Toxicity evaluation of ultra-diluted methotrexate (12c and 30c): an acute and subacute oral toxicity study in animals.

Autoimmune diseases exhibit active and remission phases. Tissues may retain an 'inflammatory memory', increasing relapse risk after stopping treatment, necessitating ongoing therapy following remission induction. Understanding inflammatory memory and remission-to-recurrence transitions could guide strategies for achieving lasting remission while minimising long-term medication use and costs. Integrative approaches may address autoimmune conditions' multifaceted pathophysiology. Literature suggests utilising modern medicine in ultra-diluted form, with chemotherapy drugs like methotrexate underg

Toxicity in relation to mode of action for the nematode Caenorhabditis elegans: Acute-to-chronic ratios and quantitative structure-activity relationships.

Acute-to-chronic ratios (ACRs) and quantitative structure-activity relationships (QSARs) are of particular interest in chemical risk assessment. Previous studies focusing on the relationship between the size or variation of ACRs to substance classes and QSAR models were often based on data for standard test organisms, such as daphnids and fish. In the present study, acute and chronic toxicity tests were performed with the nematode Caenorhabditis elegans for a total of 11 chemicals covering 3 substance classes (nonpolar narcotics: 1-propanol, ethanol, methanol, 2-butoxyethanol; metals: copper, …

Triethylene glycol HO(CH2CH2O)3H.

TEG is a liquid higher glycol of very low vapor pressure with uses that are primarily industrial. It has a very low order of acute toxicity by i.v., i.p., peroral, percutaneous and inhalation (vapor and aerosol) routes of exposure. It does not produce primary skin irritation. Acute eye contact with the liquid causes mild local transient irritation (conjunctival hyperemia and slight chemosis) but does not induce corneal injury. Animal maximization and human volunteer repeated insult patch tests studies have shown that TEG does not cause skin sensitization. A study with Swiss-Webster mice demons

Changes in activity of non-specific esterases in cadmium treated Lymantria dispar larvae.

Many biochemical, physiological and histological criteria have been used as indicators of exposures and effects of the contaminants. These changes can indicate the response of an organism to a specific environmental stressor. In the present paper, the effect of the acute and chronic exposure to cadmium as well as recovery from two cadmium concentrations (10 and 30 μgCd/g dry food) on gypsy moth (Lymantria dispar) midgut esterases was investigated. The influence of cadmium on trait plasticity was also examined. Esterases showed great sensitivity to low metal concentrations during acute and chro

Historical control data of rare events: Issues, chronological patterns and their relevance for toxicological evaluations.

Historical control data (HCD) give context for a measurement by providing a biological reference frame. HCD are used in the evaluation of toxicological bioassays for quality and performance control, informal statistical false discovery rate mitigation, and to estimate the biological relevance of observed potentially adverse findings. The current commentary shortly highlights 5 points that should be considered when working with HCD of rare events: 1) HCD database (HCDB) size, 2) the issue of rare events, 3) potential chronological patterns, 4) using point estimates to summarize HCD and 5) indep

Toxic effects of the Emamectin Benzoate exposure on cultured human bronchial epithelial (16HBE) cells.

Pesticides pollution has caused serious environmental problems in recent years, and mounting evidence has shown that more and more insecticides have serious risk in human health. Emamectin Benzoate formally regarded as a highly safety insecticide based on its exclusive targets, but the cytotoxicity to human lung was ignored for a long time. In the present study, bioassay experiments were used to assess the toxicity of the Emamectin Benzoatein on human non-target cells including cell viability assay, DNA damage assay, flow cytometer assay and western blotting assay. The results indicated that E

In vitro and in vivo toxicity evaluation of non-neuroleptic phenothiazines, antitubercular drug candidates.

The phenothiazine-derived antipsychotic drugs, such as chlorpromazine and thioridazine, are bactericidal against drug-sensitive and drug-resistant strains of Mycobacterium tuberculosis, but produce undesirable side effects at clinically relevant doses. We have previously modified four novel phenothiazines and maintained their antimycobacterial activity. This study evaluated the pharmacological and toxicity profiles of these novel non-neuroleptic phenothiazines, PTZ3, PTZ4, PTZ31 and PTZ32, for their metabolic stability, kinetic solubility and potential cytotoxic effects in vitro. Acute and chronic effects from pulse exposure of D. magna to silver and copper oxide nanoparticles.

Aquatic toxicity testing of nanoparticles (NPs) is challenged by their dynamic behavior in test suspensions. The resulting difficulties in controlling and characterizing exposure concentrations are detrimental to the generation of concentration-response data needed for hazard identification of NPs. This study explores the applicability of short-term (1, 2 and 3h) pulse exposures as means to keep the exposure stable and at the same time disclose acute and chronic effects of AgNPs and CuONPs in D. magna. Dissolution, agglomeration and sedimentation were found to have less influence on exposure c
