Cocaine esterase occurrence in global wastewater microbiomes and potential for biotransformation of novel psychoactive substances.

The analysis of drugs in wastewater for forensic purposes has been constantly increasing and the investigation of the potential interaction between drugs or metabolites and sewage microbiota is important. The results demonstrated that cocaine esterase genes were widely distributed in 1142 global wastewater samples collected from 64 countries and linked to several bacterial species. In addition, in silico predictions indicated that carfentanil, 4F-MDMB-BINACA, 5F-MDMB-PICA, MDMB-4en-PINACA and mitragynine might also undergo microbial hydrolysis, in a similar fashion of cocaine degradation by co

[Emerging and re-emerging infectious diseases--a challenge for public health].

We have recently seen a worldwide explosion of infectious diseases: emerging diseases like the HIV/AIDS pandemic, or old diseases like cholera, tuberculosis, diphteria, plague, yellow fever, dengue, or malaria. These reemerging diseases are on the surge because of multiple factors: environmental changes, transformation of ecosystems, ongoing socioeconomic degradation and deterioration of public health systems in many countries. The increasing bacterial resistance to antibiotics or virologic resistance to antiviral drugs are becoming a serious problem today. This global danger needs a global re

Production of (R)-3-Chloro-1,2-Propanediol from Prochiral 1,3-Dichloro-2-Propanol by Corynebacterium sp. Strain N-1074.

The production of (R)-3-chloro-1,2-propanediol [(R)-MCP] from prochiral 1,3-dichloro-2-propanol (DCP) was examined with a bacterial strain identified as a Corynebacterium strain. The addition of glycerol as a carbon source or some chlorinated alcohols to a medium was effective for the induction of activity catalyzing the transformation of DCP into MCP. The optimum pH for (R)-MCP production by the resting cell reaction was around 8.0. The optical purity of (R)-MCP formed was improved by keeping the level of DCP in the reaction mixture at a low concentration. (R)-MCP was obtained from 77.5 mM DC

Complementary-synergistic Fe-Ce bimetallic nanozyme enhances antimicrobial efficacy through intermodule cycling catalysis.

In contemporary antimicrobial research, the ability to comprehensively modulate bacterial redox metabolism stands at the forefront of improving therapeutic efficacy and mitigating antibiotic resistance. Single metal ions often exhibit limited capacity for redox modulation, which may accelerate resistance emergence. Bimetallic catalysts, by contrast, confer synergistic benefits in antimicrobial applications. Here, we present a bimetallic Fe/Ce system, constructed as a dual-enzyme-mimetic catalytic module (Fe/Ce-BTC), that harnesses the complementary advantages of both metals to enhance catalyti

The mechanism by which Enteromorpha Linza polysaccharide promotes Bacillus subtilis growth and nitrate removal.

In this study, we discussed the relationship between Entermorpha linza polysaccharide (EP) and Bacillus subtilis, which can transform nitrate. A sole carbon source experiment showed that Bacillus subtilis could utilize EP, and the bacterial density was maximally increased by 54.43% in the EP groups. The results of reducing sugar determination proved the secretion of polysaccharide-degrading enzymes. Scanning electron microscopy (SEM) showed that the EP groups had fewer spores and shrunken bacteria, indicating that EP could improve the growth environment and maintain bacterial integrity. Additi

A deformable Bacillus subtilis with gut homing for tumor treatment via bacterial metabolism-facilitated synergistic approach for starvation, chemotherapy, and immunotherapy.

Cancer treatment remains a critical global health challenge, which often requires multimodal and targeted therapeutic strategies, yet integrating these modalities typically demands complex formulations. Oncolytic bacterial therapy offers unique opportunities for targeted drug delivery and immune modulation, yet clinical translation has been hindered by safety concerns and tedious engineering modification. Here, we report a deformable Bacillus subtilis (DBS), constructed through one-step cisplatin-induced morphological transformation that elongates the bacterium while simultaneously incorporati

Neuraminidase and nitric oxide dual response via PSA-PLGA nanoparticles: A novel approach for controlling Glaesserella parasuis inflammation and infection.

