Wolbachia infections in world populations of bean beetles (Coleoptera: Chrysomelidae: Bruchinae) infesting cultivated and wild legumes.

Wolbachia endosymbionts are widespread among insects and other arthropods, often causing cytoplasmic incompatibility and other reproductive phenotypes in their hosts. Recently, possibilities of Wolbachia-mediated pest control and management have been proposed, and the bean beetles of the subfamily Bruchinae are known as serious pests of harvested and stored beans worldwide. Here we investigated Wolbachia infections in bean beetles from the world, representing seven genera, 20 species and 87 populations. Of 20 species examined, Wolbachia infections were detected in four species, Megabruchidius

Preliminary evidence of Rhodnius prolixus (Hemiptera: Triatominae) attraction to human skin odour extracts.

Host-seeking behaviour displayed by the haematophagous triatomine Rhodnius prolixus Stål, 1859 is influenced by different cues involving heat, moisture and odours. Concentrations of volatile odours present in human sweat and breath that activate, attract or non-attract triatomines were tested behaviourally with a double choice olfactometer in adults of R. prolixus. Attraction of a mixture of substances carried by a controlled air-stream was established and compared with volatiles from human extracts obtained by rubbing face, hands or feet. Volatiles from face and feet extracts showed a higher

A predator from East Africa that chooses malaria vectors as preferred prey.

BACKGROUND: All vectors of human malaria, a disease responsible for more than one million deaths per year, are female mosquitoes from the genus Anopheles. Evarcha culicivora is an East African jumping spider (Salticidae) that feeds indirectly on vertebrate blood by selecting blood-carrying female mosquitoes as preferred prey. METHODOLOGY/PRINCIPAL FINDINGS: By testing with motionless lures made from mounting dead insects in lifelike posture on cork discs, we show that E. culicivora selects Anopheles mosquitoes in preference to other mosquitoes and that this predator can identify Anopheles by

[Aedes (Stegomyia) aegypti (Linnaeus, 1762) breeding sites in native bromeliads in Vitória City, ES].

Some insects that are vectors of human diseases have accompanied man in his migrations throughout the world and breed exclusively in the proximity of human dwellings. The mosquito Aedes aegypti has been responsible for epidemics of dengue in Brazil and its presence also constitutes a serious risk for future outbreaks of urban yellow fever. The failure of campaigns to eradicate this species justifies the search for alternative breeding sites, which may be beyond the reach of present control measures. In this study the occurrence of Aedes aegypti breeding sites in native bromeliads on rocky slop

Genome sequence and comparative analysis of a putative entomopathogenic Serratia isolated from Caenorhabditis briggsae.

BACKGROUND: Entomopathogenic associations between nematodes in the genera Steinernema and Heterorhabdus with their cognate bacteria from the bacterial genera Xenorhabdus and Photorhabdus, respectively, are extensively studied for their potential as biological control agents against invasive insect species. These two highly coevolved associations were results of convergent evolution. Given the natural abundance of bacteria, nematodes and insects, it is surprising that only these two associations with no intermediate forms are widely studied in the entomopathogenic context. Discovering analogous

Rickettsial and other tick-borne infections.

Tick bites are best prevented by people avoiding tick-infested areas. When this is not possible, tick bites may be prevented by the wearing of long trousers that are tucked into boots. The best method to avoid tick bites is twofold: application of a topical deet (N,N-diethyl-m-toluamide) repellent to exposed skin, and treatment of clothing with permethrin. This system is currently used by the US Army to protect soldiers. Ticks can crawl underneath clothing and bite untreated portions of the body; therefore, treating clothing is imperative. Permethrin is nontoxic to humans, and can be used in a

Genome-wide DNA methylation homeostasis influences jasmonate signaling and potato defenses against the potato tuber moth Phthorimaea operculella.

BACKGROUND: DNA methylation is an epigenetic mechanism involved in plant defense, yet its role in insect resistance remains unclear. Although herbivory remodels plant methylomes, the links between DNA methylation, resistance traits, and phytohormone signaling remain unresolved. RESULTS: We induced genome-wide hypomethylation in potato plants, Solanum tuberosum L., using the DNA methyltransferase inhibitor 5-azacytidine (5-azaC), and assessed their resistance to the potato tuber moth (PTM), Phthorimaea operculella Zeller. Foliar application of 100 μm 5-azaC significantly reduced global DNA met

Distinct intracellular and extracellular maturation of goose astrovirus 2 capsid protein and the potential of virus-like particles as a novel subunit vaccine.

Goose astrovirus 2 (GAstV-2) is the primary causative agent of gosling gout in China, causing severe economic losses, yet no commercial vaccines are available. Virus-like particles (VLPs) represent a promising subunit vaccine platform, but the capsid maturation mechanism and VLP immunogenicity of GAstV-2 remain unknown. Herein, we expressed the GAstV-2 ORF2 capsid protein utilizing the baculovirus/insect cell system and systematically characterized its intracellular and extracellular maturation dynamics. Western blot analysis with domain-specific antibodies revealed distinct maturation pathway

New tools for an old pest: HiPlex targeted sequencing for Bt corn resistance screening in Ostrinia nubilalis (Lepidoptera: Crambidae) from Canada.

For almost three decades, crystal (Cry) insecticidal proteins from the bacterium Bacillus thuringiensis (Bt) have been used as plant-incorporated protectants (PIPs) in transgenic corn, resulting in areawide suppression of the European corn borer (Ostrinia nubilalis). Control of this once-considered most damaging pest of corn in North America is now threatened by the evolution of practical resistance to PIPs in Bt corn. Practical resistance to the Cry1F protein has been detected in eastern Canada, and cases of resistance to the other available Bt PIPs (Cry1Ab, Cry1A.105, and Cry2Ab) are startin

CRISPR/Cas9-mediated knockout of CsRDL1 decreases the susceptibility of Chilo suppressalis to cyproflanilide.

