Narrative Review of the Pathogenesis of Stasis Dermatitis: An Inflammatory Skin Manifestation of Venous Hypertension.

INTRODUCTION: Stasis dermatitis (SD), also known as venous dermatitis, is a form of inflammatory dermatitis of the lower extremities that typically occurs in older individuals and represents a cutaneous manifestation of venous hypertension. Venous hypertension (also known as sustained ambulatory venous pressure) is most often due to retrograde blood flow, which occurs due to calf muscle pump failure. This failure is most commonly secondary to incompetent venous valves, valve destruction, or obstruction of the venous system. Many of the common symptoms associated with SD are caused by inflammat

TLR-1, TLR-2, and TLR-6 MYD88-dependent signaling pathway: A potential factor in the interaction of high-DNA fragmentation human sperm with fallopian tube epithelial cells.

OBJECTIVE: The DNA integrity of spermatozoa that attach to fallopian tube (FT) cells is higher than spermatozoa that do not attach. FT epithelial cells can distinguish normal and abnormal sperm chromatin. This study investigated the effects of sperm with a high-DNA fragmentation index (DFI) from men with unexplained repeated implantation failure (RIF) on the Toll-like receptor (TLR) signaling pathway in human FT cells in vitro. METHODS: Ten men with a RIF history and high-DFI and 10 healthy donors with low-DFI comprised the high-DFI (>30%) and control (<30%) groups, respectively. After fresh

Increased Nucleosomes and Neutrophil Activation Link to Disease Progression in Patients with Scrub Typhus but Not Murine Typhus in Laos.

Cell-mediated immunity is essential in protection against rickettsial illnesses, but the role of neutrophils in these intracellular vasculotropic infections remains unclear. This study analyzed the plasma levels of nucleosomes, FSAP-activation (nucleosome-releasing factor), and neutrophil activation, as evidenced by neutrophil-elastase (ELA) complexes, in sympatric Lao patients with scrub typhus and murine typhus. In acute scrub typhus elevated nucleosome levels correlated with lower GCS scores, raised respiratory rate, jaundice and impaired liver function, whereas neutrophil activation correl

Integrated Transcriptomic Analysis Identifies ENO3 as a Candidate Lactate Metabolism-Associated Biomarker in Colorectal Cancer Metastasis.

BACKGROUND AND OBJECTIVES: Metastasis is the primary cause of mortality in colorectal cancer (CRC), a leading global malignancy. Although metabolic reprogramming is a hallmark of cancer, the stage-associated transcriptional features of metabolic pathways during CRC progression remain incompletely understood. This study aimed to characterize transcriptionally inferred metabolic heterogeneity during CRC progression and identify candidate molecular factors associated with metastasis. METHODS: We performed an integrated transcriptomic analysis using TCGA and GEO datasets and compared metabolic pa

Reprogramming Transcriptional Networks via CREB1 Lactylation at K122 Activates HMGB1-Mediated NETosis and Chemoresistance.

Cisplatin resistance remains a major obstacle in ovarian cancer treatment. While lactate-rich tumor microenvironments promote chemoresistance, the role of lysine lactylation (Kla) in this process remains poorly understood. Here, we identify CREB1 lactylation at K122 as a pivotal epigenetic driver of cisplatin resistance. Through quantitative lactyl-proteomics, we found CREB1 K122 as a hyperlactylated site specifically enriched in cisplatin-resistant ovarian cancer cells and patient tissues. This modification is dynamically regulated by the opposing activities of p300 (writer) and SIRT1 (eraser

Involvement of Btk in Cardiovascular Disease and Its Therapeutic Targeting.

Btk (Bruton's tyrosine kinase), a Tec-family kinase initially recognized for its role in B-cell signaling, has emerged as a critical player in thrombosis and cardiovascular disease. Beyond the established therapeutic effects of Btk inhibitors in B-cell malignancies, its expression in platelets, macrophages, and neutrophils implicates Btk in platelet activation, atherothrombosis, and innate immunity. This state-of-the-art review synthesizes the current understanding of Btk's mechanistic contributions to thrombosis and cardiovascular disease, evaluates the evolution of Btk inhibitors (BTKi), and

SPTLC2-driven sphingolipid reprogramming of neutrophils impairs anti-tumour immunity and drives liver cancer progression.

Liver cancer exhibits limited and heterogeneous responses to immune checkpoint blockade, underscoring the need to elucidate tumour microenvironmental mechanisms sustaining immune dysfunction. Here, we integrated single-cell transcriptomic annotation with BayesPrism deconvolution across multiple bulk cohorts and combined these analyses with in vivo liver cancer models, functional assays, and mechanistic investigations to define tumour-associated neutrophil (TAN) programmes linked to clinical outcome. We identified a neutrophil subset characterized by elevated sphingolipid metabolism as the pred

Serum YB-1 links dyslipidemia to NET-mediated vascular calcification in hemodialysis.