The overexpression of virulence factors and the induced inflammatory cytokine storm, resulting in tissue damage, represent significant challenges in treating antibiotic-resistant bacterial infections with conventional antibiotics. Herein, we have developed a bifunctional nanoparticle loaded with antibiotics (PSA@PLGA-TD/SMT) designed for precise response to the virulence proteins of drug-resistant bacteria while protecting the host from excessive inflammatory damage. This is achieved by modifying polylactic-co-glycolic acid (PLGA) nanoparticles with polysialic acid (PSA). Tildipirosin (TD), th

Bacterial resistance to antimicrobial agents and its impact on veterinary and human medicine.

BACKGROUND: Antimicrobial resistance has become a major challenge in veterinary medicine, particularly in the context of bacterial pathogens that play a role in both humans and animals. OBJECTIVES: This review serves as an update on acquired resistance mechanisms in bacterial pathogens of human and animal origin, including examples of transfer of resistant pathogens between hosts and of resistance genes between bacteria. RESULTS: Acquired resistance is based on resistance-mediating mutations or on mobile resistance genes. Although mutations are transferred vertically, mobile resistance genes

Novel nanoparticle vaccines for Listeriosis.

In recent years, nanomedicine has transformed many areas of traditional medicine, and enabled fresh insights into the prevention of previously difficult to treat diseases. An example of the transformative power of nanomedicine is a recent nano-vaccine against listeriosis, a serious bacterial infection affecting not only pregnant women and their neonates, but also immune-compromised patients with neoplastic or chronic autoimmune diseases. There is a major unmet need for an effective and safe vaccine against listeriosis, with the challenge that an effective vaccine needs to generate protective T

Exacerbation of virulence of multi-drug resistant Escherichia coli O104:H4 by subinhibitory concentrations of ampicillin.

Little is known about how subinhibitory concentrations of antibiotics to which bacteria are resistant affect bacterial virulence. In this study, the effect of subinhibitory concentrations of ampicillin on the virulence of E. coli O104:H4 was analyzed. Bacteria were pre-exposed to 0.1, 0.3, or 0.5 mg/mL of ampicillin in LB media and incubated for 4 h at 37 °C. Transformation capacity (using plasmids and PCR-amplified DNA sequences), swarming motility, biofilm production, curli formation, and virulence gene expression were determined. Ampicillin increased the transformation of E. coli O104:H4, w

IRF6 C-terminal phosphorylation links inflammation and cell-cell adhesion in oral keratinocytes: implications for oral lichen planus.

BACKGROUND: Oral Lichen Planus (OLP) is classified by the World Health Organization as an Oral Potentially Malignant Disorder (OPMD). OLP is characterized by chronic inflammation, which may contribute to its malignant transformation. In this study, we examined whether Interferon Regulator Factor 6 (IRF6), a key regulator of immune responses and epithelial homeostasis, modulates inflammation in oral keratinocytes via Toll-Like Receptor 2 (TLR2). This regulation may contribute to the malignant transformation of OLP. METHODS: Primary Human Oral Keratinocytes (HOKs) were exposed to lipopolysaccha

Trichloromethane dechlorination by a novel Dehalobacter sp. strain 8M reveals a third contrasting C and Cl isotope fractionation pattern within this genus.

Trichloromethane (TCM) is a pollutant frequently detected in contaminated aquifers, and only four bacterial strains are known to respire it. Here, we obtained a novel Dehalobacter strain capable of transforming TCM to dichloromethane, which was denominated Dehalobacter sp. strain 8M. Besides TCM, strain 8M also completely transformed 1,1,2-trichloroethane to vinyl chloride and 1,2-dichloroethane. Quantitative PCR analysis for the 16S rRNA genes confirmed growth of Dehalobacter with TCM and 1,1,2-trichloroethane as electron acceptors. Carbon and chlorine isotope fractionation during TCM transfo

Antibiotic pollution in the Katari subcatchment of the Titicaca Lake: Major transformation products and occurrence of resistance genes.

