First detection of Leishmania donovani in sand flies from Cameroon and its epidemiological implications.

OBJECTIVES: A sound knowledge of the vector-host-parasite transmission dynamics is a prerequisite for adequate control measures of vector-borne diseases. To achieve this, an entomological investigation was conducted in the cutaneous leishmaniasis (CL) endemic focus of Mokolo District, northern Cameroon to identify the insect vector(s) of the disease. METHODS: Phlebotomine sand flies were collected in and around Mokolo using New Standard CDC Miniature Light Traps. Individual sand flies were used for morphological species identification, and the remainder of the body for DNA analysis. Sand flie

Expression Levels of Odorant Receptor Genes in the Savanna Tsetse Fly, Glossina morsitans morsitans.

Tsetse flies (Glossina) are vectors of African trypanosomiasis. Olfaction plays a critical role in Glossina behavior, including larviposition, feeding, and reproduction. Odorant receptors (ORs) are important in insect chemoreception as they bind volatile odorants and transport them to olfactory receptor neurons to elicit behavioral response. To better understand Glossina chemoreception, we used quantitative polymerase chain reaction to examine the expression levels of ORs in female and male Glossina morsitans morsitans Wiedemann, 1850 (Diptera: Glossinidae) antennae and legs. Results showed th

The Role of Host Cholesterol During Flavivirus Infection.

In recent years the emergence and resurgence of arboviruses have generated a global health alert. Among arboviruses, Dengue (DENV), Zika (ZIKV), Yellow Fever (YFV), and West Nile (WNV) virus, belong to the genus Flavivirus, cause high viremia and occasionally fatal clinical disease in humans. Given the genetic austerity of the virus, they depend on cellular factors and organelles to complete its replication. One of the cellular components required for flavivirus infection is cholesterol. Cholesterol is an abundant lipid in biomembranes of eukaryotes cells and is necessary to maintain the cellu

Short-distance dispersal of Hypothenemus hampei (Ferrari) females (Coleoptera: Curculionidae: Scolytidae) during the coffee tree fruiting period.

The coffee berry borer (CBB), Hypothenemus hampei (Ferrari), is a multivoltine species closely associated with coffee crops worldwide, causing severe damage to the bean. In Mexico, as in all tropical regions, CBB survives during the inter-harvest period in residual berries on the ground or in dry berries remaining on the branches, and then disperses in search of the first suitable berries. In this study, we investigated how CBB dispersed from the first infested nodes during the fruiting period of Coffea canephora Pierre, which provides a favourable trophic level for this insect. Forty-five bra

Nanotechnology-based recent approaches for sensing and remediation of pesticides.

Pesticides are meant to control and destroy the pests and weeds. They are classified into different categories on the basis their origin and type of pest they target. Chemical pesticides such as insecticides, herbicides and fungicides are commonly used in agricultural fields. However, the excessive use of these agrochemicals have adverse effects on environment such as reduced population of insect pollinators, threat to endangered species and habitat of birds. Upon consumption; chemical pesticides also cause various health issues such as skin, eye and nervous system related problems and cancer

Cloning and expression of long neuropeptide F and the role of FMRFamide-like peptides in regulating egg production in the Chagas vector, Rhodnius prolixus.

Long neuropeptide F (NPF) is a neuropeptide implicated in the control of feeding, digestion and reproduction in various insect species. Here we have isolated the cDNA sequence encoding NPF in Rhodnius prolixus (RhoprNPF). The RhoprNPF gene is composed of 3 exons and 2 introns, one of which is present in the peptide coding region. RhoprNPF is 42 amino acids long and has the characteristic RFamide C-terminus, which is common of FMRFamide-like peptides (FLPs). Quantitative PCR (qPCR) shows that RhoprNPF mRNA is present in higher amounts in fifth instars than in adults, implying that it may play a

Zoophthora radicans (Entomophthorales), a fungal pathogen of Bagrada hilaris and Bactericera cockerelli (Hemiptera: Pentatomidae and Triozidae): Prevalence, pathogenicity, and interplay of environmental influence, morphology, and sequence data on fungal identification.

