Reflex sympathetic dystrophy: early treatment and psychological aspects.

We report the results of two prospective studies of early treatment and psychological aspects in a series of 26 patients with sympathetic reflex dystrophy of the hand in which treatment was started within 3 months after diagnosis. Ismelin blocks is an often used therapy in sympathetic reflex dystrophy but a probable better therapy in the first stage of the dystrophy was also investigated. Thirteen patients were treated with Regional Intravenous Ismelin blocks and 13 other patients were treated with a hydroxyl radical scavenger, dimethylsulfoxide (DMSO). After 9 weeks there was a better result

Glyceraldehyde-3-phosphate dehydrogenase/1,3-bisphosphoglycerate-NADH as key determinants in controlling human retinal endothelial cellular functions: Insights from glycolytic screening.

Maintaining barrier integrity, along with cell adhesion to the extracellular matrix and the subsequent process of cell spreading, are essential functions of endothelial cells, including human retinal endothelial cells (HRECs). Disruptions in these processes can lead to vision-threatening conditions like diabetic retinopathy. However, the bioenergetic mechanisms that regulate HREC barrier function and cell spreading remain incompletely understood. This study investigates the role of lower glycolytic components in modulating these critical functions of HRECs. In vitro, Electric Cell-Substrate Im

Nanoencapsulated Syzygium aromaticum oil alleviates acetic acid-induced ulcerative colitis in rats by influencing critical redox, NF-κB/iNOS, and Keap1/Nrf2/HO-1 signaling pathways.

Ulcerative colitis (UC) is an autoimmune inflammatory condition characterized by significant mucosal destruction. Although Syzygium aromaticum (Clove oil; CO) oil is well-known for its antioxidant and anti-inflammatory properties, its high volatility, toxicity, and hydrophobicity can compromise its biological efficacy. Therefore, nanoencapsulation is a feasible approach for boosting its therapeutic potential, including stability, bioavailability, and target delivery. Herein, this study was designed to minimize acetic acid-mediated UC using CO alone or encapsulated in nano-vehicles including PC

Biochemical properties of glycerol kinase from the hypersaline-adapted archaeon Haloferax volcanii.

Extremophilic microorganisms are promising candidates for industrial and analytical biocatalysis. Haloferax volcanii, a halophilic archaeon that prefers glycerol over glucose, channels this substrate into central metabolism through glycerol kinase (GK). Here, we report the biochemical properties of H. volcanii GK and its potential for biotechnological applications. An N-terminal His-tagged GK was functional in vivo and yielded 3 mg/L culture-4.5 times more enzyme than a C-terminal StrepII-tagged version. Size exclusion chromatography revealed a glycerol-induced oligomeric shift from homodimer

Mixtures of Diethyl Azelate as Novel Nonopioid Modalities for Pain Management.

Introduction Effective pain management is essential for improving the quality of life. Currently, we have medications to address both mild and severe pain, but there remains a therapeutic gap for pain that is not adequately managed by over-the-counter (OTC) or prescription non-steroidal anti-inflammatory drugs (NSAIDs). Prescription opioids can lead to addiction, respiratory suppression, and even death. OTC pain relief options often lack the potency required to alleviate more intense pain, while stronger treatments, though effective, carry risks of addiction and other adverse effects, limiting

Expression profiling of Wnt/β-catenin, antimicrobial peptides, endoplasmic reticulum stress, and apoptosis signaling pathways and antioxidant/oxidant status in Oreochromis niloticus following Proteus mirabilis challenge: the palliative role of naringenin flavanone.

The most prevalent diseases in fish farming are bacterial infections, which drive considerable financial losses to the aquaculture sector. Figuring out an innovative, eco-friendly antimicrobial is becoming more crucial, especially in light of the escalating resistance of pathogens to the existing pharmaceuticals. Accordingly, this work is the inaugural attempt to look into the antibacterial attributes of a flavanone [naringenin (NG) dissolved in dimethyl sulfoxide (DMSO)] in assisting Oreochromis niloticus (Nile tilapia) to combat a bacterial challenge (Proteus mirabilis). This study was carri

Functional characterization of TGFβ/BMP receptors in Echinococcus granulosus sensu stricto: implications for parasite survival and therapeutic targeting.

Cystic echinococcosis (CE), a zoonosis caused by the larval stage of the tapeworm Echinococcus granulosus sensu lato (E. granulosus s.l.), remains a global health burden. Despite the conserved role of the transforming growth factor beta (TGFβ)/Smad signaling in cellular regulation across species, the molecular mechanisms of TGFβ/bone morphogenetic proteins (BMP) receptors in E. granulosus sensu stricto (E. granulosus s.s.) are poorly understood. The TGFβ/BMP receptors of E. granulosus s.s. were cloned and molecularly characterized. The positive binding interaction between receptors and their l

Determination of HER2 binding domain in antigen-antibody complexes based on chemical crosslinking mass spectrometry.

