Evaluation of root and canal morphology of permanent maxillary molars: A CBCT study with a new classification approach in a Saudi Arabian subpopulation.

BACKGROUND: A thorough understanding of root canal anatomy is essential for achieving predictable endodontic outcomes, particularly in maxillary molars, which are anatomically complex. The presence of additional canals, especially in the mesiobuccal root, remains a frequent cause of missed anatomy and treatment failure. Despite the growing use of cone-beam computed tomography (CBCT), population-specific data using contemporary classification systems remain limited. AIM: The present study aimed to assess the root and canal configurations of permanent maxillary molars in a Saudi Arabian subpopu

Single-cell and spatial transcriptomics analysis of osteoarthritis: pathway regulation, cell interaction networks, and therapeutic translation.

BACKGROUND: Osteoarthritis (OA) transcends the outdated paradigm of mere "wear and tear" cartilage loss. It is now recognized as a complex, whole-organ disease driven by multifaceted interactions among diverse cell populations across synovium, cartilage, subchondral bone, and infrapatellar fat pad. The profound heterogeneity within these tissues and the spatiotemporal dynamics of pathological processes have, until recently, remained a "black box," limiting our understanding of disease initiation and progression. The convergence of single-cell RNA sequencing (scRNA-seq) and spatial transcriptom

A Systematic Pipeline for Evaluating Factors Affecting Radar-Based Microwave Breast Imaging Quality.

Microwave imaging (MWI) has emerged as a promising non-ionizing, cost-effective technique for breast cancer detection. However, the influence of key system design choices remains poorly characterized. This study aims to systematically investigate how antenna array geometry, tumor characteristics, and tissue dielectric properties influence radar-based image reconstruction using a robust computational pipeline. A total of 575 different scenarios were simulated and analyzed, combining 25 distinct breast models with varying tumor configurations and 23 antenna configurations (including 20 randomly

Cell line-specific gene network enrichment analysis for interpreting continuous phenotypes.

Gene network enrichment analysis (GNEA) offers a robust approach for interpreting the complex molecular mechanisms underlying phenotypic variability. Despite its utility, prevailing GNEA methodologies are predominantly optimized for binary phenotypes, leading to substantial information loss when applied to continuous biological traits such as drug sensitivity, cancer progression, etc. Additionally, traditional enrichment strategies often exhibit conceptual inconsistency between their null models and test hypotheses, as they rely on permuting phenotype labels rather than genes. To address these

Integrated Network Pharmacology and Experimental Validation Approach to Systematically Elucidate the Cardioprotective Mechanisms of Shengmai Injection.

Core to advancing the study of traditional Chinese medicine (TCM) mechanisms is the integrated "computational prediction-experimental verification" strategy, which enables the precise identification of therapeutic targets through multilevel technological integration. Within this framework, this study systematically investigates the therapeutic mechanisms of Shengmai injection (SMI) against heart failure following myocardial infarction (MI). Initially, using network pharmacology approaches, we systematically screened TCMSP, BATMAN, and disease databases to identify 237 common targets of SMI and

Effect of psychopharmacological combinations on psychopathological symptom burden and BMI trajectories in patients with eating disorders.

INTRODUCTION: Eating disorders (EDs), particularly Anorexia Nervosa (AN), remain one of the most severe and treatment-resistant eating disorders, with high relapse rates and limited pharmacological options. While second-generation antipsychotics and antidepressants are commonly prescribed as adjuncts to nutritional rehabilitation, their real-world impact on weight restoration and psychopathological symptom severity remains unclear. METHODS: We conducted a prospective, naturalistic observational study of 127 inpatients diagnosed with restrictive anorexia nervosa (AN-r), binge-purge anorexia ne

Anatomy of the Soft Palate and Its Role in Upper Airway Function: A Narrative Review.

The soft palate is a complex anatomical structure, located at the posterior aspect of the oral and nasal cavities, able to dynamically create valve closure of the nasal and the oral cavities. Its precise movement is critical for speech, swallowing, and respiration. Despite its functional importance, detailed characterization of its constituent muscles-the levator veli palatini (LVP), palatopharyngeus (PP), tensor veli palatini (TVP), musculus uvulae (MU), and palatoglossus (PG)-as well as the palatine aponeurosis (PA) remains limited and inconsistent. This narrative review synthesizes findings

Accuracy of Preoperative Positron Emission Tomography - Computed Tomography for Mediastinal Lymph Node Staging in Non-Small Cell Lung Cancer.

BACKGROUND: Accurate mediastinal lymph node staging is essential in non-small cell lung cancer (NSCLC) treatment. While 18F-fluorodeoxyglucose positron emission tomography/computed tomography (FDG-PET/CT) is widely used for noninvasive staging, its diagnostic reliability, particularly for nodal assessment, remains debated. METHODS: A retrospective multicenter analysis included 278 patients with histologically confirmed NSCLC who underwent FDG-PET/CT followed by surgery with systematic lymphadenectomy between 2015 and 2021. PET/CT-based nodal staging was compared with histopathology. Diagnosti

Repurposing Alzheimer's and ovarian cancer drugs as sonosensitizers for glioblastoma via a positive-unlabeled learning and 3D bioprinting-based new approach methodology (NAM).

Glioblastoma (GBM) remains a lethal primary brain tumor, in part because therapeutic efficacy is limited by the blood-brain barrier (BBB) and the complex tumor microenvironment (TME). Sonodynamic therapy (SDT), i.e., use of ultrasound to activate chemical sensitizers and generate cytotoxic stress, offers a non-invasive strategy for treating deep-seated intracranial disease, but progress is constrained by the scarcity of validated sonosensitizers and the inefficiency of conventional in vitro screening methods. Here, we introduce a New Approach Methodology (NAM) that couples a neural network-bas

Evaluation of Hand Gesture Classification Based on Motor Units Activation for Prosthetic Application.

Recent advancements in upper-limb prosthetics have enabled users to control multi-degree-of-freedom prostheses using pattern recognition algorithms applied to electromyography (EMG). These methods are built on the extraction of EMG features that treat EMG signals as colored noise, neglecting their biological origin. In contrast, recent works have explored the use of features based on the decomposition of EMG signal into the activity of the motor units. These features hold great potential for control applications as they reflect the actual neural drive to the muscles. Here, we evaluate the feas