The exotic bagrada bug or painted bug, Bagrada hilaris, and the native potato/tomato psyllid, Bactericera (=Paratrioza) cockerelli, are key pests of horticulture in western North America. In 2014-2015, adult and juvenile B. hilaris and B. cockerelli killed by fungi in the genus Zoophthora were detected near Saltillo, northeastern Mexico. We report the field prevalence and observations of Zoophthora on these hosts. The morphology and growth characteristics of field-collected specimens and pure in vitro cultures, as well as molecular markers (ITS1 and ITS4) were analyzed to identify these Zoopht

Disruption of Ethylene Responses by Turnip mosaic virus Mediates Suppression of Plant Defense against the Green Peach Aphid Vector.

Plants employ diverse responses mediated by phytohormones to defend themselves against pathogens and herbivores. Adapted pathogens and herbivores often manipulate these responses to their benefit. Previously, we demonstrated that Turnip mosaic virus (TuMV) infection suppresses callose deposition, an important plant defense induced in response to feeding by its aphid vector, the green peach aphid (Myzus persicae), and increases aphid fecundity compared with uninfected control plants. Further, we determined that production of a single TuMV protein, Nuclear Inclusion a-Protease (NIa-Pro) domain,

The Wiggle Index: An Open Source Bioassay to Assess Sub-Lethal Insecticide Response in Drosophila melanogaster.

Toxicological assays measuring mortality are routinely used to describe insecticide response, but sub-lethal exposures to insecticides can select for resistance and yield additional biological information describing the ways in which an insecticide impacts the insect. Here we present the Wiggle Index (WI), a high-throughput method to quantify insecticide response by measuring the reduction in motility during sub-lethal exposures in larvae of the vinegar fly Drosophila melanogaster. A susceptible wild type strain was exposed to the insecticides chlorantraniliprole, imidacloprid, spinosad, and i

Transgenic plants expressing ω-ACTX-Hv1a and snowdrop lectin (GNA) fusion protein show enhanced resistance to aphids.

Recombinant fusion proteins containing arthropod toxins have been developed as a new class of biopesticides. The recombinant fusion protein Hv1a/GNA, containing the spider venom toxin ω-ACTX-Hv1a linked to snowdrop lectin (GNA) was shown to reduce survival of the peach-potato aphid Myzus persicae when delivered in artificial diet, with survival <10% after 8 days exposure to fusion protein at 1 mg/ml. Although the fusion protein was rapidly degraded by proteases in the insect, Hv1a/GNA oral toxicity to M. persicae was significantly greater than GNA alone. A construct encoding the fusion protein

Corallopyronin A - a promising antibiotic for treatment of filariasis.

Lymphatic filariasis and onchocerciasis are diseases of severe morbidity that affect the poorest of the poor in the world. The diseases are caused by filarial nematodes that are transmitted by mosquitoes or biting blackflies and are endemic to more than 80 countries worldwide, mainly in the tropics and sub-tropics. Current control programs aim to eliminate the diseases by distributing antifilarial drugs. However, the primary effect of the drugs is to kill the microfilariae in the blood or skin, thus preventing uptake by the obligate insect vector. Since the adult worms live 10 years or longer,

The role of basophil activation test in special populations with mastocytosis and reactions to hymenoptera sting.

BACKGROUND: Systemic mastocytosis (SM) may be associated with hymenoptera allergy. In such cases, immunotherapy is a life-saving treatment, but a circumstantiated diagnosis is needed for its prescription. Patients with SM and previous reactions to stings, but with negative tests represent a diagnostic dilemma. The basophil activation test (BAT) may be helpful in refining the diagnosis. OBJECTIVE: We assessed the usefulness of BAT in subpopulations of mastocytosis patients, including those with negative tests for insect allergy. METHODS: Within a population of patients with mastocytosis and p

The role of the ventrolateral caudoputamen in predatory hunting.

