enters into the downstream ecosystem in the melt waters, threatening human health and ecosystem security in the region. Methylmercury (MeHg), which has higher toxicity than Hg itself, is converted from inorganic Hg. However, little is known about the process of Hg methylation and, in particular, microbial Hg methylation in high altitude mountain glaciers. We combined Hg speciation measurements and metagenomic analysis of 6 sample types from the terminus of Laohugou No.12 glacier to elucidate po…

Contamination of wastewater with Echinococcus multilocularis - possible implications for drinking water resources in the QTP China.

Echinococcus multilocularis is a parasite that causes a dangerous zoonosis, alveolar echinococcosis (AE). Its presence in water sources, however, has scarcely been studied heretofore. Accordingly, 222 samples of different origin including wastewater from wastewater treatment plants (WWTPs) (n = 137), slaughterhouse (n = 49) as well as water from rivers (n = 26) and a cattle farm (n = 10) were collected from Xining City and a rural area in Qinghai-Tibet Plateau (QTP), an endemic area. Material obtained after processing of 10 L volume samples was subsequently analysed using three molecular detec…

Ultraviolet radiation rather than inorganic nitrogen increases dissolved organic carbon biodegradability in a typical thermo-erosion gully on the Tibetan Plateau.

Permafrost thaw could lead to frozen carbon (C) being laterally transferred to aquatic systems as dissolved organic carbon (DOC). If this part of DOC has high biodegradability, it could be decomposed during the delivery process, release greenhouse gases to the atmosphere and trigger positive C-climate feedback. Thermokarst is an abrupt permafrost thaw process that can enhance DOC export and also impact DOC processing through increased inorganic nitrogen (N) and ultraviolet (UV) light exposure. Especially on the Tibetan Plateau, where thermokarst develops widely and suffers from serious UV radi…

Tuberculosis in Tibetan refugee settlements in India: a human rights perspective.

The statelessness and political vulnerability of Tibetans-in-exile in India are crucial drivers of tuberculosis (TB) in this population. The violent and forced displacement of Tibetans from Tibet into India in 1959 led to overcrowded and poorly resourced settlement camps that sparked a micro-epidemic of TB among exiled Tibetans, which persists to this day. This paper focuses on the diversity and precarity in legal status among Tibetans in exile in India, human rights implications and impacts on drivers of TB particularly access to housing, health care, nutrition, and employment. The absence of…

Small intestinal morphology and sugar transporters expression when consuming diets of different energy levels: comparison between Tibetan and small-tailed Han sheep.

Some non-structural carbohydrates, especially starch, escape ruminal fermentation, are converted into glucose, and are absorbed from the small intestine. This glucose provides an important source of energy, and its usage is more efficient than glucose from carbohydrates which are fermented as short chain fatty acids in the rumen and, subsequently, undergo hepatic gluconeogenesis. Tibetan sheep graze on the harsh Qinghai-Tibetan Plateau (QTP) all year round and their carbohydrate and energy intakes fluctuate greatly with seasonal forage availability. Consequently, a high capacity to absorb gluc…

Cryptosporidium infection induced the dropping of SCFAS and dysbiosis in intestinal microbiome of Tibetan pigs.

The infection of Cryptosporidium in pigs causes digestive system ailments, diarrhea and weight loss serving as an economic burden, especially in newborn animals. The bacterial fermentation products of short-chain fatty acids have important roles in immune function, microbiota regulation, osmotic balance and metabolism. However, till now little knowledge is available about the effect of Cryptosporidium infection on microbiota and SCFAs in plateau pigs. Hence, we performed this study to explore the response of microbiota and SCFAs in the natural infection of Cryptosporidium in Tibetan pigs. Cryp…

A Lytic Yersina pestis Bacteriophage Obtained From the Bone Marrow of Marmota himalayana in a Plague-Focus Area in China.

A lytic Yersinia pestis phage vB_YpP-YepMm (also named YepMm for briefly) was first isolated from the bone marrow of a Marmota himalayana who died of natural causes on the Qinghai-Tibet plateau in China. Based on its morphologic (isometric hexagonal head and short non-contractile conical tail) and genomic features, we classified it as belonging to the Podoviridae family. At the MOI of 10, YepMm reached maximum titers; and the one-step growth curve showed that the incubation period of the phage was about 10 min, the rise phase was about 80 min, and the lysis amount of the phage during the lysis…