The novel meta-diamide insecticide cyproflanilide represents a promising tool for pest control, yet its precise molecular target remains uncharacterized. The RDL1 encodes a subunit of insect ionotropic GABA receptors (GABAr), which is targeted by meta-diamide pesticides. This study investigated the functional role of the resistance to dieldrin 1 (RDL1) subunit in Chilo suppressalis, a major rice pest, by generating a homozygous CsRDL1 knockout strain (CsRDL1-/-, lacking 101 bp) via CRISPR/Cas9-mediated gene editing. Bioassay results showed that the CsRDL1-/- strain exhibited a 2.67-fold increa

Balancing Barcoding and Genomics: gDNA Quality in Insect Vouchers After HotSHOT DNA Extraction.

DNA extraction with an alkaline buffer system called 'HotSHOT' is widely used for barcoding because it is rapid, inexpensive, and voucher preserving, but it remains unclear whether sufficient genomic DNA (gDNA) remains in small vouchers for downstream use in genomics. We here evaluate gDNA quality and quantity before and after HotSHOT treatment of 11 insect families representing six orders. Some specimens were flash frozen immediately after collection, while others were kept for 1 week at tropical temperatures in ethanol to mimic Malaise trap conditions. Encouragingly, we show that gDNA of suf

Readdressing methods for evaluating New World screwworm (Diptera: Calliphoridae) strain performance.

For the sterile insect technique to successfully control New World Screwworm (NWS), Cochliomyia hominivorax (Coquerel, 1858), released sterile males must be healthy and reproductively competitive with wild populations. In mass-rearing, daily evaluations of developmental and physical metrics are used to evaluate production efficiency, output, and to identify problems with production processes. These observations can detect major strain or production issues, but they may not be sufficient to evaluate how a strain will perform in the field. Here we evaluate novel mating assays, locomotor activity

Processed animal proteins in poultry nutrition: nutritional value, digestibility, safety, and applications.

Processed animal proteins (PAPs) are concentrated protein ingredients derived from animal by-products through rendering or hydrolytic processes and are increasingly reconsidered in poultry nutrition due to their nutritional and sustainability advantages. This review provides a comprehensive evaluation of the composition, digestibility, functional properties, safety, and practical applications of PAPs in poultry feeding systems. Nutritional value varies considerably depending on raw material origin and processing conditions, with excessive heat treatment reducing amino acid availability through

Evaluation of acute toxicity and enzyme activity of commonly used pesticides in rice fields to Chironomus kiiensis and Chironomus javanus.

Various chemical pesticides are commonly applied in rice field ecosystems to control insect pests, diseases, and weeds. However, pesticide residues and the resulting environmental pollution pose serious threats to the safety of farmland ecosystems. This study evaluated the acute toxicity of five insecticides (chlorantraniliprole, abamectin, pymetrozine, nitenpyram, and indoxacarb), five fungicides (trifloxystrobin, validamycin, tebuconazole, thiamethoxam, and propiconazole), and three herbicides (pretilachlor, cyhalofop-butyl, and penoxsulam) on the fourth-instar larvae of two chironomid speci

First record of Haematobia irritans (L.) (Diptera: Muscidae) infesting cattle in Northeastern Algeria.

Haematobia irritans (L.) is a haematophagous ectoparasite considered one of the most economically damaging pests of cattle, as it causes a decrease in milk production, growth retardation in calves due to reduced weaning weight, and weight loss in adult cattle. These flies can also transmit pathogenic agents. This study reports the first record of H. irritans in northeastern Algeria, in the wilayas of Mila and Guelma, during the active season of the insect from May to September in 2021 and 2022. A total of 181 cattle of various breeds, sexes, and ages were examined, and 1040 adult H. irritans s

High-throughput image-based pupal sex classification in Aedes aegypti using convolutional neural network models for sterile insect technique applications.

Aedes aegypti, the primary vector of dengue, yellow fever, Zika, and chikungunya, poses serious public health threats, especially in tropical regions. With limited vaccine availability, innovative control strategies like the Sterile Insect Technique (SIT) are essential, requiring efficient pupal sex separation to release only non-biting sterile males. This study developed a high-throughput, low-cost imaging pipeline using 96-well plates with optimized conditions (fixed camera stand, backlighting, refrigeration for immobilization, and black-painted wells) to capture mosquito pupal images using

The RNA promoter for pathogenic orthoflaviviruses replication is universal and serves as target for viral inhibition.

Orthoflaviviruses comprise a diverse genus of positive-strand RNA viruses that includes major human pathogens such as dengue, yellow fever, Zika, Japanese encephalitis, tick-borne encephalitis, and West Nile viruses. Despite their global impact, the molecular constraints that preserve viral replication across distinct vectors and host environments remain incompletely understood. Viral RNA replication depends on Stem-Loop A (SLA), a structured RNA element located at the 5' end of the genome that recruits the viral polymerase NS5. Here, we examine the structural diversity of this RNA promoter ac

Genome-wide investigation and functional analysis of 20S proteasome subunits in Locusta migratoria.

The 20S proteasome is a core component of the ubiquitin-proteasome system, participating in various biological processes such as cell cycle regulation, signal transduction, apoptosis, and protein homeostasis. However, its roles in mammals are well-documented, its function in the insect intestine remains largely unexplored. In this study, we identified 14 20S proteasome subunits, including 7 α-subunits and 7 β-subunits in Locusta migratoria, a worldwide agricultural pest. Phylogenetic tree revealed conservation of amino acid sequences among different species. While the conserved domains and mot
