Comparative proteomic analysis reveals functional and evolutionary diversity in five Montivipera snake venoms.

Proteomic characterization of snake venoms is essential for understanding the molecular basis of their evolution and for identifying bioactive compounds of therapeutic interest. The Montivipera species endemic to the Near and Middle East region remain poorly studied despite their interesting biological activities. Previous analyses of Montivipera venoms have provided only partial proteomic profiles, with notable discrepancies between studies. To address this gap, we conducted a proteomic analysis of five Montivipera species, including M. bornmuelleri, M. bulgardaghica, M. albizona, M. raddei a

Antivenomic and Proteomic Assessment of Inter- and Intrapopulation Venom Variations in Nikolsky's Adder Vipera nikolskii: Comparison to Common Adder Vipera berus.

Snake venom variation has important clinical implications, yet individual-level venomics remains limited. We investigated inter- and intrapopulation variability in forest-steppe adder Vipera nikolskii and its recognition by commercial V. berus antivenom using proteomic and immunological approaches. Venoms from 12 individual V. nikolskii specimens representing two geographically distinct populations (BG and KM), together with three pooled V. nikolskii and one pooled V. berus samples were analyzed by LC-MS/MS, ELISA, Western blot, and pull-down assays. Multivariate analysis revealed relative hom

Ontogenetic variation in Milos viper (Macrovipera schweizeri) venom composition and biochemical activity.

Ontogenetic shifts in diet are common in venomous snakes, with juveniles and adults often feeding on different prey types. These dietary changes are frequently paralleled by age-related variation in venom composition, likely reflecting adaptive optimisation for prey with different physiologies, particularly ectothermic versus endothermic organisms. Here, we investigated age-dependent differences in venom composition and biochemical activity in the Milos viper (Macrovipera schweizeri), a medically relevant venomous snake species that shifts from a lizard-based to an avian-based diet during grow

Importance of the Cysteine-Rich Domain of Snake Venom Prothrombin Activators: Insights Gained from Synthetic Neutralizing Antibodies.

Snake venoms are cocktails of biologically active molecules that have evolved to immobilize prey, but can also induce a severe pathology in humans that are bitten. While animal-derived polyclonal antivenoms are the primary treatment for snakebites, they often have limitations in efficacy and can cause severe adverse side effects. Building on recent efforts to develop improved antivenoms, notably through monoclonal antibodies, requires a comprehensive understanding of venom toxins. Among these toxins, snake venom metalloproteinases (SVMPs) play a pivotal role, particularly in viper envenomation

Clinical features of snake envenomation in South Korea.

INTRODUCTION: Three venomous snakes of the Gloydius genus belonging to the Viperidae family cause most snake envenomations in South Korea. Envenomation signs often include local swelling, coagulopathy, and rhabdomyolysis. The benefit of additional antivenom after the initial does is unclear. METHODS: This retrospective study divided patients into four groups according to the presence of rhabdomyolysis (creatine kinase ≥1000 IU/L) and coagulopathy, which were defined using the Korean Society on Thrombosis and Hemostasis disseminated intravascular coagulation score (rhabdomyolysis, coagulopathy…

Thrombocytopenic purpura following envenomation by the nose-horned viper (Vipera ammodytes ammodytes): Two case reports.

RATIONALE: Two clinical cases are reported of envenomation by the nose-horned viper (Vipera ammodytes ammodytes) venom of a 9-year-old boy and of an 84-year-old woman. PATIENT CONCERNS: Both patients had been bitten on their extremities by such a snake in August near Split, a town in southern Croatia. DIAGNOSES: Clinical manifestation of envenomation was severe in the case of the boy, being characterized by a severe coagulopathy. This was only just apparent in the case of the elderly woman, who suffered extensive local edema and hematoma at the site of the bite, together with a neurotoxic ef

Accelerated evolution of toxin genes: Exonization and intronization in snake venom disintegrin/metalloprotease genes.

Toxin genes in animals undergo accelerated evolution compared to non-toxin genes to be effective and competitive in prey capture, as well as to enhance their predator defense. Several mechanisms have been proposed to explain this unusual phenomenon. These include (a) frequent mutations in exons compared to introns and nonsynonymous substitutions in exons; (b) high frequency of point mutations are due to the presence of more unstable triplets in exons compared to introns; (c) Accelerated Segment Switch in Exons to alter Targeting (ASSET); (d) Rapid Accumulation of Variations in Exposed Residues

Comparative Profiling of Three Atheris Snake Venoms: A. squamigera, A. nitschei and A. chlorechis.

A proteomic and transcriptomic comparative analysis of the venoms of three Atheris species (A. squamigera, A. nitschei and A. chlorechis) was carried out by size exclusion liquid chromatography, gel electrophoresis, mass spectrometry, and mRNA sequencing. The improved proteomic profiling utilised in this work was combined with transcript studies, advancing our insights into venom composition, protein distribution and inter-species variation among the three bush vipers. Crude venoms of all three samples contained at least 10-20 protein components, ranging in size from ≤ 3 to > 98 kDa. Both appr

Twenty-four-hour ambulatory electrocardiography characterization of heart rhythm in Vipera berus-envenomed dogs.

