Great advances in the field of transplantation have been made in the last half of this century. However, the severe scarcity of donor organs, especially in the pediatric population, has become a major limitation. A new field, tissue engineering, applies the principles of biology and engineering toward the development of biological substitutes that restore, maintain, or improve tissue function. This article discusses the groundwork and challenges of this interdisciplinary field and its attempts to provide solutions to create new tissue for transplantation and other fields of reconstructive surg…

Cell therapy and tissue engineering may well likely dominate medical therapeutics in the next century. Growing a functional glomerular filter and tubule reabsorber from a combination of cells, biomaterials, and synthetic polymers to replace renal excretory and regulatory functions is a specific example of these evolving technologies. The kidney was the first organ whose function was substituted by an artificial device. The kidney was also the first organ to be successfully transplanted. The ability to replace renal function with these revolutionary technologies in the past was due to the fact …

Tracheal fistula is a life-threatening disorder for which no current therapy can simultaneously provide immediate airway rescue and long-term functional regeneration. Here we report a completely biodegradable, cell-free, off-the-shelf therapeutic construct that enables spatiotemporally matched repair by dynamically adapting to the evolving physiological demands of healing. The system integrates dynamic covalent and non-covalent interactions with distinct response timescales across spatially stratified layers, thereby supporting acute functional substitution, intermediate regenerative integrati

Biological tissues exhibit nonlinear strain-stiffening behavior with distinct toe (EToe) and heel (EHeel) moduli essential for load-bearing function. However, synthetic materials lack a unified strategy and mechanistic basis for independently tuning EToe and EHeel while maintaining high-fidelity 3D printability. Here, we introduce a 3D-printable strain-stiffening double-network granular hydrogel (SDGH) that enables region-specific control of EToe and EHeel through modulation of secondary-network monomer concentration and microgel packing density, respectively. We elucidate the mechanism of str

Three-dimensional (3D) spheroid models have emerged as advanced in vitro systems that overcome the limitations of conventional two-dimensional (2D) culture by better recapitulating native tissue architecture, cell-cell and cell-extracellular matrix interactions, and physiologically relevant gradients of oxygen, nutrients, and signaling factors, making them valuable platforms for tissue engineering and regenerative medicine. Realizing their full potential, however, requires rational control over cellular composition and spatial organization. In this review, we classify spheroids by cellular com…

Tissue injury and organ dysfunction remain major clinical challenges, as conventional therapies often fail to achieve functional regeneration. Chimeric antigen receptor (CAR) technology endows macrophages with the ability to specifically recognize and clear pathological targets, making CAR-macrophages (CAR-M) a promising tool in tissue engineering and regenerative medicine. However, the efficient, safe, and controllable engineering of CAR-M still depends on advanced chemical delivery systems. This review systematically summarizes five major platforms for CAR-M engineering, including viral vect…

UNLABELLED: Type 1 diabetes results from autoimmune destruction of pancreatic β-cells, and while whole-organ pancreas and islet transplantation can restore endogenous insulin secretion, both are limited by donor scarcity, early inflammatory injury, hypoxia, and the need for lifelong immunosuppression. Pancreatic tissue engineering has therefore emerged as a strategy to generate scalable, transplantable constructs capable of reestablishing physiological glucose regulation. However, as native islets depend on a robust microvascular network for oxygen delivery, nutrients and coordinated hormone s…

The potential of the porous scaffolds lies in its dynamic architectural features, such as porosity, pore size & shape, interconnectivity, and spatial distribution which are highly essential in tissue engineering applications. The porous structure of the scaffolds influences the biological and physicochemical activities in the tissue regeneration process. The porosity also allows better distribution of nutrients and diffusion of biological fluids thereby dispersing the seeding material uniformly. Generally, damaged tissues can be regenerated on their own but many times due to unavoidable factor…

The application of additive manufacturing techniques has increased over the years in almost all production fields, thanks to the possibility of creating objects from scratch and with the desired shape, with no need for molds or complex machinery typical of subtractive manufacturing. This success has concerned the biomedical world as well, where melt-based methods represent the golden standard to produce scaffolds for hard-tissue engineering. Despite the large number of studies present in the literature on scaffold production, the fabrication process is still affected by drawbacks and limitatio…

Soft-tissue loss resulting from trauma or oncologic resection is a significant problem worldwide. Surgical reconstruction using adipose tissue has long been the gold-standard solution. However, these surgeries are often highly morbid, not always feasible in patients with insufficient adipose, and can have unpredictable results. Engineered soft-tissue replacements present a promising alternative. Many cell types, such as adipose-derived stem cells, have been recognized as a viable starting platform upon which new avenues in tissue engineering can be built. Additionally, efforts to develop scaff…

Cardiovascular disease (CVD) is a leading cause of death worldwide. CVD includes conditions such as myocardial infarction (MI), arrhythmias, valvular heart disease, and cardiomyopathy. The limitations of heart treatment are related to the inability of damaged cells to regenerate, which leads to an increasing demand for new therapies. Cardiac tissue engineering (CTE) aims to efficiently regenerate damaged cardiac tissues by combining cells and biomaterials to address these diseases. Various cell types have been used in CTE research, including adult and pluripotent stem cells, the latter differe…

Cornea tissue engineering is strictly dependent on the development of biomaterials that fulfill the strict biocompatibility, biomechanical, and optical requirements of this organ. In this work, we generated novel biomaterials from the squid gladius (SG), and their application in cornea tissue engineering was evaluated. Results revealed that the native SG (N-SG) was biocompatible in laboratory animals, although a local inflammatory reaction was driven by the material. Cellularized biomaterials (C-SG) demonstrated that the SG provides an adequate substrate for cell attachment and growth, and cor…

Chronic Kidney Disease (CKD) is a growing worldwide problem, leading to end-stage renal disease (ESRD). Current treatments for ESRD include haemodialysis and kidney transplantation, but both are deemed inadequate since haemodialysis does not address all other kidney functions, and there is a shortage of suitable donor organs for transplantation. Research in kidney tissue engineering has been initiated to take a regenerative medicine approach as a potential treatment alternative, either to develop effective cell therapy for reconstruction or engineer a functioning bioartificial kidney. Currentl…

Discogenic pain is associated with deep nerve ingrowth in annulus fibrosus tissue (AF) of intervertebral disc (IVD). To model AF nerve ingrowth, primary bovine dorsal root ganglion (DRG) micro-scale tissue units are spatially organised around an AF explant by mild hydrodynamic forces within a collagen matrix. This results in a densely packed multicellular system mimicking the native DRG tissue morphology and a controlled AF-neuron distance. Such a multicellular organisation is essential to evolve populational-level cellular functions and in vivo-like morphologies. Pro-inflammatory cytokine-pri
