Acute distal colitis impairs gastric emptying in rats via an extrinsic neuronal reflex pathway involving the pelvic nerve.

BACKGROUND AND AIMS: Patients with inflammatory bowel disease often present with abnormal gut motility away from the inflammatory site. We studied remote motility disturbances and their pathophysiology in a rat model of colitis. METHODS: Colitis was induced 72 h prior to experiments using trinitrobenzene sulphate (TNBS) instillation. Inflammation was verified using histology and myeloperoxidase (MPO) measurements. To assess gut motility, we determined gastric emptying, distal front and geometric centre (GC) of intestinal transit 30 min after intragastric administration of a semiliquid Evans b

Comparison of metabotropic glutamate receptor responses at segmental and descending inputs to motoneurons in neonatal rat spinal cord.

We compared the contribution of metabotropic glutamate receptors (mGluRs) to the generation and modulation of synaptic responses elicited in intracellularly recorded L5 motoneurons from neonatal rats by segmental and descending fibers. Dorsal root (DR) stimulation at high intensity (C-fiber strength) evoked long latency (2-5-s) depolarization in addition to early monosynaptic and polysynaptic responses. Stimulation of the descending ventrolateral funiculus (VLF) failed to evoke a late response in the same motoneuron. The mGluR antagonist (+)-alpha-methyl-4-carboxyphenylglycine (MCPG; 0.4 mM) s

Connections of higher order visual relays in the thalamus: a study of corticothalamic pathways in cats.

Axonal markers injected into layers 5 and 6 of cortical areas 17, 18, or 19 labeled axons going to the lateral geniculate nucleus (LGN), the lateral part of the lateralis posterior nucleus (LPl), and pulvinar (P). Area 19 sends fine axons (type 1, Guillery [1966] J Comp Neurol 128:21-50) to LGN, LPl, and P, and thicker, type 2 axons to LPl and P. Areas 17 and 18 send type 1 axons to LGN, and a few type 1, but mainly type 2 axons to LPl and P. Type 1 and 2 axons from a single small cortical locus distribute to distinct, generally nonoverlapping parts of LP and P; type 1 axons have a broader dis

Opioid and GABA modulation of accumbens-evoked ventral pallidal activity.

The principle output of the nucleus accumbens innervates the ventral pallidum and rostral substantia innominata. GABA and opioid peptides are among the neurotransmitter candidates for this projection. The goal of the present experiments was to delineate further the physiology and pharmacology of the accumbens projection to the ventral pallidum. The trans-synaptic responsiveness of ventral pallidal and rostral substantia innominata neurons to electrical stimulation of the nucleus accumbens was examined concurrently with the ability of microiontophoretically applied morphine (an opioid agonist),

Developmental dysphasia: clinical importance and underlying neurological causes.

This survey deals with two aspects of developmental dysphasia which are relevant to child psychiatry; the early diagnosis and treatment of children with developmental dysphasia, which may prevent the progression of learning and behaviour disorders, and the underlying biological causes of this neuro-developmental disorder. The pathophysiology of developmental dysphasia is complex and age-related. In the pre-verbal and early verbal stage, the severity of the clinical picture is primarily determined by concomitant motor pathology (motor dysfunction, dysarthria, general and oral dyspraxia) and by

Passive Blood Flow Restriction as a Countermeasure Against Disuse-Induced Muscle Atrophy: Physiological Mechanisms and Translational Evidence.

Passive blood flow restriction (BFR)-intermittent vascular occlusion applied without voluntary contraction-has been proposed as a countermeasure against disuse-induced muscle atrophy in patients unable to perform conventional exercise. However, a comprehensive mechanistic synthesis integrating translational evidence with clinical applicability is lacking. This narrative review evaluated the physiological rationale, translational evidence, and safety considerations for passive BFR in load-restricted populations. PubMed/MEDLINE, Embase, and Web of Science were searched through April 2026. Studie

The role of C-C motif chemokine ligand 2 in the preservation of myogenic tone in the kidney microvasculature of Dahl Salt-Sensitive rats.

INTRODUCTION: Mechanisms of kidney injury in hypertension are incompletely understood. Here, we investigate the role of C-C motif chemokine ligand 2 (CCL2) in kidney microvascular function and hypertension-associated kidney injury using male Dahl salt-sensitive (SS) rats and SS rats lacking Ccl2 (SSCcl2-/-). METHODS: SS and SSCcl2-/- rats were examined to determine changes in blood pressure and kidney function with increased sodium chloride intake. Kidney microvasculature was examined for changes in expression of key regulatory contractile proteins and autoregulatory function in vivo and effe

Guanfacine as a potential preventive therapy for chronic migraine: a mechanistic hypothesis.

BACKGROUND: Despite the recent therapeutic advances, approximately one-third of patients with chronic migraine fail to achieve clinically meaningful benefit, and existing oral preventive therapies remain limited by tolerability and off-target effects. There is, therefore, a pressing need for new, well-tolerated, mechanistically distinct preventive treatments for chronic migraine. This hypothesis article evaluates the pharmacological and biological rationale supporting guanfacine as a potential preventive therapy for chronic migraine. HYPOTHESIS: We hypothesize that guanfacine, a selective α2A

Overactive Bladder as a Bioenergetic Disorder: A Sex-Convergent Mitochondrial-Oxidative Framework.

AIMS: Overactive bladder (OAB) is an urgency-based symptom syndrome that increases with aging in both sexes, but its explanations remain fragmented around benign prostatic hyperplasia/bladder outlet obstruction in men, menopause/estrogen loss in women, and metabolic, sleep-related, or neurogenic mechanisms. We examine whether these diverse associations can be organized within a sex-convergent mitochondrial-oxidative framework while distinguishing OAB from urodynamic detrusor overactivity (DO) and broader lower urinary tract dysfunction (LUTD). METHODS: This hypothesis-generating Debate articl

Atypical clinical and MRI features in Leber hereditary optic neuropathy: a case series of four patients.

