Transcription-coupled recruitment of SMC complexes in yeast.

Despite their distinct biological functions, structural maintenance of chromosomes (SMC) complexes, including cohesin, condensin, and SMC5/6, preferentially accumulate at transcriptionally active loci, with recent studies revealing that SMC5/6 also shares the loop extrusion mechanism previously considered a hallmark of cohesin and condensin. Using yeast as a model system, this review discusses how RNA polymerases I, II, and III, together with their associated transcriptional machinery, contribute to the recruitment of SMC complexes to specific genomic sites. We examined the reciprocal interpla

Bypass of the canonical biotin synthesis pathway in yeast.

Biotin serves as an essential cofactor for several carboxylation enzymes that catalyze reactions vital for cell survival and proliferation. While bacterial biotin synthesis pathways are well characterized, the mechanisms in eukaryotic organisms remain poorly understood. In Saccharomyces cerevisiae, Bio1 catalyzes the initial and rate-limiting step in the yeast biosynthetic pathway. Previous workers demonstrated that the Bio1 ortholog from the related yeast, Cyberlindnera fabianii, functionally substituted for S. cerevisiae Bio1 and greatly increased growth in the absence of biotin. However, we

Observing intersubunit dynamics in single yeast ribosomes.

Translation is accompanied by the rotation of the small and large ribosomal subunits relative to each other. Here, we use single-molecule Förster resonance energy transfer between fluorophores introduced into ribosomal proteins uS15 and eL30 to follow the intersubunit dynamics of Saccharomyces cerevisiae ribosomes. Similar to their bacterial counterparts, yeast ribosomes are observed to sample two predominant FRET states corresponding to the nonrotated (NR) and rotated (R) conformations. Our data yield further evidence that intersubunit rotation is coupled to tRNA transitions between the class

Alterations to the yeast chromatin landscape during hypoxia.

When cells encounter low oxygen conditions, signaling events target key transcription factors to reprogram gene expression to promote cell survival and adaptation under this challenging environmental stress. The substantial metabolic changes that occur in hypoxia also drive changes to the chromatin environment due to altered cofactor availability and direct oxygen sensitivity of chromatin-modifying enzymes, among other factors. Given the links between hypoxic cellular environments and numerous pathophysiological processes, including tumor progression and metastasis, cardiovascular disorders, a

On the rate of aneuploidy reversion in a wild yeast model.

Aneuploidy, arising from the gain or loss of chromosomes due to nondisjunction, is a special class of mutation. It can create significant phenotypic changes by altering the abundance of hundreds of genes in a single event, providing material for adaptive evolution. But it can also incur large fitness costs relative to other types of mutations. Understanding the mutational dynamics of aneuploidy is important for modeling its impact in nature, but aneuploidy rates are difficult to measure accurately. One challenge is that aneuploid karyotypes may revert back to euploidy, biasing forward mutation

Loss of Heterozygosity associated with ubiquitous environments in yeast.

The effect of ubiquitous environmental conditions on mutational mechanisms, particularly loss of heterozygosity (LOH) remains poorly understood. Environment induced LOH can rapidly alter the genome and promote disease progression. Using mutation accumulation (MA) lines, we analysed the effect of ubiquitous environmental conditions on mutational mechanisms in a diploid hybrid (S288c/YJM789) baker's yeast strain. These included blue light, low glucose (calorie restriction), oxidative stress (H2O2), high temperature (37°C), ethanol, and salt (NaCl). The frequency of LOH increased significantly in

Yeast proteomics: Advances and applications in alcoholic fermentation.

Yeast proteomics has become a crucial approach for elucidating molecular mechanisms underlying alcoholic fermentation, widely applied in winemaking, brewing, and bioethanol production. Advances in high-throughput techniques, such as two-dimensional electrophoresis and mass spectrometry, have enabled detailed characterization of protein expression dynamics in fermenting yeast strains. These studies have improved strain classification, optimized fermentation conditions, and identified protein biomarkers linked to flavor and aroma profiles. Key findings include the upregulation of enzymes involve

De novo biosynthesis of taxifolin in yeast peroxisomes.