Chemical crosslinking (XL) of non-covalent antigen-antibody complexes followed by mass spectrometric identification (MS) of inter-protein crosslinks can provide spatial constraints between relevant residues, which are valuable structural information associated with the molecular binding interface. To highlight the potential of XL/MS in the biopharmaceutical industry, we herein developed and validated an XL/MS workflow that employed a zero-length linker, 1,1'‑carbonyldiimidazole (CDI), and a widely used medium-length linker, disuccinimidyl sulfoxide (DSSO), for fast, accurate determination of a

Protective Effect of the Viola spathulata Extract on NCX3 Gene Expression in an Animal Model of Cerebral Ischemia.

INTRODUCTION: Viola plant has been used traditionally to treat neurological disorders. We aimed at determining whether pretreatment with Viola spathulata extract can alleviate the severity of ischemic-reperfusion damages and exert its protective effects through the regulation of a sodium/calcium exchanger (NCX3) gene expression in a rat brain. METHODS: Male Wistar rats were divided into two main groups: one main group for evaluating Neurologic Deficit Score (NDS) and Infarct Volume (IV) and the other group for the evaluation of NCX3 gene expression in the brain tissue. The latter group was su

AMPK-dependent phosphorylation is required for transcriptional activation of TFEB and TFE3.

Increased macroautophagy/autophagy and lysosomal activity promote tumor growth, survival and chemo-resistance. During acute starvation, autophagy is rapidly engaged by AMPK (AMP-activated protein kinase) activation and MTOR (mechanistic target of rapamycin kinase) complex 1 (MTORC1) inhibition to maintain energy homeostasis and cell survival. TFEB (transcription factor E3) and TFE3 (transcription factor binding to IGHM enhancer 3) are master transcriptional regulators of autophagy and lysosomal activity and their cytoplasm/nuclear shuttling is controlled by MTORC1-dependent multisite phosphory

Modulating FKBP5/FKBP51 and autophagy lowers HTT (huntingtin) levels.

Current disease-modifying therapies for Huntington disease (HD) focus on lowering mutant HTT (huntingtin; mHTT) levels, and the immunosuppressant drug rapamycin is an intriguing therapeutic for aging and neurological disorders. Rapamycin interacts with FKBP1A/FKBP12 and FKBP5/FKBP51, inhibiting the MTORC1 complex and increasing cellular clearance mechanisms. Whether the levels of FKBP (FK506 binding protein) family members are altered in HD models and if these proteins are potential therapeutic targets for HD have not been investigated. Here, we found levels of FKBP5 are significantly reduced

[Determination of absolute mobility and dissociation constant of lovastatin using capillary electrophoresis and empirical equation of ion mobility].

In capillary electrophoresis, determination of the basic physical and chemical properties of compounds, such as absolute mobility (m0) and dissociation constant (pKa), is of great practical significance. This is because the aforementioned properties are often used for the qualitative or quantitative analyses of the relevant compounds toward their application as potential drugs. Lovastatin is a potential drug candidate that can reduce the levels of cholesterol and low-density lipoprotein cholesterol in the blood, as well as prevent atherosclerosis and coronary heart disease. For a more convenie

Inhibition of the SEC61 translocon by mycolactone induces a protective autophagic response controlled by EIF2S1-dependent translation that does not require ULK1 activity.

The Mycobacterium ulcerans exotoxin, mycolactone, is responsible for the immunosuppression and tissue necrosis that characterizes Buruli ulcer. Mycolactone inhibits SEC61-dependent co-translational translocation of proteins into the endoplasmic reticulum and the resultant cytosolic translation triggers degradation of mislocalized proteins by the ubiquitin-proteasome system. Inhibition of SEC61 by mycolactone also activates multiple EIF2S1/eIF2α kinases in the integrated stress response (ISR). Here we show mycolactone increased canonical markers of selective macroautophagy/autophagy LC3B-II, ub

Interaction between Streptococcus pneumoniae and Staphylococcus aureus Generates ·OH Radicals That Rapidly Kill Staphylococcus aureus Strains.

Streptococcus pneumoniae rapidly kills Staphylococcus aureus by producing membrane-permeable hydrogen peroxide (H2O2). The mechanism by which S. pneumoniae-produced H2O2 mediates S. aureus killing was investigated. An in vitro model that mimicked S. pneumoniae-S. aureus contact during colonization of the nasopharynx demonstrated that S. aureus killing required outcompeting densities of S. pneumoniae Compared to the wild-type strain, isogenic S. pneumoniae ΔlctO and S. pneumoniae ΔspxB, both deficient in production of H2O2, required increased density to kill S. aureus While residual H2O2 activi

Toxic masking and synergistic modulation of the estrogenic activity of chemical mixtures in a yeast estrogen screen (YES).

BACKGROUND, AIM AND SCOPE: Estrogenic and non-estrogenic chemicals typically co-occur in the environment. Interference by non-estrogenic chemicals may confound the assessment of the actual estrogenic activity of complex environmental samples. The aim of the present study was to investigate whether, in which way and how seriously the estrogenic activity of single estrogens and the observed and predicted joint action of estrogenic mixtures is influenced by toxic masking and synergistic modulation caused by non-estrogenic chemical confounders.