BACKGROUND: Leber's hereditary optic neuropathy (LHON) is a mitochondrial disorder typically presenting with subacute, painless, bilateral vision loss in young individuals. While often recognizable, atypical presentations can complicate diagnosis. Notably, optic nerve lesions on MRI are uncommon in LHON and are typically associated with inflammatory optic neuropathies, increasing the risk of misdiagnosis. METHODS: We retrospectively analyzed four patients who initially presented with suspected optic neuritis but were later diagnosed with LHON. Demographic, genetic, clinical, and multimodal da

Trigeminal neuroplasticity after pulpitis and oral administration ofparacetamol in rats.

UNLABELLED: Experimental stimulation of dental pulp nociceptors induces plastic changes in the trigeminal subnucleus caudalis (Vc) through increased intracellular calcium levels and discharge of nociceptive afferent fibers. Acetaminophen (paracetamol) is a potent and selective inhibitor of COX-2 within the central nervous system (CNS) and may act in association with the cannabinoid system and the descending serotonergic pathway. AIM: In this context, the main objective of this study is to analyze the plastic changes induced by paracetamol in the nociceptive circuit of the Vc following unilate

Spinal Dorsal Horn Neurons Receiving Descending Input from the Primary Somatosensory Cortex Contribute to Aβ Fiber-Induced Neuropathic Allodynia in Male Rats.

Mechanical allodynia is the predominant symptom of neuropathic pain following peripheral nerve injury (PNI) and is characterized by pain evoked by innocuous sensory signals transmitted through low-threshold mechanoreceptive primary afferents, including Aβ fibers. However, the underlying neural mechanisms remain insufficiently understood. Previous studies have suggested that the pathological conversion of tactile input into nociceptive signals involves maladaptive alterations in neural circuits and function within the spinal dorsal horn (SDH). Somatosensory processing and transmission in the SD

Trigeminal nerve-driven neurogenic inflammation linking migraine to glioblastoma invasion: a literature review.

Migraines are among the most common neurological disorders, disabling nearly one in seven people worldwide, whereas glioblastoma (GBM) is the most aggressive primary brain tumour, with median survival scarcely beyond 15 months. Historically considered distinct, these conditions are increasingly linked by trigeminal nerve-driven neurogenic inflammation. Activation of trigeminovascular afferents provokes antidromic release of calcitonin gene-related peptide (CGRP), substance P (SP), and pituitary adenylate cyclase-activating polypeptide (PACAP); beyond mediating migraine pain, these peptides rem

Renal nerve stimulation modulates renal blood flow in a frequency-dependent manner.

BACKGROUND: Chronic overactivity of the renal nerves is a key pathophysiological attribute of drug-resistant hypertension. Indeed, catheter-based renal denervation can lower blood pressure by severing the brain-kidney (efferent nerves) and kidney-brain (afferent nerves) link, but its irreversibility and potential nerve reconnection limits adaptability and longevity, highlighting the need for alternative treatments. Kilohertz-frequency electrical stimulation is an approach known to reversibly inhibit peripheral nerve activity and has potential to reversibly modulate renal blood flow. METHODS:

Revisiting the dermatomal recruitment of, and pressure-dependent influences on, human eccrine sweating.

Herein we describe two experiments in which the recruitment and pressure-induced modifications of human eccrine sweating were investigated. In one experiment, the longstanding belief that glandular recruitment follows a gradual, caudal-to-rostral (dermatomal) recruitment pattern was re-evaluated. The onset of sweating was simultaneously determined (ventilated capsules) from four spinal (dermatomal) segments (forehead, dorsal hand, lower chest and dorsal foot) during the passive heating of supine participants (N = 8). No evidence was found to support either dermatomal or simultaneous glandular

Differential activation of ascending noxious pathways associated with trigeminal nerve injury.

Trigeminal spinal subnucleus caudalis (Vc) neurons that project to the ventral posteromedial thalamic nucleus (VPM) and parabrachial nucleus (PBN) are critical for orofacial pain processing. We hypothesized that persistent trigeminal nerve injury differentially alters the proportion of Vc neurons that project to VPM and PBN in a modality-specific manner. Neuroanatomical approaches were used to quantify the number of Vc neurons projecting to VPM or PBN after chronic constriction injury of the infraorbital nerve (ION-CCI) and subsequent upper-lip stimulation. Male rats received injections of ret

Shifts in developmental timing, and not increased levels of experience-dependent neuronal activity, promote barrel expansion in the primary somatosensory cortex of rats enucleated at birth.

Birth-enucleated rodents display enlarged representations of whiskers (i.e., barrels of the posteromedial subfield) in the primary somatosensory cortex. Although the historical view maintains that barrel expansion is due to incremental increases in neuronal activity along the trigeminal pathway during postnatal development, recent evidence obtained in experimental models of intramodal plasticity challenges this view. Here, we re-evaluate the role of experience-dependent neuronal activity on barrel expansion in birth-enucleated rats by combining various anatomical methods and sensory deprivatio

Infantile and acquired nystagmus in childhood.

Nystagmus is an involuntary, periodic eye movement caused by a slow drift of fixation which is followed by a fast refixation saccade (jerk nystagmus) or a slow movement back to fixation (pendular nystagmus). In childhood most cases are benign forms of nystagmus: idiopathic infantile, ocular or latent nystagmus. They arise at the age of 3 months, without oscillopsia and show the absence of the physiologic opto-kinetic nystagmus. A full ophthalmologic evaluation is all that is needed in most cases: albinism, macular or optic nerve hypoplasia and congenital retinal dystrophies are the most common