The ventrolateral caudoputamen (VLCP) is well known to participate in the control of orofacial movements and forepaw usage accompanying feeding behavior. Previous studies from our laboratory have shown that insect hunting is associated with a distinct Fos up-regulation in the VLCP at intermediate rostro-caudal levels. Moreover, using the reversible blockade with lidocaine, we have previously suggested that the VLCP implements the stereotyped actions seen during prey capture and handling, and may influence the motivational drive to start attacking the roaches, as well. However, considering that

Proteomic analysis of Glossina pallidipes salivary gland hypertrophy virus virions for immune intervention in tsetse fly colonies.

Many species of tsetse flies (Diptera: Glossinidae) can be infected by a virus that causes salivary gland hypertrophy (SGH). The genomes of viruses isolated from Glossina pallidipes (GpSGHV) and Musca domestica (MdSGHV) have recently been sequenced. Tsetse flies with SGH have reduced fecundity and fertility which cause a serious problem for mass rearing in the frame of sterile insect technique (SIT) programmes to control and eradicate tsetse populations in the wild. A potential intervention strategy to mitigate viral infections in fly colonies is neutralizing of the GpSGHV infection with speci

Innate immune responses regulate trypanosome parasite infection of the tsetse fly Glossina morsitans morsitans.

Tsetse flies transmit the protozoan parasite African trypanosomes, the agents of human sleeping sickness in sub-Saharan Africa. Parasite transmission in the insect is restricted by a natural resistance phenomenon (refractoriness). Understanding the mechanism of parasite resistance is important as strengthening fly's response(s) via transgenic approaches can prevent parasite transmission and lead to the development of novel vector control strategies. Here, we investigated the role of one of the two major pathways regulating innate immunity in invertebrates, the immunodeficiency (Imd) pathway, f

Effects of the olfactory environment and nutrition on the ability of male Mediterranean fruit flies to endure starvation.

The Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae), is targeted for control using the sterile insect technique (SIT). For this technique to succeed, released males must be able to compete with wild males for copulations. Male success is mediated by survival in the field often in adverse conditions. Manipulation of the postteneral environment experienced by sterile males before release has been shown to affect male sexual success and survival. The objectives of this study were to determine how various diets, combined with exposure to volatiles containing alpha-co

Factors affecting abundance and oviposition rates of a field population of the Old World screw-worm fly, Chrysomya bezziana (Diptera: Calliphoridae).

Sentinel cattle and a grid of swormlure-baited sticky traps were used to monitor a Malaysian population of the Old World screw-worm fly, Chrysomya bezziana Villeneuve. Observations were carried out on an isolated cattle station at monthly intervals during the period August 1996 to June 2000. The number of flies caught was unaffected by weather conditions at the time of trapping, but was positively correlated with the total rainfall and the average daily air temperature prevailing 15-28 days earlier, when trapped flies were still juveniles. Trap catches were biased in favour of females, but dai

Effects of formulation on the run-off of imidacloprid from turf.

Imidacloprid is a pesticide often used to control insect pests on residential lawns and golf courses. To investigate its potential to be transported into non-target aquatic systems by rainfall events, imidacloprid was applied as 5.0 g kg-1 GR and 750 g kg-1 WP to 12 plots planted with bermuda grass set on a 5% slope. The dimethylamine salt of the herbicide 2,4-D was applied simultaneously to the plots in order to normalize the results to prior trials conducted with other pesticides. At a rate of 2.5 cm h-1, 5-cm rainfall events were simulated at 24 and 48 h after application and 2.5-cm events

[Effect of a sublethal dose of an insect growth regulator on the physiology of onchocercosis vectors].

In most of the large scale vector control programs, the larviciding operations are not fully successful, resulting in a surviving vector population due to the use of sublethal doses. This problem leads the authors to study the effect of such doses upon the future of the residual population in order to know if it represents an epidemiologically dangerous population. One insect growth regulator was selected to study this phenomenon using Onchocerca volvulus as the parasite and Simulium damnosum s.l. as the vector. The experiment was conducted under field conditions by treating a river of a savan