BACKGROUND: Vascular calcification (VC) is highly prevalent in patients undergoing maintenance hemodialysis (MHD) and is associated with cardiovascular morbidity. However, traditional lipid and mineral markers have limited predictive value. Y-box binding protein-1 (YB-1), a regulator of lipid metabolism and inflammation, may provide additional mechanistic and clinical insight. METHODS: Serum YB-1 was measured in 209 MHD patients (30-month follow-up) who were stratified into hyperlipidemia and control groups. Receiver Operating Characteristic (ROC) and decision curve analysis (DCA, threshold r

Multi-Omics and Experimental Validation Reveal the Protective Effect of Paeoniflorin Against Coronary Heart Disease in Mice via Inhibiting the C3-Cfd-C3aR Pathway.

Coronary heart disease (CHD) is a global cardiovascular disease with high morbidity and mortality, and its complex pathological mechanism poses great challenges to clinical prevention and treatment. Paeoniflorin (PA), a monoterpene glycoside active ingredient from Ranunculaceae plants, has shown potential in cardiovascular protection, but its specific anti-CHD molecular targets and systematic regulatory networks remain unclear. In this study, a mouse model of CHD was established, and a multi-omics strategy combining label-free quantitative proteomics and metabolomics was adopted to explore the

Lactate and lactylation in sepsis: regulation of immune-metabolic crosstalk and organ injury.

Sepsis is a life-threatening organ dysfunction caused by a dysregulated host response to infection and remains a leading cause of death worldwide. Hyperlactatemia, a hallmark metabolic disorder in sepsis, has recently been recognized as an epigenetic modulator via lysine lactylation. This Review synthesizes the evolving understanding of lactate-from a prognostic biomarker to a pathogenic mediator and, most recently, to an epigenetic modulator through lysine lactylation (Kla). Sepsis induces persistent Warburg-like glycolytic reprogramming in immune and parenchymal cells, generating lactate tha

Plasma NET-associated readouts and a multidomain neuroimmune injury axis after experimental subarachnoid hemorrhage: response to dual-pathway modulation.

BACKGROUND: Early neurological dysfunction defines early brain injury after subarachnoid hemorrhage (SAH). Neutrophil extracellular trap (NET) formation increasingly appears to operate as a neuroimmune amplifier rather than a passive marker. We tested whether dual-pathway attenuation of NETosis is associated with suppression of immunothrombotic, redox, and blood-CSF barrier injury and with improved early neurological function after experimental SAH. METHODS: Male Sprague-Dawley rats underwent endovascular perforation SAH and were allocated to Sham+Vehicle, SAH + Vehicle, SAH + DNase-L (2.5 mg

Deciphering transcriptome alterations in bone marrow immune cells at single-cell resolution under denosumab treatment.

BACKGROUND: Denosumab, an anti-RANKL antibody, effectively inhibits bone resorption, increases bone mass, and reduces fracture risk; however, its immunomodulatory effects raise concerns about possible immune imbalances during the treatment. Given the key role of bone marrows in immune cell development, this study aimed to evaluate alternations in the bone marrow osteoimmune microenvironment under denosumab treatment. METHODS: Adult female mice were administered 10 mg/kg anti-RANKL antibody via intraperitoneal injections twice weekly for 4 weeks. Bone marrow cells from both treated and control

Multi-omics profiling reveals epidermal growth factor as a potential biomarker and therapeutic target in lupus nephritis and ANCA-associated vasculitis with rapidly progressive glomerulonephritis.

The kidney is a primary target organ in systemic lupus erythematosus (SLE) and ANCA-associated vasculitis (AAV), which frequently manifest as lupus nephritis (LN) and renal AAV, respectively. A severe acute presentation of both diseases is rapidly progressive glomerulonephritis (RPGN), leading to rapid loss of renal function. However, the shared molecular mechanisms underlying this aggressive phenotype remain poorly defined. This study aimed to identify key biomarkers and pathological processes in acute autoimmune-related glomerulonephritis, with a focus on LN and AAV presenting as RPGN. Throu

Role of gut microbiome modulation in the attenuation of aging-related neuroinflammation in early brain injury after subarachnoid hemorrhage.

