Non-canonical Platelet activation by oral squamous cell carcinoma extracellular vesicles through cooperative thrombin formation.

Coagulation contributes to cancer mortality, and tumor cells activate platelets to release growth factors. However, this activation is non-canonical, employs unknown agonists and, uniquely, is not immediate. Platelets are activated by human cell lines derived from Oral Squamous Cell Carcinomas (OSCC) with this atypical delay. The stimulatory activity was not a soluble factor, but rather was completely recovered in spontaneously shed extracellular vesicles (EV). EV did not contain established platelet agonists. Instead, chemical and pharmacologic inhibitors show OSCC EV interacted with quiescen

ANCA-associated vasculitis: From immunopathogenesis to diagnosis and toward precision therapy.

Anti-neutrophil cytoplasmic antibody (ANCA)-associated vasculitis (AAV) comprises a group of rare, potentially life-threatening autoimmune disorders characterized by necrotizing inflammation of small vessels. Over recent decades, remarkable advances have been made in unraveling its immunopathogenesis, emphasizing the interplay between genetic predisposition, environmental triggers, loss of immune tolerance, neutrophil extracellular trap formation, and complement-mediated neutrophil activation. This review provides an integrative synthesis of recent mechanistic and clinical insights into AAV, b

Xing-nao-sheng-jiang powder alleviates ischemic stroke in rats by inhibiting pyroptosis-related microglial ETosis: An emerging perspective on microglial ETosis.

BACKGROUND: Stroke imposes a substantial global health burden. Microglial pyroptosis promotes acute cerebral ischemia. Gasdermin D (GSDMD), the principal executor of pyroptosis, has been implicated in a newly characterized form of proinflammatory cell death known as extracellular trap death (ETosis). Thus, targeting GSDMD may represent a promising therapeutic strategy to mitigate ischemic stroke. Xing-nao-sheng-jiang powder (XNSJP) was demonstrated to inhibit GSDMD-mediated pyroptosis in cerebral ischemia-reperfusion injury (CIRI). Based on these findings, we hypothesize that XNSJP may inhibit

Peripheral Blood Neutrophils and Monocytes Predict Disease Progression in Stage III Melanoma Patients Treated With Anti Programmed Cell Death Protein-1 (PD-1) Inhibitors.

Melanoma is a global health issue, with an increasing incidence in recent years. Immune checkpoint inhibitors (ICIs, e.g., antiprogrammed cell death protein 1 [PD-1]) and targeted therapies (e.g., BRAF/MEK inhibitors) have revolutionized treatment for melanoma patients (MPs), but prognostic markers of drug response and disease progression remain elusive. We conducted a prospective baseline and follow-up study to investigate peripheral blood neutrophils and monocytes as immune biomarkers in stage III MPs receiving either anti-PD-1 therapy or B-type rapidly accelerated fibrosarcoma (BRAF)/mitoge

Post-translational modification crosstalk in acute pancreatitis: linking acinar stress to innate immune amplification and tissue repair.

Acute pancreatitis (AP) is a sterile inflammatory disease initiated by acinar cell stress and amplified through innate immune, vascular, and inter-organ responses, yet effective disease-modifying therapies remain unavailable. Although premature digestive enzyme activation, organelle stress, sterile inflammation, and microvascular failure are well-recognized features of AP, the mechanisms linking early acinar stress to coordinated injury across epithelial, immune, stromal, and vascular compartments remain insufficiently integrated. Emerging evidence suggests that post-translational modification

The immunothrombosis pathway linking pulmonary disease to stroke and neurodegeneration: mechanisms and therapeutic implications.

Pulmonary diseases increasingly reshape vascular biology and coagulation beyond the lung. Across acute infections and acute respiratory distress syndrome (ARDS), chronic airway inflammation (e.g., COPD), sleep-disordered breathing, fibrotic interstitial lung disease, and particulate air pollution, convergent immune programs couple inflammation to coagulation through immunothrombosis. Physiologic immunothrombosis can confine pathogens within the microvasculature, but dysregulated thromboinflammation drives endotheliopathy, platelet-leukocyte cooperation, neutrophil extracellular trap (NET) form

Atg5/Autophagy inactivation in mouse bone microenvironment promotes tumor development.

Bone is an attractive site for cancer colonization, both for primary tumors such as osteosarcoma and for metastases of various malignancies. Preventing bone metastasis, which is associated with a poor prognosis, is a major challenge and identifying the factors involved in skeletal tumoral development is crucial to improve survival. In the present work, we showed that inactivation of the macroautophagy/autophagy-essential gene Atg5 in osteoblasts, the cells in charge of bone formation, stimulates osteosarcoma and breastbone metastasis growth as well as metastatic dissemination. We determined th

Uncovering immune dysfunction in ACLF: Cellular mechanisms, molecular pathways, and therapeutic frontiers.

