Response characteristics of barrel cortical neurons in layers IV/V of juvenile rats with autism-like traits after tactile stimulation.

The barrel cortex is a specialized region of the primary somatosensory cortex that processes tactile information from whiskers. This study investigates how tactile stimulation (TS) affects excitatory receptive fields and surrounds suppression in barrel cortex neurons of male and female autistic-like rats, using various whisker displacement protocols. The animals were categorized into control, Valproic acid pre-treated (Val), and Val-TS treatment groups. In male Val-TS rats, TS reduced layer IV ON/OFF amplitudes for principal and adjacent whisker displacements, while only OFF response latency t

Early postnatal whisker deprivation cross-modally modulates prefrontal cortex myelination and leads to social novelty deficit.

Sensory experience affects not only the corresponding primary sensory cortex, but also synaptic and neural circuit functions in other brain regions in a cross-modal manner. However, it remains unclear whether oligodendrocyte (OL) generation and myelination can also undergo cross-modal modulation. Here, we report that while early life short-term whisker deprivation from birth significantly reduces in the number of mature of OLs and the degree of myelination in the primary somatosensory cortex(S1) at postnatal day 14 (P14), it also simultaneously affects the primary visual cortex (V1), but not t

Multiple Morphological Factors Underlie Experience-Dependent Cross-Modal Plasticity in the Developing Sensory Cortices.

Sensory experience regulates the structural and functional wiring of sensory cortices. In previous work, we showed that whisker deprivation (WD) from birth not only reduced excitatory synaptic transmission of layer (L) 2/3 pyramidal neurons of the correspondent barrel cortex in mice, but also cross-modally reduced synaptic transmission of L2/3 pyramidal neurons in other sensory cortices. Here, we used in utero electroporation, in combination with optical clearing, to examine the main morphological components regulating neural circuit wiring, namely presynaptic bouton density, spine density, as

Development of the interscutularis model as an outcome measure for facial nerve surgery.

INTRODUCTION: Animal models for the study of facial paralysis have been well developed, but concern has arisen regarding the accuracy of eye closure and whisker movement as outcome measures due to new data regarding interconnectivity between facial nerve branches and autonomic innervation. The posterior auricular nerve (PAN) is an isolated branch of the facial nerve which has been confirmed as the sole motor innervat or of the interscutularis muscle. This study was designed to develop a model for facial nerve palsy utilizing the PAN and interscutularis muscle. METHODS: A custom-made automated

The effect of electrical and mechanical stimulation on the regenerating rodent facial nerve.

OBJECTIVES/HYPOTHESIS: Investigators have long sought realistic methods to accelerate regeneration following nerve injury. Herein, we investigated the degree to which manual target muscle manipulation and brief electrical stimulation of the facial nerve, alone or in combination, affects recovery following rat facial nerve injury. STUDY DESIGN: Prospective, randomized animal study. METHODS: Sixty rats were randomized to three groups: brief electrical stimulation (BES), mechanical stimulation of the whisker pad (MEC), or both (COMBO). Animals underwent facial nerve transection and immediate mi

[Changes in voltage-gated potassium currents in the trigeminal neurons after a chronic constriction of infraorbital nerve.].

The purpose of this study was to establish a model of trigeminal neuralgia (TN) through an approach from lower edge of cheekbone and to observe the functional changes in the voltage-gated potassium currents in the cultured trigeminal ganglion (TG) neurons. Thirty Sprague-Dawley male rats were divided into two groups, the sham-operated (sham) group and the operated group. The TN model was carried out by using a chronic constriction injury of the infraorbital nerve (ION-CCI) from lower edge of cheekbone. Peripheral pain threshold test and whole-cell patch clamp recording were used to determine t

Regenerative process of the facial nerve: rate of regeneration of fibers and their bifurcations.

After the main trunk of the mouse facial nerve was injured by crushing, a fiber tracing method was used to quantify the facial motor neurons that extended regenerating nerve fibers to the specific site of the facial nerve branch. The total number of motor neurons retrogradely labeled with a fluorescent tracer, Fluoro-Gold (FG), were 0 on postsurgical days (PSDs) 1 and 2, 75+/-25 on PSD3, 264+/-21 on PSD4, 378+/-19 on PSD6, 428+/-19 on PSD8, 491+/-13 on PSD12 and 532+/-15 on PSD16. Biased Inter-columnar Communication and Short-Term Plasticity in Mouse Barrel Cortex.