Early brain injury (EBI) critically determines post-subarachnoid hemorrhage (SAH) outcome, with worse prognosis in older patients. Aging confers exaggerated inflammation, possibly via gut microbiome (GM) alterations. We investigated whether GM alterations contribute to aging-related worsening of post-SAH EBI and whether GM modulation can reduce this inflammatory vulnerability. Young (8-12 weeks) and aged (17-20 months) male C57BL/6 mice underwent experimental SAH. Neurological scores, brain water content, neuronal degeneration, blood-brain barrier permeability, and inflammation were assessed.

Transcriptomic and bioinformatics analysis reveals the host response feature and potential treatment strategy of patients with dengue fever.

BACKGROUND: The dengue virus (DENV) can cause various clinical syndromes and organ damage, known as dengue fever, with the probability of developing severe dengue. However, the underlying mechanisms of host response against DENV infection remain unclear, and there is still no specific medicine for dengue fever. In the present study, we revealed the transcriptomic features of the host factor in patients with DENV infection and explored potential therapeutic medication. METHODS: The peripheral blood samples were taken from 42 people with dengue fever and 23 healthy volunteers. Transcriptome seq

Anti-IL-5 treatment, but not neutrophil interference, attenuates inflammation in a mixed granulocytic asthma mouse model, elicited by air pollution.

INTRODUCTION: Diesel exhaust particles (DEP) have been proven to aggravate asthma pathogenesis. We previously demonstrated that concurrent exposure to house dust mite (HDM) and DEP in mice increases both eosinophils and neutrophils in bronchoalveolar lavage fluid (BALF) and also results in higher levels of neutrophil-recruiting chemokines and neutrophil extracellular trap (NET) formation compared to sole HDM, sole DEP or saline exposure. We aimed to evaluate whether treatment with anti-IL-5 can alleviate the asthmatic features in this mixed granulocytic asthma model. Moreover, we aimed to unra

Prolonged immune activation in post-acute sequelae of SARS-CoV-2: neutrophil dynamics and therapeutic insights.

Post-acute sequelae of SARS-CoV-2 (PASC) is characterized by persistent symptoms such as fatigue, respiratory complications and cognitive dysfunction, affecting approximately 13.5% of SARS-CoV-2-infected individuals. Despite its clinical significance, the mechanisms driving PASC remain poorly understood. Here, to address this, we utilized a Phodopus roborovskii hamster model to investigate the long-term effects of SARS-CoV-2 infection compared with influenza A virus. While 46.25-47.50% of hamsters survived SARS-CoV-2 or influenza A virus H1N1 infection, 13.75% of SARS-CoV-2 survivors exhibited

Myeloid PDLIM2 repression as a common mechanism of infection susceptibility in lung diseases.

INTRODUCTION: The PDZ-LIM domain-containing protein PDLIM2 serves as a unique tumor suppressor and immune modulator. Its repression in either lung epithelial or myeloid cells has been shown to promote lung cancer and therapy resistance. However, whether PDLIM2 plays a broader role in other lung diseases remains unclear. METHODS: Gene expression data on human samples were exploited to investigate if PDLIM2 is repressed in the lung of patients with chronic obstructive pulmonary disease (COPD) or interstitial lung disease (ILD/idiopathic pulmonary fibrosis (IPF). PDLIM2 conditional knockout (KO)

Regulating Neutrophil PAD4/NOX-Dependent Cerebrovasular Thromboinflammation.

Background: Neutrophil extracellular trap (NET) production has been implicated in the pathogenesis of thromboinflammatory conditions such as Sickle Cell Disease (SCD), contributing to heightened risk for ischemic stroke. NETs are catalyzed by the enzyme Peptidyl Arginine Deiminase 4 (PAD4) and neutrophil derived reactive oxygen species (ROS), especially NADPH oxidase (NOX) which interacts with PAD4 and is therefore critical for neutrophil function. However, the role that NOX-dependent ROS and NETs play in the accelerated cerebral microvascular thrombosis associated with thromboinflammatory con

Myeloid Poldip2 Contributes to the Development of Pulmonary Inflammation by Regulating Neutrophil Adhesion in a Murine Model of Acute Respiratory Distress Syndrome.

Background Lung injury, a severe adverse outcome of lipopolysaccharide-induced acute respiratory distress syndrome, is attributed to excessive neutrophil recruitment and effector response. Poldip2 (polymerase δ-interacting protein 2) plays a critical role in regulating endothelial permeability and leukocyte recruitment in acute inflammation. Thus, we hypothesized that myeloid Poldip2 is involved in neutrophil recruitment to inflamed lungs. Methods and Results After characterizing myeloid-specific Poldip2 knockout mice, we showed that at 18 hours post-lipopolysaccharide injection, bronchoalveol