Acute-on-chronic liver failure (ACLF) is a life-threatening condition characterised by acute hepatic decompensation, multi-organ failure, and high short-term mortality in patients with cirrhosis. A hallmark of ACLF is profound immune dysfunction, which contributes to excessive organ-specific inflammation and impaired host defence, predisposing patients to infection, multi-organ failure and death. This review aims to elucidate the cellular and molecular mechanisms implying systemic immune dysfunction in ACLF, highlighting key pathophysiological pathways and their clinical significance. We provi

Chemotherapy-photothermal liposomes modified by DNA-protein network inhibit oral squamous cell carcinoma progression basing on regulating tumor microenvironment.

Oral squamous cell carcinoma (OSCC) is primarily treated by surgery, radiotherapy, and chemotherapy, yet high recurrence/metastasis rates and treatment-associated toxicity remain major challenges. A key barrier is the immunosuppressive tumor microenvironment (TME) rich in cancer-associated fibroblasts (CAFs), which limits drug penetration and compromises therapeutic synergy. To overcome these obstacles, we developed a composite biomimetic nanodelivery system comprising two distinct formulations: platelet membrane-coated liposomes encapsulating berberine (BBR) and ginsenoside Rg3 (RG3) (B@R/PL)

Decoding Low-Density Neutrophils in Breast and Lung Cancer: From Bystanders to Drivers.

Neutrophils, once viewed as short-lived and functionally uniform antimicrobial cells, are now recognized as highly plastic immune regulators with context-dependent roles in cancer. Among circulating neutrophil subsets, low-density neutrophils (LDNs) have emerged as a clinically relevant population in oncology due to their distinct phenotypic, potent immunosuppressive activity, and strong association with disease progression. LDNs are rare in healthy individuals but markedly expanded in patients with solid tumors. LDNs display enhanced activation, increased expression of immunoregulatory molecu

NETosis in the Kidney, Liver, and Lung of Mice With Cecal Ligation and Puncture-Induced Sepsis and the Ameliorating Effects of Adipose-Derived Stem Cell Exosomes.

Sepsis-induced organ failure involves dysregulated neutrophil responses, including neutrophil extracellular trap (NET) formation, via NETosis. Exosomes generated from adipose-derived stem cells (ADSCs) have shown potential in sepsis treatment. This study characterized organ-specific NETosis in the kidney, liver, and lungs using a murine cecal ligation and puncture (CLP) model and assessed the modulatory effects of ADSC exosomes. Male C57BL/6 mice were grouped as follows: control, CLP, and CLP with ADSC exosome therapy (n = 6 per group). Plasma cell-free DNA (cfDNA) and tissue citrullinated his

Neutrophils and NETosis in polycystic ovary syndrome: unraveling the immuno-metabolic thromboinflammatory axis.

Polycystic ovary syndrome (PCOS), traditionally defined by its endocrine and reproductive hallmarks-hyperandrogenism, oligo-anovulation, and polycystic ovarian morphology-is increasingly viewed as a complex disorder associated with chronic low-grade immuno-metabolic inflammation. Emerging evidence suggests that neutrophils, the most abundant innate immune cells, may play a contributory role in linking metabolic stress, inflammatory signaling, and vascular dysfunction in PCOS. In women with PCOS, hyperandrogenism, insulin resistance, and adipose-derived inflammatory cues have been associated wi

Spatial distribution and structural discrimination of quorum sensing metabolites in Pseudomonas aeruginosa by MALDI-MSI.

Bacterial biofilms are complex bacterial communities embedded within an extracellular polymeric substance (EPS) that protects the cells and increases survival. Once established on an orthopaedic implant, a biofilm can cause difficult-to-treat and severe infections. Pseudomonas aeruginosa is of particular concern due to its strong biofilm-forming capability. Within biofilm communities, quorum sensing (QS) networks regulate biofilm formation, virulence, and intercellular communication. This is achieved through the production of signalling molecules, QS metabolites. In P. aeruginosa, quinolones s

Baicalein Ameliorates Experimental Ulcerative Colitis Recurrency by Downregulating Neonatal Fc Receptor via the NF-κB Signaling Pathway.