An increasing number of studies pointed out the ubiquitous presence of medical residues in surface and ground water as well as in soil compartments. Not only antibiotics can be found in the environment but also their transformation products about which little information is generally available. The development of bacterial resistance to antibiotics is particularly worrying as it can lead to sanitary and health problems. Studies about the dissemination of antibiotics and associated resistances in the Bolivian Altiplano are scarce. We provide baseline information on the occurrence of Sulfamethox

Promoting effect of ammonia oxidation on sulfur oxidation during composting: Nitrate as a bridge.

Ammonia (NH3) and hydrogen sulfide (H2S) are the main odor components in the composting process. Controlling their emissions is very important to reduce environmental pollution and improve the quality of composting products. This study explored the effects of functional bacteria on nitrogen and sulfur metabolism in the composting process of food waste (FW) by adding ammonia-oxidizing bacteria (AOB, A treatment), sulfur-oxidizing bacteria (SOB, S treatment), and combined AOB and SOB (AS treatment), respectively. The key bacterial species involved in nitrogen and sulfur transformation were ident

Application of UHPLC-MS/MS method to monitor the occurrence of sulfonamides and their transformation products in soil in Silesia, Poland.

Sulfonamides circulating in the environment lead to disturbances in food chains and local ecosystems, but most importantly contribute to development of resistance genes, which generate problems with multidrug-resistant bacterial infections treatment. In urban areas, sources of sulfonamide distribution in soils have received comparatively less attention in contrast to rural regions, where animal-derived manure, used as a natural fertilizer, is considered the main source. The aim of this study was to determine eight sulfonamides (sulfadiazine, sulfamerazine, sulfamethazine, sulfamethizole, sulfa

Efficient biotransformation of sulfide in anaerobic sequencing batch reactor by composite microbial agent: performance optimization and microbial community analysis.

Sulfur-containing wastewater is very common as an industrial waste, yet a high-efficiency composite microbial agent for sulfur-containing wastewater treatment is still lacking. In this work, three novel and efficient desulfurizing bacteria were isolated from the sewage treatment tank of Zhejiang Satellite Energy Co., Ltd. They were identified as Brucella melitensis (S1), Ochrobactrum oryzae (S8), and Achromobacter xylosoxidans (S9). These three strains of bacteria were responsible for the oxidative metabolism of sodium sulfide via a similar polythionate pathway, which could be expressed as fol

Utilization of glyphosate as phosphate source: biochemistry and genetics of bacterial carbon-phosphorus lyase.

After several decades of use of glyphosate, the active ingredient in weed killers such as Roundup, in fields, forests, and gardens, the biochemical pathway of transformation of glyphosate phosphorus to a useful phosphorus source for microorganisms has been disclosed. Glyphosate is a member of a large group of chemicals, phosphonic acids or phosphonates, which are characterized by a carbon-phosphorus bond. This is in contrast to the general phosphorus compounds utilized and metabolized by microorganisms. Here phosphorus is found as phosphoric acid or phosphate ion, phosphoric acid esters, or ph

Oxytetracycline interactions at the soil-water interface: effects of environmental surfaces on natural transformation and growth inhibition of Azotobacter vinelandii.

The mechanism of oxytetracycline (OTC) adsorption to a silty clay loam soil was investigated using sorption isotherm experiments, Fourier transform infrared spectroscopy (FTIR), and X-ray diffraction spectroscopy (XRD). Sorption data fit well to a cation-exchange capacity sorption model. Spectroscopic data indicate that the interactions between oxytetracycline and silty clay loam soil were primarily through electrostatic interactions between the protonated dimethylamino group of OTC and the negatively charged moieties on the surface of the soil. Based on XRD results, OTC adsorption appeared to