BACKGROUND: Yeast peroxisomes have been engineered as ideal synthetic compartments to enhance the heterologous biosynthesis of natural products, particularly terpenoids and fatty acid derivatives. This advantage is primarily attributed to the rich acetyl-CoA pool generated from the spatially specific fatty acid β-oxidation within peroxisomes. However, their potential for flavonoid biosynthesis has been largely underexplored, primarily due to limited knowledge regarding precursor transport, cofactor availability, and the redox environment in peroxisomes. RESULTS: In this study, we successfully

Yeast-amoeba interaction influences murine cryptococcosis.

The virulence of Cryptococcus spp. is modulated in the natural environment through interaction with abiotic and biotic factors, and this can occasionally have implications for the progression of cryptococcosis in mammals. Hence, we evaluated whether the prior interaction of highly virulent Cryptococcus gattii strain R265 with Acanthamoeba castellanii influenced the progression of cryptococcosis. The influence of the capsule on endocytosis was evaluated using amoeba and yeast morphometrics. Mice were intratracheally infected with yeast re-isolated from the amoeba (Interaction), yeast without pr

From Plant to Yeast-Advances in Biosynthesis of Artemisinin.

Malaria is a life-threatening disease. Artemisinin-based combination therapy (ACT) is the preferred choice for malaria treatment recommended by the World Health Organization. At present, the main source of artemisinin is extracted from Artemisia annua; however, the artemisinin content in A. annua is only 0.1-1%, which cannot meet global demand. Meanwhile, the chemical synthesis of artemisinin has disadvantages such as complicated steps, high cost and low yield. Therefore, the application of the synthetic biology approach to produce artemisinin in vivo has magnificent prospects. In this review,

The Use of Iron-Enriched Yeast for the Production of Flatbread.

The most common cause of iron deficiency is an improperly balanced diet, in which the body's need for iron cannot be met by absorption of this element from food. Targeted iron supplementation and food fortification may be the main treatments for iron deficiency in the population. However, many iron-rich supplements and foods have low bioavailability of this element. In our study, we used yeast enriched with iron ions to produce flatbread. The yeast cells accumulated iron ions from the medium supplemented with Fe(NO3)3·9H2O, additionally one of the cultures was treated with pulsed electric fiel

YPIBP: A repository for phosphoinositide-binding proteins in yeast.

Phosphoinositides (PIs) are a family of eight lipids consisting of phosphatidylinositol (PtdIns) and its seven phosphorylated forms. PIs have important regulatory functions in the cell including lipid signaling, protein transport, and membrane trafficking. Yeast has been recognized as a eukaryotic model system to study lipid-protein interactions. Hundreds of yeast PI-binding proteins have been identified, but this research knowledge remains scattered. Besides, the complete PI-binding spectrum and potential PI-binding domains have not been interlinked. No comprehensive databases are available t

Extracellular Vesicles from Sporothrix Yeast Cells.

Sporotrichosis is an emerging deep mycosis and a public health concern in Brazil. This mycosis is caused by the traumatic inoculation of several species of Sporothrix from nature. However, when cats transmit the disease to humans as zoonotic transmission, severe extracutaneous manifestations are observed. Therefore, effective control of the disease requires the engagement of host receptors by pathogen-derived molecules to stimulate the immune response. In this context, extracellular vesicles from Sporothrix sp contribute to intracellular communication with host cells. In addition, however, ext

Anthelmintic Activity of Yeast Particle-Encapsulated Terpenes.

Soil-transmitted nematodes (STN) infect 1-2 billion of the poorest people worldwide. Only benzimidazoles are currently used in mass drug administration, with many instances of reduced activity. Terpenes are a class of compounds with anthelmintic activity. Thymol, a natural monoterpene phenol, was used to help eradicate hookworms in the U.S. South circa 1910. However, the use of terpenes as anthelmintics was discontinued because of adverse side effects associated with high doses and premature stomach absorption. Furthermore, the dose-response activity of specific terpenes against STNs has been