Each barrel of the barrel cortex (BC) receives thalamic input primarily from a single whisker. Barrels then communicate through intracortical circuitry to integrate input from multiple whiskers and perform processing functions such as spatial filtering, motion sensing, and object localization. To investigate the circuits that enable barrels to communicate independently of extracortical and inter-areal influences, we prepared slices of somatosensory cortex from mice of both sexes with a hybrid voltage sensor (hVOS) targeted to Scnn1a excitatory neurons in cortical layer 4 (L4). We then imaged v

Impact of buccal and marginal mandibular branch transection on neuromuscular junction integrity in a rodent model of facial nerve paralysis.

BACKGROUND: Facial nerve paralysis is a complex and devastating condition, with translational research primarily relying on animal models. Models include injuries to the main trunk or distal branches of the facial nerve, such as the buccal and marginal mandibular branches. While recovery is well described following main trunk injury, this study investigates the impact of distal branch injuries on facial nerve recovery and neuromuscular junction (NMJ) innervation. NEW METHOD: Adult rats underwent transection of the buccal branch (BB) or of the buccal and marginal mandibular branches (BB+MM) of

Developmental divergence of sensory stimulus representation in cortical interneurons.

Vasocative-intestinal-peptide (VIP+) and somatostatin (SST+) interneurons are involved in modulating barrel cortex activity and perception during active whisking. Here we identify a developmental transition point of structural and functional rearrangements onto these interneurons around the start of active sensation at P14. Using in vivo two-photon Ca2+ imaging, we find that before P14, both interneuron types respond stronger to a multi-whisker stimulus, whereas after P14 their responses diverge, with VIP+ cells losing their multi-whisker preference and SST+ neurons enhancing theirs. Additiona

Bacterial cellulose tubes as a nerve conduit for repairing complete facial nerve transection in a rat model.

PURPOSE: Functionality of the facial nerve is cosmetically important. While many techniques have been investigated, early and effective treatment for traumatic facial nerve paralysis remains challenging. Here, we aim to examine bacterial cellulose (BC) as a new tubularization material for improving facial nerve regeneration. METHODS: Our study was performed on 40 female Sprague Dawley rats. Rats were randomly divided into four groups, with 10 rats per group. In all rats, the main trunk of the facial nerve was completely cut 8 mm before the branching point. For repairing the facial nerve, in g

The role of whiskers in compensation of visual deficit in a mouse model of retinal degeneration.

Sensory deprivation in one modality can enhance the development of the remaining modalities via mechanisms of synaptic plasticity. Mice of the C3H strain suffer from RD1 retinal degeneration that leads to visual impairment at weaning age. We examined a role of whiskers in compensation of the visual deficit. In order to differentiate the contribution of the whiskers from other mechanisms that can take part in the compensation, we investigated the effect of both chronic and acute tactile deprivation. Three-month-old mice were used. We examined motor skills (rotarod, beam walking test), gait cont

Neural correlates of long-term object memory in the mouse anterior cingulate cortex.

Damage to the hippocampal formation results in a profound temporally graded retrograde amnesia, implying that it is necessary for memory acquisition but not its long-term storage. It is therefore thought that memories are transferred from the hippocampus to the cortex for long-term storage in a process called systems consolidation (Dudai and Morris, 2000). Where in the cortex this occurs remains an open question. Recent work (Frankland et al., 2005; Vetere et al., 2011) suggests the anterior cingulate cortex (ACC) as a likely candidate area, but there is little direct electrophysiological evid

Reorganization of adult rat barrel cortex intrinsic signals following kainic acid induced central lesion.

A plasticity model studying the adult rat barrel cortex intrinsic signal after a central lesion was developed. Repeated optical imaging studies of the barrel cortex of five rats were performed over variable periods of time (2 days to 6 weeks) after intracortical injection of kainic acid. The signal of the elicited principal whisker corresponding to the injected barrel in the repeat studies relocated to the perimeter of the lesion. The area of the signals of this principal whisker and of surrounding whiskers were larger in the first two weeks studies than those obtained before injection (P<0.01