BACKGROUND: Vipera berus has a worldwide distribution and causes high morbidity in dogs annually. A complication to envenomation may be cardiac arrhythmias. The purpose of this study was to investigate the prevalence, types, and timing of arrhythmias, using 24-h ambulatory electrocardiography (24-AECG), in dogs bitten by V. berus in the first 24-32 h after envenomation. In addition, this study aimed to investigate if there were differences in selected clinical and hematological- and biochemical variables (including cardiac troponin I) at admission between V. berus-envenomed dogs with and witho

Antivenom for European Vipera species envenoming.

BACKGROUND: European viper bite is relatively uncommon but can cause serious envenoming, particularly swelling and hemorrhage spreading from limb to trunk that can cause long term disability. Systemic features are relatively mild compared to many other venomous species. Moderate-to-severe envenoming requires antivenom, which is given many hundreds of times each year across the continent. Several Vipera spp antivenoms are produced in Europe, but there is little comparative information available for the antivenoms and none is licensed with the European Medicines Agency. We aimed to collect descr

[Lymphoedema after an adder bite].

Bites from the adder, Vipera berus, are well known for causing local and general toxicity. We report a case of a 63-year-old female who was bitten in one of her lower extremities. She was seen twice in the emergency department with symptoms and signs consistent with severe envenomation. Despite treatment with antihistamines and corticosteroids the oedema persisted. A lymphoscintigraphy was performed showing dermal backflow in the right leg, confirming the diagnosis of lymphoedema. Based on the present case report and review of the literature antivenom therapy should be considered more actively

[Adder bite. A rare dermatologic emergency].

Bites by poisonous snakes are rare dermatologic emergencies. A 43-year-old woman was bitten on her right forefinger by an adder during a hiking vacation in Austria. Her right arm and chest wall became swollen and then hemorrhagic, but she responded well to analgesics and immobilization. Administration of a specific antiserum was not necessary because of the lack of a systemic reaction.

[Accidents with venomous and poisonous animals in Central Europe].

Central Europe is largely safe from accidents with venomous and poisonous animals. The regions where European vipers are regularly found are shrinking. Today accidents with jellyfish and stings of venomous fish afflicted during leisure activities at the sea side play the dominant role. Life threatening accidents in Europe are mainly due to exotic snakes held in captivity. A system useful in daily medical practice is explained to classify and stage accidents due to poisonous and venomous animals. The important poisonous and venomous animals of Central Europe and the specific therapeutics, the a

Snakes of medical significance in India: the first reported case of envenoming by the Levantine viper (Macrovipera lebetina).

This article reports for the first time the medical significance of the Levantine viper (Macrovipera lebetina) in India. This snake is found in India in the mountainous regions of Jammu and Kashmir and is capable of causing necrosis and hemostatic manifestations, as seen in the case reported here. It is thus a class II snake of medical significance, as defined by the World Health Organization grading scale. This finding reinforces the recent decision by the Indian Government to abandon the term "Big Four" to describe important venomous species in India. Levantine viper venom is not included in

Abruptio placentae and fetal death following a Malayan pit viper bite.

Reports of venomous snakebites during pregnancy are uncommon. Little is known about maternal and fetal outcome following the venomous snakebite of a pregnant woman, and there is no consensus for proper management. In southern Thailand, Malayan pit viper (Calloselasma rhodostoma) bites are quite common, and we have some experience with bites of pregnant women. With these victims, the toxicity is most severely expressed in a bleeding disorder that is a significant cause of both morbidity and mortality in both the gravid woman and the fetus. Herein, because there are few such published reports, w

Viral association with the elusive rickettsia of viper plague from Ghana, West Africa.

We previously reported a rickettsial heartwater-like disease in vipers from Ghana that resembled heartwater in its gross lesions, was apparently transmitted by ticks (Aponomma and Amblyomma), and responded clinically favorably to early treatment with tetracycline. Cell culture showed consistent cytopathic effects in bovine endothelial cells, viper cells, and mouse cells, and inhibition of cytopathic effect by tetracycline in vitro. A type D retrovirus was observed in vacuoles in all infected cells. The virus and rickettsia infection was associated with transfer of cytopathic effect, regardless

Identification of a novel thrombin-like phospholipase A2 from Gloydius ussuriensis snake venom.

The coagulant effects of phospholipase A2 with Gln at 49 sites (Gln49-PLA2), purified from Gloydius ussurensis snake venom, were investigated on human citrated plasma and fibrinogen. Gln49-PLA2 clotted human plasma dose-dependently from 180.67 +/- 1.86 s to 19.00 +/- 0.58 s, and reduced the re-calcification time from 7.46 +/- 1.17 to 0.75 +/- 0.33 min and the prothrombin time from 12.4 +/- 0.29 s to 6.95 +/- 0.20 s, but it could not activate factor XIII, and the procoagulant effects were inhibited by heparin. The specific clotting activities of Gln49-PLA2 were equivalent to 1100 NIH thrombin U