Ulcerative colitis (UC) is a chronic autoimmune disease (AID) that causes mild to moderate unpredictable symptoms, including diarrhea and abdominal pain. Against neonatal Fc receptor (FcRn) has been proven to be a unique AID treatment strategy by decreasing the effects of pathogenic autoantibody. Our previous study revealed that FcRn inhibition is beneficial in UC treatment through reducing colonic neutrophil extracellular trap (NET) formation via accelerating serum antineutrophil cytoplasm antibodies (ANCA) clearance. In this study, we initially confirmed the specific impact of downregulating

Humanization of CD47 enables development of functional human neutrophils via postirradiation remodeling of the bone marrow.

Murine and human immune systems differ significantly, particularly within the myeloid lineage. Humanized mice, generated by transplanting human hematopoietic stem, progenitor cells into genetically modified mice, are invaluable to study human immune development and function in vivo. However, a major limitation of current models is suboptimal myelopoiesis, particularly lack of functional human neutrophils, hampering the modeling of human immune responses and chronic diseases. Here, we describe a humanized mouse model, named MaGIC for genes replaced, in the C57Bl/6 N strain, which improves human

Impact of chronic hyperglycaemia on the coronary microcirculation - benefits of targeting IL-36 and diet reversal.

Following myocardial infarction (MI), patients with type 2 diabetes mellitus (T2DM) have poorer prognosis which may be linked to increased susceptibility of coronary microvessels to injury. Interleukin-36 (IL-36) may mediate this injury but its role in the microcirculation of the chronically hyperglycaemic injured heart is unknown. Intravital and laser speckle imaging of the anaesthetised mouse beating heart evaluated the impact of a 16-week high fat diet (HFD)-induced hyperglycaemia ± myocardial ischaemia-reperfusion injury (IR) injury on coronary microvessels. Neutrophil/platelet recruitment

Inhibition of S100A8/A9 ameliorates neuroinflammation by blocking NET formation following traumatic brain injury.

Traumatic brain injury (TBI) triggers a robust inflammatory response that is closely linked to worsened clinical outcomes. S100A8/A9, also known as calprotectin or myeloid-related protein-8/14 (MRP8/14), is an alarmin primarily secreted by activated neutrophils with potent pro-inflammatory property. In this study, we explored the roles of S100A8/A9 in modulating neuroinflammation and influencing TBI outcomes, delving into the underlying mechanisms. S100A8/A9-enriched neutrophils were present in the injured brain tissue of TBI patients, and elevated plasma levels of S100A8/A9 were correlated wi

Dual B-cell targeting in systemic lupus erythematosus: The role of combined and sequential therapy with rituximab and belimumab.

Systemic lupus erythematosus (SLE) is a complex autoimmune disease characterized by immune dysregulation and autoantibody production. Despite advances in treatment, achieving sustained disease control remains challenging. Rituximab (RTX) and belimumab (BELI) are two B-cell-targeting biologics with complementary mechanisms of action, leading to increasing interest in their combination as a therapeutic strategy for refractory SLE. RTX depletes CD20+ B cells, whereas BELI inhibits B-lymphocyte stimulator (BLyS), reducing the survival of autoreactive B cells. Sequential therapy with these agents m

Ultrasmall PtIr Bimetallic Nanozyme Treats Myocardial Infarction via Ischemic/Inflammatory Cardiac Microenvironment Remodeling.

Myocardial infarction (MI) poses a serious threat to human health. MI induces oxidative damage and inflammation, leading to myocardial death, scarring, and ventricular remodeling. Nanozymes have shown potential to alleviate reactive oxygen species (ROS)-induced damage and treat cardiovascular diseases. In this study, we developed an ultrasmall PtIr bimetallic nanozyme to treat MI. The PtIr nanozyme exhibited robust superoxide dismutase- and catalase-mimicking catalytic activities, modulating the conversion of excessive ROS into harmless products. Furthermore, PtIr nanozyme treatment reduced RO

Forsythoside B protects cartilage and subchondral bone in osteoarthritis by regulating the Nrf2/NF - κ B signaling pathway.

BACKGROUND: Joint diseases like osteoarthritis (OA) are common, however, there is still room for progress in the drug treatment of this disease. FTS ·B acts as an anti-inflammatory and antioxidant by blocking the NF-κB signal pathway and stimulating Nrf2, which are linked to cartilage and subchondral bone. PURPOSE: At present, the main treatment of OA is symptomatic therapy, including anti-inflammatory analgesia and surgical excision, while there are fewer treatments in terms of mechanism. Our aim is to find a novel compound that acts as a therapeutic effect on OA by blocking the mechanical p
